Heterolinker MOFs synergized with entropy-driven DNA amplification for precision dual-mode sensing of Microcystin-LR.

To overcome the high risk of false results in single-mode detection for environmental toxins, a self-validating dual-mode biosensing platform integrating photoelectrochemical (PEC) and fluorescence (FL) signals was developed for the ultrasensitive detection of Microcystin-LR (MC-LR). The sensor of core innovation lies in a heterolinker-engineered metal-organic framework (MOF), where the incorporation of 1,4-diazabicyclo [2.2.2] octane (Dabco) transformed a two-dimensional Zn-tetrakis (4-carboxyphenyl) ethylene (Zn-TCBPE) layer into a stable three-dimensional Zn-TCBPE-Dabco architecture. Negative effects of microcystin-LR on larval zebrafish: Focus on visual function, behavior, and circadian rhythm regulation.

Microcystin-LR (MC-LR) is the most prevalent and toxic microcystin congeners, posing a significant threat to aquatic organisms as well as humans; however, its underlying toxic mechanisms remain incompletely elucidated. In this study, the negative impacts of MC-LR and the underlying mechanisms in zebrafish larvae were investigated. The results demonstrated that MC-LR could penetrate zebrafish larvae and induce developmental toxicity, characterized by reduced heart rate, decreased body length, and smaller eye area. H&E staining revealed that MC-LR exposure significantly reduced the thickness of

Mechanistic investigation of the hepatotoxicity and potential carcinogenicity of microcystin-LR, -LA, and -RR in the HepaRG cell line model.

Microcystins (MCs) are cyanobacterial toxins found in water and food and are suspected to pose significant health risks, particularly to the liver. To gain insight into the mechanisms of hepatotoxicity and the potential carcinogenicity of MCs, this study investigated the effects of MC-LR, -LA, and -RR (congeners differing in cellular uptake and detoxification efficiency) on PP2A activity, cell proliferation, and gene expression profiling focused on toxicity pathways and carcinogenesis in HepaRG cells. The three MCs similarly inhibited PP2A activity; however, only MC-LR and -LA significantly in

Microcystin-LR drives hepatic meta-inflammation and insulin resistance by hijacking the PP2A-JNK signaling axis.

Chronic low-grade inflammation is a pivotal driver of metabolic diseases, yet the environmental triggers that initiate this process are not fully defined. Here, we identify the ubiquitous freshwater cyanotoxin, Microcystin-LR (MC-LR), as a potent inducer of hepatic meta-inflammation and subsequent insulin resistance. We demonstrate that chronic exposure to MC-LR in mice provokes a robust pro-inflammatory state in the liver, characterized by the activation of the NF-κB signaling pathway and elevated production of key cytokines such as TNF-α, IL-1β, and IL-6. This inflammatory milieu was tightly

Unraveling the Toxicity of a Non-Microcystin-Producing Strain (CCIBt3106) of Microcystis aeruginosa: Ecotoxicological Effects on Aquatic Invertebrates.

Cyanobacterial blooms are becoming increasingly frequent and intense worldwide, often dominated by Microcystis aeruginosa, a species capable of producing a wide array of bioactive metabolites beyond microcystins. This study evaluates the ecotoxicological potential of a non-microcystin-producing strain, M. aeruginosa CCIBt3106, using acute immobilization assays with three microcrustacean species: Daphnia similis, Artemia salina, and Parhyale hawaiensis. Biomass was extracted using solvents of varying polarity, and selected extracts (aqueous and 50% methanol) were further fractionated and analyz

Key drivers of microcystin-producing cyanobacteria in South Korean eutrophic waters determined with data-driven models.

The rise in cyanobacterial harmful algal blooms (CHABs), driven by eutrophication and climate change, necessitates understanding cyanotoxin conditions to mitigate risks. However, limited studies have explored the influencing factors of cyanobacterial community and cyanotoxins in freshwaters using large datasets and data-driven models (DDMs). This study examines cyanobacterial composition across regions and relates it to microcystin-LR, -RR, -LA, -YR, -LF, and -LY as well as to the taste and odor compounds geosmin and 2-methylisoborneol. It also assesses the impact of environmental factors usin

Rapid microcystin-LR detection using antibody-based electrochemical biosensors with a simplified calibration curve approach.

Harmful algal blooms (HABs) can release cyanotoxins such as microcystins (MCs), especially, microcystin-leucine-arginine (MC-LR) which is one of the commonest and most toxic, into our water bodies and can lead to several acute or chronic diseases such as liver diseases and respiratory irritation in humans. There is an increasing need for rapid and simple detection of MC-LR in water bodies for early warning of HABs. In this study, we developed an innovative on-site screening electrochemical impedance spectroscopy (EIS) biosensor with a simplified calibration curve that can rapidly detect blooms

Mutations at Two Key Sites in PP2A Safeguard Caenorhabditis elegans Neurons from Microcystin-LR Toxicity.

Microcystin-LR (MC-LR) is a secondary metabolite produced by cyanobacteria, globally renowned for its potent hepatotoxicity. However, an increasing body of research suggests that it also exhibits pronounced neurotoxicity. PP2A is a fundamental intracellular phosphatase that plays a pivotal role in cell development and survival. Although extensive research has focused on the binding of MC-LR to the C subunit of PP2A, few studies have explored the key amino acid sites that can prevent the binding of MC-LR to PP2A-C. Due to the advantages of Caenorhabditis elegans (C. elegans), such as ease of ge

A Förster Resonance Energy Transfer (FRET)-Based Immune Assay for the Detection of Microcystin-LR in Drinking Water.

