Acid-base regulation in the toad Bufo marinus during environmental hypoxia.

Gas exchange and the correlated changes in blood and tissue metabolic and acid-base status were investigated during long term exposure of the toad Bufo marinus to graded levels of hypoxia. During hypoxia, PCO2 values in blood and tissues fell, leading to a transient alkalosis in the extracellular but not in the intracellular space. A reduction in blood perfusion of the skin during hypoxia may explain why PCO2 was low in sartorius muscle under normoxia, but approached the PCO2 values found in other tissues (gastrocnemius muscle, ventricle) under hypoxia. At PO2 values below the critical PO2, la

Plant In Vitro Production of Phenolic Bioactives: Molecular Regulation, Functional Equivalence and Translational Challenges.

Phenolic compounds are important plant secondary metabolites with broad biological activity and potential applications in pharmaceutical, food, cosmetic, nutraceutical, and veterinary sectors. Conventional production from field-grown plants is limited by environmental variability, seasonality, and difficulties in standardizing metabolite composition. Plant in vitro cultures provide controlled systems for modulating secondary metabolism and producing phenolic compounds under defined conditions. This review summarizes current advances in plant in vitro platforms for phenolic production and discu

Physiological and metabolic response to isolated closed-head injury. Part 1: Basal metabolic state: correlations of metabolic and physiological parameters with fasting and stressed controls.

Studies of the metabolic and physiological response to closed-head injury have intimated the presence of persistent hypermetabolism. To more fully define and evaluate the metabolic response to head trauma, a prospective study was conducted in patients with isolated closed-head injuries. Metabolic and cardiopulmonary data were obtained for a 7-day period. Patients with multiple injuries or infections, or those who received steroids, were excluded. The basic treatment regimen utilized hyperventilation, bed rest with head elevation, intracranial pressure monitoring, mild fluid restriction, and ma

The challenge to reach nutritional adequacy for vitamin A: β-carotene bioavailability and conversion--evidence in humans.

β-Carotene is an important dietary source of vitamin A for humans. However, the bioavailability and vitamin A equivalency of β-carotene are highly variable and can be affected by food- and diet-related factors, including the food matrix, food-processing techniques, size of the dose of β-carotene, and the amounts of dietary fat, fiber, vitamin A, and other carotenoids in the diet as well as by characteristics of the target population, such as vitamin A status, nutrient deficiencies, gut integrity, and genetic polymorphisms associated with β-carotene metabolism. The absorption of β-carotene from

Use of epidermal equivalents generated from follicular outer root sheath cells in vitro and for autologous grafting of chronic wounds.

During wound healing, outer root sheath (ORS) cells of hair follicles can substitute for interfollicular epidermal keratinocytes and thus act as precursor cells for interfollicular epidermal keratinocytes. Owing to improved culture techniques, ORS cells can be induced to develop highly differentiated epidermal equivalents, which are close to the normal human epidermis in terms of histological, ultrastructural, biochemical and immunohistological criteria. Such epidermal equivalents provide a versatile system for various applications in vitro, e.g. the study of epidermal homeostasis, cell intera

Remarks on the Hormonal Background of the Male Equivalent of Polycystic Ovary Syndrome.

The hypothesis that the most common female endocrine disease, the polycystic ovarian syndrome (PCOS), has a male equivalent, has recently become more widely accepted. The male form of PCOS is marked by alterations in the secretion of gonadotropins, increased insulin resistance, and changes of the levels of several steroid hormones, with clinical manifestations including premature androgenic alopecia (AGA). Because these symptoms are not always found in men with genetic predispositions, knowledge of the male equivalent of PCOS needs to be supplemented by measurements of adrenal 11-oxygenated C1

Parathyroid hormone effects on serum 1,25-dihydroxyvitamin D levels in patients with X-linked hypophosphatemic rickets: evidence for abnormal 25-hydroxyvitamin D-1-hydroxylase activity.

Patients with X-linked hypophosphatemic rickets (XLH) have normal or marginally low serum 1,25-dihydroxyvitamin D [1,25-(OH)2D] levels despite manifesting hypophosphatemia and phosphate depletion, which increase 1,25-(OH)2D production in many animal species. These data are consistent with the possibility that regulation of vitamin D metabolism is abnormal in XLH. However, controversy concerning the role of phosphate in the regulation of 25-hydroxyvitamin D-1-hydroxylase activity in man has raised doubt about this proposed defect. The presence of a defect in vitamin D metabolism could be establ

Metabolism, toxicity, and carcinogenicity of trichloroethylene.

Lifetime cancer or unit risk estimates for TRI have been calculated by the EPA on the basis of metabolized dose-tumor incidence relationships. Previously, it was common practice to directly extrapolate exposure dose-tumor incidence data from laboratory animal studies to predict cancer risks in humans. Such direct species-to-species extrapolations, however, do not take into account potentially important species differences in systemic uptake, tissue distribution, metabolism, deposition at the site(s) of action, and elimination. The consideration and use of pharmacokinetic and metabolic data can

Conductive Fe₃O₄ drives a stress-dependent shift in electron-transfer strategies during anaerobic tetramethylammonium hydroxide degradation.

Anaerobic treatment of tetramethylammonium hydroxide (TMAH) wastewater is often constrained by metabolic disruption under high TMAH loading. Conductive Fe₃O₄ has shown promise for enhancing anaerobic bioprocesses; however, the dominant mechanism governing its enhancement under different metabolic conditions remains unclear. Here, anaerobic reactors were operated under stepwise increases in TMAH concentration (1-10 g/L) to compare the effects of soluble Fe²⁺ and conductive Fe₃O₄ using reactor performance, electrochemical characterization, intracellular reducing-equivalent measurements, and inte

The influence of traumatic transfemoral amputation on metabolic cost across walking speeds.

