This study identifies GmRAMOSA1 as a key regulator of adventitious shoot regeneration in soybean and uncovers its regulatory network via comparative transcriptome profiling, providing new insights into the molecular mechanism of soybean shoot organogenesis. Soybean genetic transformation is strongly constrained by genotype-dependent in vitro regeneration capacity, particularly in adventitious shoot formation. Here, we compared the highly regenerable cultivar Dongnong 50 (DN50) with the low regenerative capacity Sichuan landrace Gongqiudou 5 (GQD5) using a cotyledonary node-based adventitious s

We uncovered a previously unrecognized role of asparagine synthetase 1 (ASN1) in regulating rice root elongation. The plant root system is crucial for water and nutrient acquisition. Amino acids play pivotal roles in regulating root development. In this study, we uncovered a role of asparagine synthetase 1 (ASN1) in regulating rice root development. Histological analysis of asn1 mutants showed a shortened root length resulting from reduced cell proliferation rates in the root apical meristem. Metabolite profiling indicated that jasmonic acid (JA) metabolism exhibited the most significant alter

During the domestication of Brassica campestris (syn. Brassica rapa), significant variations in bolting time have emerged, critically impacting crop yield and quality. To elucidate the genetic basis of bolting time regulation in B. campestris, we integrated selective-sweep analysis on 365 resequenced accessions with genome-wide association studies. This identified genes including BcLMI1-LF (LATE MERISTEM IDENTITY 1), an HD-ZIP I subfamily gene in the least fractionated subgenome of B. campestris. Further, gene ontology enrichment analysis indicated that BcLMI1-LF was a key candidate regulator

Many plant species can give rise to embryos from somatic cells after a simple hormone treatment, illustrating the remarkable developmental plasticity of differentiated plant cells. However, many species are recalcitrant to somatic embryo formation for unknown reasons, which poses a significant challenge to agriculture, where somatic embryogenesis is an important tool to propagate desired genotypes. The micro-RNA394 (miR394) promotes shoot meristem maintenance in Arabidopsis thaliana, but the underlying mechanisms have remained elusive. We analyzed whether miR394 affects indirect somatic embryo

Legumes have unique features, such as compound inflorescences and a complex floral ontogeny. Thus, the study of regulatory genes in these species during inflorescence and floral development is essential to understand their role in the evolutionary origin of developmental novelties. The SUPERMAN (SUP) gene encodes a C2H2 zinc-finger transcriptional repressor that regulates the floral organ number in the third and fourth floral whorls of Arabidopsis thaliana. In this work, we present the functional characterization of the Medicago truncatula SUPERMAN (MtSUP) gene based on gene expression analysi

The architecture of a plant's root system, established postembryonically, results from both coordinated root growth and lateral root branching. The plant hormones auxin and cytokinin are central endogenous signaling molecules that regulate lateral root organogenesis positively and negatively, respectively. Tight control and mutual balance of their antagonistic activities are particularly important during the early phases of lateral root organogenesis to ensure continuous lateral root initiation (LRI) and proper development of lateral root primordia (LRP). Here, we show that the early phases of

Maize is a monoecious species that produces imperfect (unisexual), highly derived flowers called florets. Within the spikelet, the basic repeating unit of the maize inflorescence, the spikelet meristem gives rise to an upper and a lower floret. Although initially bisexual, floret unisexuality is established through selective organ elimination. In addition, the lower floret of each ear spikelet is aborted early in its development, leaving the upper floret to mature as the only pistillate floret. Expression from the cytokinin-synthesizing isopentenyl transferase (IPT) enzyme under the control of

Members in the YABBY gene family of proteins are plant-specific transcription factors that play critical roles in determining organ polarity. We have isolated a cDNA clone from rice that encodes a YABBY protein. This protein, OsYAB1, is similar to Arabidopsis YAB2 (50.3%) and YAB5 (47.6%). It carries a zinc-finger motif and a YABBY domain, as do those in Arabidopsis . A fusion protein between OsYAB1 and GFP is located in the nucleus. RNA gel-blot analysis showed that the OsYAB1 gene is preferentially expressed in flowers. In-situ hybridization experiments also indicated that the transcript acc

The lifespan of annual plants is tightly linked to stem cell senescence and the cessation of proliferative activity in the inflorescence meristem (IM). A "senescence factor" originating in developing fruits has long been proposed to trigger proliferative arrest in the IM. Here, we show that jasmonic acid (JA) accumulates in the aging IM, where the JA-responsive transcription factor MYC2 directly represses WUSCHEL (WUS) expression and cytokinin accumulation. We demonstrate that sucrose originating in the siliques triggers local JA biosynthesis in the IM and induces stem cell senescence and meri

Developmental transitions occur in the life cycles of all multicellular organisms. Despite their fundamental relevance, the underlying dynamics remain poorly understood. In plants, floral transition is a key developmental process whereby the shoot apical meristem changes from producing leaves to forming flowers. Using quantitative imaging, developmental genetics, and dynamical systems theory, we show that a time-dependent bistable switch between expression of APETALA2, a key floral inhibitor, and the floral activators SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1 and FRUITFULL, can explain the dy