Cyanobacteria bloom is the term used to describe an abnormal and rapid growth of cyanobacteria in aquatic ecosystems such as lakes, rivers, and oceans as a consequence of anthropic factors, ecosystem degradation, or climate change. Cyanobacteria belonging to the genera Microcystis, Anabaena, Planktothrix, and Nostoc produce and release toxins called microcystins (MCs) into the water. MCs can have severe effects on human and animal health following their ingestion and inhalation. The MC structure is composed of a constant region (composed of five amino acid residues) and a variable region (comp

Hepatotoxicity of cyanotoxin microcystin-LR in human: Insights into mechanisms of action in the 3D culture model Hepoid-HepaRG.

Microcystin-LR (MC-LR) is a potent hepatotoxin produced by harmful cyanobacterial blooms (CyanoHABs). MC-LR targets highly differentiated hepatocytes expressing organic anion transporting polypeptides OATP1B1 and OATP1B3 that are responsible for hepatocellular uptake of the toxin. The present study utilized an advanced 3D in vitro human liver model Hepoid-HepaRG based on the cultivation of collagen-matrix embedded multicellular spheroids composed of highly differentiated and polarized hepatocyte-like cells. 14-d-old Hepoid-HepaRG cultures showed increased expression of OATP1B1/1B3 and sensitiv

Sub-chronic microcystin-LR renal toxicity in rats fed a high fat/high cholesterol diet.

Microcystin-LR (MCLR) is a liver and kidney toxin produced by cyanobacteria. Recently, it was demonstrated that MCLR exposure drives the progression of high fat/high cholesterol (HFHC) induced nonalcoholic fatty liver disease (NAFLD) to a more severe state. NAFLD is also a risk factor for chronic kidney disease (CKD), and the current study investigated MCLR renal toxicity in the context of an HFHC diet. Sprague Dawley rats were fed either a control diet or an HFHC diet for 10 weeks. After 6 weeks of diet, animals were administered either vehicle, 10 μg/kg, or 30 μg/kg MCLR via intraperitoneal

Dual-Modal Split-Type Immunosensor for Sensitive Detection of Microcystin-LR: Enzyme-Induced Photoelectrochemistry and Colorimetry.

Microcystins, the lethal cyanotoxins from Microcystis aeruginosa, can inhibit the activity of protein phosphatase and promote liver tumors. Herein, a dual-modal split-type immunosensor was constructed to detect microcystin-LR (MC-LR), based on the photocurrent change of CdS/ZnO hollow nanorod arrays (HNRs) and the blue shift of the surface plasmon resonance peak from Au nanobipyramids@Ag. By using mesoporous silica nanospheres as the carrier to immobilize secondary antibody and DNA primer, a hybridization chain reaction was adopted to capture alkaline phosphatase, while its catalytic reaction

Hepatotoxic microcystin removal using pumice embedded monolithic composite cryogel as an alternative water treatment method.

Microcystins are the most commonly encountered water-borne cyanotoxins which present short- and long-term risks to human health. Guidelines at international and national level, and legislation in some countries, have been introduced for the effective health risk management of these potent hepatotoxic, tumour-promoters. The stable cyclic structure of microcystins and their common production by cyanobacteria in waterbodies at times of high total dissolved organic carbon content presents challenges to drinking water treatment facilities, with conventional, advanced and novel strategies under eval

Removal of microcystin-LR from drinking water using a bamboo-based charcoal adsorbent modified with chitosan.

A new kind of low-cost syntactic adsorbent from bamboo charcoal and chitosan was developed for the removal of microcystin-LR from drinking water. Removal efficiency was higher for the syntactic adsorbent when the amount of bamboo charcoal was increased. The optimum dose ratio of bamboo charcoal to chitosan was 6:4, and the optimum amount was 15 mg/L; equilibrium time was 6 hr. The adsorption isotherm was non-linear and could be simulated by the Freundlich model (R2 = 0.9337). Adsorption efficiency was strongly affected by pH and natural organic matter (NOM). Removal efficiency was 16% higher a

Promotion of oxidative stress in the aquatic macrophyte Ceratophyllum demersum during biotransformation of the cyanobacterial toxin microcystin-LR.

Cyanobacterial toxins have been shown to have adverse effects on mammals, birds and fish and are therefore being increasingly recognised as a potent stress and health hazard factor in aquatic ecosystems. Microcystins, which are cyclic heptapeptides and a main group of the cyanotoxins, are mainly retained within the producer-cells during cyanobacterial bloom development. However, these toxins are released into the surrounding medium by senescence and lysis of the blooms. The released toxins could then come into contact with a wide range of aquatic organisms including invertebrates, fish and aqu

Comparative toxicity evaluation of cyanobacterial cyclic peptide toxin microcystin variants (LR, RR, YR) in mice.

The cyclic peptide toxins microcystins and nodularins are the most common and abundant cyanotoxins present in diverse water systems. They have been the cause of human and animal health hazards and even death. Over 60 microcystin variants have been reported so far. We report here the results of our study on comparative toxicity evaluation of three most predominant microcystins, MC-LR, MC-RR and MC-YR in mice. The mice were administered one LD(50) dose of MC-LR, RR and YR (43, 235.4 and 110.6 micro g/kg body weight, respectively), and biochemical and histological variables were determined at 30

Are microcystin-LR and cylindrospermopsin neurotoxic for human cells? An in vitro study on induced pluripotent stem cells-derived models.

Microcystin LR (MC LR) and cylindrospermopsin (CYN) are cyanobacterial toxins commonly detected during harmful algal blooms in freshwater systems. Their increasing occurrence raises concerns about water quality and potential risks to human health. Although the liver is their target organ, these cyanotoxins can also affect the nervous system. The potential neurotoxicity of MC-LR and CYN is still scarcely investigated, particularly for CYN. In this study, the effects of both cyanotoxins on neural progenitor cells were evaluated. MC-LR did not significantly affect cell viability at concentrations
