We have developed a compact, chemical ionization mass spectrometer for the sensitive detection of atmospheric trace gases using a commercial residual gas analyzer (RGA). A high-pressure ion molecule reactor interface incorporating two radiofrequency (RF)-only octupoles, powered by Wisconsin Oscillator RF power supplies, is used to focus ions through three stages of pumping before analysis via a modified RGA. We operated this instrument with benzene cation cluster reagent ions generated using a 210Po α emitter to enable the efficient ionization, transfer, and detection of analyte ions and ion-m

Primary aldosteronism (PA) is the most common form of secondary hypertension. In many cases, somatic mutations in ion channels and pumps within adrenal cells initiate the pathogenesis of PA, and this mechanism might explain why PA is so common and suggests that milder and evolving forms of PA must exist. Compared with primary hypertension, PA causes more end-organ damage and is associated with excess cardiovascular morbidity, including heart failure, stroke, nonfatal myocardial infarction, and atrial fibrillation. Screening is simple and readily available, and targeted therapy improves blood p

Magnesium and potassium are the 2 major intracellular cations. The intracellular concentrations of these 2 ions appear to be closely correlated, but the existence of a relationship between the plasma concentrations of these ions has been controversial. A major function of potassium is to maintain the excitability of nerve and muscle tissue. Alterations in either the intracellular or extracellular potassium concentration affect membrane excitability by alterations in the resting membrane potential. The critical factor is not the actual potassium concentration in either compartment but rather th

Archaerhodopsin-3 (AR-3) and its mutants have attracted large attention for applications in optogenetics since they display a voltage-sensitive, near-infrared fluorescence. Recent quantum chemistry modeling of the electrostatic interactions between the protein environment and the protonated Schiff base retinal gave a detailed insight about the origin of a mutation-induced excited-state barrier, which hinders the usual subpicosecond photo-isomerization and increases the fluorescence quantum yield (Φf). However, despite the large range of mutations tested, the Φf rarely exceeds 1%, which is in l

Bacteriorhodopsin (BR) is a light-driven outward proton pump found mainly in halophilic archaea. A BR from an archaeon Haloquadratum walsbyi (HwBR) was found to pump protons under more acidic conditions compared with most known BR proteins. The atomic structural study on HwBR unveiled that a pair of hydrogen bonds between the BC and FG loop in its periplasmic region may be a factor in such improved pumping capability. Here, we further investigated the retinal-binding pocket of HwBR and found that Trp94 contributes to the higher acid tolerance. Through single mutations in a BR from Halobacteriu

Proteins of the CLC gene family assemble to homo- or sometimes heterodimers and either function as Cl(-) channels or as Cl(-)/H(+)-exchangers. CLC proteins are present in all phyla. Detailed structural information is available from crystal structures of bacterial and algal CLCs. Mammals express nine CLC genes, four of which encode Cl(-) channels and five 2Cl(-)/H(+)-exchangers. Two accessory β-subunits are known: (1) barttin and (2) Ostm1. ClC-Ka and ClC-Kb Cl(-) channels need barttin, whereas Ostm1 is required for the function of the lysosomal ClC-7 2Cl(-)/H(+)-exchanger. ClC-1, -2, -Ka and -

We introduce the concept of a Floquet gauge pump whereby a dynamically engineered Floquet Hamiltonian is employed to reveal the inherent degeneracy of the ground state in interacting systems. We demonstrate this concept in a one-dimensional XY model with periodically driven couplings and transverse field. In the high-frequency limit, we obtain the Floquet Hamiltonian consisting of the static XY and dynamically generated Dzyaloshinsky-Moriya interaction (DMI) terms. The dynamically generated magnetization current depends on the phases of complex coupling terms, with the XY interaction as the re

Treatment with thiazides and loop diuretics increase the urinary excretion of potassium and magnesium and the body content of these ions are reduced after long-term treatment. The diuretic-induced magnesium deficiency influences the potassium metabolism. Magnesium is a necessary activator of Na-K-ATPase, which supplies the Na-K pump with energy. Lack of magnesium will therefore impair the pumping of sodium out of the cell and of potassium into the cell. The change of the relationship between extra and intracellular potassium may induce cardiac arrhythmias. Certain groups of patients, such as p

Therapy with most kinds of diuretics causes increased urinary excretion of both potassium and magnesium and after long term treatment the skeletal muscle content of these ions is reduced. Magnesium is known activator of Na-K-ATP:ase, which provides the energy for the sodium-potassium pump. Thus, lack of magnesium may impair the pumping of sodium out from the cell and potassium into the cell. Three studies are presented which agree with this theory. In one study the correlation between potassium in serum and potassium in muscle was, among other factors, found to be dependent on the content of m

Sickle cell disease, a highly debilitating disease affecting millions worldwide, is caused by the homozygous inheritance of the mutant hemoglobin S (HbS), a malaria-stabilized gene conferring protection against lethal cerebral malaria. When sickle red blood cells traverse deoxygenated bloodstreams, HbS rapidly nucleates into polymers that keep the sickle cell Piezo1 channels open for the duration of deoxy transits. On transition to oxy streams, Piezo1 channels close immediately and remain closed. Downgradient ion fluxes ("leaks") through open deoxy-Piezo1 channels change the intracellular conc

