Essential roles of YAP-TEAD complex in adult stem cell development during thyroid hormone-induced intestinal remodeling of Xenopus laevis.

During amphibian metamorphosis which is triggered by thyroid hormone (TH), the small intestine is extensively remodeled from the larval to adult form. In the Xenopus laevis intestine, some of the larval epithelial cells dedifferentiate into adult stem cells, which newly form the adult epithelium similar to the mammalian one. We have previously shown that TH-activated Shh, Wnt and Notch signaling pathways play important roles in adult epithelial development. Here we focus on the Hippo signaling pathway, which is known to interact with these pathways in the mammalian intestine. Our quantitative

Characterization of the Mechanistic Linkages Between Iodothyronine Deiodinase Inhibition and Impaired Thyroid-Mediated Growth and Development in Xenopus laevis Using Iopanoic Acid.

Iodothyronine deiodinases (DIO) are key enzymes that influence tissue-specific thyroid hormone levels during thyroid-mediated amphibian metamorphosis. Within the larger context of evaluating chemicals for thyroid system disrupting potential, chemical activity toward DIOs is being evaluated using high-throughput in vitro screening assays as part of U.S. EPA's ToxCast program. However, existing data gaps preclude any inferences between in vitro chemical inhibition of DIOs and in vivo outcomes relevant to ecological risk assessment. This study aimed to generate targeted data in a laboratory model

Effect assessment of reclaimed water and carbamazepine exposure on the thyroid axis of X. laevis: Apical and histological effects.

There is increasing environmental concern about the constant presence of pharmaceuticals and personal care products (PPCPs) in surface water, generally attributed to water discharge from wastewater treatment plants (WWTPs) that are unable to completely remove these compounds. The slight, but continuous, presence of these contaminants in reclaimed water (RW) poses a risk of chronic and sublethal toxicity, and the thyroid axis can likely be a target of many of these PPCPs. In this work, we addressed the effects of RW on the Xenopus laevis thyroid system. The Amphibian Metamorphosis Assay (AMA te

Internal morphological changes during metamorphosis in the sheep nasal bot fly, Oestrus ovis.

During the larval stage, oestrid flies (Diptera: Oestridae) are obligate parasites, whereas during the adult stage they are free-living and do not feed. Like other cyclorrhaphous flies, oestrids undergo metamorphosis inside an opaque puparium, formed by the contracted and hardened cuticle of the third-instar larva. The present study documents the internal morphological changes taking place during metamorphosis of the sheep nasal bot fly, Oestrus ovis L., using non-invasive, micro-CT-based virtual histology and provides quantitative data of volumetric changes in specific organs. Virtual histolo

Brief exposure to intense turbulence induces a sustained life-history shift in echinoids.

In coastal ecosystems, attributes of fluid motion can prompt animal larvae to rise or sink in the water column and to select microhabitats within which they attach and commit to a benthic existence. In echinoid (sea urchin and sand dollar) larvae living along wave-exposed shorelines, intense turbulence characteristic of surf zones can cause individuals to undergo an abrupt life-history shift characterized by precocious entry into competence - the stage at which larvae will settle and complete metamorphosis in response to local cues. However, the mechanistic details of this turbulence-triggered

The odontoclasts of Ambystoma mexicanum.

The resorption of teeth in Ambystoma mexicanum during postembryonal ontogenesis and induced metamorphosis occurs by means of light-microscopic detectable giant-cells. These have morphological and functional characters similar to those of odontoclasts of other vertebrates. The multinucleated odontoclasts resorb not only the pedicel (base), but the stalk of the tooth, too. When active, the cells form a ruffled border and a sealing zone. In this way they are able to demineralize the hard tissues of teeth (dentin and mineral of the pedicel) and to dissolve the extracellular matrix. Resorption of e

Nineteenth century research on naturally occurring cell death and related phenomena.

Research on naturally occurring cell death is older than current opinion gives credit. More than 100 nineteenth century publications deal with it, and we review most of these. Soon after the establishment of the cell theory by Schleiden and Schwann, Carl Vogt (1842) reported cell death in the notochord and adjacent cartilage of metamorphic toads. Subsequent landmark discoveries included the massive cell death that occurs in pupating diptera (Weismann 1864), chondrocyte death during endochondral ossification (Stieda 1872), phagocytosis associated with cell death in the muscles of metamorphic to

Ultrastructural characteristics of glomus cells in the external carotid artery during larval development and metamorphosis in bullfrogs, Rana catesbeiana.

Electron microscopic observations of the external carotid artery in the larvae of the bullfrog, Rana catesbeiana, showed that glomus cells are present in the subendothelial stroma of the septum between the expanded region of the external carotid artery and the carotid arch. There were some differences in the ultrastructure of the glomus cells at each stage of larval development. At the early stages (stages I, III, V, X), most glomus cells were isolated and free from the covering of a supporting cell. The cytoplasm of the glomus cells contained fewer dense-cored vesicles. No synaptic junctions

Pattern regulation in the ventral histoblasts of the housefly: induction of sternal pattern abnormalities by mechanical wounding of larval epidermal cells.

In higher Diptera, two nests of diploid cells called the ventral histoblasts, located one on either side of each abdominal segment among the polytene larval epidermal cells, give rise to the sternite and its surrounding pleura. During metamorphosis of the insect, these two groups of cells migrate and meet with each other in the midventral region of the developing adult. The cuticular pattern elements and pigmentation in the fifth sternite of the male housefly, when compared to those of other segments as well as the tergites of both sexes, are quite distinct. The above-mentioned features, coupl

Unveiling the novel role of several Flavobacteriaceae isolates: Cultivable bacteria associated with Porolithon onkodes induce Pocillopora damicornis larval settlement and metamorphosis.

