Fostering aquaculture is a promising strategy to meet the food demands of a growing population. Yet, marine aquaculture farms are situated primarily in nearshore regions that coincide with major mercury reservoirs─coastal sediments─wherein microbial methylation is most dynamic. Sustainable aquaculture calls for safe practices that minimize unintended stimulation of microbial production of neurotoxic bioaccumulative methylmercury. Here, we show that antibiotics released from aquaculture farms significantly boost methylmercury production by sulfate-reducing bacteria. Mercury methylation risk was

Microbial mercury (Hg) methylation drives the formation of methylmercury (MeHg) hotspots in natural environments, but the cellular pathways that determine MeHg fate and isotopic signatures remain largely unresolved. In this study, we cultured the model sulfate-reducing bacterium Pseudodesulfovibrio hydrargyri BerOc1 anaerobically under fumarate respiration for 30 h and quantified Hg speciation and fraction- and species-specific Hg isotopic compositions. The results showed that intracellular MeHg was rapidly exported and dominated the extracellular pool by 30 h (> 80%). Exported MeHg bound to s

Methylmercury (MeHg) is a hazardous neurotoxin, predominantly formed by microbial transformation of inorganic mercury in oxygen-depleted aquatic and terrestrial ecosystems. The ongoing deoxygenation of aquatic ecosystems due to global warming is likely to expand microbial niches for MeHg production. Although mercury methylators have also been reported to thrive in oxyge-deficients conditions in a few marine and freshwater ecosystems, there is a lack of comprehensive understanding of how they are distributed in freshwater systems. In this study, we retrieved hgcA genes, genomic marker for mercu

A five-compartment hybrid model that simulates the pharmacokinetic behaviour of methyl mercury (MeHg) and its biotransformation (demethylation) product inorganic mercury (Hg(II)) in humans is described. This model accounts for demethylation of MeHg at two sites: one which is within the body and a second which is extracorporeal (presumably the gut lumen). Simulations indicate that approximately 85% of demethylation occurs in the gut lumen and 15% in body tissues. The two-compartment model for Hg(II) pharmacokinetics developed by Farris et al. serves as the basis for the five-compartment model and

Methylmercury is primarily responsible for most food mercury pollution cases. However, most biosensors developed for mercury pollution analysis can only detect mercury ions. Although oxidative strong-acid digestion or microwave-assisted digestion can convert methylmercury into mercury ions, it is unsuitable for on-site detection. This study designed a bio-digestion gene circuit and integrated it into a mercury ion whole-cell biosensor, creating a novel on-site methylmercury detection method. Five alkyl mercury lyases from different bacterial genomes were screened via bioinformatics analysis, of

The origin of microbial mercury methylation has long been a mystery. Here, we employed genome-resolved phylogenetic analyses to decipher the evolution of the mercury-methylating gene, hgcAB, constrain the ancestral origin of the hgc operon, and explain the distribution of hgc in Bacteria and Archaea. We infer the extent to which vertical inheritance and horizontal gene transfer have influenced the evolution of mercury methylators and hypothesize that evolution of this trait bestowed the ability to produce an antimicrobial compound (MeHg+) on a potentially resource-limited early Earth. We specu

Methylmercury (MeHg) is a potent neurotoxin and has great adverse health impacts on humans. Organisms and sunlight-mediated demethylation are well-known detoxification pathways of MeHg, yet whether abiotic environmental components contribute to MeHg degradation remains poorly known. Here, we report that MeHg can be degraded by trivalent manganese (Mn(III)), a naturally occurring and widespread oxidant. We found that 28 ± 4% MeHg could be degraded by Mn(III) located on synthesized Mn dioxide (MnO2-x) surfaces during the reaction of 0.91 μg·L-1 MeHg and 5 g·L-1 mineral at an initial pH of 6.0 fo

Anaerobic digestion (AD) with municipal wastewater contained heavy metal mercury (Hg) highly affects the utilization of activated sludge, and poses severe threat to the health of human beings. However, the biogeochemical transformation of Hg during AD remains unclear. Here, we investigated the biogeochemical transformation and environmental characteristics of Hg and the variations of dominant microbes during AD. The results showed that Hg(II) methylation is dominant in the early stage of AD, while methylmercury (MeHg) demethylation dominates in the later stage. Dissolved total Hg (DTHg) in the

Methylmercury (MeHg) is converted to inorganic mercury (iHg) in several organs; however, its impact on tissues and cells remains poorly understood. Previously, we established a bacterial organomercury lyase (MerB)-expressing mammalian cell line to overcome the low cell permeability of iHg and investigate its effects. Here, we elucidated the cytotoxic effects of the resultant iHg on autophagy and deciphered their relationship. Treatment of MerB-expressing cells with MeHg significantly increases the mRNA and protein levels of LC3B and p62, which are involved in autophagosome formation and substr

Nanoparticulate mercury (Hg-NPs) are ubiquitous in nature. However, the lack of data on their concentration in soils impedes reliable risk assessments. This is due to the analytical difficulties resulting from low ambient Hg concentrations and background interferences of heterogeneous soil components. Here, coupled to single particle inductively coupled plasma-mass spectrometry (spICP-MS), a standardized protocol was developed for extraction and quantification of Hg-NPs in natural soils with a wide range of properties. High particle number-, particle mass-, and total mass-based recoveries were

