Selective binding of soybean agglutinin to the olfactory system of Xenopus.

The binding patterns of four different lectins conjugated to horseradish peroxidase were investigated in the nervous system of juvenile Xenopus borealis. Only the lectin soybean agglutinin revealed a very selective binding pattern, which was restricted to the olfactory system. The olfactory and vomeronasal epithelia, the olfactory and accessory olfactory nerves and the olfactory and accessory olfactory bulbs were all labelled. The ventral portions of the olfactory nerve and bulb were however more intensely labelled than their dorsal portions. The rest of the brain and spinal cord did not bind

Assessment of reinforcement enhancing effects of toluene vapor and nitrous oxide in intracranial self-stimulation.

RATIONALE: Despite widespread abuse, there are few validated methods to study the rewarding effects of inhalants. One model that may have utility for this purpose is intracranial self-stimulation (ICSS). OBJECTIVES: This study aims to compare and contrast the ICSS reward-facilitating effects of abused inhalants to other classes of abused drugs. Compounds were examined using two different ICSS procedures in mice to determine the generality of each drug's effects on ICSS and the sensitivity of the procedures. METHODS: Male C57BL/6J mice with electrodes implanted in the medial forebrain bundle

The kappa-opioid agonist U69,593 blocks cocaine-induced enhancement of brain stimulation reward.

BACKGROUND: Increasing evidence indicates that brain kappa-opioid receptors (KORs) are involved in regulation of mood states. In animal models often used to study psychiatric illness, KOR agonists produce depressive-like effects (e.g., anhedonia), whereas KOR antagonists produce antidepressant- and anxiolytic-like effects. The ability of KOR agonists to produce anhedonia-like signs in laboratory animals raises the possibility that this class of drugs might be useful to ameliorate states characterized by excess reward or motivation, such as mania or stimulant intoxication. METHODS: We examined

Apomorphine-induced turning behavior in 6-hydroxydopamine lesioned rats is increased by histidine and decreased by histidine decarboxylase, histamine H1 and H2 receptor antagonists, and an H3 receptor agonist.

The role of histamine and its receptors in basal ganglia neurocircuitry was assessed in apomorphine-induced turning behavior. Rats with unilateral 6-hydroxydopamine lesions of the substantia nigra pars compacta and medial forebrain bundle were administered histaminergic agents, and apomorphine-induced turning behavior was tested on Days 7 and 14 post-lesion. Compared with saline-treated rats, histidine (500 mg/kg, i.p.), a precursor of histamine, increased turning behavior (p<0.05), while alpha-fluoromethylhistidine (alpha-FMH, 25 microg, i.c.v.), an irreversible inhibitor of histidine decarbo

Alterations in striatal dopamine overflow during rotational behavior induced by amphetamine, phencyclidine, and MK-801.

Rats lesioned unilaterally in the medial forebrain bundle with 6-OHDA rotated ipsilateral to the lesion following injections of amphetamine, phencyclidine (PCP), and MK-801. Concurrent measurement of striatal dopamine (DA) in the intact striatum with in vivo microdialysis revealed a dissociation between rotational behavior and alterations in DA overflow induced by the three drugs. Amphetamine produced robust ipsilateral rotational behavior and a substantial elevation in striatal DA (approximately 130% increase at asymptote). PCP produced comparable increases in rotational behavior, but only ap

Hypothalamic substrates of self-stimulation in cats.

The purpose of this study was to gather anatomical data concerning sites for self-stimulation in the lateral hypothalamus in the cat. The study was conducted on 25 adult cats. In each cat, one to three monopolar stimulating electrodes were implanted bilaterally in the lateral hypothalamus in a region between sections Fr 10.0 and Fr 13.0, L 2.0 and L 5.0, and H -2.0 and H -6.0. A reference electrode was placed in the calvaria over the frontal sinus. Twenty-two of these cats learned to press a lever when each press was rewarded by a brief (0.3 s) electrical stimulus (2.0 to 7.0 V, 100/s, 1 ms du

Paraventricular organ of the lungfish Protopterus dolloi: morphology and projections of CSF-contacting neurons.

