N-Acetyl Cysteine and Vitamin C Modulate the Antibiotic Efficacy Against Escherichia coli Cells.

Supplements with their own beneficial effect on hosts are consumed by us. N-acetyl cysteine (NAC) and Vitamin C (Vit C) are antioxidants and supplements, consumed for their beneficial properties. The present investigation evaluates the effect of their antioxidant property on antibiotic efficacy against Escherichia coli cells from different physiological states, including exponential and stationary-phase, cell aggregates, and in-vitro stress-induced persister cells. Survival was measured in cfu/mL by cfu (colony-forming unit) counting, with efficacy determined by log-fold change in survival by

A screening system using minimal media identifies a flavin-competing inhibitor of Porphyromonas gingivalis growth.

Chronic periodontitis is caused by dysbiosis of human oral commensals and especially by increase in Porphyromonas gingivalis. Inhibitors of P. gingivalis growth are expected to serve as effective drugs for the periodontal therapy. In the present study, we isolated new growth inhibitors of P. gingivalis using minimal media for P. gingivalis. The minimal media included the previously reported Globulin-Albumin (GA) and the newly developed Lactalbumin-Ferric chloride (LF) and Globulin-Calcium chloride (GC); all supported growth of the wild-type strain of P. gingivalis but did not support the growt

Synergistic Activity of Niclosamide in Combination With Colistin Against Colistin-Susceptible and Colistin-Resistant Acinetobacter baumannii and Klebsiella pneumoniae.

Colistin is among the few antibiotics effective against multidrug-resistant Acinetobacter baumannii and Klebsiella pneumoniae clinical isolates. However, in the last few years, colistin-resistant A. baumannii and K. pneumoniae strains have emerged. Therefore, combination therapies, between colistin and other old drugs, restoring the activity of colistin are required. The main objective of this study was to analyse the activity of niclosamide, an anthelmintic drug, in combination with colistin against colistin-susceptible (Col-S) and colistin-resistant (Col-R) A. baumannii and K. pneumoniae. In vitro activity of artemisone and artemisinin derivatives against extracellular and intracellular Helicobacter pylori.

The in vitro activity of the new artemisinin derivative artemisone as well as other molecules of the same class against Helicobacter pylori and their effects when combined with standard antibiotics were evaluated. Since H. pylori can be internalised into gastric epithelial cells, the effects of artemisinin, dihydroartemisinin and artemisone against intracellular H. pylori were also investigated. Bacteriostatic [minimum inhibitory concentration (MIC)] and bactericidal [minimum bactericidal concentration (MBC)] activities were assessed against 24 clinical strains of H. pylori with different anti

Searching for new strategies against polymicrobial biofilm infections: guanylated polymethacrylates kill mixed fungal/bacterial biofilms.

OBJECTIVES: Biofilm-related human infections have high mortality rates due to drug resistance. Cohabitation of diverse microbes in polymicrobial biofilms is common and these infections present additional challenges for treatment compared with monomicrobial biofilms. Here, we address this therapeutic gap by assessing the potential of a new class of antimicrobial agents, guanylated polymethacrylates, in the treatment of polymicrobial biofilms built by two prominent human pathogens, the fungus Candida albicans and the bacterium Staphylococcus aureus. METHODS: We used imaging and quantitative met

Relation between antibiotic susceptibility and ultrastructure of Staphylococcus aureus biofilms on surgical suture.

BACKGROUND: Infectious biofilms are recalcitrant to antimicrobial therapy, but the mechanism(s) responsible for the greater resistance are unclear. Experiments were designed to clarify the association between antibiotic resistance and biofilm ultrastructure. METHODS: Staphylococcus aureus was cultivated for 24 h on silk suture, where robust biofilms formed. Initial experiments compared the susceptibilities of planktonic (free-living) cells and mechanically dispersed biofilm cells to ampicillin, oxacillin, and vancomycin. Antibiotics in bactericidal concentrations were then incubated overnight

Biofilm formation directly correlates with cell viability in Candida tropicalis on polypropylene.

Candida tropicalis, the most prevalent non-Candida albicans Candida species, is an emerging pathogen forming robust biofilms on medical devices, contributing to biofouling, virulence, and antifungal resistance. In this study, growth conditions for six C. tropicalis clinical isolates (C4, U873, U951, U1179, U1309, U1360) and a standard strain (MTCC-184) were optimized on polypropylene using central composite design-based response surface methodology. The parameters tested included temperature, pH, shaker speed, inoculum size, and incubation time, with biofilm formation quantified by crystal vio

Evaluation of the efficacy of inactivated fecal transplants against Clostridioides difficile: an in vitro study.

Fecal microbiota transplantation (FMT) requires rigorous donor screening, limiting donor eligibility and increasing costs. Recent evidence suggests that bacterial viability may not be essential for treating recurrent Clostridioides difficile infections. This study aimed to evaluate whether inactivated fecal preparations retain anti-C. difficile (CD) activity in vitro. Stool samples from three donors underwent eight different inactivation protocols, including mechanical, thermal, and chemical treatments. Bacterial viability and anti-CD activity were assessed by culture and inhibition assays. Me

A photoactivation-free viability PCR reagent for rapid, culture-independent detection of live bacteria.

