Effects of mineralocorticoids on α-Na+/K+ ATPase expression and parasite morphology in T. crassiceps WFU cysticerci.

Mineralocorticoids are corticosteroid hormones that, in vertebrates, regulate extracellular fluid volume and salt balance by modulating the expression and activity of Na+/K+ ATPase, the enzyme that generates and maintains Na+ and K+ gradients across plasma membranes. However, the function of mineralocorticoids in invertebrates has not been well studied. Cysticerci (larvae) of the parasite Taenia crassiceps WFU synthesize the corticosteroids 11-deoxycorticosterone (DOC) and corticosterone in vitro. Here, we searched the genome and transcriptome of T. crassiceps for the presence of α-Na+/K+ ATPa

wingless inhibits morphogenetic furrow movement in the Drosophila eye disc.

Differentiation of the Drosophila eye imaginal disc is an asynchronous, repetitive process which proceeds across the disc from posterior to anterior. Its propagation correlates with the expression of decapentaplegic at the front of differentiation, in the morphogenetic furrow. Both differentiation and decapentaplegic expression are maintained by Hedgehog protein secreted by the differentiated cells posterior to the furrow. However, their initiation at the posterior margin occurs prior to hedgehog expression by an unknown mechanism. We show here that the wingless gene contributes to the correct

Evolution of the AKH/corazonin/ACP/GnRH receptor superfamily and their ligands in the Protostomia.

In this review we trace the evolutionary connections between GnRH receptors from vertebrates and the receptors for adipokinetic hormone (AKH), AKH/corazonin-related peptide (ACP), and corazonin from arthropods. We conclude that these G protein-coupled receptors (GPCRs) are closely related and have a common evolutionary origin, which dates back to the split of Proto- and Deuterostomia, about 700 million years ago. We propose that in the protostomian lineage, the ancestral GnRH-like receptor gene duplicated as did its GnRH-like ligand gene, followed by diversification, leading to (i) a corazonin

Neurons projecting to the retrocerebral complex of the adult blow fly, Protophormia terraenovae.

Anatomical study of neurons projecting to the retrocerebral complex of the adult blow fly, Protophormia terraenovae, was done by NiCl2 filling and immunocytochemistry. Retrograde filling through the cardiac-recurrent nerve labeled three groups of neurons in the brain/subesophageal ganglion: (1) paramedial clusters of the pars intercerebralis, (2) neurons in each pars lateralis, and (3) neurons in the subesophageal ganglion. The pars intercerebralis neurons send prominent axons into the median bundle and exit from the brain via the contralateral nervus corporis cardiaci. Based on the projection

Advances in 3D culture systems for maintaining prostate tissue architecture and functional ex vivo prostate organ culture for biomedical applications.

Model systems are inevitable for biomedical research, and advanced prostate three dimensional (3D) models help bridge the gap between in vitro and in vivo systems. Prostate diseases are biologically complex and regulated by hormone signaling, stromal-epithelial interactions, immunological infiltration, and glandular microenvironmental factors. Traditional two-dimensional (2D) cell culture techniques and prostatic cell lines exhibit limitations for translational research, as they reflect varied phenotypic and genotypic characteristics compared to native prostate tissue. Recent advancements in p

The upregulation of immune responses in tyrosine hydroxylase (TH) silenced Litopenaeus vannamei.

Catecholamines (CAs) play a crucial role in maintaining physiological and immune homeostasis in invertebrates and vertebrates under stressful conditions. Tyrosine hydroxylase (TH) is the first and rate-limiting enzyme in CA synthesis. To develop an effective CA-related immunological defense system against stress and pathogen infection, various criteria, were evaluated in TH double-stranded (ds) RNA-injected white shrimp, Litopenaeus vannamei. Specifically, the relative transcript quantification of TH, dopamine β-hydroxylase (DBH), crustacean hyperglycemic hormone (CHH), and other immune-relate

Activation of Ah receptor by pure humic acids.