The mer operon encodes enzymes that transform and detoxify methylmercury (MeHg) and/or inorganic mercury [Hg(II)]. Organomercurial lyase (MerB) and mercuric reductase (MerA) can act sequentially to demethylate MeHg to Hg(II) and reduce Hg(II) to volatile elemental mercury (Hg0) that can escape from the cell, conferring resistance to MeHg and Hg(II). Most identified mer operons encode either MerA and MerB in tandem or MerA alone; however, microbial genomes were recently identified that encode only MerB. However, the effects of potentially producing intracellular Hg(II) via demethylation of MeHg

Microbial reduction of inorganic divalent mercury (Hg2+) and methylmercury (MeHg) demethylation is performed by the mer operon, specifically by merA and merB genes, respectively, but little is known about the mercury tolerance capacity of marine microorganisms and its prevalence in the ocean. Here, combining culture-dependent analyses with metagenomic and metatranscriptomic data, we show that marine bacteria that encode mer genes are widespread and active in the global ocean. We explored the distribution of these genes in 290 marine heterotrophic bacteria (Alteromonas and Marinobacter spp.) is

Particulate matter (PM) impregnated with methlymercury (MeHg+) was analyzed using atomic absorption spectrometry coupled to a customized dielectric barrier discharge (DBD) device. Chemical vapor generation (CVG) was applied to generate methylmercury hydride and the DBD device promoted bond cleavage with subsequent release of free Hg atoms to the gas phase. Hydride generation was carried out using a lab-made syringe-based device in batch mode using argon as a carrier gas. Optimized conditions included the use of 1.0 mL of a 0.05% m/v NaBH4 solution and 1.0 mL of a 10% v/v HCl solution. This sys

Investigation of mercury (Hg) from atmospheric precipitation is important for evaluating its ecological impacts and developing mitigation strategies. Western China, which includes the Tibetan Plateau and the Xinjiang Uyghur Autonomous Region, is one of the most remote region in the world and is understudied in regards to Hg precipitation. Here we report seesaw-like patterns in spatial variations of precipitation Hg in Western China, based on Hg speciation measurements at nine stations over this remote region. The Hg fraction analyzed included total Hg (HgT), particulate-bound Hg (HgP) and meth

Aerobic methanotrophy is strongly controlled by copper, and methanotrophs are known to use different mechanisms for copper uptake. Some methanotrophs secrete a modified polypeptide-methanobactin-while others utilize a surface-bound protein (MopE) and a secreted form of it (MopE*) for copper collection. As different methanotrophs have different means of sequestering copper, competition for copper significantly impacts methanotrophic activity. Herein, we show that Methylomicrobium album BG8, Methylocystis sp. strain Rockwell, and Methylococcus capsulatus Bath, all lacking genes for methanobactin

Reducing health risk of mercury (Hg)/methylmercury (MeHg) in sewage sludge is vital to its land application. This study revealed that thermal hydrolysis reduced MeHg content both during pretreatment process and subsequent anaerobic digestion (AD), which resulted in decrease of MeHg content from 4.24 ng/g to 0.95 ng/g after thermal hydrolysis (150 ℃) and further decreased to 0.39 ng/g after AD. Notably, thermal hydrolysis at high temperature (120 ℃ and 150 ℃) promoted both Hg methylation and MeHg demethylation rather than the control or at low temperature (100 ℃). Hg methylation dominated in hy

The Baihua Reservoir (Guizhou Province, Southwest China) has a history of mercury contamination associated with past acetic acid production activities at the Guizhou Organic Chemical Plant (GOCP). Soil and plant samples collected from riparian zones were analyzed for total Hg (THg) and methylmercury (MeHg) using cold vapor atomic fluorescence spectroscopy. The concentrations of THg and MeHg in soil samples were in the range of 109-371 ng g-1 and 0.32-1.80 ng g-1, respectively. Soils in the riparian zones close to the pollution source (the GOCP) presented higher Hg contamination, with relativel

Elevated methylmercury (MeHg) production in mining-contaminated paddy soils, despite the high fraction of refractory HgS(s), has been frequently reported, while the underlying mechanisms are not fully understood. Here, we hypothesized that sulfate input, via fertilization, rainfall, and irrigation, is critical in mobilizing refractory HgS(s) and thus enhancing Hg methylation in mining-contaminated paddy soils. To test this hypothesis, the effects of sulfate amendment on Hg methylation and MeHg bioaccumulation in mining-contaminated soil-rice systems were examined. The results indicated 28-61%

In natural freshwater and sediments, mercuric mercury (Hg(II)) is largely associated with particulate minerals and organics, but it remains unclear under what conditions particulates may become a sink or a source for Hg(II) and whether the particulate-bound Hg(II) is bioavailable for microbial uptake and methylation. In this study, we investigated Hg(II) sorption-desorption characteristics on three organo-coated hematite particulates and a Hg-contaminated natural sediment and evaluated the potential of particulate-bound Hg(II) for microbial methylation. Mercury rapidly sorbed onto particulates

Four commercially available sorbents (BioChar (BC), ThiolSAMMS® (TS), SediMite (SM), and Organoclay™ PM-199 (OC-199)) were tested for their ability to sorb methylmercury (MeHg) and MeHg complexed with dissolved organic matter (DOM). Testing sorption behavior with DOM is more representative of the environmental conditions and mercury speciation expected during in-situ remediation efforts. Isotherms were fit using a robust, iterative re-weighting scheme. This fitting approach improves upon the traditionally used indirect sorption method by removing the dependence between aqueous and solid phase
