Diseases caused by mutations in luteinizing hormone/chorionic gonadotropin receptor.

Accumulating evidence showed that the luteinizing hormone/chorionic gonadotropin receptor (LHCGR) is an essential regulator of sexual development and reproduction from zebrafish to human. Activating and inactivating mutations of LHCGR gene have been identified from patients of different phenotypes. Familial male-limited precocious puberty, Leydig cell hypoplasia, and empty follicle syndrome are caused by LHCGR mutations. More than 50 mutations have been reported from subjects of different ethnic backgrounds. Functional analyses of the mutant LHCGR revealed multiple defects, including cell surf

Lutein facilitates physiological revascularization in a mouse model of retinopathy of prematurity.

BACKGROUND: Retinopathy of prematurity is one of the leading causes of childhood blindness worldwide, with vessel growth cessation and vessel loss in phase I followed by neovascularization in phase II. Ischaemia contributes to its pathogenesis, and lutein protects against ischaemia-induced retinal damages. We aimed to investigate the effects of lutein on a murine model of oxygen-induced retinopathy. METHODS: Mouse pups were exposed to 75% oxygen for 5 days and returned to room air for another 5 days. Vascular obliteration, neovascularization and blood vessel leakage were examined. Immunohisto

Hypothalamic Prolactin Regulation of Luteinizing Hormone Secretion in the Female Rat.

Prolactin (PRL) levels increase in response to long-term antipsychotic treatment that disrupts reproductive function. Recent evidence suggests that activation of central PRL receptors (PRLR) inhibits LH secretion and in ovariectomized rats. However, the mechanisms involved, the mode of LH secretion affected and relevance to hyperprolactinemia remain unknown. We therefore investigated the contribution of central PRL/PRLR signaling to the control of estradiol-induced surges of LH and PRL and pulsatile LH secretion under basal and hyperprolactinemic conditions. First, by subjecting ovariectomized

In vitro antiviral activity of lutein against hepatitis B virus.

Despite the availability of an effective vaccine, the hepatitis B virus (HBV) infection and its treatment remains one of the foremost public health problems in the world. The present study was performed in order to investigate the anti-HBV activity of lutein in vitro. The antiviral activity of lutein was examined by detecting the levels of HBsAg, HBeAg and extracellular HBV DNA in stable HBV-producing human hepatoblastoma HepG2 2.2.15 cells. It was found that lutein effectively suppressed the secretion of HBsAg from HepG2 2.2.15 cells in a dose-dependent manner, and it also suppressed the amou

The pleiotropic effects of excessive luteinizing hormone secretion in transgenic mice.

Luteinizing hormone (LH) is member of the glycoprotein hormone family of gonadotropins, which also includes the highly related human chorionic gonadotropin and follicle-stimulating hormone. The necessity of these factors for sustaining human fertility has been known for decades. In addition, elevated serum levels of LH have been associated with polycystic ovarian syndrome, suggesting that the appropriate balance of LH is critical for maintaining reproductive function. To dissect the biological consequences of aberrant LH signaling in vivo, several genetically engineered mouse models have been

Constitutively active luteinizing hormone receptors: consequences of in vivo expression.

Activating mutations in the luteinizing hormone receptor (LHR) gene are one of the most common mutations found in the gonadotropin receptor genes. Human males with these mutations exhibit precocious puberty while females do not have an obvious phenotype. To better understand the pathophysiology of premature LHR activation, transgenic mice have been generated with an activating mutation in LHR and a genetically engineered ligand-activated LHR. This review will summarize the major findings obtained with these two genetically modified mouse models and briefly discuss the similarities and differen

Facilitation of lordosis in rats by a metabolite of luteinizing hormone releasing hormone.

In the female rat, ovulation is preceded by a marked increase in the release of the decapeptide, LHRH, culminating in a preovulatory LH surge, which coincides with a period of sexual receptivity. The decapeptide, LHRH, is processed by a zinc metalloendopeptidase EC 3.4.24.15 (EP24.15) that cleaves the hormone at the Tyr(5)-Gly(6) bond. We have previously reported that the autoregulation of LHRH gene expression can also be mediated by its metabolite, LHRH-(1-5). Given the central function of LHRH in reproduction and reproductive behavior, we examined the role of the metabolite, LHRH-(1-5), in m

Extragonadal luteinizing hormone receptors in the reproductive tract of domestic animals.

Binding sites for LH/hCG and/or its mRNA are found in the uterus of several species, including human, primate, pig, cow, and turkey. Activation of LH receptors around Day 15 of the estrous cycle is associated with increased prostaglandin F(2alpha) production in the bovine, porcine, and ovine uterus. Activation of uterine LH receptors is also associated with increased levels of prostaglandins in human and primate. The presence of gonadotropin receptors with a dynamic pattern in the oviduct, endometrium, myometrium, and cervix of different species provides evidence that gonadotropins play a subs

Dietary lutein reduces ultraviolet radiation-induced inflammation and immunosuppression.