Humic substances (HS) are ubiquitous in the environment. However, some studies indicate that HS could induce direct adverse effects on human health and hormone-like effects in fish, amphibians, and invertebrates. In this study we investigated a possible biochemical mechanism of HS toxicity via activation of the aryl hydrocarbon receptor (AhR). AhR mediates the toxic and biological effects of environmental contaminants such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), but a number of structurally diverse compounds has also been found to activate AhR. Alkali solutions of humic acids (HA) were

D-Amino Acids in Peptides from Animals, Including Human: Occurrence, Structure, Bioactivity and Pharmacology.

All life forms typically possess homochirality, with rare exceptions. In the case of peptides and proteins, only L-amino acids are known to be encoded by genes. Nevertheless, D-amino acids have been identified in a variety of peptides, synthesized by animal cells. They include neuroexcitatory and neuroprotective peptides, cardioexcitatory peptides, hyperglycemic hormones, opioid peptides, antimicrobial peptides, natriuretic and defensin-like peptides, and fibrinopeptides. This article is a review of their occurrence, structure and bioactivity. It further explores the pharmacology and potential

Bursicon-α subunit modulates dLGR2 activity in the adult Drosophila melanogaster midgut independently to Bursicon-β.

Bursicon is the main regulator of post molting and post eclosion processes during arthropod development. The active Bursicon hormone is a heterodimer of Burs-α and Burs-β. However, adult midguts express Burs-α to regulate the intestinal stem cell niche. Here, we examined the potential expression and function of its heterodimeric partner, Burs-β in the adult midgut. Unexpectedly, our evidence suggests that Burs-β is not significantly expressed in the adult midgut. burs-β mutants displayed the characteristic developmental defects but showed wild type-like adult midguts, thus uncoupling the devel

Dietary Intake of Vitamin D from Dairy Products Reduces the Risk of Osteoporosis.

Vitamin D and calcium are important dietary compounds that affect bone mass, even if other minerals (potassium, zinc, etc.) and vitamins (A, C and K) are also involved. Vitamin D and certain minerals, in fact, play an important role in calcium homeostasis and calcium absorption. Hip fracture incidence is higher in Europe and the United States, where calcium is frequently included in the human diet; while the occurrence of these fractures is lower in developing countries, where diets are often poor in calcium. This condition is named the "calcium paradox", and may be partially explained by phos

Crystal structure of the human alpha-thrombin-haemadin complex: an exosite II-binding inhibitor.

The serine proteinase alpha-thrombin plays a pivotal role in the regulation of blood fluidity, and therefore constitutes a primary target in the treatment of various haemostatic disorders. Haemadin is a slow tight- binding thrombin inhibitor from the land-living leech Haemadipsa sylvestris. Here we present the 3.1 A crystal structure of the human alpha-thrombin- haemadin complex. The N-terminal segment of haemadin binds to the active site of thrombin, forming a parallel beta-strand with residues Ser214-Gly216 of the proteinase. This mode of binding is similar to that observed in another leech-

Neuroendocrine secretory protein 7B2: structure, expression and functions.

7B2 is an acidic protein residing in the secretory granules of neuroendocrine cells. Its sequence has been elucidated in many phyla and species. It shows high similarity among mammals. A Pro-Pro-Asn-Pro-Cys-Pro polyproline motif is its most conserved feature, being carried by both vertebrate and invertebrate sequences. It is biosynthesized as a precursor protein that is cleaved into an N-terminal fragment and a C-terminal peptide. In neuroendocrine cells, 7B2 functions as a specific chaperone for the proprotein convertase (PC) 2. Through the sequence around its Pro-Pro-Asn-Pro-Cys-Pro motif, i

RNA interference analysis of potential functions of cyclin A in the reproductive development of male oriental river prawns (Macrobrachium nipponense).

