Metabolic engineering of Corynebacterium glutamicum for production of myo-Inositol and D-chiro-Inositol.

D-chiro-inositol (DCI), a key signaling molecule in glucose and lipid metabolism and steroid synthesis, plays important roles in ameliorating human metabolic disorders. To achieve de novo DCI biosynthesis in Corynebacterium glutamicum, the inositol catabolism-blocked chassis strain DCI‑1 was engineered by dynamically downregulating pgi, pfkA, and zwf genes. The most effective strain DPa‑1 produced 2.89 g/L myo-inositol (MI), the direct precursor of DCI, and 0.83 g/L DCI, with the total inositol over four times the control. Subsequently, to promote the conversion of MI to DCI and the supply of

The isopentenol utilization pathway: enzyme engineering, metabolic integration, and applications in microbial terpenoid biosynthesis.

Terpenoids are among the most structurally diverse and commercially important natural products, with applications in pharmaceuticals, flavors, fragrances, and biofuels. Controllable supply of the universal precursors isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP) remains a central limitation for microbial terpenoid production. The native mevalonate (MVA) and methylerythritol phosphate (MEP) pathways are deeply integrated with sterol homeostasis, central carbon metabolism and redox balance. Their distributed regulation, cofactor demands and scale-dependent bottlenecks compl

In Vivo Metabolic Engineering of Bladder Cancer-Derived Extracellular Vesicles for Noninvasive Cancer Detection.

Early and precise diagnosis of bladder cancer (BC) is crucial for improving patient prognosis. BC-derived extracellular vesicles (BC-EVs), which carry specific molecular information from their parent tumor cells, are directly released into the urine to serve as noninvasive liquid biopsy targets. However, the clinical utility of BC-EVs is largely hindered by their low urinary abundance and high interference from massive nontumor EVs. To overcome this barrier, we present an in vivo metabolic engineering strategy for BC-EVs with a tumor-targeting reactive oxygen species (ROS)-responsive micelle p

Metabolic engineering of Escherichia coli to modulate hydrogen sulfide levels in the mammalian gut.

Hydrogen sulfide (H2S) is a microbiota-derived metabolite in the gastrointestinal tract implicated in a number of diseases. Its volatility and reactivity make experimentally controlling H2S concentration in vivo difficult, limiting our ability to interrogate its dose-dependent effects on host physiology. Engineered bacteria present a compelling solution, yet most probiotic metabolic engineering approaches have focused on in vitro optimization, failing to account for the complex intestinal environment. Here, we engineered Escherichia coli strains to produce or consume H2S in specific intestinal

Metabolic engineering of Bacillus subtilis for enhanced p-Coumaric acid production and antimicrobial applications.

p-Coumaric acid (p-CA) is widely utilized in the food, pharmaceutical and other industries, and has traditionally been derived from plant extraction or chemical synthesis. However, p-CA synthesized by the safe B. subtilis remains poorly explored. In this study, we first engineered a recombinant B. subtilis strain (PBK) capable of synthesizing p-Coumaric acid, achieving an initial yield of 3.81 mg L-1. A high-yielding strain PBnprE was then developed through promoter substitution, with a yield reaching 60.92 mg L-1, and the yield of PBnprE was further increased to 304.04 mg L-1 by optimizing fe

Insights into the methanol utilization capacity of Y. lipolytica and improvements through metabolic engineering.

Methanol is a promising sustainable alternative feedstock for green biomanufacturing. The yeast Yarrowia lipolytica offers a versatile platform for producing a wide range of products but it cannot use methanol efficiently. In this study, we engineered Y. lipolytica to utilize methanol by overexpressing a methanol dehydrogenase, followed by the incorporation of methanol assimilation pathways from methylotrophic yeasts and bacteria. We also overexpressed the ribulose monophosphate (RuMP) and xylulose monophosphate (XuMP) pathways, which led to significant improvements in growth with methanol, re

De novo biosynthesis of β-Arbutin in Komagataella phaffii based on metabolic engineering strategies.

