Stimulation of bronchus-associated lymphoid tissue in rats by repeated inhalation of aerosolized lipopeptide MALP-2.

OBJECTIVE: Bronchus-associated lymphoid tissue (BALT) is a part of the integrated mucosal immune system. It may play an important functional role for antigen uptake and induction of specific immune reactions. The aim of this study was to investigate whether it is possible to induce or modulate BALT by the repetitive inhalation of the synthetic lipopeptide MALP-2. METHODS: Female Lewis rats (245 +/- 19 g) inhaled 25 microg of MALP-2 six times at intervals of 1 week. One week after the last inhalation, they were sacrificed. Cells of the bronchoalveolar lavage and the left lung were investigated

Therapeutic potential of Bacillus-derived lipopeptides in controlling enteropathogens and modulating immune responses to mitigate post-weaning diarrhea in swine.

Post-weaning diarrhea (PWD) is a major concern for pig producers, as stress and early weaning increase susceptibility to enteropathogens like enterotoxigenic Escherichia coli (ETEC) and Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium). This study explores the immunomodulatory and antimicrobial properties of lipopeptide extracts (LPEs) from Bacillus strains which contain surfactin (SF) - an immunomodulatory lipopeptide - and their potential in mitigating PWD. Pre-incubation with LPEs from B. subtilis TC12, containing 1, 40 and 60 µg/ml SF, stimulated TNF-α production sig

Effects of critical medium components on the production of antifungal lipopeptides from Bacillus amyloliquefaciens Q-426 exhibiting excellent biosurfactant properties.

In this study, influence of three critical parameters nitrogen sources, initial pH and metal ions was discussed in the production of antifungal lipopeptides from Bacillus amyloliquefaciens Q-426. The results revealed that lipopeptide biosynthesis might have relations with the population density of strain Q-426 and some special amino acids. Also, the alkali-resistant strain Q-426 could grow well in the presence of Fe(2+) ions below 0.8 M l(-1) and still maintain the competitive advantage below 0.2 M l(-1). Moreover, lipopeptides exhibited significant inhibitory activities against Curvularia lun

Dual-targeting amphiphilic lipopeptides combat multidrug-resistant Gram-positive pathogens by inhibiting type I signal peptidase and disrupting bacterial membrane.

Antimicrobial resistance poses a significant threat to global health, particularly with multidrug-resistant (MDR) Gram-positive pathogens evading last-line treatments. We herein designed and synthesized novel amphiphilic lipopeptide derivatives to exert dual antibacterial mechanisms: SPase I inhibition and bacterial membrane disruption. Among these, compound A09 demonstrated potent in vitro activity against a series of MDR Gram-positive pathogens. Biochemical characterization revealed potent inhibition of Escherichia coli SPase I (LepB) with an IC50 of 4.475 μM and a Kd of 16.3 ± 11.4 μM. Mech

Antifungal Activity of Linear and Disulfide-Cyclized Ultrashort Cationic Lipopeptides Alone and in Combination with Fluconazole against Vulvovaginal Candida spp.

Vulvovaginal candidiasis (VVC) occurs in over 75% of women at least once during their lifetime and is an infection that significantly affects their health. Candida strains resistant to standard azole antifungal therapy and relapses of VVC are more and more common. Hypothetically, biofilm is one of the main reasons of relapses and failure of the therapy. Ultrashort cationic lipopeptides (USCLs) exhibit high antimicrobial activities. Our previous study on USCLs revealed that disulfide cyclization can result in selective antifungal compounds. Therefore, four USCL were selected and their antifunga

Are cyclic lipopeptides produced by Bacillus amyloliquefaciens S13-3 responsible for the plant defence response in strawberry against Colletotrichum gloeosporioides?

UNLABELLED: The antagonistic strain Bacillus amyloliquefaciens strain S13-3 decreased the severity of strawberry anthracnose caused by Colletotrichum gloeosporioides. The foliar application of S13-3 triggered the expression of pathogenesis-related proteins, chitinase and β-1,3-glucanase, in strawberry leaves. We identified lipopeptide antibiotics, including iturin A, fengycin, mixirin, pumilacidin and surfactin, produced and secreted by S13-3. Iturin A and surfactin elicited the gene expression of the pathogenesis-related proteins in strawberry leaves, suggesting that antagonistic strain S13-3

Iturin and fengycin lipopeptides inhibit pathogenic Fusarium by targeting multiple components of the cell membrane and their regulative effects in wheat.

Biocontrol microorganisms and their derived metabolites with antagonistic activity represent promising alternatives to chemical fungicides in managing plant pathogens. The lipopeptides (LPs) iturin and fengycin derived from Bacillus amyloliquefaciens S76-3 exhibit highly inhibitory effects against pathogenic fungi, especially Fusarium graminearum (Fg), the primary pathogen causing Fusarium head blight (FHB) in cereals. However, the specific target of iturin and fengycin in Fg and the underlying mechanism of antagonistic activity remain unclear. Here, global transcriptome sequencing, combined w

pH-induced transition of sponge-phase nanoparticles to micelles: The case of Leu-enkephalin-squalene lipopeptide prodrug.

