Gut-heart-kidney communication in cardiovascular disease: from a single-metabolite hypothesis to a context-dependent metabolite network.

Gut microbiota-derived metabolites are increasingly recognized as key mediators of interorgan communication in cardiovascular disease. However, the field has often relied on a single-metabolite hypothesis, particularly centered on trimethylamine N-oxide (TMAO), which does not fully capture the complexity of host-microbe-organ interactions. In this review, we argue that cardiovascular risk is better understood as a network of interacting gut-derived molecules, in which TMAO functions as a node within a broader system that includes phenylacetylglutamine, indole-related metabolites, short-chain f

Photovoltaic Gas Sensor Based on Double-Heterojunction Synergy for Self-Powered Trimethylamine Detection.

Trimethylamine (TMA), a representative volatile organic amine, is crucial for the precise and rapid evaluation of food freshness and environmental safety. However, existing gas sensors face challenges, such as low system integration and limited operational lifetime, which restrict their practical application in the Internet of Things (IoT). In this work, we fabricated a photovoltaic gas sensor by constructing a PANI:PSS/MoO3 multifunctional layer on silicon nanowire arrays. Through precise energy-band engineering and dual-heterojunction structure, we optimized the separation, transport, and tr

Gut Microbe-Generated Metabolite Trimethylamine-N-Oxide and Ischemic Stroke.

Trimethylamine-N-oxide (TMAO) is a gut microbiota-derived metabolite, the production of which in vivo is mainly regulated by dietary choices, gut microbiota, and the hepatic enzyme flavin monooxygenase (FMO), while its elimination occurs via the kidneys. The TMAO level is positively correlated with the risk of developing cardiovascular diseases. Recent studies have found that TMAO plays an important role in the development of ischemic stroke. In this review, we describe the relationship between TMAO and ischemic stroke risk factors (hypertension, diabetes, atrial fibrillation, atherosclerosis,

The Role of the Gut Microbiome and Trimethylamine Oxide in Atherosclerosis and Age-Related Disease.

The gut microbiome plays a major role in human health, and gut microbial imbalance or dysbiosis is associated with disease development. Modulation in the gut microbiome can be used to treat or prevent different diseases. Gut dysbiosis increases with aging, and it has been associated with the impairment of gut barrier function leading to the leakage of harmful metabolites such as trimethylamine (TMA). TMA is a gut metabolite resulting from dietary amines that originate from animal-based foods. TMA enters the portal circulation and is oxidized by the hepatic enzyme into trimethylamine oxide (TMA

Interplay between diet and gut microbiome, and circulating concentrations of trimethylamine N-oxide: findings from a longitudinal cohort of US men.

OBJECTIVES: Gut-produced trimethylamine N-oxide (TMAO) is postulated as a possible link between red meat intake and poor cardiometabolic health. We investigated whether gut microbiome could modify associations of dietary precursors with TMAO concentrations and cardiometabolic risk markers among free-living individuals. DESIGN: We collected up to two pairs of faecal samples (n=925) and two blood samples (n=473), 6 months apart, from 307 healthy men in the Men's Lifestyle Validation Study. Diet was assessed repeatedly using food-frequency questionnaires and diet records. We profiled faecal meta

Gut microbiota dependent trimethylamine N-oxide aggravates angiotensin II-induced hypertension.

Gut microbiota produce Trimethylamine N-oxide (TMAO) by metabolizing dietary phosphatidylcholine, choline, l-carnitine and betaine. TMAO is implicated in the pathogenesis of chronic kidney disease (CKD), diabetes, obesity and atherosclerosis. We test, whether TMAO augments angiotensin II (Ang II)-induced vasoconstriction and hence promotes Ang II-induced hypertension. Plasma TMAO levels were indeed elevated in hypertensive patients, thus the potential pathways by which TMAO mediates these effects were explored. Ang II (400 ng/kg-1min-1) was chronically infused for 14 days via osmotic minipumps

[Trimethylamine-N-oxide and cardiovascular events in chronic kidney disease].

Trimethylamine-N-oxide (TMAO) is a biologically active molecule which is metabolized by intestinal microflora and excreted by the kidney. The incidence and mortality of cardiovascular disease (CVD) in patients with chronic kidney disease (CKD) are significantly increased, which seriously affects the prognosis of patients with CKD. Recent studies have shown that TMAO can cause myocardial fibrosis and cardiovascular damage. Plasma TMAO levels are elevated in CKD patients, and TMAO was an independent predictor for all-cause mortality in patients with CKD, including CVD events. As a "uremic toxin"

Density variations of TMAO solutions in the kilobar range: Experiments, PC-SAFT predictions, and molecular dynamics simulations.

