Direct Infusion Acoustic Droplet Ejection Mass Spectrometry: Enabling High-Throughput Shotgun Lipidomics.

High-throughput lipidomics is increasingly important for large-scale studies and clinical applications. While shotgun lipidomics enables rapid analysis, it suffers from limitations such as carryover, ion suppression, and limited structural specificity. Acoustic droplet ejection mass spectrometry (ADE-MS) presents a novel approach, enabling touchless nanoliter-scale sample introduction at high speed, precision, and accuracy. Initially designed for single-droplet injection, ADE-MS was adapted for direct infusion with stable signals. In this study, we developed and benchmarked a scalable workflow

A large, multisite investigation into the lipidomics of survival in dairy cows.

Identifying physiological determinants of dairy cow survival and their potential modulation by parity may reveal opportunities to improve herd health and longevity. This multisite, prospective, observational study investigated culling and mortality hazards using targeted lipidomic and standard metabolite assays. Blood samples, stratified by parity, were collected from 2 cow cohorts (1) dry and (2) peak-milk, from across 29 commercial Australian farms (14 pasture-based, 15 confinement-based). There were 717 nonlactating, late-pregnant, dry cows (∼27 d prepartum) and 794 peak-milk cows (∼58 DIM)

Resolving Spectral Complexity in 4D Lipidomics Using a Two-Dimensional Deconvolution Framework.

The application of data-independent acquisition (DIA) in 4D lipidomics has been constrained by spectral interference due to fragment ion overlap, a bottleneck that existing one-dimensional deconvolution methods fail to fully resolve. Here, we overcome this limitation by introducing a two-dimensional liquid chromatography-ion mobility (LC-IM) deconvolution framework that unlocks the full potential of 4D lipidomics. By mathematically modeling the orthogonal LC-IM separation dimensions, our method reconstructs high-quality MS/MS spectra from highly complex DIA data, effectively disentangling co-e

Optimization of untargeted lipidomics analysis of human plasma after Dangguijakyak-san administration.

Untargeted lipidomics enables comprehensive characterization of lipid profiles; however, analytical variability in sample preparation, chromatographic conditions, and data processing often limits reproducibility. Thus, this study aimed to optimize untargeted lipidomics analysis by evaluating chromatographic columns, mobile-phase additives, and lipid extraction methods to maximize feature detection, reproducibility, and overall data quality. Chromatographic performance was compared between ethylene-bridged hybrid (BEH) and charged-surface hybrid columns. For mobile-phase additives, 10 mM ammoni

Lipidomics Reveals Circulating Lipid Biomarkers for Retinopathy of Prematurity in Preterm Infants.

OBJECTIVE: Retinopathy of prematurity (ROP) is a multifactorial eye disease affecting children born premature and is a leading cause of blindness in preterm infants worldwide. Although it has primarily been associated with high oxygen supplementation from respiratory support, there are indications that additional metabolic factors, like circulating lipids, may play a role in the disease's pathophysiology. DESIGN: An exploratory study on the development of ROP in preterm infants was conducted in Denmark during 2018 and 2019. Infants who developed a maximum of stage 1 ROP were classified as hav

Deciphering Lipidomics Landscape of Urine Extracellular Vesicles in Alcohol Use Disorder Patients.

Urine extracellular vesicles (EVs) have garnered increasing interest in recent years, as their lipid content represents a promising source of noninvasive biomarkers. Alcohol use disorder (AUD) is one of the most common psychiatric disorders and is a major contributor to morbidity and mortality. Accordingly, there is growing interest in assessing prior alcohol consumption and evaluating the severity of liver or brain injury, particularly through noninvasive methods. In this study, we employed a novel approach based on urine EVs and a highly sensitive lipidomics strategy to characterize lipid sp

Unraveling pyrrolizidine alkaloid-induced liver damage with an integrative spatial lipidomics framework.

Pyrrolizidine alkaloids (PAs), a class of secondary metabolites widely distributed in plants and the accidental ingestion or improper use of foods and herbs containing PAs, can lead to irreversible liver damage. Considering that the toxic mechanism of PAs is closely associated with metabolism, the hepatotoxicity was analyzed from the perspective of lipid metabolism. An integrated analytical approach was employed, combining mass spectrometry imaging (MSI) with liquid chromatography-mass spectrometry (LC-MS), to comprehensively investigate the spatial and temporal dynamics of lipid metabolites d

Lipidomics analysis of bone marrow in a mouse model of postmenopausal osteoporosis.

Postmenopausal osteoporosis (PMOP) is a major public health problem worldwide, afflicting many postmenopausal women. Although many studies have focused on the biological role of individual lipids in osteoporosis, no studies have systematically elucidated the lipid profile of osteoporosis. In this study, liquid chromatography-tandem mass spectrometry (LC-MS/MS) technology based on multiple reaction monitoring (MRM) method was used to compare the levels of lipid molecules in bone marrow cells of osteoporotic mice (OVX) group and sham-operation (Sham) group. Principal component analysis (PCA) was

Lipidomics in pathogenesis, progression and treatment of nonalcoholic steatohepatitis (NASH): Recent advances.

Nonalcoholic fatty liver disease (NAFLD) is a chronic liver disease affecting up to 30% of the general adult population. NAFLD encompasses a histological spectrum ranging from pure steatosis to non-alcoholic steatohepatitis (NASH). NASH can progress to cirrhosis and is becoming the most common indication for liver transplantation, as a result of increasing disease prevalence and of the absence of approved treatments. Lipidomic readouts of liver blood and urine samples from experimental models and from NASH patients disclosed an abnormal lipid composition and metabolism. Collectively, these cha

Serum lipidomics reveal the mechanism of memory disorder improvement by Qifu decoction.

