The decline in malaria across Africa has been largely attributed to vector control using long-lasting insecticidal nets (LLINs). However, this intervention has prompted widespread insecticide resistance (IR) and been associated with changes in mosquito behaviour that reduce their contact with LLINs. The relative importance and rate at which IR and behavioural adaptations emerge are poorly understood. We conducted surveillance of mosquito behaviour and IR at 12 sites in Burkina Faso to assess the magnitude and temporal dynamics of insecticide, biting and resting behaviours in vectors in the 2-y

BACKGROUND: In Benin, Indoor Residual Spraying (IRS) and long-lasting insecticidal nets (LLINs) are the cornerstones of malaria prevention. In the context of high resistance of Anopheles gambiae to pyrethroids, The National Malaria Control Program (NMCP) has undertaken a full coverage of IRS in a no-flood zone in the Oueme region, coupled with the distribution of LLINs in a flood zone. We assessed the impact of this campaign on phenotypic resistance, kdr (knock-down resistance) and ace-1(R) (insensitive acetylcholinesterase) mutations.
METHODS: Insecticides used for malaria vector control int

BACKGROUND: Nicotinic acetylcholine receptors (nAChRs) of insects play a key role in fast excitatory neurotransmission. Several classes of insecticides target insect nAChRs, which are composed of subunit members of a family of multiple subunit encoding genes. Alternative splicing and RNA A-to-I editing can add further to receptor diversity. Native and recombinant receptors have been explored as sites of insecticide action using radioligands, electrophysiology and site-directed mutagenesis.
METHODS: We have reviewed the properties of native and recombinant insect nAChRs, the challenges of func

The susceptibility of local head lice to permethrin, sumithrin, deltamethrin, and carbaryl was determined by laboratory bioassays in field-collected colonies. Head lice collected from the infested heads of children 6-12 yr old were tested within 3 h of collection. The longest survival of control insects in the laboratory was obtained by keeping them in the dark at 18 degrees C and 70-80% RH. The base line susceptibility data obtained for insects collected from children not treated for lice, the reference colony, showed that deltamethrin caused the highest mortality of the insecticides tested (

Nilaparvata lugens has evolved high resistance to neonicotinoid insecticides such as nitenpyram (NTP), driven by detoxification-associated genes. RNA interference (RNAi) can suppress these genes and restore insecticide susceptibility, but delivering double-stranded RNA (dsRNA) in the field remains a major challenge because naked dsRNA degrades within hours. To overcome this barrier, a layered double hydroxide (LDH)-based nanocarrier for co-delivery of dsRNA and NTP (dsRNA@NTP@LDH) was developed. The nanoplatform was synthesized by one-step co-precipitation and electrostatic dsRNA adsorption, t

BACKGROUND: Alkaloids such as caffeine can be toxic for insects. However, although mosquitoes feed on many plants with nectar-containing alkaloids, the impact of these alkaloids on the vectorial capacity of mosquitoes is not known, in particular regarding increased resistance to insecticides.
METHODS: The effects of caffeine on Anopheles coluzzii female mosquitoes in terms of resistance to the insecticide deltamethrin, the rate at which mosquitoes are knocked-down during insecticide exposure, mortality within 48 h of insecticide exposure and longevity following exposure were studied. We also

BACKGROUND: Long-lasting insecticidal nets are the primary malaria vector control measure in coastal Kenya. In 2018, phenotypic resistance to pyrethroids and low frequency of L1014S kdr mutation were reported in the Anopheles gambiae complex. Since then, additional pyrethroid-treated nets were distributed in 2021. The objectives of this study were to determine the insecticide resistance profiles of An. gambiae and Anopheles funestus from Kwale County and evaluate potential resistance mechanisms.
METHODS: From July 2023 to May 2024, adult and larval collections of An. funestus and An. gambiae

Dengue is the primary arbovirus transmitted by Aedes aegypti mosquitoes. Effective management of dengue demands a multidisciplinary approach. The aim of this study was to conduct an integrated analysis of dengue transmission, focusing on its vector mosquitoes, to establish a baseline for dengue control and prevention in an endemic region of Colombia. The study was conducted from 2015 to 2018 across four municipalities in the Vichada department near the Venezuelan border. Five complementary approaches were employed: (1) determining the natural infection rate and circulation of various dengue vi

Anopheles stephensi invasion in Ethiopia poses a risk of increased malaria disease burden in the region. Thus, understanding the insecticide resistance profile and population structure of the recently detected An. stephensi population in Fiq, Ethiopia, is critical to inform vector control to stop the spread of this invasive malaria species in the country. Following entomological surveillance for An. stephensi in Fiq, Somali region, Ethiopia, we confirmed the presence of An. stephensi morphologically and molecularly in Fiq. Characterization of larval habitats and insecticide susceptibility test