BACKGROUND: β-Arbutin, found in the leaves of bearberry, stands out as one of the globally acknowledged eco-friendly whitening additives in recent years. However, the natural abundance of β-Arbutin is low, and the cost-effectiveness of using chemical synthesis or plant extraction methods is low, which cannot meet the requirements. While modifying the β-Arbutin synthesis pathway of existing strains is a viable option, it is hindered by the limited synthesis capacity of these strains, which hinders further development and application. RESULTS: In this study, we established a biosynthetic pathwa

Engineering Tropane Alkaloid Production Based on Metabolic Characterization of Ornithine Decarboxylase in Atropa belladonna.

Ornithine decarboxylase (ODC) plays an important role in various biological processes; however, its role in plant secondary metabolism, especially in the biosynthesis of tropane alkaloids (TAs) such as pharmaceutical hyoscyamine, anisodamine, and scopolamine, remains largely unknown. In this study, we characterized the physiological and metabolic functions of the ODC gene of Atropa belladonna (AbODC) and determined its role in TA production using metabolic engineering approaches. Feeding assays with enzyme inhibitors indicated that ODC, rather than arginine decarboxylase (ADC), plays a major r

Development of Transcription Factor-Based Designer Macrolide Biosensors for Metabolic Engineering and Synthetic Biology.

Macrolides are a large group of natural products that display broad and potent biological activities and are biosynthesized by type I polyketide synthases (PKSs) and associated enzymatic machinery. There is an urgent need to access macrolides and unnatural macrolide derivatives for drug discovery, drug manufacture, and probe development. Typically, efforts to engineer the biosynthesis of macrolides and macrolide analogues in various microbial hosts are hampered by the complexity of macrolide biosynthetic pathways and our limited ability to rationally reprogram type I PKSs and post-PKS machiner

Metabolic engineering of Escherichia coli for the production of cadaverine: a five carbon diamine.

A five carbon linear chain diamine, cadaverine (1,5-diaminopentane), is an important platform chemical having many applications in chemical industry. Bio-based production of cadaverine from renewable feedstock is a promising and sustainable alternative to the petroleum-based chemical synthesis. Here, we report development of a metabolically engineered strain of Escherichia coli that overproduces cadaverine in glucose mineral salts medium. First, cadaverine degradation and utilization pathways were inactivated. Next, L-lysine decarboxylase, which converts L-lysine directly to cadaverine, was am

Metabolic engineering as therapy for inborn errors of metabolism--development of mice with phenylalanine hydroxylase expression in muscle.

Treatment of many inherited liver enzyme deficiencies requires the removal of toxic intermediate metabolites from the blood of affected individuals. We propose that circulating toxins can be adequately cleared and disease phenotype influenced by enzyme expressed in tissues other than the liver. Phenylalanine hydroxylase (PAH) activity was constitutively expressed in skeletal and cardiac muscle of transgenic mice which carried the PAH cDNA under the transcriptional control of the mouse muscle creatine kinase promoter. Muscle PAH-expressing mice were bred to liver PAH-deficient, hyperphenylalani

Metabolic engineering of Caldicellulosiruptor bescii for 2,3-butanediol production from unpretreated lignocellulosic biomass and metabolic strategies for improving yields and titers.

The platform chemical 2,3-butanediol (2,3-BDO) is used to derive products, such as 1,3-butadiene and methyl ethyl ketone, for the chemical and fuel production industries. Efficient microbial 2,3-BDO production at industrial scales has not been achieved yet for various reasons, including product inhibition to host organisms, mixed stereospecificity in product formation, and dependence on expensive substrates (i.e., glucose). In this study, we explore engineering of a 2,3-BDO pathway in Caldicellulosiruptor bescii, an extremely thermophilic (optimal growth temperature = 78°C) and anaerobic bacte
