Mild phenotype in Molybdenum cofactor deficiency: A new patient and review of the literature.

BACKGROUND: Molybdenum cofactor deficiency (MoCD) is a rare autosomal-recessive disorder that results in the combined deficiency of molybdenum-dependent enzymes. Four different genes are involved in Molybdenum cofactor biosynthesis: MOCS1, MOCS2, MOCS3, and GEPH. The classical form manifests in the neonatal period with severe encephalopathy, including intractable seizures, MRI changes that resemble hypoxic-ischemic injury, microcephaly, and early death. To date, an atypical phenotype with late-onset has been reported in the literature in 13 patients. METHODS: We describe a late-onset and a re

Note: Fabrication and characterization of molybdenum tips for scanning tunneling microscopy and spectroscopy.

We present a method for the preparation of bulk molybdenum tips for Scanning Tunneling Microscopy and Spectroscopy and we assess their potential in performing high resolution imaging and local spectroscopy by measurements on different single crystal surfaces in UHV, namely, Au(111), Si(111)-7 × 7, and titanium oxide 2D ordered nanostructures supported on Au(111). The fabrication method is versatile and can be extended to other metals, e.g., cobalt.

Identification of a bis-molybdopterin intermediate in molybdenum cofactor biosynthesis in Escherichia coli.

The molybdenum cofactor is an important cofactor, and its biosynthesis is essential for many organisms, including humans. Its basic form comprises a single molybdopterin (MPT) unit, which binds a molybdenum ion bearing three oxygen ligands via a dithiolene function, thus forming Mo-MPT. In bacteria, this form is modified to form the bis-MPT guanine dinucleotide cofactor with two MPT units coordinated at one molybdenum atom, which additionally contains GMPs bound to the terminal phosphate group of the MPTs (bis-MGD). The MobA protein catalyzes the nucleotide addition to MPT, but the mechanism o

Evidence for nifU and nifS participation in the biosynthesis of the iron-molybdenum cofactor of nitrogenase.

The nifU and nifS genes encode the components of a cellular machinery dedicated to the assembly of [2Fe-2S] and [4Fe-4S] clusters required for growth under nitrogen-fixing conditions. The NifU and NifS proteins are involved in the production of active forms of the nitrogenase component proteins, NifH and NifDK. Although NifH contains a [4Fe-4S] cluster, the NifDK component carries two complex metalloclusters, the iron-molybdenum cofactor (FeMo-co) and the [8Fe-7S] P-cluster. FeMo-co, located at the active site of NifDK, is composed of 7 iron, 9 sulfur, 1 molybdenum, 1 homocitrate, and 1 uniden

Kinetics and mechanism of the reaction of cyanide with molybdenum nitrogenase from Azotobacter vinelandii.

The steady-state kinetic behavior of the six-electron reduction of N2 by nitrogenase is known to differ markedly from the six-electron reduction of cyanide in two ways. First, on extrapolation to infinite concentration of cyanide, the H2 evolution reaction is almost completely suppressed whereas at extrapolated infinite concentration of N2, H2 evolution continues. Second, as the ratio of the Fe protein to the MoFe protein increases, the reduction of N2 is favored over H2 evolution, whereas the reduction of cyanide becomes less favored relative to H2 evolution. We have extended these steady-sta

Molybdenum at levels of 2, 20, 40, and 60 mg/kg dry matter was added to a sheep ration containing 82 copper and 3, 8 g sulphur per kg dry matter. At the 2 higher levels of molybdenum, a substantial accumulation of copper and molybdenum was observed in the kidney cortices of the South African Mutton Merinos (reported elsewhere). The effect of these treatments on the health of the sheep was investigated. Observations on the 3 molybdenum supplemented treatments were: Serum enzyme levels remained normal throughout the trial. No evidence of kidney damage due to treatment could be obtained from kidn

One Hundred Years of Sulfido Ligand in the Molybdenum Cofactor: New Perspectives.

