The Role of Mitophagy in Various Neurological Diseases as a Therapeutic Approach.

Mitochondria are critical to multiple cellular processes, from the production of adenosine triphosphate (ATP), maintenance of calcium homeostasis, synthesis of key metabolites, and production of reactive oxygen species (ROS) to maintain necrosis, apoptosis, and autophagy. Therefore, proper clearance and regulation are essential to maintain various physiological processes carried out by the cellular mechanism, including mitophagy and autophagy, by breaking down the damaged intracellular connections under the influence of various genes and proteins and protecting against various neurodegenerativ

The role of mitophagy in the regulation of mitochondrial energetic status in neurons.

Mitochondria are the main cellular energy powerhouses and supply most of the energy in the form of ATP to fuel essential neuronal functions through oxidative phosphorylation (OXPHOS). In Alzheimer disease (AD), metabolic and mitochondrial disruptions are an early feature preceding any histopathological and clinical manifestations. Mitochondrial malfunction is also linked to synaptic defects in early AD. Mitophagy serves as a key cellular quality control mechanism involving sequestration of damaged mitochondria within autophagosomes and their subsequent degradation in lysosomes. However, it rem

Reduced mitophagy in the cochlea of aged C57BL/6J mice.

An increase in mitochondrial damage has been associated with a decline in the ability to mitigate damage through mitophagy in age-related pathologies. The present study aimed to investigate the changes of mitophagy in a mouse model with age-related hearing loss. C57BL/6J mice were divided into two groups: young (1 month) and aged (12 months). Hearing tests were conducted through the measurement of auditory brainstem response (ABR). Mitochondrial DNA copy number, the level of mitochondrial DNA damage, mitochondrial biogenesis, and mitophagy-related genes and proteins were investigated using rea

Ubiquitin and Receptor-Dependent Mitophagy Pathways and Their Implication in Neurodegeneration.

Selective autophagy of mitochondria, or mitophagy, refers to the specific removal and degradation of damaged or surplus mitochondria via targeting to the lysosome for destruction. Disruptions in this homeostatic process may contribute to disease. The identification of diverse mitophagic pathways and how selectivity for each of these pathways is conferred is just beginning to be understood. The removal of both damaged and healthy mitochondria under disease and physiological conditions is controlled by either ubiquitin-dependent or receptor-dependent mechanisms. In this review, we will discuss t

RNF10 deficiency in cardiomyocytes impairs mitophagy and drives pathological hypertrophy.

Mitophagy-mediated mitochondrial quality control is essential for normal cardiac physiology. In this study, we observed that cardiac RNF10 expression was induced by multiple chronic stressors, including aging, angiotensin II (Ang II) exposure, and obesity. Cardiac-specific RNF10 knockout (RNF10-CKO) mice developed cardiac hypertrophy with aging, characterized by cardiomyocyte enlargement, exacerbated myocardial fibrosis, and impaired cardiac function. Aged RNF10-CKO mice exhibited elevated reactive oxygen species (ROS) levels and reduced mitochondrial membrane potential in cardiomyocytes. Tran

Mitophagy in inflammatory bowel disease: regulatory mechanisms and therapeutic potential.

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC), is a chronic inflammatory disorder of the gastrointestinal tract driven by complex interactions among genetic susceptibility, barrier dysfunction, and immune dysregulation. Mitophagy, the selective autophagic clearance of damaged mitochondria, has emerged as a key regulator of intestinal homeostasis and immune balance. Impaired mitophagy compromises epithelial barrier integrity, amplifies inflammatory signaling, and promotes disease progression. This review summarizes the molecular mechanisms of mi

Procyanidins A1 and B1 Suppress PEDV CV777 by Modulating Mitophagy.

Porcine epidemic diarrhea virus (PEDV) causes severe enteric disease and high mortality in piglets, yet effective therapeutic options remain scarce. To identify novel antivirals, we evaluated the inhibitory potential and molecular mechanisms of four procyanidin subtypes (A1, A2, B1, and B2) in Vero cells. Our results indicate that procyanidins A1 and B1 possess superior anti-PEDV activity compared with their analogs, acting primarily through direct virucidal inactivation (p < 0.001) and by blocking viral adsorption (p < 0.001), internalization (p < 0.001), and replication (p < 0.05). The SI of

FTMT-mediated suppression of mitophagy links iron accumulation to osteoporosis.