Self-assembled bioconjugates into nanoparticles (NPs) represent a powerful strategy to enhance drug efficacy. In the case of Leu-enkephalin-squalene (LENK-SQ) bioconjugate, self-assembly leads to spherical NPs internally organized into a sponge phase, which exhibit significant analgesic activity, in contrast to the free Leu-enkephalin (LENK) peptide, whose therapeutic use is severely limited by its rapid degradation and poor bioavailability. However, the presence of an ionizable group in the LENK peptide part raises questions regarding the structural evolution of these nano-assemblies under ph

Novel Synthesis of the Antifungal Cyclic Lipopeptide Iturin A and Its Fluorinated Analog for Structure-Activity Relationship Studies.

The rapid development of antifungal resistance poses a threat to the health and agricultural sectors. Iturin A, a cyclic lipopeptide with pronounced antifungal properties produced by Bacillus sp., holds promise against several pathogens. Here, a novel synthesis of iturin A is presented, which enables access to different analogs for the study of its mode of action. The route includes the enantioselective synthesis of the β-amino fatty acid present in the lipopeptide structure, followed by solid-phase peptide synthesis and on-resin cyclization. This robust synthesis was used to obtain natural it

N-acetylcysteine inhibits bacterial lipopeptide-mediated neutrophil transmigration through the choroid plexus in the developing brain.

The etiology of neurological impairments associated with prematurity and other perinatal complications often involves an infectious or pro-inflammatory component. The use of antioxidant molecules have proved useful to protect the neonatal brain from injury. The choroid plexuses-CSF system shapes the central nervous system response to inflammation at the adult stage, but little is known on the neuroimmune interactions that take place at the choroidal blood-CSF barrier during development. We previously described that peripheral administration to neonatal mice of the TLR2 ligand PAM3CSK4 (P3C), a

Design and study of lipopeptide inhibitors on preventing aggregation of human islet amyloid polypeptide residues 11-20.

Type 2 diabetes mellitus, a kind of conformational disease, has become an epidemic disease, which seriously endangers the quality of life and health of human beings. The deposition of human islet amyloid polypeptide (hIAPP) has been considered as one of the major pathological features of type 2 diabetes mellitus. As lipopeptides have some hydrophobic groups, which are similar to the reported aggregation inhibitors, and some lipopeptides could prevent cells from depositing of amyloid fibrils, several potential lipopeptide inhibitors have been engineered and synthesized, which have been assessed

Development of a dendritic cell-targeting lipopeptide as an immunoadjuvant that inhibits tumor growth without inducing local inflammation.

Materials used for the past 30 years as immunoadjuvants induce suboptimal antitumor immune responses and often cause undesirable local inflammation. Some bacterial lipopeptides that act as Toll-like receptor (TLR) 2 ligands activate immune cells as immunoadjuvants and induce antitumor effects. Here, we developed a new dendritic cell (DC)-targeting lipopeptide, h11c (P2C-ATPEDNGRSFS), which uses the CD11c-binding sequence of intracellular adhesion molecule-1 to selectively and efficiently activate DCs but not other immune cells. Although the h11c lipopeptide activated DCs similarly to an artifi

Lipopeptide vaccines illustrate the potential role of subtype-crossreactive T cells in the control of highly virulent influenza.

BACKGROUND: The best form of protection against influenza is high-titred virus-neutralizing antibody specific for the challenge strain. However, this is not always possible to achieve by vaccination due to the need for predicting the emerging virus, whether it be a drift variant of existing human endemic influenza type A subtypes or the next pandemic virus, for incorporation into the vaccine. By activating additional arms of the immune system to provide heterosubtypic immunity, that is immunity active against all viruses of type A influenza regardless of subtype or strain, it should be possibl

Engineering lipopeptide-gramicidin S conjugates to combat multidrug-resistant Acinetobacter baumannii and other ESKAPE pathogens.

Gramicidin S (GS) is a cyclic decapeptide that primarily targets Gram-positive bacteria and due to its hemolytic toxicity, is mainly used for the topical treatment of eye, ear, and throat infections caused by S. aureus. Due to its unique mechanism of action, no clinically significant resistance to GS has been reported to date. With the aim of expanding the therapeutic scope of GS to Gram-negative bacteria, we developed novel GS analogues by conjugating with cationic amino acid residues and lipopeptides on the ornithine (Orn) side-chain. A total of 14 peptides were synthesized and assessed for

The Discovery of Cyclic Lipopeptide Olenamidonins in a Deepsea-Derived Streptomyces Strain by Knocking Out a DtxR Family Regulator.

Three new cyclic lipopeptides, olenamidonins A-C (1-3), in addition to two previously reported metabolites (4 and 5), were accumulated in the ΔdtxRso deletion mutant of deepsea-derived Streptomyces olivaceus SCSIO 1071. The structures of these cyclic lipopeptides were determined by a combination of spectroscopic methods and circular dichroism (CD) measurement. The antibacterial assay results showed that compounds 1-5 displayed different degrees of growth inhibition against multidrug-resistant (MDR) bacterial strains Enterococcus faecalis CCARM 5172 and Enterococcus faecium CCARM 5203 with mini
