Acute and long-term effects of electroacupuncture alter frontal and insular cortex activity and functional connectivity during resting state.

Electroacupuncture (EA) is a safe method for treating obesity; however, its underlying neural mechanisms remain unclear. We employed resting-state-functional-magnetic-resonance-imaging (RS-fMRI) and amplitude-of-low-frequency-fluctuation (ALFF) to investigate acute/long-term effects of EA on brain activity and resting-state-functional-connectivity (RSFC) in overweight/obesity subjects who received real/Sham stimulation. For acute effects, 26 and 19 overweight/obesity subjects were included in EA and Sham groups respectively. There were significant time effects on ALFF in the right insula (INS)

The anterior insular cortex in the rat exerts an inhibitory influence over the loss of control of heroin intake and subsequent propensity to relapse.

The anterior insular cortex (AIC) has been implicated in addictive behaviour, including the loss of control over drug intake, craving and the propensity to relapse. Evidence suggests that the influence of the AIC on drug-related behaviours is complex as in rats exposed to extended access to cocaine self-administration, the AIC was shown to exert a state-dependent, bidirectional influence on the development and expression of loss of control over drug intake, facilitating the latter but impairing the former. However, it is unclear whether this influence of the AIC is confined to stimulant drugs

Structural brain correlates of executive engagement in working memory: children's inter-individual differences are reflected in the anterior insular cortex.

Although the development of executive functions has been extensively investigated at a neurofunctional level, studies of the structural relationships between executive functions and brain anatomy are still scarce. Based on our previous meta-analysis of functional neuroimaging studies examining executive functions in children (Houdé, Rossi, Lubin, and Joliot, (2010). Developmental Science, 13, 876-885), we investigated six a priori regions of interest: the left anterior insular cortex (AIC), the left and the right supplementary motor areas, the right middle and superior frontal gyri, and the le

Efferent connections of the lamina terminalis, the preoptic area and the insular cortex to submandibular and sublingual gland of the rat traced with pseudorabies virus.

Neurones situated in the lamina terminalis (organum vasculosum of the lamina terminalis, median preoptic nucleus and subfornical organ) as well as within medial and lateral parts of the preoptic area and in the insular cortex become transneuronally labelled following pseudorabies virus injections into the submandibular or the sublingual gland. These neurones are efferently connected to a chain of central neurones directed to secretory or vascular tissue of the submandibular or the sublingual gland. By varying the postinoculation time a stepwise infection of different forebrain nuclei was regis

Calretinin-Positive Neurons in Central Medial Thalamus Drive Nociception and Anxiety via Projections to Anterior Insular Cortex in male mice.

The central medial thalamic nucleus (CM), a key component of the thalamic intralaminar nuclei, receives nociceptive inputs from the lateral parabrachial nucleus and is implicated in pain processing. However, the specific neuronal subtypes and downstream pathways that mediate these functions remain poorly defined. Here, we combined tract tracing, immunofluorescence, in situ hybridization, chemogenetics, fiber photometry, and behavioral tests to investigate the role of calretinin-positive (CR+) neurons in the CM (CMCR+) and their projections to the anterior insular cortex (aIC) in nociceptive tr

Neural dynamics between anterior insular cortex and right supramarginal gyrus dissociate genuine affect sharing from perceptual saliency of pretended pain.

Empathy for pain engages both shared affective responses and self-other distinction. In this study, we addressed the highly debated question of whether neural responses previously linked to affect sharing could result from the perception of salient affective displays. Moreover, we investigated how the brain network involved in affect sharing and self-other distinction underpinned our response to a pain that is either perceived as genuine or pretended (while in fact both were acted for reasons of experimental control). We found stronger activations in regions associated with affect sharing (ant

Thinning of the temporal and insular cortex is associated with negative symptoms and impaired attention in Chinese chronic schizophrenia patients with deficit syndrome.

BACKGROUND: The considerable clinical heterogeneity of schizophrenia poses significant challenges for elucidating its neurobiology. The concept of deficit schizophrenia (DS) is a valuable framework for addressing the heterogeneity of schizophrenia. Growing evidence suggests notable differences between deficit (DS) and nondeficit (NDS) schizophrenia, indicating that DS could represent a separate disease entity. METHODS: We aimed to use FreeSurfer to identify specific changes in cortical thickness among NDS patients and healthy controls (HCs) in a Chinese sample. Furthermore, we examined the po

A case of superficial hemisensory dysfunction due to operculo-insular infarction: radiological depiction of thalamocortical projections to the secondary somatosensory cortex.

A 64-year-old obese man developed hypesthesia in the left arm and leg. Neurological examination revealed decreased senses of pain, touch, and temperature in the left face, arm, trunk, and leg. Remaining functions were normal. Electrocardiogram showed atrial fibrillation. Somatosensory-evoked potentials using the stimulation in the median nerve were normal on both sides. Brain magnetic resonance imaging revealed acute infarction in the right parietal operculum and insula. There were no pathognomonic lesions in the postcentral gyrus, the thalamus, or the brain stem. Cardioembolic operculo-insula

Self-injuring adolescent girls exhibit insular cortex volumetric abnormalities that are similar to those seen in adults with borderline personality disorder.