Salinity stress is among the most pervasive abiotic factors limiting plant growth and agricultural productivity worldwide, currently affecting more than 20% of irrigated croplands. Elevated salt concentrations induce osmotic imbalance, ionic toxicity, and oxidative stress, leading to disruptions in photosynthesis, metabolism, and plant development. To counteract these deleterious effects, plants have evolved intricate, highly coordinated defense systems comprising structural and functional cellular protectors. At the structural level, the cell wall, plasma membrane, vacuole, and peroxisome pla

Copper is an essential element in cells; it can act as either a recipient or a donor of electrons, participating in various reactions. However, an excess of copper ions in cells is detrimental as these copper ions can generate free radicals and increase oxidative stress. In multicellular organisms, copper metabolism involves uptake, distribution, sequestration, and excretion, at both the cellular and systemic levels. Mammalian enterocytes take in bioavailable copper ions from the diet in a Ctr1-dependent manner. After incorporation, cuprous ions are delivered to ATP7A, which pumps Cu+ from ent

Data on the interaction of reflux disease and Helicobacter pylori infection are limited in scope and rigour, controversial and difficult to interpret. Despite this, a framework of understanding is emerging, which is consistent with known effects on gastric acid secretion. In patients with moderate to severe H. pylori-induced corpus gastritis, eradication can increase substantially impaired gastric acid secretion sufficiently to precipitate reflux disease in people with pre-existing sub-clinical defective gastro-oesophageal competence. By contrast, reflux disease in duodenal ulcer patients prob

Oxidant injury to pulmonary vascular endothelium is an important factor in the pathogenesis of acute lung injury. Oxidant injury to other cell types has been reported to alter the function of Na-K-adenosinetriphophatase (ATPase) an enzyme important in maintenance of cellular ionic homeostasis and in transport of ions across biological membranes. We investigated the effect of H2O2 (0.001-10 mM) or xanthine (X) (15.2 micrograms/ml) plus xanthine oxidase (XO) (0.0153 U/ml) on the Na-K pump activity of cultured bovine pulmonary arterial endothelial cells (PAECs). We used a functional assay, using

Magnesium has a significant role in the regulation of ion transport. Marginal deficiency of Mg can therefore affect myocardial excitability and contractility. This study was taken up with the objective of examining the inotropic response of the myocardium to variation in extracellular [Mg]o and identifying the ion channels and pumps mediating the inotropic changes. Electrically stimulated rat papillary muscle was used as the experimental model and mechanical changes were recorded using a physiograph. Channel specific antagonists were used to identify the channels mediating the functional chang

Corneal transparency is dependent on regulation of the hydration of the corneal stroma. Water is driven into the cornea across the epithelial and endothelial cell layers by the stromal swelling pressure. This fluid leak into the cornea is counterbalanced by the corneal fluid pump, which is predominantly attributed to the ion and fluid transport capacity of the endothelial cell layer. Primary and secondary active transport mechanisms are responsible for generating a net ion flux from the stromal to anterior chamber side of the endothelium; however, the identity and location of all the component

The intracellular human malaria parasite, Plasmodium falciparum, uses the PfATP4 cation pump to maintain Na+ and H+ homeostasis in parasite cytosol. PfATP4 is the target of advanced antimalarial leads, which produce many poorly understood metabolic disturbances within infected erythrocytes. Here, we expressed the mammalian ligand-gated TRPV1 ion channel at the parasite plasma membrane to study ion regulation and examine the effects of cation leak. TRPV1 expression was well-tolerated, consistent with negligible ion flux through the nonactivated channel. TRPV1 ligands produced rapid parasite dea

In the present report, the effects exerted by dexamethasone on transepithelial, electrogenic Na+ transport across A6 cell monolayers grown on permeable support were further characterized in terms of time course and relationship to the rate of Na+ transport; furthermore this agonist was compared to vasopressin and insulin. (Na+ + K+)-ATPase activity and density of ouabain binding sites were measured in cell homogenates and on dispersed cells, respectively, after documenting transepithelial electrical parameters of the preparations. Na+ transport, measured by short-circuit current (Isc), was inc

Microbial rhodopsins are photoactive membrane proteins known for transporting small inorganic ions such as H+, Cl-, and Na+. Their compact structure─comprising seven transmembrane helices─has long been thought to limit their substrate range to such ions. Here, we report that several anion-pumping rhodopsins can also transport organic anions. In particular, a rhodopsin from cyanobacteria transports bulky organic anions, including those with a benzene ring, with volumes up to ∼120 Å3─five times larger than Cl-. These anions bind in the dark state and are translocated upon photoactivation, via a

Strong field ionization injects a transient vacancy in the atom which is entangled to the outgoing photoelectron. When the electron is finally detached, the ion is populated at different excited states with part of coherence information lost. The preserved coherence of matter after interacting with intense short pulses has important consequences on the subsequent nonequilibrium evolution and energy relaxation. Here we employ attosecond transient absorption spectroscopy to measure the time-delay of resonant transitions of krypton vacancy during their creation. We have observed that the absorpti