Crustose coralline algae (CCA) are pivotal for coral larval settlement and metamorphosis, yet the functional roles of their associated bacteria in this critical process remain underexplored. Here, we systematically isolated 554 cultivable bacteria from the dominant CCA species Porolithon onkodes and assessed their ability to induce larval settlement and metamorphosis of Pocillopora damicornis. The isolates were dominated by Pseudomonadota (50.72%) and Bacteroidota (32.85%), with 57.40% exhibiting < 98.60% 16S rRNA gene similarity to their closest type-strain matches, suggesting that Por. onkod

Temperature reorganises developmental time during thyroid hormone-driven fish metamorphosis.

The development of organisms with complex life cycles relies on precise endocrine coordination to link environmental cues with morphogenetic programs. In teleost fish, thyroid hormones (TH) play a central role in post-embryonic development by orchestrating the onset, pacing, and consistency of metamorphosis. The influence of climate change-induced warming on developmental coordination in fish remains poorly understood. Here, we investigated how elevated temperature reshapes TH-driven developmental trajectories during fish metamorphosis using a transcriptomic time series spanning six developmen

Mechanisms of midgut injury induced by chronic trace concentrations of chlorantraniliprole in silkworm (Bombyx mori).

The diamide insecticide chlorantraniliprole (CAP) is widely applied for pest control, and its environmental residues pose toxic risks to non-target species. However, the mechanisms underlying the developmental toxicity of environmentally relevant chronic exposure to trace concentrations of CAP in non-target lepidopteran insects remain poorly characterized. Here, we used the silkworm (Bombyx mori) as a lepidopteran model to investigate the toxic effects of chronic exposure to trace concentrations of CAP on development and physiology. Chronic exposure to trace concentrations of CAP significantly

Systemic positive feedback regulation of ecdysone biosynthesis via sugar metabolism in insects.

Insects are recognized as the most diverse species in the world. They are classified into three separate groups based on their developmental characteristics: ametabola, hemimetabola, and holometabola. Holometabolous insects have attracted the attention of many developmental biologists because of their dramatic metamorphosis as a key model for studying regulatory mechanisms of development. Central to this process is ecdysone, a steroid hormone that governs insect growth and maturation. Extensive research has clarified ecdysone's structure, functions, and signaling pathways. In particular, the r

Developing under artificial light is not so bright: Oxidative stress as a physiological response to light pollution across amphibian life stages.

Light pollution has emerged as a new environmental challenge. Natural light has long been a selecting force in shaping physiological, morphological and behavioral traits across species. The introduction of artificial light at night (ALAN) disrupts the natural light-dark cycle, affecting a broad range of biological processes. In amphibians, ALAN has been suggested as a potential driver behind demographic declines in recent years. In this study, we investigated how exposure to cold LED light affects the development, body length, and oxidative status of the European tree frog (Hyla arborea). Indi

Micro-computed tomography of pupal metamorphosis in the solitary bee Megachile rotundata.

Insect metamorphosis involves a complex change in form and function. In this study, we examined the development of the solitary bee, Megachile rotundata, using micro-computed tomography (μCT) and volume analysis. We describe volumetric changes of brain, tracheae, flight muscles, gut, and fat bodies in prepupal, pupal, and adult M. rotundata. We observed that individual organ systems have distinct patterns of developmental progression, which vary in their timing and duration. This has important implications for commercial management of this agriculturally relevant pollinator.

Possible involvement of thyrotropin-releasing hormone receptor 3 in the release of prolactin in the metamorphosing bullfrog larvae.

In amphibians, thyrotropin (TSH), corticotropin (ACTH) and prolactin (PRL) are regarded as the major pituitary hormones involved in metamorphosis, their releasing factors being corticotropin-releasing factor (CRF), arginine vasotocin (AVT), and thyrotropin-releasing hormone (TRH), respectively. It is also known that thyrotropes and corticotropes are equipped with CRF type-2 receptor and AVT V1b receptor, respectively. As for PRL cells, information about the type of receptor for TRH (TRHR) through which the action of TRH is mediated to induce the release of PRL is lacking. In order to fill this

Diapause in ticks of the medically important Ixodes ricinus species complex.

Four members of the Ixodes ricinus species complex, Ixodes pacificus, Ixodes persulcatus, Ixodes ricinus and Ixodes scapularis, have, between them, a worldwide distribution within the northern hemisphere. They are responsible for the transmission of several animal and human pathogens, including the causal agents of Lyme borreliosis, tick-borne encephalitis, human granulocytic anaplasmosis and human babesiosis. Despite the importance of these ticks as vectors, the knowledge and understanding of the role that diapause plays in their complex life cycles are confused and incomplete. In view of the

Respecified larval proleg and body wall muscles circulate hemolymph in developing wings of Manduca sexta pupae.

Most larval external muscles in Manduca sexta degenerate at pupation, with the exception of the accessory planta retractor muscles (APRMs) in proleg-bearing abdominal segment 3 and their homologs in non-proleg-bearing abdominal segment 2. In pupae, these APRMs exhibit a rhythmic 'pupal motor pattern' in which all four muscles contract synchronously at approximately 4 s intervals for long bouts, without externally visible movements. On the basis of indirect evidence, it was proposed previously that APRM contractions during the pupal motor pattern circulate hemolymph in the developing wings and
