Strigolactones (SLs) are a class of plant hormones with multifaceted roles in rice (Oryza sativa L.) growth and development. The F-box protein DWARF3 (D3) is a core component of the SL signalling pathway, coupling SL perception to the downstream responses. However, whether D3 also contributes to rice immunity and how it functions in this process remain unclear. Here, we analysed D3 allelic mutants carrying deletions in F-box- or C-terminal helix (CTH) domains. These mutants exhibited increased susceptibility to Magnaporthe oryzae, and differentially affected the expression of PHENYLALANINE AMM

BACKGROUND: Rice blast, which is caused by the fungal pathogen Magnaporthe oryzae, is one of the most devastating diseases on rice worldwide. The effectiveness of chemical fungicides is being increasingly challenged by pathogen resistance and environmental pollution, underscoring the urgent need for eco-friendly biocontrol alternatives. Herein, we report the antifungal potential of bioactive components in Saussurea costus against M. oryzae infection and explore the underlying mechanism.
RESULTS: Essential oil (EO) from Saussurea costus roots was screened from the plant EOs library, and exhibi

Nucleotide-binding leucine-rich repeat (NLR) proteins play critical roles in plant immunity. However, how NLRs are regulated and activate defense signaling is not fully understood. The rice (Oryza sativa) NLR receptor Piz-t confers broad-spectrum resistance to the fungal pathogen Magnaporthe oryzae and the RING-type E3 ligase AVRPIZ-T INTERACTING PROTEIN 10 (APIP10) negatively regulates Piz-t accumulation. In this study, we found that APIP10 interacts with two rice transcription factors, VASCULAR PLANT ONE-ZINC FINGER 1 (OsVOZ1) and OsVOZ2, and promotes their degradation through the 26S protea

Folate-dependent one‑carbon (1C) metabolism is vital for plant development and stress responses, but its specific role in immunity remains unclear. Arabidopsis Methylenetetrahydrofolate reductase 2 (MTHFR2), a prominent enzyme in 1C metabolism, is crucial in regulating methionine biosynthesis and controlling broad-spectrum immunity. Here, we show by HRMS profiling that loss-of-function mutation in MTHFR2 disturbs methionine and homocysteine concentrations, which lead to differential immune responses against a broad-spectrum of pathogens, including Magnaporthe oryzae, Rhizoctonia solani, and Ps

BACKGROUND: WRKY proteins are one of the largest gene families and are well-known for their regulatory roles in many aspects of plant development, including plant response to both biotic and abiotic stresses. Although the roles of WRKY proteins in leaf blast resistance have been well-documented in rice, their functions in panicle blast, the most destructive type of blast disease, are still largely unknown.
RESULTS: Here, we identified that the transcription of OsWRKY67 was strongly activated by leaf and panicle blast infection. OsWRKY67 is ubiquitously expressed and sub-localized in the nucle

Members of the EPIDERMAL PATTERNING FACTOR (EPF) and EPF-Like (EPFL) families perform diverse regulatory functions in plant tissue morphogenesis, controlling the development of stomata, awns, shoot apical meristems (SAMs), and inflorescences. Nevertheless, the biological functions of OsEPF/EPFL family members in mediating responses to biotic/abiotic stresses are not yet widely characterized. Here, we demonstrated abundant cis-acting elements in the putative promoters of OsEPF/EPFL genes, including those associated with dehydration-, MeJA-, MYB binding site for drought, stress-, and ABA-respons

Rising global populations due to medicinal advancements increases the patient population susceptible to superficial and severe fungal infections. Fungi often implicated in these diseases includes the dermatophytes (Microsporum spp., Epidermophtyon spp., Trichophyton spp.) as well as species of the Candida spp., Aspergillosis spp. and Cryptococcus spp. genera. In addition, increasing global populations leads to increasing agricultural demands. Thus, fungal infections of preharvested crops and stored food by plant pathogens such as Magnaporthe oryzae and Fusarium oxysporum can have detrimental s

Gibberellin (GA) 20-oxidase (GA20ox) catalyses consecutive steps of oxidation in the late part of the GA biosynthetic pathway. A T-DNA insertion mutant (17S-14) in rice, with an elongated phenotype, was isolated. Analysis of the flanking sequences of the T-DNA insertion site revealed that an incomplete T-DNA integration resulted in enhanced constitutively expression of downstream OsGA20ox3 in the mutant. The accumulation of bioactive GA(1) and GA(4) were increased in the mutant in comparison with the wild-type plant. Transgenic plants overexpressing OsGA20ox3 showed phenotypes similar to those

