Ultrahigh antibacterial efficacy of meropenem-loaded chitosan nanoparticles in a septic animal model.

Antimicrobial resistance is one of the most significant health challenges worldwide. Meropenem is a broad spectrum beta-lactam antibiotic that possesses high activity against Gram-positive and Gram-negative bacteria. However, it has a short plasma half-life, and thus requiring frequent administration of high doses. For the first time, meropenem-loaded chitosan nanoparticles were prepared and evaluated as a potential tool to overcome antimicrobial resistance and to improve pharmacokinetics of the drug. Spherical nanosized particles were prepared and demonstrated ultrahigh encapsulation efficien

Drug-Induced Cholestasis Secondary to Meropenem in an Elderly Patient: Report of A Rare Case.

Meropenem is a broad-spectrum carbapenem antibiotic commonly used in hospitals to treat moderate-to-severe infections. Although rare, meropenem can cause drug-induced liver disease, including mild transaminase elevation, cholestasis, or hepatic failure. This report presents an 80-year-old woman admitted after a mechanical fall and diagnosed with a catheter-associated urinary tract infection. Intravenous meropenem was initiated following urine culture confirmation of Citrobacter koseri in the context of worsening confusion and elevated inflammatory markers. Six days after starting meropenem, th

Antibacterial gold nanoclusters combined with meropenem for the treatment of sepsis by alleviating the cytokine storm.

Sepsis is a life-threatening condition characterized by systemic organ dysfunction caused by a cytokine storm mediated by inflammatory mediators. Antibacterial gold nanoclusters (AuDAMP) were synthesized via a one-pot method. The therapeutic efficacy of AuDAMP was systematically evaluated in two sepsis mouse models: the cecum ligation and puncture (CLP) model and the lipopolysaccharide (LPS)-induced sepsis-associated acute lung injury model. Remarkably, AuDAMP demonstrated potent anti-septic effects through antimicrobial activity and attenuation of the cytokine storm. Furthermore, AuDAMP exhib

PBPK modelling for the evaluation of drug-drug interaction between meropenem and valproic acid.

AIMS: The present study aimed to quantitatively investigate the potential drug-drug interaction (DDI) mechanism between meropenem (MEPM) and valproic acid (VPA). METHODS: In the present study, we firstly developed a physiologically based pharmacokinetic (PBPK) model of MEPM and VPA. The verified PBPK model was then used to quantitatively investigate the potential DDI between MEPM and VPA. The effect of genetic polymorphisms of acylpeptide hydrolase (APEH) on DDI was also evaluated. RESULTS: In our simulation, the plasma concentration of hydrolysate of VPAG decreased to 63% when combined with

Evaluation of meropenem alone and combined with rifampin in the guinea pig model of pneumococcal meningitis.

Meropenem is a broad-spectrum carbapenem antibiotic that is highly active against the pathogens causing meningitis. The aims of this study was to determine the efficacies of meropenem alone and combined with rifampin against two Streptococcus pneumoniae strains with different susceptibility to beta-lactams using the guinea pig meningitis model and compare them with the standard ceftriaxone plus vancomycin therapy. All treatments except rifampin were bactericidal from 6 h. The addition of rifampin did not improve the activity of meropenem alone. Our results provide good evidence of the efficacy

Meropenem Exposure During Extracorporeal Membrane Oxygenation: What Should Intensivists Know? A Scoping Review.

BackgroundExtracorporeal membrane oxygenation (ECMO) is increasingly used in adults with severe respiratory and/or cardiac failure. Because ECMO may alter drug pharmacokinetics (PK), optimizing antimicrobial dosing in this population remains challenging. Meropenem is widely administered in intensive care units (ICUs), yet its disposition during ECMO is not fully understood. This scoping review aimed to characterize population PK methodologies and synthesize current evidence on meropenem pharmacokinetics and pharmacodynamics (PK/PD) in critically ill adults receiving ECMO.MethodsA scoping revie

Restoring Meropenem Activity Against VIM-1-Producing Klebsiella pneumoniae Through Zinc-Binding Ligands.

The growing prevalence of metallo-β-lactamase (MBL)-producing bacteria threatens the clinical efficacy of carbapenems such as meropenem. To investigate potential strategies for restoring β-lactam activity, a panel of compounds was evaluated as putative MBL inhibitors and antibiotic adjuvants. The panel included FDA-approved drugs (carbonic anhydrase inhibitors, an antiviral agent, and a metal chelator), and two novel histidine-pyridine derivatives (D- and L-2-amino-3-(1H-imidazol-4-yl)-N-[2-(pyridin-2-yl)ethyl]propenamides). The latter were synthesized and characterized by NMR spectroscopy and

Stability Study of Meropenem 50 mg/mL Eye Drops in Polypropylene Dropper Bottles.

Background/Objectives: Meropenem is a broad-spectrum carbapenem antibiotic with demonstrated efficacy against multidrug-resistant Gram-negative pathogens. Although its use as an ophthalmic formulation is off-label, growing clinical evidence supports its application in severe ocular infections such as keratitis and endophthalmitis. However, the intrinsic instability of meropenem in aqueous solutions and the absence of standardized ophthalmic preparations limit its routine use. Furthermore, no stability studies are currently available for meropenem 50 mg/mL eye drops stored in polypropylene (PP)