The morphology and projections of neurons in the paraventricular organ (PVO) were studied by means of silver impregnation after intraocular application of cobaltous lysine in the lungfish Protopterus dolloi. Cobalt-labeled neurons were found exclusively in the PVO in the dorsal and infundibular hypothalamus. These bipolar neurons possess one CSF-contacting process that protrudes into the ventricular lumen with a club-shape ending and a thick, ramifying process directed into the hypothalamic neuropil; the ependymofugal processes form intra- and extrahypothalamic projections. Impregnated fibers

Melatonin is protective against 6-hydroxydopamine-induced oxidative stress in a hemiparkinsonian rat model.

Melatonin is known to reduce detrimental effects of free radicals by stimulating antioxidant enzymes; however, its role has not been studied in 6-hydroxydopamine (6-OHDA)-induced rat model of Parkinson's disease (PD). Therefore, we aimed to elucidate the effects of melatonin on motor activity and oxidative stress parameters in 6-OHDA-induced rat model of PD. Three-month-old male Wistar rats were divided into 5 groups: vehicle (V), melatonin-treated (M), 6-OHDA-injected (6-OHDA), 6-OHDA-injected + melatonin-treated (6-OHDA-Mel), and melatonin-treated + 6-OHDA-injected (Mel-6-OHDA) group. Reinforcing versus anticonvulsant drugs: effects on intracranial self-stimulation rate-frequency M50 indices.

Drugs of abuse, such as amphetamine and morphine, produce reward-related shifts on intracranial self-stimulation (ICSS) thresholds. The facilitatory effects on ICSS thresholds of drugs that act through the GABAergic system, however, are reported to be attributed to their antiseizure and anticonvulsant effects, rather than their reinforcing effects. Using a rate-frequency ICSS paradigm, we examined the effects of amphetamine (a reinforcing drug of abuse that acts via the catecholaminergic system), pentobarbital (a GABA(A) receptor agonist and reinforcing barbiturate with anticonvulsant properti

Persisting expression of galanin in axotomized mamillary and septal neurons of adult rats labeled for c-Jun and NADPH-diaphorase.

In adult male rats, the expression of the neuropeptide galanin and its co-localization with the c-Jun transcription factor and the NADPH-diaphorase, the marker enzyme for the nitric oxide synthase (NOS), was investigated by immunohistochemistry in axotomized neurons following unilateral stereotaxic transection of the (a) mamillo-thalamic tract, (b) medial forebrain bundle, (c) fimbria fornix bundle and (d) sciatic nerve. This surgical procedure resulted in axotomy of neurons of (a) mamillary ncl. (MnM), (b) substantia nigra compacta (SNC) and paraventricular ncl. of thalamic (PF) neurons, (c)

The hypothalamus: anatomy and functions.

The neuroanatomy of the human hypothalamus is reviewed with special interest focused on its neuroendocrine role. The magnocellular neurons in the supraoptic and paraventricular nuclei are the site of synthesis of the nonapeptides antidiuretic hormone and oxytocin and their carriers, the neurophysins. They are in close relation with the posterior lobe of the pituitary which contains their axonal neurosecretory endings. The parvocellular neurons are scattered around the third ventricle, from the preoptic area towards the infundibulum. They control the adenohypophysis by the releasing hormones fo

Rotational behavior mediated by dopaminergic and nondopaminergic mechanisms after intranigral microinjection of specific mu, delta and kappa opioid agonists.

The regulation of motor behavior by mu, delta and kappa opiate receptors in the substantia nigra was examined. Unilateral microinjections of specific mu (DAGO), delta (DPDPE) and kappa (U-50,488H) ligands into the substantia nigra pars reticulata of rats produced dose-dependent contralateral turning. The opiate antagonist naloxone blocked these effects, suggesting that the circling was mediated through opiate receptors. The involvement of midbrain dopaminergic systems in this behavior was tested in two ways. Unilateral 6-hydroxydopamine lesions of the medial forebrain bundle decreased the circ

Investigation of nerve pathways mediating colorectal dysfunction in Parkinson's disease model produced by lesion of nigrostriatal dopaminergic neurons.

