Molecular imaging reveals unique degenerative changes in experimental glaucoma.

Experimentally induced changes in the central visual pathway were studied by using positron emission tomography in monkeys with unilateral hypertension glaucoma. In 2-[18F]fluoro-2-deoxy-glucose studies, monocular visual stimulation of the affected eye yielded significantly reduced neural responses in the occipital visuocortical areas. The response reduction was limited to the visual cortex ipsilateral to the affected eye, indicating the unique vulnerability of ipsilateral visual cortex in experimental unilateral glaucoma. In addition, in [11C]PK11195 positron emission tomography and immunohis

Ambient molecular imaging by desorption electrospray ionization mass spectrometry.

Desorption electrospray ionization (DESI) allows the direct analysis of ordinary objects or pre-processed samples under ambient conditions. Among other applications, DESI is used to identify and record spatial distributions of lipids and drug molecules in biological tissue sections. This technique does not require sample preparation other than production of microtome tissue slices and does not involve the use of ionization matrices. This greatly simplifies the procedure and prevents the redistribution of analytes during matrix deposition. Images are obtained by continuously moving the sample r

Structural and Molecular Imaging of Aging Brain: A Focus on MR Imaging and PET Modalities.

The process of aging in the brain is reflective of various factors including the environment, lifestyle, genetics, and management of concurrent chronic conditions. Aging in the brain leads to observable structural changes on neuroimaging, such as brain volume reduction, neuronal atrophy, and synaptic loss, which affect higher cognitive functions. Positron emission tomography imaging can help visualize these changes earlier before structural changes even take place and the associated decline in brain function, revealing important insights into how the brain ages and the impact on neural connect

Illuminating the Invisible: Translational Molecular Imaging in Cardiovascular Medicine.

Cardiovascular disease remains the predominant cause of global morbidity and mortality, with recent epidemiological trends indicating a resurgence in adverse outcomes. Conventional imaging modalities-including coronary computed tomography angiography, echocardiography, and cardiac magnetic resonance imaging-are indispensable for anatomic and functional assessment, yet they lack the sensitivity to detect subclinical molecular perturbations that precede overt pathology. The advent of molecular imaging, leveraging nuclear, magnetic resonance, computed tomography, contrast-enhanced ultrasound, and

Multitarget Molecular Imaging-based Characterization of Gastrointestinal Stromal Tumor.

The imaging arsenal for nuclear medicine physicians continues to grow rapidly with multiple radiopharmaceuticals now being used routinely in clinical practice. More specifically, PET imaging targeting various aspects of tumor biology has demonstrated utility in diagnosing incidental/synchronous malignancies as well as biochemical profiling of tumors for prognostication (ie, in the setting of prostate and neuro-endocrine cancers). These concepts are illustrated in this case of an incidental gastric gastrointestinal stromal tumor (GIST) which was imaged with 3 separate PET radiopharmaceuticals.

Molecular Imaging of Human Brain Organoids Using Mass Spectrometry.

Brain organoid models serve as a powerful tool for studying human brain development and function. Mass spectrometry imaging (MSI), a cutting-edge technology, allows us to map the spatial distribution of diverse molecules such as lipids, sugars, amino acids, drugs, and their metabolites within these organoids, all without the need for specific molecular probes. High-quality MSI data hinge on meticulous sample preparation. Fixatives play a pivotal role, but conventional options such as glutaraldehyde, paraformaldehyde, and cryopreserving such as sucrose may inadvertently impact tissue metabolite

Atypical Parkinsonian Syndromes: Structural, Functional, and Molecular Imaging Features.

Atypical parkinsonian syndromes, also known as Parkinson-plus syndromes, are a heterogeneous group of movement disorders, including dementia with Lewy bodies (DLB), progressive supranuclear palsy (PSP), multisystem atrophy (MSA), and corticobasal degeneration (CBD). This review highlights the characteristic structural, functional, and molecular imaging features of these complex disorders. DLB typically demonstrates parieto-occipital hypometabolism with involvement of the cuneus on FDG-PET, whereas dopaminergic imaging, such as [123I]-FP-CIT SPECT (DaTscan) or fluorodopa (FDOPA)-PET, can be uti

Curcumin-based molecular probes for fluorescence imaging of fungi.

Fluorescence imaging is a powerful and widely used method to visualize and study living organisms. However, fungi are notoriously difficult to visualize using fluorescence microscopy, given that their cell wall represents a diffusion barrier, and the synthetic organic dyes available are very limited when compared to molecular probes available for other organisms. Moreover, these dyes are usually available in only one colour, preventing co-staining experiments. To fill this gap, curcumin-based molecular probes were designed based on the rationale that curcumin is fluorescent and has moderate to

Molecular Imaging of Valvular Diseases and Cardiac Device Infection.

