Keratocytes migrate against flow with a roly-poly-like mechanism.

While cell migration can be directed by various mechanical cues such as force, deformation, stiffness, or flow, the associated mechanisms and functions may remain elusive. Single cell migration against flow, repeatedly reported with leukocytes, is arguably considered as active and mediated by integrin mechanotransduction, or passive and determined by a mechanical bias. Here, we reveal a phenotype of flow mechanotaxis with fish epithelial keratocytes that orient upstream or downstream at shear stresses around tens of dyn cm-2. We show that each cell has an intrinsic orientation that results fro

The speed of the hair cell mechanotransducer channel revealed by fluctuation analysis.

Although mechanoelectrical transducer (MET) channels have been extensively studied, uncertainty persists about their molecular architecture and single-channel conductance. We made electrical measurements from mouse cochlear outer hair cells (OHCs) to reexamine the MET channel conductance comparing two different methods. Analysis of fluctuations in the macroscopic currents showed that the channel conductance in apical OHCs determined from nonstationary noise analysis was about half that of single-channel events recorded after tip link destruction. We hypothesized that this difference reflects a

Hemodynamic Forces, Endothelial Mechanotransduction, and Vascular Diseases.

Cells in the tissues and organs of a living body are subjected to mechanical forces, such as pressure, friction, and tension from their surrounding environment. Cells are equipped with a mechanotransduction mechanism by which they perceive mechanical forces and transmit information into the cell interior, thereby causing physiological or pathogenetic mechano-responses. Endothelial cells (ECs) lining the inner surface of blood vessels are constantly exposed to shear stress caused by blood flow and a cyclic strain caused by intravascular pressure. A number of studies have shown that ECs are sens

Plasma membrane disruption (PMD) formation and repair in mechanosensitive tissues.

Mammalian cells employ an array of biological mechanisms to detect and respond to mechanical loading in their environment. One such mechanism is the formation of plasma membrane disruptions (PMD), which foster a molecular flux across cell membranes that promotes tissue adaptation. Repair of PMD through an orchestrated activity of molecular machinery is critical for cell survival, and the rate of PMD repair can affect downstream cellular signaling. PMD have been observed to influence the mechanical behavior of skin, alveolar, and gut epithelial cells, aortic endothelial cells, corneal keratocyt

Adenylyl cyclase 3 regulates osteocyte mechanotransduction and primary cilium.

Osteocytes are accepted as the primary mechanosensing cell in bone, but how they translate mechanical signals into biochemical signals remains unclear. Adenylyl cyclases (AC) are enzymes that catalyze the production of second messenger cyclic adenosine monophosphate (cAMP). Osteocytes display a biphasic, cAMP response to fluid shear with an initial decrease in cAMP concentrations and then an increased concentration after sustained mechanical stimulation. To date, AC6, a calcium-inhibited AC, is the primary isoform studied in bone. Since osteocytes are calcium-responsive mechanosensors, we aske

The potential role of mechanically sensitive ion channels in the physiology, injury, and repair of articular cartilage.

Biomechanical factors play an extremely important role in regulating the function of articular chondrocytes. Understanding the mechanical factors that drive chondrocyte biological responses is at the heart of our interpretation of cascade events leading to changes in articular cartilage osteoarthritis. The mechanism by which mechanical load is transduced into intracellular signals that can regulate chondrocyte gene expression remains largely unknown. The mechanically sensitive ion channel (MSC) may be one of its specific mechanisms. This review focuses on four ion channels involved in the mech

New Therapeutic Targets for Osteoarthritis Pain.

Osteoarthritis (OA), the most common form of arthritis, causes pain and disability, as well as emotional distress. While total joint replacement is one of the most effective treatments available for improving the quality of life in people with severe OA, it is not suitable for all patients and all joints. Current pharmacological analgesics have limited efficacy, and their use is often restricted by adverse events. Medications that might reduce pain by slowing or preventing structural disease remain elusive. Our increasing understanding of the complex mechanisms that underlie OA pain offers a w

The chondrocyte primary cilium.

UNLABELLED: The presence and role of primary, or non-motile, cilia on chondrocytes has confused cartilage researchers for decades. Initial explanations attributed a vestigial nature to chondrocyte cilia. Evidence is now emerging that supports the role of the chondrocyte primary cilium as a sensory organelle, in particular, in mechanotransduction and as a compartment for signaling pathways. Early electron microscopy images depicted bent cilia aligned with the extracellular matrix (ECM) in a manner that suggested a response to mechanical forces. Molecules known to be mechanotransducers in other

Endothelial CD44 cleavage by ADAM17 impairs shear stress mechanotransduction.

In individuals with type 2 diabetes (T2D), blood flow-mediated increases in endothelial shear stress fail to elicit a robust vasodilatory response. This defective flow-mediated dilation (FMD) is associated with loss of the endothelial glycocalyx, a mechanosensitive extracellular structure that lines the luminal side of blood vessels. Hyaluronan (HA), a polysaccharide constituent of the glycocalyx, is anchored to the plasma membrane by its primary cell-surface receptor, CD44, which is implicated in mechanotransduction of shear stress. Critically, CD44 is also a substrate of a disintegrin and me

Targeting Matrix Stiffness and Mechanotransduction in Breast Cancer: Implications for Emerging Therapies.

Breast cancer remains a leading cause of mortality among women worldwide. The inherent heterogeneity in tumors among patients with breast cancer poses a challenge to effective therapeutic management. The extracellular matrix (ECM) is an important structural component of the tumor microenvironment (TME) that regulates cellular behavior. When the ECM adopts a stiff configuration, this coincides with biochemical remodeling in response to biomechanical cues that drive tumor cell invasion, immune evasion, and metastatic spread in breast cancer. Emerging studies suggest that patient ancestry signifi

A panoramic analysis of keloid pathogenesis: multidimensional network regulation of genetics-immunity-metabolism-mechanical force.