BACKGROUND: Gastrointestinal (GI) dysfunction, including constipation, is a common non-motor symptom of Parkinson's disease (PD). The toxin 6-hydroxydopamine (6OHDA) produces the symptoms of PD, surprisingly including constipation, after it is injected into the medial forebrain bundle (MFB). However, the mechanisms involved in PD-associated constipation caused by central application of 6OHDA remain unknown. We investigated effects of 6OHDA lesioning of the MFB on motor performance and GI function. METHODS: Male Sprague Dawley rats were unilaterally injected with 6OHDA in the MFB. Colorectal p

Afferents of the ventral tegmental area in the rat-anatomical substratum for integrative functions.

The ventral tegmental area (VTA) is critically important to an organism's capacity to detect rewards and novelty and to enlist appropriate behavioral responses. Although there has been substantial progress concerning information processing at the single cell and molecular levels in the VTA, our knowledge of its overall afferent connections is based principally on the benchmark description by Phillipson ([1979] J. Comp. Neurol. 187:117-144). Given that, since then, the sensitivity of tracing methods and knowledge about the organization of brain structures have increased considerably, we underto

Globus pallidus, but not entopeduncular nucleus, 6-OHDA-induced lesion attenuates L-Dopa-induced dyskinesia in the rat model of Parkinson's disease.

Although extrastriatal dopaminergic (DAergic) systems are being recognized as contributors to Parkinson's disease (PD) pathophysiology, the role of extrastriatal DA depletion in L-Dopa-induced dyskinesia (LID) is still unknown. In view of the physiologic actions of DA on pallidal neuronal activity and the effects on motor behavior of local injection of DA drugs, the loss of the external (GPe, GP in rodents) and internal (GPi, entopeduncular nucleus (EP) in rodents) pallidal DAergic innervation might differentially contribute to LID. A role of pallidal serotonergic (SER) terminals in LID has be

Dopaminergic manipulations and its effects on neurogenesis and motor function in a transgenic mouse model of Huntington's disease.

Huntington's disease (HD) is an inherited neurodegenerative disorder that is classically defined by a triad of movement and cognitive and psychiatric abnormalities with a well-established pathology that affects the dopaminergic systems of the brain. This has classically been described in terms of an early loss of dopamine D2 receptors (D2R), although interestingly the treatments most effectively used to treat patients with HD block these same receptors. We therefore sought to examine the dopaminergic system in HD not only in terms of striatal function but also at extrastriatal sites especially

Behavioral differences in a rotenone-induced hemiparkinsonian rat model developed following intranigral or median forebrain bundle infusion.

A mitochondrial complex-I inhibitor, rotenone was unilaterally infused into the substantia nigra pars compacta (SNpc) or median forebrain bundle (MFB) to create hemiparkinsonian animal models and investigated spontaneous and drug-induced stereotypic rotations, as well as certain postural behaviors in Sprague-Dawley rats. Animals infused intranigrally, but not intra-MFB, with rotenone exhibited spontaneous contralateral rotations immediately after recovery from anesthesia. Head position bias and elevated body swing test showed insignificant contralateral bias in animals with nigral damage but a

The role of the medial olfactory pathways in olfaction: behavioral and electrophysiological data.

The role of the olfactory inputs transmitted by the medial olfactory pathways --i.e. anterior limb of the anterior commissure and medial forebrain bundle-was studied in rats submitted either to a bilateral lesion of the anterior commissure (bAC group) or to a bilateral transection of the lateral olfactory tract followed by an anterior commissure bilateral lesion (bCL group). In the bCL group, the characteristic emotional reactions usually observed in sham-operated rats to biologically meaningful odorants (odor of a predator or of conspecifics) were no longer observed; in this group, the modula