The use of positron emission tomography imaging with 18F-fluorodeoxyglucose in the diagnostic workup of patients with suspected prosthetic valve endocarditis and cardiac device infection (implantable electronic device and left ventricular assist device) is gaining momentum in clinical practice. However, in the absence of prospective randomized trials, guideline recommendations about 18F-fluorodeoxyglucose positron emission tomography in this setting are currently largely based on expert opinion. Translational molecular imaging and drug development in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that primarily affects elderly people and constitutes a major source of disability worldwide. Notably, the neuropathological hallmarks of PD include nigrostriatal loss and the formation of intracellular inclusion bodies containing misfolded α-synuclein protein aggregates. Cardinal motor symptoms, which include tremor, rigidity and bradykinesia, can effectively be managed with dopaminergic therapy for years following symptom onset. Nonetheless, patients ultimately develop symptoms that no longer fully respond to dopaminergic t

Multimodality Molecular Imaging in Arthropathy Associated with Multiple Myeloma.

We report a patient with multiple myeloma (MM) and polyarthritis of large joints. During the staging of the disease, bone marrow diffusely involved by MM was clearly demonstrated by 99mTc-2-methoxy-isobutyl-isonitrile (MIBI) single-photon emission computed tomography/computed tomography (SPECT/CT) but not by 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography/CT images. On the other hand, a very intense uptake of 18F-FDG was detected in periarticular tissues of multiple joints, with nonabnormal 99mTc-MIBI accumulation. Rheumatology tests were negative. A subsequent bone scintigraphy

Molecular imaging of myocardial necrosis: an updated mini-review.

Acute myocardial infarction (AMI) remains the most severe and common cardiac emergency among various ischaemic heart diseases. Both unregulated (necrosis) and regulated (apoptosis, autophagy and necroptosis et al.) forms of cell death can occur during AMI. Non-invasive imaging of cardiomyocyte death represents an attractive approach to acquire insights into the pathophysiology of AMI, track the temporal and spatial evolution of MI, guide therapeutic decision-making, evaluate response to therapeutic intervention and predict prognosis. Although several forms of cell death have been identified du

Molecular and structural imaging in surgically induced murine osteoarthritis.

Preclinical imaging in osteoarthritis is a rapidly growing area with three principal objectives: to provide rapid, sensitive tools to monitor the course of experimental OA longitudinally; to describe the temporal relationship between tissue-specific pathologies over the course of disease; and to use molecular probes to measure disease activity in vivo. Research in this area can be broadly divided into those techniques that monitor structural changes in tissues (microCT, microMRI, ultrasound) and those that detect molecular disease activity (positron emission tomography (PET), optical and optoa

Molecular Imaging Probes Based on Matrix Metalloproteinase Inhibitors (MMPIs).

Matrix metalloproteinases (MMPs) are a family of zinc- and calcium-dependent endopeptidases which are secreted or anchored in the cell membrane and are capable of degrading the multiple components of the extracellular matrix (ECM). MMPs are frequently overexpressed or highly activated in numerous human diseases. Owing to the important role of MMPs in human diseases, many MMP inhibitors (MMPIs) have been developed as novel therapeutics, and some of them have entered clinical trials. However, so far, only one MMPI (doxycycline) has been approved by the FDA. Therefore, the evaluation of the activ

Molecular Pathogenesis and Diagnostic Imaging of Metastatic Jaw Tumors.

Metastasis is the spread of malignant cells from a primary tumor to distant sites through lymphatics or blood vessels. Malignant lesions metastasizing to the oral and perioral region are a rarity indeed. Malignant lesions could metastasize to both soft tissue of oral cavity and the hard tissues of the jaws and recent meta-analysis showed that metastasis is more common in the jaws than oral soft tissues because of rich vascular supply. The incidence is very low when compared to the incidence of primary oral cancers; nevertheless, one has to include in the diagnostic workup, metastatic malignant

Molecular Magnetic Resonance Imaging of Tumor Response to Therapy.

Personalized cancer medicine requires measurement of therapeutic efficacy as early as possible, which is optimally achieved by three-dimensional imaging given the heterogeneity of cancer. Magnetic resonance imaging (MRI) can obtain images of both anatomy and cellular responses, if acquired with a molecular imaging contrast agent. The poor sensitivity of MRI has limited the development of activatable molecular MR contrast agents. To overcome this limitation of molecular MRI, a novel implementation of our caspase-3-sensitive nanoaggregation MRI (C-SNAM) contrast agent is reported. C-SNAM is trig

Functional and molecular imaging techniques in aortic aneurysm disease.

PURPOSE OF REVIEW: Functional and molecular aortic imaging has shown great promise for evaluation of aortic disease, and may soon augment conventional assessment of aortic dimensions for the clinical management of patients. RECENT FINDINGS: A range of imaging techniques is available for evaluation of patients with aortic disease. Magnetic resonance blood flow imaging can identify atherosclerosis prone aortic regions and may be useful for predicting aneurysm growth. Computational modeling can demonstrate significant differences in wall stress between abdominal aortic aneurysms of similar size

Recent advances in molecular, multimodal and theranostic ultrasound imaging.

Ultrasound (US) imaging is an exquisite tool for the non-invasive and real-time diagnosis of many different diseases. In this context, US contrast agents can improve lesion delineation, characterization and therapy response evaluation. US contrast agents are usually micrometer-sized gas bubbles, stabilized with soft or hard shells. By conjugating antibodies to the microbubble (MB) surface, and by incorporating diagnostic agents, drugs or nucleic acids into or onto the MB shell, molecular, multimodal and theranostic MBs can be generated. We here summarize recent advances in molecular, multimoda
