BACKGROUND & OBJECTIVE: Potato tuber moth (PTM), Phthorimaea operculella Zeller is a widely distributed, devastating pest of potatoes attacking the foliage and infest the tubers in both field and store causing serious economic damage. As application of PTM granulovirus (PTM-GV) has shown significant reduction in damage, attempts were made to develop a new cell line from this insect to grow PTM-GV for use as a biopesticide.
METHODS: Approximately 100 mg of insect eggs were collected, surface sterilized and crushed gently in a boiling tube aseptically. The tissues were washed with physiological

The catecholamine dopamine (DA) plays a fundamental role in the regulation of behavior and neurodevelopment across animal species. Uncovering the embryonic origins of neurons that express DA opens a path for a deeper understanding of how DA expression is regulated and, in turn, how DA regulates the activities of the nervous system. In a well-established insect model, Manduca sexta, we identified the putative homologue of the embryonic grasshopper "H-cell" using intracellular techniques, laser scanning confocal microscopy, and immunohistochemistry. In both species, this neuron possesses four ax

Two distinct opsin-encoding cDNAs, designated MANOP1 and MANOP2, were isolated as 3' fragments from the sphingid moth Manduca sexta. They were obtained by reverse transcription of retinal RNA and amplification with the polymerase chain reaction (PCR) using a degenerate primer designed to an amino-acid sequence conserved in arthropod opsins. The cDNA fragments labelled bands at approximately 1.8 kb on Northern blots of retinal RNA extracts. Levels of opsin message were compared in retinas from normal moths, whose diets were fortified with carotenoid precursors of the Manduca rhodopsin chromopho

Of the trivalent (CrSO4) and hexavalent (K2Cr2O7) chromium compounds, only the hexavalent produced significant deleterious effects on development and fertility of E. cautella, when eggs were reared on laboratory medium supplemented with different concentrations of these salts.

Pericopina is divided into 36 genera, with the greatest diversity recorded in the Neotropics. Studies on the taxonomy and systematics of pericopines are scarce, and one of the few phylogenetic hypotheses available in the literature places some Afro-Indo-Malayan lineages among the Neotropical representatives. Through a survey of adult morphological characters, we analyzed the phylogenetic relationships in Pericopina including Nyctemera and other Afro-Indo-Malayan genera as putative members of this subtribe. Additionally, we tested the monophyly of Dysschema, a key genus to understanding the evo

Tyramine signaling orchestrates essential physiological processes in insects, yet the functional diversification of tyramine receptors (TARs) and the evolutionary significance of receptor gene duplication remain poorly understood. While most lepidopterans harbor a single TAR1, the diamondback moth Plutella xylostella uniquely possesses two TAR1 paralogs (PxTAR1 and PxTAR1A) alongside PxTAR2, providing an exceptional model to investigate sub-functionalization. However, functional characterization of PxTARs has been limited to TAR1. Here, we uncover striking functional differentiation of TARs th

The Australian giant wood moth, Endoxyla cinereus (Cossidae), is a major forestry pest whose later instars bore into the trunks and branches of Eucalyptus, but the early larval stages have remained unknown for over a century. Using DNA analysis, we identified larvae collected from galls formed by scale insects (Apiomorpha spp.), as En. cinereus. This relationship was tested by rearing over 100 000 neonate larvae on alternative potential food resources including active galls of scale insects (Apiomorpha munita and A. crispa). Only larvae assigned to galls fed and developed into later instars. T

AbstractThe energy needed for insect flight varies with the size and body proportions of different species or individuals. Wingbeat frequency during flight usually decreases as size increases, though these changes depend on the proportions of the wings and flight muscles. Metabolic rate during flight often changes with wingbeat frequency. Nutrition during development plays a key role in determining an individual's size and body proportions. We used an intermittent feeding regime during the fifth larval instar of the hawk moth (Manduca sexta) to investigate how nutrition-induced size variation

The diamondback moth, Plutella xylostella (L.), is a major pest of cruciferous crops with a remarkable capacity to develop insecticide resistance. The present study evaluated the susceptibility, resistance ratios, and detoxification mechanisms of field populations collected from Andhra Pradesh, India, against tolfenpyrad 15 EC and emamectin benzoate 5 SG. Laboratory selection studies revealed declining LC50 values over successive generations reared without insecticide pressure, indicating partial recovery of susceptibility. Field bioassays showed resistance ratios ranging from 9.8 to 13.1 for

Abamectin has been used for decades as an insecticide and acaricide in arthropod pest management. However, there is no direct evidence from CRISPR/Cas9 studies confirming the involvement of GSTs in insect resistance to abamectin. The oriental fruit moth, Grapholita molesta, is a destructive pest of fruit trees worldwide. The role of GSTs in the oriental fruit moth remains unclear. In this study, an abamectin-resistant strain (AB-R) was derived from a susceptible laboratory strain (AB-S) of G. molesta. Synergist bioassays showed that the GST inhibitor diethyl maleate (DEM) significantly increas