UNLABELLED: Detection of viable bacteria in complex samples is a long-standing challenge in microbiology and public health. Traditional culture methods are slow and often fail to capture viable but non-culturable cells, while PCR-based methods can overestimate bacterial loads by amplifying DNA from non-viable (dead) cells. To address this challenge, we evaluated a novel, photoactivation-free viability PCR (vPCR) reagent from Promega Corporation that selectively crosslinks DNA from non-viable cells, rendering it unamplifiable by qPCR. This approach enables accurate, culture-independent quantifi

Investigation of a viable but non-culturable state in Porphyromonas gingivalis and host cell invasion.

Porphyromonas gingivalis (P. gingivalis) is a gram-negative, black-pigmented, anaerobic pathogen known for its biofilm formation and its central role in periodontal disease. More recently, P. gingivalis has been implicated in various systemic conditions, including atherosclerosis, Alzheimer's disease, and certain types of cancer, such as pancreatic and oral cancer. This bacterium employs several mechanisms to evade environmental stress, thereby contributing to its pathogenicity. The viable but non-culturable (VBNC) state is characterized by bacteria that remain viable but have reduced metaboli

Microbial Survival, Sublethal Injury, and Recovery in Frozen Food Systems: Implications for Safety, Quality Deterioration, and Cold-Chain Control.

Freezing is widely regarded as a microbiologically protective process that suppresses growth and extends shelf life but rarely eliminates microorganisms. Microbial risk in frozen foods is, therefore, determined not only by survival during storage at -18°C but by post-thaw repair and growth during thawing and handling. This review synthesizes freeze-induced injury mechanisms, including membrane damage, osmotic stress, oxidative injury, and macromolecular disruption, and links them to physiological state transitions, lag phase behavior, and recovery potential. Rather than treating sublethal inju

Yeasticidal efficacy of far-UV-C radiation with 233 nm peak wavelength for inactivating Candida spp. with focus on the clinically relevant species C. auris for potential application on the skin.

BACKGROUND: Candida spp. are significant pathogens for hospital-acquired infections. Infections with Candida auris especially have gained increased attention due to their transmissibility and resistance properties, making new decolonization tools necessary. Far-ultraviolet-C (UV-C) radiation might be a new tool to inactivate Candida spp. independent of their resistance profile. The use of light-emitting diodes (LEDs) allows a flexible application of the radiation enabling, for example, the treatment of areas that are not accessible by the use of common antiseptics such as the throat or the mid

Mycobacterium tuberculosis induces delayed lipid droplet accumulation in dendritic cells depending on bacterial viability and virulence.

Tuberculosis remains a global health threat with high morbidity. Dendritic cells (DCs) participate in the acute and chronic inflammatory responses to Mycobacterium tuberculosis (Mtb) by directing the adaptive immune response and are present in lung granulomas. In macrophages, the interaction of lipid droplets (LDs) with mycobacteria-containing phagosomes is central to host-pathogen interactions. However, the data available for DCs are still a matter of debate. Here, we reported that bone marrow-derived DCs (BMDCs) were susceptible to Mtb infection and replication at similar rate to macrophages

Helicobacter pylori Dormant States Are Affected by Vitamin C.

Helicobacter pylori colonizes human gastric mucosa, overcoming stressful conditions and entering in a dormant state. This study evaluated: (i) H. pylori's physiological changes from active to viable-but-non-culturable (VBNC) and persister (AP) states, establishing times/conditions; (ii) the ability of vitamin C to interfere with dormancy generation/resuscitation. A dormant state was induced in clinical MDR H. pylori 10A/13 by: nutrient starvation (for VBNC generation), incubating in an unenriched medium (Brucella broth) or saline solution (SS), and (for AP generation) treatment with 10xMIC amo

Combined laser and ozone therapy for onychomycosis in an in vitro and ex vivo model.

In order to develop a fast combined method for onychomycosis treatment using an in vitro and an ex vivo models, a combination of two dual-diode lasers at 405 nm and 639 nm wavelengths, in a continuous manner, together with different ozone concentrations (until 80 ppm), was used for performing the experiments on fungal strains growing on PDA agar medium or on pig's hooves samples. In the in vitro model experiments, with 30 min combined treatment, all species are inhibited at 40 ppm ozone concentration, except S. brevicaulis, which didn't show an inhibition in comparison with only ozone treatmen

Identification of a reliable fixative solution to preserve the complex architecture of bacterial biofilms for scanning electron microscopy evaluation.

Bacterial biofilms are organized sessile communities of bacteria enclosed in extracellular polymeric substances (EPS). To analyze organization of bacteria and EPS in high resolution and high magnification by scanning electron microscopy (SEM), it is important to preserve the complex architecture of biofilms. Therefore, fixation abilities of formalin, glutaraldehyde, and Methacarn (methanol/chloroform/acetic acid-6:3:1) fixatives were evaluated to identify which fixative would best preserve the complex structure of bacterial biofilms. Economically important Gram-negative Mannheimia haemolytica,

Dihydroartemisinin inhibits multiplication of Brucella suis vaccine strain 2 in murine microglia BV2 cells via stimulation of caspase‑dependent apoptosis.

Brucellosis, caused by a facultative intracellular parasite Brucella species, is the most common bacterial zoonotic infection worldwide. Brucella can survive and proliferate in several phagocytic and non‑phagocytic cell types. Human brucellosis has similar clinical symptoms with systemic diseases, which may lead to delay of diagnosis and increasing of complications. Therefore, investigating the proliferation of Brucella in host cells is important to understand the pathogenesis of the disease. Dihydroartemisinin (DHA), a semi‑synthetic derivative of artemisinin, has been recommended by World He
