Long-term results of multi-antigen stimulated cell therapy-I, alone or in combination with chemotherapy, as first-line maintenance therapy in advanced sarcoma: a multicenter, phase 1 trial.

PURPOSE: Advanced sarcomas lack effective maintenance therapies after first-line chemotherapy. We present long-term phase 1 results evaluating multi-antigen stimulated cell therapy-I (MASCT-I), a novel immunotherapy integrating dendritic cell vaccination with adoptive T-cell transfer, as maintenance treatment (NCT03034304). PATIENTS AND METHODS: Thirty-one patients with disease control after chemotherapy received MASCT-I alone (n = 17) or with ifosfamide (n = 14). The primary endpoint was safety. Secondary endpoints included progression-free survival (PFS), overall survival (OS), time to prog

Case Report: Bevacizumab combined with chemotherapy followed by PARP inhibitor maintenance therapy in POLE-mutated primary fallopian tube carcinoma: a case of precision treatment in a rare gynecologic malignancy.

BACKGROUND: Primary fallopian tube carcinoma (PTC) is a rare epithelial malignancy of gynecologic origin. The adenosquamous carcinoma variant with POLE mutations is particularly uncommon, and current treatment strategies are limited by its elusive pathogenesis. Given similarities in biological behavior and clinical characteristics to ovarian cancer, PTC staging and management follow ovarian cancer guidelines. Previous studies suggest that anti-angiogenic therapy and poly (ADP-ribose) polymerase (PARP) inhibitors may be potential therapeutic options. CASE PRESENTATION: We report a case of bila

Efficacy and Safety of Platinum-based Chemotherapy With Bevacizumab Followed by Bevacizumab Maintenance for Recurrent Ovarian, Fallopian Tube, and Primary Peritoneal Cancer During PARP Inhibitor Therapy: A Multicenter Retrospective Study.

BACKGROUND/AIM: In recent years, the usefulness of poly ADP-ribose polymerase (PARP) inhibitors as subsequent maintenance therapy with poly ADP-ribose polymerase (PARP) inhibitors has been reported. However, it has been reported shown that platinum-based chemotherapy has a low response rate and short progression-free survival for recurrent platinum-sensitive ovarian cancer during treatment with PARP inhibitor therapy. This retrospective study evaluated platinum-based chemotherapy with bevacizumab (BEV) followed by BEV maintenance in these recurrent patients. PATIENTS AND METHODS: Efficacy and

Adjuvant immunotherapy plus chemotherapy and maintenance immunotherapy for pulmonary lymphoepithelioma-like carcinoma with hepatitis B virus infection, KRAS mutation and high expression of programmed death ligand 1: A case report.

Pulmonary lymphoepithelioma-like carcinoma (PLELC) is a rare subtype of non-small cell lung cancer, often associated with Epstein-Barr virus (EBV) infection. It typically exhibits distinct epidemiological and molecular features compared to other lung malignancies. However, the clinical behavior and optimal therapeutic strategies for PLELC, particularly in the presence of concomitant viral infections or specific genetic mutations, remain poorly defined. This report comprehensively outlines the treatment process of a 58-year-old female patient diagnosed with PLELC in the right middle lobe of the

A phase II study in newly diagnosed stage III/IV epithelial ovarian cancer evaluating carboplatin/taxol/pembrolizumab in patients receiving neoadjuvant chemotherapy followed by pembrolizumab maintenance with or without olaparib.

BACKGROUND: Pembrolizumab, a PD-1 immune checkpoint inhibitor, has shown promise in various cancers, but its role in ovarian cancer remains under investigation. While the current standard of care (SOC) is initially effective for epithelial ovarian cancer (EOC), approximately 70% of patients relapse within 3 years of completing upfront treatment, highlighting an unmet need for therapies that extend time to first progression. The survival benefit observed with hyperthermic intraperitoneal chemotherapy (HIPEC) at interval debulking surgery (IDS), combined with the potential for checkpoint inhibit

A phase III randomized clinical trial of Gemcitabine and Nab-Paclitaxel as switch maintenance versus continuation of modified FOLFIRINOX as first-line chemotherapy in patients with advanced pancreatic cancer: The PANThEON Study.