The key regulator of salicylic acid (SA)-mediated resistance, NPR1, is functionally conserved in diverse plant species, including rice (Oryza sativa L.). Investigation in depth is needed to provide an understanding of NPR1-mediated resistance and a practical strategy for the improvement of disease resistance in the model crop rice. The rice genome contains five NPR1-like genes. In our study, three rice homologous genes, OsNPR1/NH1, OsNPR2/NH2 and OsNPR3, were found to be induced by rice bacterial blight Xanthomonas oryzae pv. oryzae and rice blast Magnaporthe grisea, and the defence molecules

Wheat blast, caused by Magnaporthe oryzae Triticum (MoT), is a devastating fungal disease threatening global food security. Emergence of fungicide-resistant MoT populations necessitates safe, eco-friendly alternatives. This study evaluates the biorational potential of 3-methylpentanoic acid (3-MP), a volatile organic compound (VOC) produced by Bacillus safensis NI2B against MoT. In vitro assays demonstrated that 3-MP inhibits MoT mycelial growth, conidiogenesis, conidial germination, and appressorium formation in a dose-dependent manner, achieving complete suppression at 100-125 µM. The most devastating diseases in rice (Oryza sativa) are sheath blight caused by the fungal necrotroph Rhizoctonia solani, rice blast by hemibiotrophic fungus Magnaporthe oryzae, and leaf blight by bacterial biotroph Xanthomonas oryzae (Xoo). It has been reported that the Class III acyl-CoA-binding proteins (ACBPs) such as those from dicots (Arabidopsis and grapevine) play a role in defence against biotrophic pathogens. Of the six Arabidopsis (Arabidopsis thaliana) ACBPs, AtACBP3 conferred protection in transgenic Arabidopsis against Pseudomonas syringae, but not the necrotrophic fungus, Botry

Septins are a conserved family of GTPases that regulate important cellular processes such as cell wall integrity, and septation in fungi. The requirement of septins for virulence has been demonstrated in the human pathogenic yeasts Candida albicans and Cryptococcus neoformans, as well as the plant pathogen Magnaporthe oryzae. Aspergillus spp. contains five genes encoding for septins (aspA-E). While the importance of septins AspA, AspB, AspC, and AspE for growth and conidiation has been elucidated in the filamentous fungal model Aspergillus nidulans, nothing is known on the role of septins in g

Our results indicate that overexpression of OsSPL1 in transgenic tobacco plants attenuated disease resistance and facilitated programmed cell death. Long-chain base phosphates including sphingosine-1-phosphate have been shown to act as signaling mediators in regulating programmed cell death (PCD) and stress responses in mammals. In the present study, we characterized a rice gene OsSPL1, encoding a putative sphingosine-1-phosphate lyase that is involved in metabolism of sphingosine-1-phosphate. Expression of OsSPL1 was down-regulated in rice plants after treatments with salicylic acid, benzothi

Kasugamycin (KSM), a unique aminoglycoside antibiotic, has been used in agriculture for many years to control not only rice blast caused by the fungus Magnaporthe grisea but also rice bacterial grain and seedling rot or rice bacterial brown stripe caused by Burkholderia glumae or Acidovorax avenae subsp. avenae, respectively. Since both bacterial pathogens are seed-borne and cause serious injury to rice seedlings, the emergence of KSM-resistant B. glumae and A. avenae isolates highlights the urgent need to understand the mechanism of resistance to KSM. Here, we identified a novel gene, aac(2')

It has long been suggested that the essential and ubiquitous enzyme fructose 1,6-bisphosphate (FBP) aldolase could be a good drug target against bacteria and fungi, since lower organisms possess a metal-dependant (Class II) FBP aldolase, as opposed to higher organisms which possess a Schiff-base forming (Class I) FBP aldolase. We have tested the capacity of derivatives of the metal-chelating compound dipicolinic acid (DPA), as well a thiol-containing compound, to inhibit purified recombinant Class II FBP aldolases from Mycobacterium tuberculosis, Pseudomonas aeruginosa, Bacillus cereus, Bacill

The complex network of plant defense mechanisms in response to various stress conditions is often tightly regulated by multiple transcription factors (TFs). They can alter their target gene expression upon binding to their respective cognate cis-elements. A number of studies have functionally characterized the role of different WRKY TFs in various stress conditions in plants such as Arabidopsis, rice, and tobacco. However, their role in a major C4 crop such as pearl millet remains elusive. In our previous study, we showed that the ectopic expression of the group IId member PgWRKY44 in Arabidop
