Whole Exome Sequencing of 23 Multigeneration Idiopathic Scoliosis Families Reveals Enrichments in Cytoskeletal Variants, Suggests Highly Polygenic Disease.

Adolescent idiopathic scoliosis (AIS) is a lateral spinal curvature >10° with rotation that affects 2-3% of healthy children across populations. AIS is known to have a significant genetic component, and despite a handful of risk loci identified in unrelated individuals by GWAS and next-generation sequencing methods, the underlying etiology of the condition remains largely unknown. In this study, we performed exome sequencing of affected individuals within 23 multigenerational families, with the hypothesis that the occurrence of rare, low frequency, disease-causing variants will co-occur in dis

Polygenic hazard score, amyloid deposition and Alzheimer's neurodegeneration.

Mounting evidence indicates that the polygenic basis of late-onset Alzheimer's disease can be harnessed to identify individuals at greatest risk for cognitive decline. We have previously developed and validated a polygenic hazard score comprising of 31 single nucleotide polymorphisms for predicting Alzheimer's disease dementia age of onset. In this study, we examined whether polygenic hazard scores are associated with: (i) regional tracer uptake using amyloid PET; (ii) regional volume loss using longitudinal MRI; (iii) post-mortem regional amyloid-β protein and tau associated neurofibrillary t

Opioid Addiction, Genetic Susceptibility, and Medical Treatments: A Review.

Opioid addiction is a chronic and complex disease characterized by relapse and remission. In the past decade, the opioid epidemic or opioid crisis in the United States has raised public awareness. Methadone, buprenorphine, and naloxone have proven their effectiveness in treating addicted individuals, and each of them has different effects on different opioid receptors. Classic and molecular genetic research has provided valuable information and revealed the possible mechanism of individual differences in vulnerability for opioid addiction. The polygenic risk score based on the results of a gen

Polygenic risk assessment reveals pleiotropy between sarcoidosis and inflammatory disorders in the context of genetic ancestry.

Sarcoidosis is a complex disease of unknown etiology characterized by the presence of granulomatous inflammation. Though various immune system pathways have been implicated in disease, the relationship between the genetic determinants of sarcoidosis and other inflammatory disorders has not been characterized. Herein, we examined the degree of genetic pleiotropy common to sarcoidosis and other inflammatory disorders to identify shared pathways and disease systems pertinent to sarcoidosis onset. To achieve this, we quantify the association of common variant polygenic risk scores from nine comple

Genetics of Depression: Progress at Last.

PURPOSE OF REVIEW: We will describe the success of recent genome-wide association studies that identify genetic variants associated with depression and outline the strategies used to reduce heterogeneity and increase sample size. RECENT FINDINGS: The CONVERGE consortium identified two genetic associations by focusing on a sample of Chinese women with recurrent severe depression. Three other loci have been found in Europeans by combining cohorts with clinical diagnosis and measures of depressive symptoms to increase sample size. 23andMe identified 15 loci associated with depression using self-

[Study of the genetic causes of polygeneticallv determined endocrinopathies--patience can bring success].

Complex endocrinopathies, such as diabetes mellitus, obesity, polycystic ovary syndrome and osteoporosis belong to the most common diseases but their pathogenesis is still not fully explained. Environmental fadors along with genetic factors contribute to their occurrence and development. The study of genetic background is based on different strategies, mostly on linkage analysis and candidate gene approach. The common forms of these endocrinopathies do not seem to be the result of a defect of one or several major genes but the search for complex gene-gene, gene-environment interactions is need

Tetralogy of Fallot in three siblings: a familial study and review of the literature.

We report a family in which three out of four siblings had tetralogy of Fallot (TOF). The family history showed TOF in the daughter of a maternal cousin, while no other congenital heart diseases were discovered. Although no teratogenic environmental agent was discovered, the absence of parental consanguinity and the presence of another affected relative suggest multifactorial inheritance. Autosomal recessive inheritance cannot be ruled out.

Familial hemifacial microsomia.

Hemifacial microsomia, a relatively distinct clinical form in the Facio-Auriculo-Vertebral Spectrum is generally thought to be a sporadic event, although infrequent familial occurrences have been reported. In this communication, we describe a family with three males and two females in two successive generations affected with hemifacial microsomia. The pedigree is most compatible with autosomal dominant inheritance, although X-linked dominant or multifactorial inheritance cannot be excluded.

Familial intestinal malrotation with midgut volvulus and facial anomalies: a disorder involving a gene controlling the normal gut rotation?

We report on 2 sisters with congenital midgut volvulus. Both had similar facial changes different from those of their parents and the other 2 unaffected sibs. Both parents had normal barium meal roentgenograms. The occurrence of these abnormalities in sibs born to parents with apparently normal gastrointestinal tract anatomy suggest autosomal recessive inheritance, although a coincidence, multifactorial or autosomal dominant inheritance with nonpenetrance in either of the parents cannot be excluded.

Mendelian inheritance of isolated nonsyndromic cleft palate.

Isolated, apparently nonsyndromic cleft palate (CP) is thought to be etiologically heterogeneous. The multifactorial threshold explanation for CP is postulated in most cases. Mendelian inheritance has been documented in 3 families. We report on three unrelated white families with 17 relatives in several generations affected with CP, including submucous CP and bifid/absent uvula. We could find no evidence of a cleft syndrome. In one family, the pattern is consistent with autosomal dominant inheritance. In 2 families, the pattern is consistent with X-linked recessive inheritance. The specific fi

Enhanced disease resistance in chickpea and canola via MAGPIE: Major-Effect Allele and Genomic Polygenic Integration for Enhancement.

