The Cyclophilin Effector TaCYPE1 From the Tomato Leaf Miner Tuta absoluta Suppresses Plant Immunity by Targeting Host RNA-Binding Proteins.

The leaf-mining insect Tuta absoluta is a highly destructive pest that deploys candidate effector proteins to modulate host defences; however, the molecular mechanisms underlying this manipulation remain poorly understood. In this study, we characterise TaCYPE1, a predicted secreted cyclophilin-like effector, and demonstrate its role in promoting insect virulence. Transient expression of TaCYPE1 in Nicotiana benthamiana suppresses pattern-triggered immunity (PTI), including INF1-induced cell death, reactive oxygen species (ROS) burst, and defence-related gene expression. Notably, this immune s

Detoxification of Insect-Derived Allergen PLA2 via Quercetin Modification: Molecular Simulation and Animal Validation.

BACKGROUND: Insect-derived proteins constitute an underutilized biological resource requiring urgent exploration to address global food protein shortages. However, their widespread application is hindered by the allergenic potential, particularly phospholipase A2 (PLA2), a highly immunoreactive allergen prevalent in edible insects such as ants and honeybees. OBJECTIVE: This study systematically investigated the molecular mechanism underlying quercetin-mediated reduction in PLA2 allergenicity, aiming to establish a novel strategy for developing hypoallergenic insect protein resources. METHODS

Characterization of three FK506-binding proteins in the entomopathogenic fungus Beauveria bassiana.

FK506 binding proteins (FKBPs) participate in regulation of diverse biological processes. However, the role of these proteins in insect-pathogenic fungi is far from well understood. To investigate the functions of FKBPs in Beauveria bassiana, a widely used entomopathogenic fungus for control of insect pests, we identify three putative FKBP genes, Bbfkbp12, Bbfkbp15, and Bbfkbp50, in the fungus. Gene-disruption experiments show that loss of Bbfkbp12 results in a significant increase of resistance of B. bassiana against the immunosuppressive compounds FK506 and rapamycin, while loss of Bbfkbp50

Arbovirus vaccines; opportunities for the baculovirus-insect cell expression system.

The baculovirus-insect cell expression system is a well-established technology for the production of heterologous viral (glyco)proteins in cultured cells, applicable for basic scientific research as well as for the development and production of vaccines and diagnostics. Arboviruses form an emerging group of medically important viral pathogens that are transmitted to humans and animals via arthropod vectors, mostly mosquitoes, ticks or midges. Few arboviral vaccines are currently available, but there is a growing need for safe and effective vaccines against some highly pathogenic arboviruses su

Insects in Pet Food Industry-Hope or Threat?

Due to the increasing global population, the world cannot currently support the well-known techniques of food production due to their harmful effects on land use, water consumption, and greenhouse gas emissions. The key answer is a solution based on the use of edible insects. They have always been present in the diet of animals. They are characterized by a very good nutritional value (e.g., high protein content and contents of essential amino acids and fatty acids, including lauric acid), and products with them receive positive results in palatability tests. Despite the existing literature dat

Efficient and Scalable Process to Produce Novel and Highly Bioactive Purified Cytosolic Crystals from Bacillus thuringiensis.

Bacillus thuringiensis (Bt) is a Gram-positive soil bacterium that is widely and safely applied in the environment as an insecticide for combatting insect pests that damage crops or are disease vectors. Dominant active ingredients made by Bt are insect-killing crystal (Cry) proteins released as crystalline inclusions upon bacterial sporulation. Some Bt Cry proteins, e.g., Cry5B (formally Cry5Ba1), target nematodes (roundworms) and show exceptional promise as anthelmintics (cures for parasitic nematode diseases). We have recently described inactivated bacteria with cytosolic crystal(s) (IBaCC)

Respiratory sensitization to insect allergens: Species, components and clinical symptoms.

Airborne insect particles have been identified as an important cause of respiratory allergies, including allergic asthma and rhinitis. In the literature, the significance of respiratory exposure to insect particles as a cause of occupational allergy has been well-documented. Indeed, many cases of occupational allergy have been reported including allergy to the larvae of flies and moths in anglers and occupationally exposed workers, to grain pests in bakers or other workers handling grains, and to crickets and/or locusts in researchers and workers in aquaculture companies. Furthermore, the prev

Culicoides allergens expressed in insect cells induce sulphidoleukotriene release from peripheral blood leukocytes of horses affected with insect bite hypersensitivity.

INTRODUCTION: Insect bite hypersensitivity (IBH) is an IgE-mediated dermatitis in horses caused by bites of Culicoides spp. The allergens are salivary gland proteins from these insects, and nine major allergens from Culicoides obsoletus have been identified and expressed in E. coli. However, proteins expressed in procaryotic systems have limitations in cellular assays, particularly in functional assays assessing the allergen-induced release of mediators in vitro, such as sulphidoleukotrienes (sLT) from basophils. The aims of the study were to produce functional Culicoides allergens in insect c

Mild hypothermia: Insights and implications for productivity in mammalian and insect cell cultures.

Mammalian cells are the preferred expression system for obtaining recombinant proteins (r-proteins) due to their ability to generate human-like glycosylation patterns. However, their slow growth and lower productivity compared to prokaryotic host cells, coupled with the rising demand for complex therapeutic proteins in the biopharmaceutical market, have driven the search for alternatives to boost productivity. In this context, mild hypothermia (MH) has emerged as a valuable tool for enhancing both the viability and productivity of r-proteins in mammalian as well as some insect-derived cell lin

Salicylic acid inducing the expression of maize anti-insect gene SPI: a potential control strategy for Ostrinia furnacalis.