We present measurements, molecular dynamics (MD) simulations, and predictions using Perturbed-Chain Statistical Associating Fluid Theory (PC-SAFT) of the density of aqueous solutions in a pressure range from 1 bar to 5000 bar, a pressure regime that is highly relevant for both biochemical applications and the fundamental understanding of solvation. The accurate determination of density data of pressurized solutions remains challenging. We determined relative density changes from the variations in X-ray absorption through the sample and developed a new water parameter set for PC-SAFT modeling t

Evaluation of a nonanal-trimethylamine lure for collection of Culex quinquefasciatus (Diptera: Culicidae) in gravid traps.

Gravid traps are useful tools for monitoring vector-borne pathogens in mosquitoes, particularly for those pathogens transmitted by Culex quinquefasciatus Say. One of the primary challenges in the use of gravid traps is the necessity of the inclusion of an oviposition attractant, usually an infusion of organic material, which changes in attractiveness over time. However, a standardized lure, using nonanal and trimethylamine (N + TMA), has been developed and is commercially available. The N + TMA lure was tested against grass infusion and tap water in Tanzania, where Cx. quinquefasciatus is a ve

Electrochemical N-Formylation of Amines: Mechanistic Insights and Sustainable Synthesis of Formamides via a Methylisocyanide Intermediate.

Electrochemical methodologies offer a transformative approach to sustainable chemical synthesis by enabling precise, energy-efficient transformations. Here, we report the selective electrochemical N-formylation of methylamine using methanol as both reagent and solvent, facilitated by a simple glassy carbon electrode. Under optimized conditions, we achieve a faradaic efficiency (FE) of 34% for methylformamide synthesis in a neutral NaClO4 electrolyte. Mechanistic insights from in situ Fourier-transform infrared spectroscopy (FTIR) and complementary synthetic experiments reveal two distinct reac

Photoinduced Processes in Rhenium(I) Terpyridine Complexes Bearing Remote Amine Groups: New Insights from Transient Absorption Spectroscopy.

Photophysical properties of two Re(I) complexes [ReCl(CO)3(R-C6H4-terpy-κ2N)] with remote amine groups, N-methyl-piperazinyl (1) and (2-cyanoethyl)methylamine (2), were investigated. The complexes show strong absorption in the visible region corresponding to metal-to-ligand charge transfer (1MLCT) and intraligand-charge-transfer (1ILCT) transitions. The energy levels of 3MLCT and 3ILCT excited-states, and thus photoluminescence properties of 1 and 2, were found to be strongly affected by the solvent polarity. Compared to the parent chromophore [ReCl(CO)3(C6H5-terpy-κ2N)] (3), both designed com

Inverted Perovskite Solar Cells With Acid-Base Neutralized Self-Assembled Molecules.

Self-assembled monolayers (SAMs) have greatly contributed to the recent rapid development of inverted perovskite solar cells (PSCs). However, the acid nature of -PO(OH)2 in SAM commonly induces severe device degradation. Here we propose a universal method to develop neutralized SAM-N molecule through typical acid-base reaction between commonly used SAM molecules and methylamine (MA). In comparison with acid SAM (pH of 3), SAM-N shows neutral nature with pH value of 7, avoiding acid-induced device degradation. Importantly, SAM-N molecules can still assemble on neighboring ITO to regulate the wo

A refined picture of the native amine dehydrogenase family revealed by extensive biodiversity screening.

Native amine dehydrogenases offer sustainable access to chiral amines, so the search for scaffolds capable of converting more diverse carbonyl compounds is required to reach the full potential of this alternative to conventional synthetic reductive aminations. Here we report a multidisciplinary strategy combining bioinformatics, chemoinformatics and biocatalysis to extensively screen billions of sequences in silico and to efficiently find native amine dehydrogenases features using computational approaches. In this way, we achieve a comprehensive overview of the initial native amine dehydrogena

Hsp70 release from peripheral blood mononuclear cells.

There are an increasing number of studies reporting the presence of Hsps in human serum. We have investigated the release of Hsp70 into blood and culture medium from peripheral blood mononuclear cells (PBMCs), and whether this release is due to cell damage or active secretion from the cells. Intact Hsp70 was released from cells within whole blood and from purified PBMCs under normal culture conditions. Hsp70 release was rapid (0.1 ng/10(6) cells/h) over the first 2 h of culture and continued at a reduced rate up to 24 h (<0.025 ng/10(6) cells/h). Using viable cell counts and lactate dehydrogen

Separation of opium alkaloids by carbon dioxide sub- and supercritical fluid chromatography with packed columns. Application to the quantitative analysis of poppy straw extracts.

The optimization of the separation of seven opium alkaloids by sub- and supercritical fluid chromatography (SFC) using packed columns and carbon dioxide as the primary mobile phase was studied. The influence of aminated polar modifiers on bare and aminopropyl-bonded silica was investigated. It was found that the presence of an amine in the mobile phase could enhance the retention of alkaloids on aminopropylsilica. Various separation schemes were possible depending on the type of analysis needed. An aminopropyl-bonded silica used with a carbon dioxide-methanol-triethylamine-water mixture (82.95