The xanthine oxidase (XO) family of mononuclear molybdoenzymes consists of well-studied enzymes like XO, xanthine dehydrogenase (XDH), and aldehyde oxidase (AO) in eukaryotes. The characteristic of this family is a terminal sulfido ligand bound to the active site Mo or W ion. Historically, these enzymes were among the first to be investigated in a purified form. Since its discovery in 1902 in cow's milk, the XO enzyme is among the best-studied molybdoenzymes. Much pioneering work has been carried out including comprehensive spectroscopic and structural analyses to study the enzymes' molybdenum

Electrochemical Interactions of Titanium and Cobalt-Chromium-Molybdenum Alloy in Different Solutions.

Pure titanium (Ti) and its alloys are the gold standard for dental implants because a stable titanium dioxide passive film provides excellent corrosion resistance in physiological environments. In this study, we aimed to examine electrochemical interactions between Ti and cobalt-chromium-molybdenum alloy (CoCrMo), and between a novel Ti-magnesium composite (BIACOM TiMg) and CoCrMo, when immersed in everyday solutions representing beverage or oral hygiene exposure. Test solutions included Coca-Cola®, lemon juice, Elmex® fluoride gel, Listerine® Cool Mint, and Sensodyne® fluoride paste. Immersio

Investigation on the antitumor and antibacterial performance of photothermally active molybdenum oxide materials.

Post-surgical breast cancer management remains limited by systemic toxicity, residual tumor burden, and infection risk. To address these issues, we developed an intelligent nanoplatform (MoOx@DOX@LNT) for the co-delivery of oxygen-deficient molybdenum oxide (MoOx) nanosheets and doxorubicin (DOX). This system enables simultaneous tumor eradication and prevention of bacterial colonization through NIR-II-triggered chemo-photothermal synergy. The hydrothermally synthesized MoOx nanosheets display strong NIR-II absorption and a high photothermal conversion efficiency of 44.88%. An electrostatic lo

Insights into the biocompatibility of biodegradable metallic molybdenum for cardiovascular applications-a critical review.

Atherosclerotic cardiovascular disease (ACD) is the leading cause of death worldwide. The gold standard of treatment is the implantation of a permanent stent implant that is often associated with complications such as thrombus formation, vascular neointimal response, and stent fracture, which altogether decrease the long-term safety and efficacy of the stent. Biodegradable metallic materials have become an attractive alternative because of the ability to facilitate a more physiological healing response while the metal degrades. Recently, Molybdenum (Mo) has been considered as a potential candi

Unlocking Phase Diagrams for Molybdenum and Tungsten Nanoclusters and Prediction of their Formation Constants.

Understanding and controlling aqueous speciation of metal oxides are key for the discovery and development of novel materials, and challenge both experimental and computational approaches. Here we present a computational method, called POMSimulator, which is able to predict speciation phase diagrams (Conc. vs pH) for multispecies chemical equilibria in solution, and which we apply to molybdenum and tungsten isopolyoxoanions (IPAs). Starting from the MO4 monomers, and considering dimers, trimers, and larger species, the chemical reaction networks involved in the formation of [H32Mo36O128]8- and

Rare cause of xanthinuria: a pediatric case of molybdenum cofactor deficiency B.

Molybdenum cofactor is essential for the activity of multiple enzymes including xanthine dehydrogenase. Molybdenum cofactor deficiencies are rare inborn errors of metabolism. Clinically, they present with intractable seizures, axial hypotonia, and hyperekplexia. They further develop cerebral atrophy, microcephaly, global developmental delay and ectopia lentis. We report a 5-year-old female with clinically, biochemically and genetically confirmed molybdenum cofactor deficiency type B due to compound heterozygous pathogenic variants in the molybdenum cofactor synthesis 2 gene found on whole exom

Investigation of molybdenum cofactor deficiency due to MOCS2 deficiency in a newborn baby.

BACKGROUND: Molybdenum cofactor deficiency (MOCD) is a severe autosomal recessive neonatal metabolic disease that causes seizures and death or severe brain damage. Symptoms, signs and cerebral images can resemble those attributed to intrapartum hypoxia. In humans, molybdenum cofactor (MOCO) has been found to participate in four metabolic reactions: aldehyde dehydrogenase (or oxidase), xanthine oxidoreductase (or oxidase) and sulfite oxidase, and some of the components of molybdenum cofactor synthesis participate in amidoxime reductase. A newborn girl developed refractory seizures, opisthotonus