Keloid, as a unique form of pathological scar, involves complex interactions across multiple dimensions in its pathogenesis, including genetic susceptibility, epigenetic regulation, immune microenvironment disorders, metabolic reprogramming, abnormal mechanical force transduction, and fine-tuning by non-coding RNAs. At the genetic level, alleles such as HLA-DRB1*15, HOX gene family, and high-frequency mutation genes including MUC4 constitute the foundation of congenital susceptibility; at the epigenetic level, reduced DNA methylation and imbalanced m6A modification promote fibrosis by regulati

Improving Nonviral Gene Delivery by Activating Mechanosensing-Dependent Endocytic Pathways.

The delivery of nucleic acids into host cells has emerged as an innovative and promising therapeutic approach for various diseases. Despite significant advances in nanoparticle delivery systems, persistent cellular barriers limit the clinical application of most existing technologies. In this study, we developed a programmable device that applies precise uniaxial cyclic stretching to cells cultured on custom polydimethylsiloxane chambers to investigate whether mechanical stimulation can enhance the transfection efficiency (TE) of gold-standard non-viral gene delivery vectors. Fibrosis and Mechanotransduction in Endometriosis: Linking ECM Remodeling and Tissue Stiffness to Pain.

Endometriosis is characterized by pain symptoms, including dysmenorrhea, chronic pelvic pain, and dyspareunia, the severity of which does not necessarily correlate with the size or extent of lesions. Recent evidence suggests that, in addition to extracellular matrix (ECM) remodeling and tissue stiffening, neural remodeling and neuroimmune interactions may contribute to pain pathophysiology in endometriotic lesions. This review focuses on the relationship between fibrosis and mechanotransduction in endometriosis. We summarize their associations with inflammation, mechanical stimuli, intracellul

Peroxisome proliferator-activated receptor gamma (PPARγ) as a mechano-metabolic transducer: coordinating lipid homeostasis through mechanical cues.

The pathogenesis of prevalent metabolic diseases such as obesity, atherosclerosis, metabolic dysfunction-associated steatotic liver disease, and diabetes is intricately linked to dysregulated lipid metabolism. Peroxisome proliferator-activated receptor gamma (PPARγ) is a key transcriptional regulator of lipid homeostasis and a well‑researched therapeutic target. Although biochemical signaling pathways have been the traditional focus, recent studies now highlight the mechanical microenvironment (matrix stiffness, fluid shear stress, and tensile strain) as a pivotal physical metabolism regulator

Conversations in collagen: multicellular cross talk shapes the fibrotic niche in pulmonary fibrosis.

Pulmonary fibrosis is often framed as a problem of excess collagen causing stiffening of the lung, but that framing misses the more dynamic reality: fibrosis is built through ongoing conversations between fibroblasts, neighboring cells, and extracellular matrix (ECM) that interpret and reinforce injury signals. Over the last 3 years, single-cell, spatial, and multiomics tools, paired with lineage tracing and functional models, have revealed distinct fibroblast states, trajectories, and spatial niches with increasing resolution. Here, we summarize emerging principles from this sharpened point o

A mechanosensitive ion channel is required for nematode predation in nematode-trapping fungi.

UNLABELLED: Mechanosensitive ion channels of small conductance-like channels (MSL) are conserved in cell-walled organisms and enable cellular responses to mechanical stimuli. Nematode-trapping fungi (NTF) rely on nematode-derived mechanical cues to form specialized predatory structures, including adhesive network and mechanically actuated constricting ring (CR) that rapidly inflate upon nematode contact. However, the sensory mechanisms underlying these processes remain unclear. Here, using the adhesive network-forming Arthrobotrys oligospora and the CR-forming Drechslerella dactyloides as repr

Nuclear mechanobiology in confined cell migration.

Nuclear deformation is a central challenge for migration of cells through confined spaces in the tissue interstitium. In this paper, we review studies on the mechanical roles of the nucleus in confined cell migration. We focus on mechanical force generation by the cytoskeleton on the nuclear surface, the properties of sub-nuclear structures in the process, and functional responses of the nucleus in response to mechanical forces, all in the context of confined cell migration. An emerging theme is that the nucleus acts not only as a barrier for confined migration, but also as a mechanoresponsive

Advancing mechanobiology from single molecules to complex cellular systems.

Cells operate as networks of proteins, membranes, condensates and compartments, each sensing and displaying distinct intracellular mechanical properties. However, cells also sense, adapt and respond to manifold mechanical properties of the environment, including adhesion, tension, stiffness, shear, viscoelasticity, plasticity, pressure and confinement. By gauging these properties at various timescales and across nano to macro length scales, cellular systems alter their collective responses. The field of mechanobiology aims to elucidate how cellular systems such as tissues, organoids or organs

Reciprocal Coupling among Extracellular Matrix Mechanics, Mechanosensitive Ion Channels, and Cell Remodeling Behaviors.

Cells actively remodel the extracellular matrix (ECM) through a repertoire of mechanically driven behaviors, including traction force generation, migration, proteolysis, and long-term deposition. While these processes are often discussed separately, emerging evidence suggests that they are coordinated through shared mechanochemical signaling modules. In parallel, advances in hydrogel-based biomaterials have revealed that ECM mechanics, including stiffness, viscoelastic dissipation, degradability, and temporal dynamics, can systematically program cellular remodeling behaviors. In this review, w