Memory disorder (MD) is a neurodegenerative disease that seriously affects the quality of life of the elderly in China. It is characterized by cognitive deficits and psychiatric symptoms. In addition to oxidative damage, neurotransmitter disorders, and other factors, Ca2+ homeostasis and lipid metabolism are among the major pathways of MD etiology. Studies have shown that Ca2+ influx, causing Ca2+ overload, leads to neuronal apoptosis and alterations in lipid metabolites at all MD stages. Qifu decoction (QFD) is one of the classic compounds for the traditional treatment of dementia, which has

BACKGROUND: Abnormal lipid metabolism is common in patients with primary biliary cholangitis (PBC). PBC and Sjögren's syndrome (SS) frequently coexist in clinical practice; however, the lipid characteristics of both diseases are unknown. Therefore, we aimed to analyze the plasma lipid profiles of both diseases. METHODS: Plasma samples from 60 PBC patients, 30 SS patients, and 30 healthy controls (HC) were collected, and untargeted lipidomics was performed using ultrahigh-performance liquid chromatography high-resolution mass spectrometry. Potential lipid biomarkers were screened through an or

Differential lipidomics of HK-2 cells and exosomes under high glucose stimulation.

Abnormal cellular lipid metabolism has a very important role in the occurrence and progression of diabetic kidney disease (DKD). However, the lipid composition and differential expression by high glucose stimulation of renal tubular cells and their exosomes, which is a vital part of the development of DKD, are largely unknown. In this study, based on targeted lipid analysis by isotope labeling and tandem mass spectrometry, a total of 421 and 218 lipid species were quantified in HK-2 cells and exosomes, respectively. More importantly, results showed that GM3 d18:1/22:0, GM3 d18:1/16:0, GM3 d18:

A Lipidomics Atlas of Selected Sphingolipids in Multiple Mouse Nervous System Regions.

Many lipids, including sphingolipids, are essential components of the nervous system. Sphingolipids play critical roles in maintaining the membrane structure and integrity and in cell signaling. We used a multi-dimensional mass spectrometry-based shotgun lipidomics platform to selectively analyze the lipid species profiles of ceramide, sphingomyelin, cerebroside, and sulfatide; these four classes of sphingolipids are found in the central nervous system (CNS) (the cerebrum, brain stem, and spinal cord) and peripheral nervous system (PNS) (the sciatic nerve) tissues of young adult wild-type mice

LC-MS lipidomics of renal biopsies for the diagnosis of Fabry disease.

INTRODUCTION: Lipidomics analysis or lipid profiling is a system-based analysis of all lipids in a sample to provide a comprehensive understanding of lipids within a biological system. In the last few years, lipidomics has made it possible to better understand the metabolic processes associated with several rare disorders and proved to be a powerful tool for their clinical investigation. Fabry disease is a rare X-linked lysosomal storage disorder (LSD) caused by a deficiency in α-galactosidase A (α-GAL A). This deficiency results in the progressive accumulation of glycosphingolipids, mostly gl

Analytical Strategies in Lipidomics for Discovery of Functional Biomarkers from Human Saliva.

Human saliva is increasingly being used and validated as a biofluid for diagnosing, monitoring systemic disease status, and predicting disease progression. The discovery of biomarkers in saliva biofluid offers unique opportunities to bypass the invasive procedure of blood sampling by using oral fluids to evaluate the health condition of a patient. Saliva biofluid is clinically relevant since its components can be found in plasma. As salivary lipids are among the most essential cellular components of human saliva, there is great potential for their use as biomarkers. Lipid composition in cells

Inborn Errors of Metabolism in the Era of Untargeted Metabolomics and Lipidomics.

Inborn errors of metabolism (IEMs) are a group of inherited diseases with variable incidences. IEMs are caused by disrupting enzyme activities in specific metabolic pathways by genetic mutations, either directly or indirectly by cofactor deficiencies, causing altered levels of compounds associated with these pathways. While IEMs may present with multiple overlapping symptoms and metabolites, early and accurate diagnosis of IEMs is critical for the long-term health of affected subjects. The prevalence of IEMs differs between countries, likely because different IEM classifications and IEM screen

Validated comprehensive metabolomics and lipidomics analysis of colon tissue and cell lines.

Current untargeted approaches for metabolic fingerprinting of colon tissue and cell lines lack validation of reproducibility and/or focus on a selection of metabolites as opposed to the entire metabolome. Yet, both are critical to ensure reliable results and pursue a fully holistic analysis. Therefore, we have optimized and validated a platform for analyzing the polar metabolome and lipidome of colon-derived cell and tissue samples based on a consecutive extraction of polar and apolar components. Peak areas of selected targeted analytes and the number of untargeted components were assessed. An

Serum lipidomics of bovine paratuberculosis: Disruption of choline-containing glycerophospholipids and sphingolipids.

OBJECTIVES: Bovine paratuberculosis is a devastating infection with Mycobacterium avium subspecies paratuberculosis that ultimately results in death from malnutrition. While the infection is characterized by a long (2-4 years) subclinical phase with immune activation, ultimately host defense mechanisms fail and the bacteria spread from the small intestine to other organs. Since both the gastrointestinal tract and liver are essential for the biosynthesis of structural glycerophospholipids, we investigated the circulating levels of these lipids in field infections and experimentally infected cat