This study introduces a novel hybrid model for accurate CO2 emissions prediction, supporting sustainable decision-making. The model integrates the Gaussian mutation and shrink mechanism-based moth flame optimization (GMSMFO) algorithm with an extreme learning machine (ELM). GMSMFO enhances population diversity and avoids local optima through Gaussian mutation (GM), while the shrink mechanism (SM) improves exploration-exploitation balance. Validated on the congress on evolutionary computation (CEC2020) benchmark suite (dimensions 30 and 50), GMSMFO demonstrated superior performance compared to

A 78-year-old patient underwent his third biological aortic valve replacement (25 mm, Inspiris Resilia™, Edwards Lifesciences LLC, Irvine, CA, USA) with COR-KNOT® (LSI Solutions, Victor, NY, USA) due to valve degeneration in 2018 at a foreign hospital. In 2021, the patient was diagnosed with severe aortic regurgitation and admitted to our hospital for his fourth surgery. Intraoperatively, a total of 7 perforations in all 3 valve leaflets have been observed, which were obviously induced by the rigid metallic fasteners. Redo isolated aortic valve replacement (29 mm, Perimount Magna Ease, Edwards

Pheromones are a kind of signal produced by an animal that evoke innate responses in conspecifics. In moth, pheromone components can be detected by specialized olfactory receptor neurons (OSNs) housed in long sensilla trichoids on the male antennae. The pheromone receptors (PRs) located in the dendrite membrane of OSNs are responsible for pheromone sensing in most Lepidopteran insects. The potato tuber moth Phthorimaea operculella is a destructive pest of Solanaceae crops. Although sex attractant is widely used in fields to monitor the population of P. operculella, no study has been reported o

BACKGROUND: Genetic manipulation of sex determination pathways in insects provides the basis for a broad range of strategies to benefit agricultural security and human health. The P-element somatic inhibitor (PSI) protein, an exon splicing silencer that promotes male-specific splicing of dsx, plays a critical role in male sexual differentiation and development. The functions of PSI have been characterized in the lepidopteran model species Bombyx mori. However, the molecular mechanism and functions of PSI in Plutella xylostella, a worldwide agricultural pest and taxonomically basal species, are

The idea of enhancing insecticide efficacy against phytophagous insects with feeding stimulators was proposed as early as the 1960s, and a number of insect feeding stimulators based on sugars, molasses, and cottonseed extracts, biologically active at relatively high (5% and higher) concentrations, have been advocated. Here, we show that an acidic amino acid, L-aspartate, stimulates feeding in codling moth neonates at much lower concentrations and acts as an effective tank-mixed additive for increasing efficacy of insecticides, reducing fruit damage, and increasing yield of the fruit. In labora

Macrocyclic lactones such as abamectin and ivermectin constitute an important class of broad-spectrum insecticides. Widespread resistance to synthetic insecticides, including abamectin and ivermectin, poses a serious threat to the management of diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), a major pest of cruciferous plants worldwide. P-glycoprotein (Pgp), a member of the ABC transporter superfamily, plays a crucial role in the removal of amphiphilic xenobiotics, suggesting a mechanism for drug resistance in target organisms. In this study, PxPgp1, a putative Pgp gene

Codling moth, Cydia pomonella (L.), is a cosmopolitan pest of apple, potentially causing severe damage to the fruit. Currently used methods of combating this insect do not warrant full success or are harmful to the environment. The use of plant-derived semiochemicals for manipulation with fruit-infesting behavior is one of the new avenues for controlling this pest. Here, we explore the potential of Ginkgo biloba and its synthetic metabolites for preventing apple feeding and infestation by neonate larvae of C. pomonella. Experiments with crude extracts indicated that deterrent constituents of g

Transcript patterns elicited in response to hosts can reveal how fungi recognize suitable hosts and the mechanisms involved in pathogenicity. These patterns could be fashioned by recognition of host-specific topographical features or by chemical components displayed or released by the host. We investigated this in three isolates of Isaria fumosoroseus which has shown considerable pathogenic ability against Plutella xylostella. Conidia inoculated to hind wings had the highest germination ability while the conidia inoculated to head regions showed the lowest germination. Conidia applied to diamo

Gypsy moth (Lymantria dispar L.) spread is dominated by stratified dispersal, and, although spread rates are variable in space and time, the gypsy moth has invaded Wisconsin at a consistently higher rate than in other regions. Allee effects, which act on low-density populations ahead of the moving population that contribute to gypsy moth spread, have also been observed to be consistently weaker in Wisconsin. Because a major cause of an Allee effect in the gypsy moth is mate-finding failure at low densities, supplementing low-density populations with immigrants that arrive through dispersal may