The global agricultural industry suffers major economic losses due to fungal diseases. Though selective breeding and introgression of resistance genes with major effects have helped to reduce the impact of these diseases, resistance breakdown occurs within a few generations. Quantitative resistance that relies on multiple small effect genes has proven to be more durable than relying on major effect genes. In this study, we present MAGPIE (Major-Effect Allele and Genomic Polygenic Integration for Enhancement), a computational framework that integrates genomic data, predictive breeding approache

PEStimate: predicting offspring disease risk after polygenic embryo screening.

MOTIVATION: Polygenic embryo screening (PES) is a new, controversial technology, whereby human in vitro fertilization embryos are screened for their genetic risk of complex, polygenic diseases. PES aims to reduce the disease burden in offspring by prioritizing the selection of low-risk embryos. However, given that polygenic diseases are usually late-onset, PES outcomes must be estimated by epidemiological modeling. The liability threshold model has been previously used to predict outcomes. However, predictions rely on complex sets of equations, some of which require numerical integration or si

Polygenic risk and germline genetics for prostate cancer in Asians: Where do we stand?

Prostate cancer exhibits marked ethnic differences in genetic architecture. Although polygenic risk scores (PRS) and germline genetic testing have shown clinical utility in European populations, their applicability to Asian populations remains limited. This review synthesizes evidence published between 2020 and 2025 on PRS and germline genetics in Asian prostate cancer cohorts, focusing on Chinese, Japanese, and Korean populations. Recent studies demonstrate that population-specific PRS models effectively stratify prostate cancer risk in Asian men, with individuals in the highest decile showin

From polygenic risk to functional genomics: a framework for precision gynecological disease modeling.

Gynecological conditions including endometriosis, polyendocrine metabolic ovarian syndrome and malignancies cause widespread chronic morbidity and infertility, yet translating genetic discoveries into biological mechanisms remains challenging. We propose a framework integrating biobank-derived human genetic data, patient-derived stem cell models, and advanced tissue engineering. By combining naturally occurring genetic variation with targeted genome engineering and high-resolution molecular analyses, this approach promises to enable mechanistic insights and therapeutic discovery. We discuss me

A genomic structural equation modelling study elucidates shared genetic architecture of polygenic traits associated with post-intensive care syndrome.

Post-intensive care syndrome (PICS) is defined by persistent psychological, cognitive and physical impairments after critical illness, yet its shared genetic basis remains unknown. We applied genomic structural equation modeling (Genomic SEM) to large-scale GWAS summary statistics for major depressive disorder (N = 217,584), post-traumatic stress disorder (N = 199,213), cognitive function (N = 257,841), memory performance (N = 152,605) and hand grip strength (N = 461,089) to construct a latent genetic factor related to PICS component phenotypes. We then performed multivariate GWAS, Bayesian fi

Clinical translation of polygenic scores for prostate cancer screening.

Interest in using polygenic scores (PGS) to improve the risk stratification for and early detection of prostate cancer is considerable. Despite the absence of clinical guidelines for the use of prostate cancer PGS in patient care, existing and emerging standards for the clinical translation and reporting of genetic testing generally and PGS specifically provide a relevant framework to help guide these efforts. This framework is intended to harmonize advances in the development of PGS clinical assays and standardization of PGS reporting in the context of prostate cancer PGS specifically. The an

Polygenic scores for dizygotic twinning: insights into the genetic architecture of female fertility.

PURPOSE: Natural dizygotic twinning (DZT) results from hyper-ovulation and is an indicator of female fertility. However, some traits linked to DZ twinning are also associated with infertility. We examined the relationship between DZT and female (in)fertility using recent GWAS findings. METHODS: We investigated the genetic architecture of DZT and compared polygenic scores (PGS) for DZT between mothers of naturally conceived DZ twin pregnancies and mothers who required fertility treatments (MAR) in the Netherlands Twin Register (NTR) and the Norwegian Mother, Father, and Child Cohort Study (MoB

Polygenic insight identifies precision biomarkers decoding protein catabolism and autophagy pathways in obstructive sleep apnea.

Obstructive sleep apnea (OSA) is a common sleep disorder characterized by recurrent upper airway obstructions, leading to substantial health burdens and socioeconomic costs. This study aimed to identify Hypoxia and Mitophagy-Related Differentially Expressed Genes (HMRDEGs) and evaluate their potential as biomarkers and therapeutic targets for OSA. Transcriptomic data from GSE135917 and GSE38792 in the GEO database were analyzed using the limma package to identify differentially expressed genes (DEGs), which were subsequently intersected with hypoxia- and mitophagy-related gene sets(HMRGs) cura

Exploring Monogenic, Polygenic, and Epigenetic Models of Common Variable Immunodeficiency.

Common variable immunodeficiency (CVID) is the most frequent symptomatic inborn error of immunity (IEI). CVID is genetically heterogeneous and occurs in sporadic or familial forms with different inheritance patterns. Monogenic mutations have been found in a low percentage of patients, and multifactorial or polygenic inheritance may be involved in unsolved patients. In the complex disease model, the epistatic effect of multiple variants in several genes and environmental factors such as infections may contribute. Epigenetic modifications, such as DNA methylation changes, are also proposed to be

Examining the interaction between prenatal stress and polygenic risk for attention-deficit/hyperactivity disorder on brain growth in childhood: Findings from the DREAM BIG consortium.

This study explored the interactions among prenatal stress, child sex, and polygenic risk scores (PGS) for attention-deficit/hyperactivity disorder (ADHD) on structural developmental changes of brain regions implicated in ADHD. We used data from two population-based birth cohorts: Growing Up in Singapore Towards healthy Outcomes (GUSTO) from Singapore (n = 113) and Generation R from Rotterdam, the Netherlands (n = 433). Prenatal stress was assessed using questionnaires. We obtained latent constructs of prenatal adversity and prenatal mood problems using confirmatory factor analyses. The partic