Cyclin A (CycA) plays essential roles in regulating multiple steps of the cell cycle, and it affects gonad development in mammals and invertebrates. Previous RNA interference (RNAi) analysis revealed that knocking-down the expression of CycA in female oriental river prawns (Macrobrachium nipponense) inhibited ovarian development. CycA was also predicted to have regulatory roles in reproductive development of male M. nipponense based on significant changes of Mn-CycA expression after eyestalk ablation. The goal of this study was to investigate the potential functions of CycA in the reproductive

Sex differences in parasite infections: patterns and processes.

Sex differences in parasite infection rates, intensities, or population patterns are common in a wide range of taxa. These differences are usually attributed to 1 of 2 causes: (1) ecological (sociological in humans); and (2) physiological, usually hormonal in origin. Examples of the first cause include differential exposure to pathogens because of sex-specific behavior or morphology. The second cause may stem from the well-documented association between testosterone and the immune system; sexually mature male vertebrates are often more susceptible to infection and carry higher parasite burdens

Relationship between seawater temperature, brain GnRH-like peptide expression, and gonadal development in wild bigfin reef squid (Sepioteuthis lessoniana).

BACKGROUND: Cephalopods are widely distributed in oceans worldwide and are important fishery resources. Most species have a lifespan of approximately one year and die after reproduction. In cephalopods, gonadal development may be influenced by seawater temperature; however, the endocrine mechanisms underlying reproductive maturity remain unclear. In recent years, gonadotropin-releasing hormone (GnRH)-like peptide has been identified in invertebrates, including cephalopods, as a possible endocrine regulator, similar to their role in vertebrates. Nevertheless, knowledge of its specific functions

Chronic exposure to environmentally relevant levels of citalopram affects crawl-out behavior and morphological antipredator plasticity in a freshwater gastropod.

Citalopram is a commonly prescribed antidepressant and selective serotonin reuptake inhibitor (SSRI). Low wastewater treatment plant removal rates facilitate its ubiquity in natural surface waters. In non-target organisms, SSRIs interact with evolutionarily conserved stress-hormone pathways alongside other neurological receptors to impact behavior, physiology and morphology. However, we still know little about the consequences of citalopram for aquatic invertebrates, including whether it impacts their ability to plastically respond to other environmental challenges. Thus, here we investigate t

Neuropeptide hierarchies and the activation of sequential motor behaviors in the hawkmoth, Manduca sexta.

In insects, the shedding of the old cuticle at the end of a molt involves a stereotyped sequence of distinct behaviors. Our studies on the isolated nervous system of Manduca sexta show that the peptides ecdysis-triggering hormone (ETH) and crustacean cardioactive peptide (CCAP) elicit the first two motor behaviors, the pre-ecdysis and ecdysis behaviors, respectively. Exposing isolated abdominal ganglia to ETH resulted in the generation of sustained pre-ecdysis bursts. By contrast, exposing the entire isolated CNS to ETH resulted in the sequential appearance of pre-ecdysis and ecdysis motor out

Signaling through the G-protein-coupled receptor Rickets is important for polarity, detachment, and migration of the border cells in Drosophila.

Cell migration plays crucial roles during development. An excellent model to study coordinated cell movements is provided by the migration of border cell clusters within a developing Drosophila egg chamber. In a mutagenesis screen, we isolated two alleles of the gene rickets (rk) encoding a G-protein-coupled receptor. The rk alleles result in border cell migration defects in a significant fraction of egg chambers. In rk mutants, border cells are properly specified and express the marker Slbo. Yet, analysis of both fixed as well as live samples revealed that some single border cells lag behind

Changes of AhR-mediated activity of humic substances after irradiation.

Humic substances (HS) and natural organic matter (NOM) are natural organic compounds ubiquitous in the environment. However, some studies indicate that both HS and NOM can act as xenobiotics, e.g. induce hormone-like effects in fish, amphibians and invertebrates. Molecules of these substances contain a number of aromatic rings and conjugated double bonds--the so called chromophores. Irradiation of dissolved HS and NOM can lead to a series of photochemical reactions which can act on these substances itself, or on other substances present in aquatic environment along with HS and NOM such as e.g.