BACKGROUND: The emergence of insecticide resistance in Aedes mosquitoes could undermine efforts to control arboviruses. The present study aims to assess in some communes of Southern Benin, the susceptibility level of Aedes aegypti (Linnaeus, 1762) and Aedes albopictus (Skuse, 1894) to insecticides commonly used in public health, as well as mechanisms involved.
METHODS: Females Ae. albopictus and Ae. aegypti collected in Ifangni, Porto-Novo, Avrankou, Adjarra and Kétou from June 2021 to October 2022, were exposed to: deltamethrin 0.05%, permethrin 0.75%, alpha-cypermethrin 0.05%, pirimiphos me

The Applying Wolbachia to Eliminate Dengue (AWED) trial was a parallel cluster randomised trial that demonstrated Wolbachia (wMel) introgression into Ae. aegypti populations reduced dengue incidence. In this predefined substudy, we compared between treatment arms, the relative abundance of Ae. aegypti and Ae. albopictus before, during and after wMel-introgression. Between March 2015 and March 2020, 60,084 BG trap collections yielded 478,254 Ae. aegypti and 17,623 Ae. albopictus. Between treatment arms there was no measurable difference in Ae. aegypti relative abundance before or after wMel-dep

Resistance to toxins in insects is generally thought of as their own genetic trait, but recent studies have revealed that gut microorganisms could mediate resistance by detoxifying phytotoxins and man-made insecticides. By laboratory experiments, we here discovered a striking example of gut symbiont-mediated insecticide resistance in a serious rice pest, Cletus punctiger. The rice bug horizontally acquired fenitrothion-degrading Burkholderia through oral infection and housed it in midgut crypts. Fenitrothion-degradation test revealed that the gut-colonizing Burkholderia retains a high degradin

BACKGROUND: Aedes albopictus is a highly invasive mosquito and has become a potential vector of dengue, chikungunya and Zika viruses. Insecticide-based mosquito interventions are the main tools for vector-borne disease control. However, mosquito resistance to insecticides is a major threat to effective prevention and control. Five Ae. albopictus populations across Hainan Province, China were investigated for susceptibility to multiple insecticide and resistance mechanisms.
RESULTS: Larval bioassays indicated that resistance to pyrethroids was common in all larval populations. Adult bioassays

BACKGROUND: Arboviruses are controlled through insecticide control of their mosquito vector. However, inconsiderate use of insecticides often results in the selection of resistance in treated populations, so that monitoring is required to optimize their usage. Here, Culex pipiens (West Nile and Rift Valley Fever virus vector) specimens were collected from four Moroccan cities. Levels of susceptibility to the organophosphate (OP) insecticide malathion were assessed using World Health Organization (WHO)-recommended bioassays. Individual mosquitoes were tested for the presence of the G119S mutati

Resistance to first-line treatments for Plasmodium falciparum malaria and the insecticides used for Anopheles vector control are threatening malaria elimination efforts. Suboptimal responses to drugs and insecticides are both spreading geographically and emerging independently and are being seen at increasing intensities. Whilst resistance is unavoidable, its effects can be mitigated through resistance management practices, such as exposing the parasite or vector to more than one selective agent. Resistance contributed to the failure of the 20th century Global Malaria Eradication Programme, an

BACKGROUND: During a dengue outbreak with co-circulation of DENV-1 and -2 in the city of Boa Vista, one patient was diagnosed with DENV-4, a serotype supposed absent from Brazil for almost 30 years. The re-emergence of DENV-4 triggered the intensification of mechanical and chemical Aedes aegypti control activities in order to reduce vector density and avoid DENV-4 dissemination throughout the country.
METHODS/PRINCIPAL FINDINGS: Vector control activities consisted of (a) source reduction, (b) application of diflubenzuron against larvae and (c) vehicle-mounted space spraying of 2% deltamethrin

BACKGROUND: The scale-up of indoor residual spraying and long-lasting insecticidal nets, together with other interventions have considerably reduced the malaria burden in The Gambia. This study examined the biting and resting preferences of the local insecticide-resistant vector populations few years following scale-up of anti-vector interventions.
METHOD: Indoor and outdoor-resting Anopheles gambiae mosquitoes were collected between July and October 2019 from ten villages in five regions in The Gambia using pyrethrum spray collection (indoor) and prokopack aspirator from pit traps (outdoor).

Certain biorational chemical agents used against insect pests impact essential stages or processes in insect life cycles when applied for pest management. Development of resistance to these agents, while involving maintenance of the natural role of the chemical agent, frequently requires the evolution of a new chemical structure by the resistant organism. When considering the process of resistance development, one could theoretically consider biorational structural determination rather than the less predictable or feasible generation of a novel replacement insecticide. At first consideration,