Stem cells are essential for the development and organ regeneration of multicellular organisms, so their infection by pathogenic viruses must be prevented. Accordingly, mammalian stem cells are highly resistant to viral infection due to dedicated antiviral pathways including RNA interference (RNAi). In plants, a small group of stem cells harbored within the shoot apical meristem generate all postembryonic above-ground tissues, including the germline cells. Many viruses do not proliferate in these cells, yet the molecular bases of this exclusion remain only partially understood. Here, we show t

The root system of potato is made up of adventitious roots (AR) that form at the base of a sprout once it emerges from the mother tuber. By definition, AR originate from dormant preformed meristems, or from cells neighboring vascular tissues in stems or leaves. This may occur as part of the developmental program of the plant (e.g., potato), or when replacing the embryonic primary roots in response to stress conditions, such as flooding, nutrient deprivation, or wounding. AR formation is studied mainly in cereals and model plants, and less is known about its developmental program in root and tu

The WUSCHEL (WUS) gene encodes a plant-specific homeodomain-containing transcriptional regulator, which plays important roles during embryogenesis, as well as in the formation of shoot and flower meristems. Here, we isolated two homologues of Arabidopsis thaliana WUS (AtWUS), GhWUS1a_At and GhWUS1b_At, from upland cotton (Gossypium hirsutum). Domain analysis suggested that the two putative GhWUS proteins contained a highly conserved DNA-binding HOX domain and a WUS-box. Expression profile analysis showed that GhWUSs were predominantly expressed during the embryoid stage. Ectopic expression of

Cadmium (Cd) contamination, through induction of oxidative stress, severely impairs plant growth. Using primary roots of Vicia faba, we investigated how a 24 h incubation in CdCl2 solution (175 µM) affects mitotic progression in meristems and assessed whether thyme essential oil (TO; 0.03%, v/v), as a natural antioxidant, can protect proliferating cells during simultaneous Cd exposure. Cd strongly inhibited root growth, reduced mitotic index tenfold (to 0.6%), induced chromatin condensation, decreased CDKA protein levels and CycB transcripts and proteins, caused pronounced microtubule bundling

Spent pot-liner (SPL) is a hazardous solid waste produced by the aluminum industry. Although its composition may vary, fluoride and cyanide salts as well as aluminum are predominant components. A seed-germination and root-elongation test was performed with Lactuca sativa seeds as a test system. SPL induced decrease of seed germination rate and root elongation. The concentration of 26.5g/L SPL was established from a regression curve as the IC50 (inhibition concentration 50%). Through chemical analyses, the concentrations of fluoride, cyanide and aluminum in SPL solutions of 26.5g/L (IC50), 39.7

Carbendazim (CBZ) is a systemic fungicide frequently detected in aquatic environments, where it coexists with multiple pollutants that may alter its toxicity. This study aimed to determine the environmental distribution of CBZ along an upstream-downstream pollution gradient in the Pazarsuyu Stream (Türkiye) and to evaluate its individual and synergistic toxic effects using the Allium cepa bioassay. Surface water samples were collected from five stations representing different agricultural impact zones, and CBZ concentrations were determined using dispersive liquid-liquid microextraction method

We studied the D3-type cyclin function during gynoecium development in Arabidopsis and how they are related to the hormone cytokinin and the transcription factor SPATULA. Growth throughout the life of plants is sustained by cell division and differentiation processes in meristematic tissues. In Arabidopsis, gynoecium development implies a multiphasic process where the tissues required for pollination, fertilization, and seed development form. The Carpel Margin Meristem (CMM) is a mass of undifferentiated cells that gives rise to the gynoecium internal tissues, such as septum, ovules, placenta,

The morphological diversification of leaf margins reflects the evolutionary reshaping of conserved developmental programs. However, fully deciphering the underlying molecular basis remains challenging due to difficulties in phenotype quantification and the incomplete characterization of complex regulatory networks. In this study, we established a computer-vision framework to decode leaf lobe trait and identified the HD-ZIP III factor PHABULOSA (BcPHB), combining genome-wide association studies (GWAS) and selective sweep analysis in Brassica campestris. We found that promoter variation defines

Plants constantly experience partial hypoxia due to varying external oxygen availability and the existence of hypoxic niches, such as the shoot apical meristem, phloem or root nodules. Waterlogging experiments indicate that hypoxic stress at the root does not only lead to a local metabolic response, such as the accumulation of hypoxia-related metabolites, but also causes metabolic alterations in the normoxic shoot. Moreover, hypoxia-related metabolites are exported from the hypoxic root towards the normoxic shoot, where they can be recycled. Maintaining import of glycolytic substrates from the

In Brassicaceae crops such as cabbage and turnip, which are harvested for their vegetative organs, floral induction can reduce the eating quality of these organs. Therefore, developing late-flowering varieties that are insensitive to vernalization is a key breeding objective. However, a major challenge in breeding these cultivars lies in the contradiction between the late-flowering trait and the need to induce flowering for crossing. To address this, we developed a flowering induction technique using grafting, in which gibberellin-induced elongated internodes are used as scions and grafted ont