Self-inflicted injury (SII) in adolescence is a serious public health concern that portends prospective vulnerability to internalizing and externalizing psychopathology, borderline personality development, suicide attempts, and suicide. To date, however, our understanding of neurobiological vulnerabilities to SII is limited. Behaviorally, affect dysregulation is common among those who self-injure. This suggests ineffective cortical modulation of emotion, as observed among adults with borderline personality disorder. In borderline samples, structural and functional abnormalities are observed in

Blood oxygenation level-dependent functional magnetic resonance imaging of bilateral but asymmetrical responses to gustatory stimulation in the rat insular cortex.

Evidence has suggested asymmetrical processing of taste in the human insular cortex, but this phenomenon has not been demonstrated in the rodent brain. Functional magnetic resonance imaging (fMRI) is a powerful tool for studying the functional organization of the brain. In this study, we established a blood oxygenation level-dependent (BOLD) fMRI method at 7 T to investigate the responses to gustatory stimulation in the insular cortex of anesthetized rats (220-310 g, n=15). BOLD signals were observed in the insular cortex in response to 0.5 M sucrose solution as the tastant but not observed in

Operculo-insular and anterior cingulate plasticity induced by transcranial magnetic stimulation in the human motor cortex: a dynamic casual modeling study.

The ability to induce neuroplasticity with noninvasive brain stimulation techniques offers a unique opportunity to examine the human brain systems involved in pain modulation. In experimental and clinical settings, the primary motor cortex (M1) is commonly targeted to alleviate pain, but its mechanism of action remains unclear. Using dynamic causal modeling (DCM) and Bayesian model selection (BMS), we tested seven competing hypotheses about how transcranial magnetic stimulation (TMS) modulates the directed influences (or effective connectivity) between M1 and three distinct cortical areas of t

The role of the anterior insular cortex in self-monitoring: A novel study protocol with electrical stimulation mapping and functional magnetic resonance imaging.

Becoming aware of one's own states is a fundamental aspect for self-monitoring, allowing us to adjust our beliefs of the world to the changing context. Previous evidence points out to the key role of the anterior insular cortex (aIC) in evaluating the consequences of our own actions, especially whenever an error has occurred. In the present study, we propose a new multimodal protocol combining electrical stimulation mapping (ESM) and functional magnetic resonance imaging (fMRI) to explore the functional role of the aIC for self-monitoring in patients undergoing awake brain surgery. Our results

Safety signals mitigate the consequences of uncontrollable stress via a circuit involving the sensory insular cortex and bed nucleus of the stria terminalis.

BACKGROUND: Safety signals exert a powerful buffering effect when provided during exposure to uncontrollable stressors. We evaluated the role of the sensory insular cortex (Si) and the extend amygdala in this "safety signal effect." METHODS: Rats were implanted with microinjection cannula, exposed to inescapable tailshocks either with or without a safety signal, and later tested for anxiety-like behavior or neuronal Fos expression. RESULTS: Exposure to the uncontrollable stressor reduced later social exploration but not when safety signals were present. Temporary inhibition of Si during stre

Ifenprodil infusion in agranular insular cortex alters social behavior and vocalizations in rats exposed to moderate levels of ethanol during prenatal development.

Moderate exposure to alcohol during development leads to subtle neurobiological and behavioral effects classified under the umbrella term fetal alcohol spectrum disorders (FASDs). Alterations in social behaviors are a frequently observed consequence of maternal drinking, as children with FASDs display inappropriate aggressive behaviors and altered responses to social cues. Rodent models of FASDs mimic the behavioral alterations seen in humans, with rats exposed to ethanol during development displaying increased aggressive behaviors, decreased social investigation, and altered play behavior. Intracellular calcium chelation and pharmacological SERCA inhibition of Ca2+ pump in the insular cortex differentially affect taste aversive memory formation and retrieval.

Variation in intracellular calcium concentration regulates the induction of long-term synaptic plasticity and is associated with a variety of memory/retrieval and learning paradigms. Accordingly, impaired calcium mobilization from internal deposits affects synaptic plasticity and cognition in the aged brain. During taste memory formation several proteins are modulated directly or indirectly by calcium, and recent evidence suggests the importance of calcium buffering and the role of intracellular calcium deposits during cognitive processes. Thus, the main goal of this research was to study the

Corticotropin-releasing factor neurotransmission in the insular cortex regulates anxiogenic-like behavior to acute restraint stress: A site-specific study in rats.

The insular cortex (IC) plays a key role in anxiety responses to stress, with functional differences along its rostrocaudal axis, but the underlying neurochemical mechanisms remain unclear. Although corticotropin-releasing factor (CRF) is present in the IC, its contribution to stress-induced anxiety has not been established. Thus, we investigated the role of CRF neurotransmission in the anterior (aIC), rostral-posterior (rpIC) and caudal-posterior (cpIC) subregions of the IC in anxiogenic-like responses to acute restraint stress. We also assessed the role of CRF neurotransmission in restraint-

Functional topography of glutamatergic and GABAergic neurotransmissions along the rostrocaudal axis of the posterior insular cortex regulating cardiovascular/autonomic responses to stress in rats.

We investigated the role of glutamatergic and GABAergic neurotransmissions within the rostral-posterior insular cortex (rpIC) and caudal-posterior IC (cpIC) subregions of the posterior IC (pIC) in cardiovascular responses and local neuronal activation evoked by acute restraint stress in male rats. We identified that treatment of the rpIC with a cocktail containing the GABAA receptor antagonist SR95531 and the GABAB receptor antagonist CGP35348 enhanced restraint-evoked tachycardia, whereas the same pharmacological approach in the cpIC decreased this response. Treatment of the rpIC with the non