Primary osteoporosis is a major age-related disease with a significant global health burden. While iron accumulation is a known risk factor, the mechanisms linking it to bone loss remain unclear. Here, we report that impaired mitophagy in bone marrow mesenchymal stem cells (BMSCs) is a hallmark of osteoporosis and is critically exacerbated by iron accumulation. We found that iron accumulation in BMSCs inhibits mitophagy, leading to mitochondrial dysfunction, increased oxidative stress, and cellular senescence, ultimately impairing osteogenic differentiation. Importantly, targeted activation of

Sirtuin 1 Activation Mitigates Murine Vasculitis Severity by Promoting Mitophagy.

BACKGROUND: SIRT1 (sirtuin 1), a nicotinamide adenine dinucleotide-dependent protein deacetylase, regulates cardiovascular inflammation by modulating cellular stress, inhibiting NLRP3 (NLR family, pyrin domain-containing 3) activation, and promoting the clearance of damaged mitochondria. However, its precise role in the pathogenesis of Kawasaki disease, a pediatric systemic vasculitis that causes coronary artery aneurysms in children, remains unclear. METHODS: Using the Lactobacillus casei cell wall extract (LCWE) murine model of Kawasaki disease, we evaluated the severity of vasculitis in mi

Stigmasterol improves diabetic retinopathy by inhibiting VAMP7-mediated autophagy/mitophagy.

Diabetic retinopathy (DR) is a disease derived from diabetes, which brings great trouble to patients' lives. Stigmasterol has demonstrated beneficial effects in the management of diabetes; however, its influence on DR and the underlying mechanisms remain to be elucidated. Here, we cultured high-glucose (HG)-induced ARPE-19 cells and constructed a DR mice model, which were subsequently treated with stigmasterol. The function and mechanism of stigmasterol on HG-damaged cells were evaluated in vivo and in vitro models. The results showed that 10 μM stigmasterol effectively alleviated the HG-induc

Triclosan inhibits testosterone synthesis in male offspring by activating mitophagy.

Triclosan (TCS), a typical endocrine disruptor, has been shown to interfere with testosterone biosynthesis in male animals. However, the effects and mechanisms of environmental TCS levels on testosterone synthesis in male mice offspring remain unclear. In this study, male mice offspring were exposed to TCS (0, 30, 300, and 3000 μg/kg/day) during prenatal and lactation periods, and Leydig (TM3) cells exposed to TCS (0, 30, 300 and 3000 nM) were used to establish an in vitro model. Results indicated that TCS exposure led to reduced testicular weight, organ coefficient, serum testosterone levels

SFXN1 promotes bladder cancer metastasis by restraining PINK1-dependent mitophagy.

Sideroflexin 1 (SFXN1), a newly identified mitochondrial serine transporter, exhibits great potential to modulate mitochondrial function and promote tumor development. However, its role in bladder cancer (BLCA) remains unclear. Our study revealed that SFXN1 was enriched in clinical BLCA tissues, and high SFXN1 expression in BLCA was positively associated with the progression and poor prognosis. Further, SFXN1 deficiency remarkably suppressed the proliferation and metastasis of BLCA cells in vitro and in vivo, indicating an oncogenic role of SFXN1 in BLCA. Additionally, our results demonstrated

NDUFB9 ameliorates CUMS-induced depression-like behavior by promoting mitophagy.

Major depressive disorder (MDD) is characterized by persistent low mood and anhedonia. Mitochondrial dysfunction is linked to MDD, but the mechanisms are unclear. In this study, transcriptomic analysis of MDD patients' peripheral blood found three key genes: TFAM, SURF1, and NDUFB9. Single-cell transcriptomic analysis of the prefrontal cortex (PFC) in MDD patients identified seven cell types. Analysis showed strong interactions between excitatory and inhibitory neurons in the PFC, with the three genes mainly in inhibitory neurons and NDUFB9 having the highest expression. We then established a

Hyperuricemia exacerbates acetaminophen hepatotoxicity via JNK activation and mitophagy reduction.

Acetaminophen (APAP) induced hepatotoxicity represents a classic form of drug-related liver injury and ranks among the most prevalent causes of acute liver failure globally. Underlying diseases can increase the risk factor of liver injury induced by APAP overdose. Hyperuricemia (HUA), a condition defined by an abnormally high serum uric acid (UA) concentration, is a recognized risk factor for various health conditions, such as gout, cardiovascular disease, metabolic syndrome, and chronic kidney disease. Additionally, HUA is also associated with liver disease, such as non-alcoholic fatty liver