BACKGROUND: Recent literature indicates equivalent costs of walking can be achieved after a transtibial amputation when the individual is young, active, and/or has extensive access to rehabilitative care. It is unknown if a similar cohort with transfemoral amputation can also achieve lower metabolic costs of walking than previously reported. OBJECTIVE: Compare metabolic cost in individuals with a transfemoral amputation to controls and to the literature across a range of walking speeds. STUDY DESIGN: Cross-sectional. METHODS: A total of 14 individuals with a unilateral transfemoral amputati

Phenolic, Flavonoid, Short-Chain Fatty Acid, and Probiotic Biomass Changes During In Vitro Digestion and Fermentation of Litchi Seed Vinegar.

Litchi processing generates large quantities of seed by-products that are rich in starch and polyphenols but remain underutilized. In this study, the bioactivity and prebiotic potential of litchi seed vinegar (EHA-B-S1), a sustainable product derived from litchi seed by-products, were systematically evaluated. EHA-B-S1 displayed measurable in vitro antioxidant activity: total antioxidant capacity (4 mmol Trolox equivalents per mL), DPPH radical-scavenging of 50 µmol Vc equivalents per mL, and ABTS radical-scavenging activity of 1.0 mmol Vc equivalents per mL. EHA-B-S1 displayed moderate inhibi

Cardiac rehabilitation in patients with diabetes.

Cardiac rehabilitation is very important since diabetes is the set of metabolic diseases characterized by chronic hyperglycemia, with alterations in the metabolism of carbohydrates, fats and proteins as a consequence of defects in the secretion and action of insulin. When diabetes is related to cardiovascular complications, they are the main cause of death due to risk factors such as dyslipidemia, obesity and hyperglycemia, thus causing atherosclerotic changes in the vascular bed, increasing the risk of a fulminant event. The prevention of heart disease in diabetics includes preventive methods

Influence of sex and aerobic fitness on blood pressure during maximal treadmill exercise in young healthy adults.

Exaggerated exercise blood pressure (BP) is a predictor of future hypertension but commonly observed in athletes despite their reduced cardiovascular risk. Normalizing exercise BP to metabolic or mechanical work may provide better discrimination between physiological and pathological responses. This study investigated the effects of sex and aerobic fitness on peak systolic BP (SBP) and peak SBP per metabolic equivalent (SBP/MET slope). One hundred and eleven healthy adults (22 ± 5 years; 51 female) performed a graded maximal treadmill exercise test (modified Bruce protocol) with brachial BP me

Impact of dual-release hydrocortisone on disease control and metabolism in congenital adrenal hyperplasia: a retrospective cohort study.

INTRODUCTION: Congenital adrenal hyperplasia (CAH) is an autosomal recessive disorder characterized by impaired cortisol secretion and excess androgen production. The primary treatment for CAH is glucocorticoid (GC) replacement, which is essential to prevent adrenal crises and manage androgen excess. Dual-release hydrocortisone (DR-HC) is a once-daily modified-release formulation of hydrocortisone approved for adrenal insufficiency, which showed to improve the metabolic profile and reduce cardiovascular risk compared to conventional GCs. However, data on its use in CAH patients are scant. AIM

Physical activity domains and risk of gastric adenocarcinoma in the MCC-Spain case-control study.

BACKGROUND: Evidence for a protective role of physical activity against development of stomach cancer is yet inconclusive. We studied the association of domain-specific physical activity and the risk of gastric adenocarcinoma (GAC), by site and histology, in the MCC-Spain case-control study. METHODS: 428 histologically confirmed GAC cases (67% men) including the gastro-esophageal region and 3225 controls were included. Cases were recruited in hospitals from 10 different Spanish regions, whereas population controls were randomly selected within the respective hospitals' catchment areas. A phys

Cerebral magnetic resonance spectroscopy - insights into preterm brain injury.

OBJECTIVE: Magnetic resonance spectroscopy (1H-MRS) may provide clinically relevant data regarding metabolic processes that govern the course of preterm brain injury. STUDY DESIGN: 46 very preterm infants (VP) were evaluated by magnetic resonance imaging and 1H-MRS at term-equivalent age. Brain injury was assessed according to the Kidokoro scale. Moreover, 17 term-born infants with hypoxic-ischemic encephalopathy (HIE) were scanned. The metabolic profile of the central nervous system was obtained from the bilateral thalamus. RESULT: The Lipids/Creatine, Choline/Creatine, N-acetyl aspartate/C

Powered hip exoskeleton improves walking economy in individuals with above-knee amputation.

Above-knee amputation severely reduces the mobility and quality of life of millions of individuals. Walking with available leg prostheses is highly inefficient, and poor walking economy is a major problem limiting mobility. Here we show that an autonomous powered hip exoskeleton assisting the residual limb significantly improves metabolic walking economy by 15.6 ± 2.9% (mean ± s.e.m.; two-tailed paired t-test, P = 0.002) in six individuals with above-knee amputation walking on a treadmill. The observed metabolic cost improvement is equivalent to removing a 12-kg backpack from a nonamputee indi

Right prefrontal hypometabolism predicts delusions in dementia with Lewy bodies.

Delusions (DEL) are frequent in dementia with Lewy bodies (DLB); however, the neural equivalent is poorly understood. The present study therefore aimed to identify the cerebral metabolic pattern of glucose of a DLB group suffering from DEL (DLB+DEL) as compared to a non-delusional group (DLB-DEL) and a control group (NL); and to determine the predictive value of the regional metabolic deficit for DEL symptomatology in comparison to other clinical variables significantly associated with DEL. Voxel-wise comparisons were conducted between the patient and control groups in SPM2. The most significa