BACKGROUND: Due to being rooted in the ground, maize (Zea mays L.) is unable to actively escape the attacks of herbivorous insects such as the Asian corn borer (Ostrinia furnacalis). In contrast to the passive damage, plants have evolved defense mechanisms to protect themselves from herbivores. Salicylic acid, a widely present endogenous hormone in plants, has been found to play an important role in inducing plant resistance to insects. In this study, we screened and identified the insect resistance gene SPI, which is simultaneously induced by SA and O. furnacalis feeding, through preliminary

The effect of infection with the entomopathogenic fungus Conidiobolus coronatus (Entomopthorales) on eighteen cytokine-like proteins in Galleria mellonella (Lepidoptera) larvae.

BACKGROUND: In response to the replace mammal research models with insects in preliminary immunological studies, interest has grown in invertebrate defense systems. The immunological response is regulated by cytokines; however, while their role in mammals is well understood, little is known of their function in insects. A suitable target for studies into insect immunology is Galleria mellonella (Lepidoptera), the wax moth: a common host for human fungal and bacterial pathogens. G. mellonella is also a perfect subject for studies into the presence of cytokine-like proteins. SPECIFIC OBJECTIVES

Establishment of Baculovirus-Expressed VLPs Induced Syncytial Formation Assay for Flavivirus Antiviral Screening.

The baculovirus-insect cell expression system has been widely used for heterologous protein expression and virus-like particles (VLPs) expression. In this study, we established a new method for antiviral screening targeting to glycoprotein E of flaviviruses based on the baculovirus expression system. ZIKV is a mosquito-borne flavivirus and has posed great threat to the public health. It has been reported that ZIKV infection was associated with microcephaly and serious neurological complications. Our study showed that either ZIKV E or prME protein expressed in insect cells can form VLPs and ind

Only one of three hydrophobins (Hyd1-3) contributes to conidial hydrophobicity and insect pathogenicity of Metarhizium robertsii.

Class I/II hydrophobins constitute a family of small amphiphilic proteins that mediate cell hydrophobicity and adhesion to host or substrata and have pleiotropic effects in filamentous fungi. Here we report that only class I Hyd1 is essential for conidial hydrophobicity and insect pathogenicity among three hydrophobins (Hyd1-3) characterized in Metarhizium robertsii, an insect-pathogenic fungus. Aerial conidiation levels of three Δhyd1 mutants were much more reduced in 5-day-old cultures than in 7-day-old cultures, which were wettable (hydrophilic), but restored to a wild-type level in 15-day-

Comprehensive Identification and Male-Biased Expression Analysis of Odorant-Binding Protein Genes in the Hawaiian Flower Thrips, Thrips hawaiiensis (Thysanoptera: Thripidae).

Olfaction is essential for key insect behaviors, such as host-seeking and mating, and is initiated by odorant-binding proteins (OBPs), which bind and transport hydrophobic odors. Thrips hawaiiensis is a major pest that infests the flowers of numerous horticultural crops, yet its chemosensory mechanisms remain poorly understood. Now, the availability of its genome assembly allows us to address this gap. To this end, we performed a comprehensive exploration and comparative analysis of its OBP genes. Our genome-wide analysis identified a total of 12 OBP genes in T. hawaiiensis, whereas the repert

Identification of a conserved gene family with an essential role in Leishmania parasite-insect vector adhesion.

Adhesion to tissues and surfaces is used by pathogens to avoid clearance, maintain infections, and facilitate transmission. An important example is the adhesion of the Leishmania parasite to the stomodeal valve of its insect vector, the sand fly, which is important for life cycle progression, and occurs through its flagellum via a cytoskeletal adhesion complex. While three essential kinetoplastid-insect adhesion proteins (KIAPs) are known, the overall proteome of the adhesion complex is obscure. Using TurboID-tagged KIAP3 combined with proteomics and light microscopy, we have identified additi

Leveraging plant virus-vector molecular interactions for innovative vector and disease control.

Although insect-transmitted plant viruses cause substantial global crop losses, knowledge of the molecular mechanisms underlying their transmission is limited. Recent advances in omics technologies, high-resolution tissue-specific profiling, and protein-protein interaction prediction are rapidly improving our ability to identify vector proteins that mediate virus acquisition, retention, and transmission. These approaches provide critical insights into the molecular mechanisms underlying virus-vector interaction. The integration of molecular discovery with the all-important functional validatio

Neuronal Wiring Receptors Dprs and DIPs Are GPI Anchored and This Modification Contributes to Their Cell Surface Organization.

The Drosophila Dpr and DIP proteins belong to the immunoglobulin superfamily of cell surface proteins (CSPs). Their hetero- and homophilic interactions have been implicated in a variety of neuronal functions, including synaptic connectivity, cell survival, and axon fasciculation. However, the signaling pathways underlying these diverse functions are unknown. To gain insight into Dpr-DIP signaling, we sought to examine how these CSPs are associated with the membrane. Specifically, we asked whether Dprs and DIPs are integral membrane proteins or membrane anchored through the addition of glycosyl