BACKGROUND: Advanced pancreatic ductal adenocarcinoma (PDAC) remains among the most lethal cancers, with >95 % of patients dying from the disease. Chemotherapy is the standard of care in advanced stages, with FOLFIRINOX and Gemcitabine-Nab-Paclitaxel (Gem-NabP) as the main first-line regimens. Both show moderate efficacy and significant toxicity. Except for the PASS-01 trial, no direct comparison exists, though observational studies suggest that specific subgroups may benefit differently. Given the modest outcomes and rapid clinical decline, most patients are ineligible for second-line therapy

Complete remission of peritoneal dissemination of colon cancer by alternate-day S-1 and oxaliplatin plus bevacizumab treatment and maintenance chemotherapy comprising alternate-day S-1 plus bevacizumab: A case report.

A 56-year-old man diagnosed with sigmoid colon cancer underwent sigmoid colectomy. Nine months later, his serum carcinoembryonic antigen (CEA) level had increased, and the diagnosis of recurrent peritoneal dissemination was made based on positron emission tomography/computed tomography (PET/CT) findings. Although systemic chemotherapy comprising S-1 and oxaliplatin (SOX) plus bevacizumab was initiated, severe diarrhea occurred on day 4 of the second cycle despite reduction in S-1 dose. By changing the daily oral intake schedule for S-1 to an alternate-day intake from the third cycle (modified

Prognostic factors in metastatic breast cancer patients obtaining objective response or disease stabilization after first-line chemotherapy with epirubicin. Evidence for a positive effect of maintenance hormonal therapy on overall survival.

Randomized trials suggest that the outcome of metastatic breast cancer (BC) patients is not affected by the currently available therapies. Although response rates per se may be associated with survival prolongation, patients experiencing objective response may be those patients fated to have the longest natural disease history. The separation of responders from progressing patients after first-line chemotherapy could allow the selection of a more homogeneous subgroup in which further treatment strategies might achieve a better control of the disease. This study investigated the influence of so

Efficacy of Enfortumab Vedotin After Chemotherapy With Anti-programmed Death-Ligand 1 (PD-L1) Maintenance Treatment in a Clear Cell Variant of Invasive Urothelial Carcinoma of the Renal Pelvis: A Case Report.

Among upper urinary tract urothelial carcinoma (UUTUC) cases, there are few reports of the clear cell variant. Systemic chemotherapy will be given according to the usual treatment for urothelial cancer unless lymph nodes or organ metastases make surgical treatment inappropriate. Here, we report a clear cell variant of UUTUC of the left renal pelvis with aortic lymph node metastasis. The patient in this case was treated with systemic chemotherapy, anti-programmed death-ligand 1 (PD-L1) maintenance treatment, radiation therapy, and enfortumab vedotin (EV) therapy. Concordance between CA-125 and RECIST progression in patients with germline BRCA-mutated platinum-sensitive relapsed ovarian cancer treated in the SOLO2 trial with olaparib as maintenance therapy after response to chemotherapy.

BACKGROUND: Limited evidence exists to support CA-125 as a valid surrogate biomarker for progression in patients with ovarian cancer on maintenance PARP inhibitor (PARPi) therapy. We aimed to assess the concordance between CA-125 and Response Evaluation Criteria in Solid Tumours (RECIST) criteria for progression in patients with BRCA mutations on maintenance PARPi or placebo. Tislelizumab plus chemotherapy followed by anlotinib maintenance therapy in a patient with extensive-stage small cell lung cancer and intracranial recurrence after palliative whole-brain radiotherapy: a case report and literature review.

Small cell lung cancer (SCLC) accounts for approximately 15% of all lung cancer cases. It is characterized by an exceptionally high proliferative rate, a strong propensity for early metastasis, and a poor prognosis. The median overall survival (OS) following brain metastases development in extensive-stage SCLC (ES-SCLC) was 10.5 months. Herein, a 74-year-old Chinese male was initially diagnosed with ES-SCLC with synchronous hepatic metastases. Following four cycles of first-line chemotherapy comprising etoposide and carboplatin, the patient underwent thoracic radiotherapy (TRT); however, proph

Impact of Cytogenetic Abnormalities, Induction and Maintenance Regimens on Outcomes After High-Dose Chemotherapy and Autologous Stem Cell Transplantation in Patients With Newly Diagnosed Multiple Myeloma: A Decade-Long Real-World Experience.

BackgroundHigh-dose chemotherapy and autologous stem cell transplant (HDT-ASCT) has become a standard of care for transplant eligible newly diagnosed multiple myeloma (NDMM) patients. While cytogenetic abnormalities have been shown to affect outcomes after HDT-ASCT in clinical trials, these trials often exclude or underrepresent elderly patients with comorbidities and those belonging to ethnic minorities. We describe our institutional experience highlighting the impact of high-risk cytogenetic abnormalities (HRCAs) on outcomes after HDT-ASCT for NDMM patients.MethodsA total of 449 patients wit