Ultraviolet radiation (UVR) promotes skin cancer development by mutagenic, immunosuppressive, and oxidative-stress-inducing mechanisms; however, certain antioxidants may counteract and prevent UVR-induced photodamage. Lutein is a xanthophyll carotenoid with potent antioxidant activity. Because reactive oxygen species (ROS) are believed to have a role in UVR-induced skin damage, we investigated whether lutein can modify UVR effects including the tissue swelling response to midrange UVR (280-320 nm, ultraviolet B (UVB) radiation) and UVB suppression of contact hypersensitivity (CHS) in both the

Regulation of the growth hormone and luteinizing hormone response to endotoxin in sheep.

Infectious disease processes cause physiological adaptations in animals to reorder nutrient partitioning and other functions to support host survival. Endocrine, immune and nervous systems largely mediate this process. Using endotoxin injection as a model for catabolic disease processes (such as bacterial septicemia), we have focused our attention on regulation of growth hormone (GH) and luteinizing hormone (LH) secretion in sheep. Endotoxin produces an increase in plasma GH and a decrease in plasma LH concentrations. This pattern can be reproduced, in part, by administration of various cytoki

Luteinizing hormone receptor mutations in disorders of sexual development and cancer.

Human male sexual development is regulated by chorionic gonadotropin (CG) and luteinizing hormone (LH). Aberrant sexual development caused by both activating and inactivating mutations of the human luteinizing hormone receptor (LHR) have been described. All known activating mutations of the LHR are missense mutations caused by single base substitution. The most common activating mutation is the replacement of Asp-578 by Gly due to the substitution of A by G at nucleotide position 1733. All activating mutations are present in exon 11 which encodes the transmembrane domain of the receptor. Const

Male pseudohermaphroditism due to inactivating luteinizing hormone receptor mutations.

Human male sexual development is regulated by chorionic gonadotropin (CG) and luteinizing hormone (LH), the action of both mediated by the LH receptor (LHR). Mutations that inactivate the LHR cause Leydig cell hypoplasia (LCH), an autosomal recessive disorder. In its mild form, LCH patients present with male hypogonadism. In its severe form, patients present with male pseudohermaphroditism, with female external genitalia, and cryptorchid testis. Mullerian derivatives are absent. Histological examination of the testis shows absence of mature Leydig cells. LCH patients have elevated plasma level

Recurrent bilateral theca lutein cysts in association with normal pregnancy.

We describe a case of a 28-year-old Saudi patient, gravida 6, para 5, at 14 weeks' gestation, who had recurrent bilateral theca lutein cysts during her last three pregnancies. The cysts were treated surgically during her fourth pregnancy because of acute abdomen at 20 weeks' gestation. Ovarian biopsy confirmed the diagnosis of theca lutein cysts and her pregnancy continued until term. In the last two pregnancies, the theca lutein cysts were managed conservatively by serial ultrasound examination and they resolved spontaneously. There was no history of molar pregnancy or use of ovulation-induci

Evidence for an altered luteinizing hormone sensitivity to naloxone in pathological hyperprolactinaemia.

OBJECTIVE: The underlying mechanisms involved in the pathogenesis of amenorrhoea in hyperprolactinaemic states still remain unclear. Conflicting information exists on the role of endogenous opiates on gonadotrophin disturbances in this pathological condition. In this study we have undertaken a detailed investigation of LH and PRL secretion before and during administration of naloxone, an opioid receptor blocker, in hyperprolactinaemic women with or without ovarian function in order to assess the role of ovarian steroids upon naloxone induced LH and PRL release. DESIGN: Five anovulatory and si

Effect of copper status on luteinizing hormone secretion in dairy steers.

Two experiments were conducted to examine the effects of Cu status on neuroendocrine regulation of LH secretion in dairy steers. In Experiment 1, 18 Holstein steers were assigned randomly to treatment groups in a completely randomized design. Treatments consisted of basal diet alone (control) or supplemented with 5 ppm of Mo or with 20 ppm of Cu. At 8 and 16 wk, Cu concentrations in liver were higher in the group receiving Cu and lower in the group receiving Mo than in the control. Pulse frequencies of LH were not affected by any treatment. Serum LH concentration at 16 wk tended to be lower in

Identification of luteinizing hormone-like proteins in the ovine pineal gland.

A chemical analysis was instigated to investigate the identity of the luteinizing hormone (LH)-like immunoactivity present in ovine pineal protein homogenates. Isolation of pineal LH-like material was accomplished using a 0.1 M ammonium sulphate (pH 4.0) extraction followed by anion-exchange chromatography. The resulting 3.0 M ammonium sulphate precipitate containing 70% of the LH-like immunoactivity was refractionated by cation-exchange and Sephadex G-100 chromatography. Analysis of the pattern of recovered LH-like immunoactivity in the Sephadex G-100 eluate indicated the presence of molecula
