GABA and the regulation of serotonin N-acetyltransferase activity in amphibian retina-I. Effects of GABA agonists and antagonists.

Retinal melatonin biosynthesis is regulated in part by changes in the activity of serotonin N-acetyltransferase (NAT), which increases at night in dark-adapted retinas, but not in light-exposed retinas. Using an in vitro preparation of Xenopus laevis (African clawed frog) eye cups, we have obtained evidence supporting the involvement of gamma-aminobutyric acid (GABA) in the regulation of NAT activity. GABA, the GABA-A receptor agonists muscimol and isoguvacine, and the GABA-B receptor agonist (?)baclofen, in the presence of 3-isobutyl-1-methylxanthine, mimicked dark adaptation by increasing th

Sensitivity to ethanol hypnosis and modulation of chloride channels does not cosegregate with pentobarbital sensitivity in HS mice.

Several findings suggest that barbiturates and alcohol produce their sedative effects through a common neural and possibly a common genetic mechanism. We tested this hypothesis by examining the correlation between ethanol and pentobarbital sedative effects in individual animals from a genetically heterogeneous population. The duration of pentobarbital-induced hypnosis (sleep-time) was unrelated to the sleep-time produced by ethanol in heterogeneous stock (HS) mice. Therefore, the present study also examined the effect of ethanol, pentobarbital, and flunitrazepam on muscimol-stimulated chloride

Role of the cat substantia nigra pars reticulata in eye and head movements. II. Effects of local pharmacological injections.

Guided and reflex eye movements were studied in cats trained to make orienting saccades toward visual and auditory targets. Injections of a GABA-agonist (Muscimol) or GABA-antagonists (Bicuculline and Picrotoxin) were made in the Substantia Nigra pars reticulata (SNpr). Bicuculline and Picrotoxin, whether unilaterally or bilaterally injected had no effect on the posture nor the oculomotor performance of the animals. Neck muscle activity remained symmetrical. Unilateral injections of Muscimol produced oro-facial akinesia, reduction of the number of eye movements, contralateral head turning, vis

The effects of reversible inactivation of the subthalamo-pallidal pathway on the behaviour of naive and hemiparkinsonian monkeys.

This study was designed to further investigate the role of the subthalamic nucleus (STN) and globus pallidus internus (GPi) in the pathophysiology of Parkinson's disease. The prevailing theory about the pathophysiology of Parkinson's disease (PD) predicts that there is overactivity of the subthalamo-pallidal pathway. In order to inactivate that pathway, naive and hemiparkinsonian monkeys were locally administered either muscimol (to reversibly inactivate the contralateral STN) or kynurenic acid (to reduce glutamatergic activity in the contralateral GPi). Three naive and 2 hemiparkinsonian monk

Effects of GABAA and GABAB receptor agonists on reticular-elicited hippocampal rhythmical slow activity.

Hippocampal rhythmical slow activity (RSA) can be elicited by stimulation of the midbrain reticular formation. All classes of anxiolytic drug so far tested reduce the frequency of this RSA. Anxiolytic barbiturates and benzodiazepines, as opposed to compounds such as buspirone, are thought to act as receptor agonists of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). In the present study muscimol (a GABAA receptor agonist) and baclofen (a GABAB receptor agonist) were injected into freely moving rats. Baclofen produced a dose-related decrease in frequency of RSA in the range 1 to

GABAergic drugs, morphine and morphine tolerance: a study in relation to nociception and gastrointestinal transit in mice.

Agonists and antagonists of gamma-aminobutyric acid, i.e. GABAergic drugs, such as muscimol, baclofen or bicuculline, alone or in combination, exhibited analgesic effects per se and enhanced the analgesia induced by morphine. The analgesic effects of GABAergic drugs were unaffected by administration of naloxone in a dose which antagonized the analgesia induced by morphine. The ED50 for the antinociceptive effect of muscimol, bicuculline, picrotoxin, gabaculline or aminooxyacetic acid (AOAA) was not affected in the morphine-tolerant group as compared to the control group, in contrast to the inc

Stress induced alterations in striatal GABAA receptor complex.

The effect of cold-immobilized stress on the gamma-aminobutylic acid (GABA)/benzodiazepine (BZP) receptor complex in the rat striatum was examined. The stressful manipulation induced a significant decrease in the amount of [3H]muscimol binding sites in the striatal particulate fraction. On the other hand, [3H]flunitrazepam (FLN) binding and the enhancing effect of FLN or secobarbital on the [3H]muscimol binding to the striatal particulate fraction were not influenced by the stress treatment. These results suggest that cold-immobilized stress may selectively change GABAA receptor binding withou

The central amygdala nucleus is critical for incubation of methamphetamine craving.

Cue-induced methamphetamine seeking progressively increases after withdrawal but mechanisms underlying this 'incubation of methamphetamine craving' are unknown. Here we studied the role of central amygdala (CeA), ventral medial prefrontal cortex (vmPFC), and orbitofrontal cortex (OFC), brain regions implicated in incubation of cocaine and heroin craving, in incubation of methamphetamine craving. We also assessed the role of basolateral amygdala (BLA) and dorsal medial prefrontal cortex (dmPFC). We trained rats to self-administer methamphetamine (10 days; 9 h/day, 0.1 mg/kg/infusion) and tested

Blood pressure is maintained during dehydration by hypothalamic paraventricular nucleus-driven tonic sympathetic nerve activity.

Resting sympathetic nerve activity (SNA) consists primarily of respiratory and cardiac rhythmic bursts of action potentials. During homeostatic challenges such as dehydration, the hypothalamic paraventricular nucleus (PVN) is activated and drives SNA in support of arterial pressure (AP). Given that PVN neurones project to brainstem cardio-respiratory regions that generate bursting patterns of SNA, we sought to determine the contribution of PVN to support of rhythmic bursting of SNA during dehydration and to elucidate which bursts dominantly contribute to maintenance of AP. Euhydrated (EH) and

GABA(A) receptor-mediated presynaptic inhibition on glutamatergic transmission.

We investigated the functional roles of presynaptic GABA(A) receptors on excitatory nerve terminals in contributing to spontaneous and action potential-evoked glutamatergic transmission to rat hippocampal CA3 pyramidal neurons. Single CA3 neurons were mechanically isolated with adherent nerve terminals, namely the 'synaptic bouton preparation', and spontaneous glutamatergic excitatory synaptic potentials (sEPSCs) and EPSCs evoked by focal electrical stimuli of a single presynaptic glutamatergic boutons (eEPSCs) were recorded using conventional whole-cell patch recordings. The involvement of GABAergic system in the antidepressant-like effect of agmatine.

Considering the involvement of GABAergic system in the action of the fast-acting antidepressant ketamine, and that agmatine may exert an antidepressant-like effect through mechanisms similar to ketamine, the purpose of the present study was to evaluate the involvement of GABAA and GABAB receptors in the antidepressant-like effect of agmatine. Inhibitory effect of the potential endogenous benzodiazepine receptor ligand, octadecaneuropeptide (ODN), on gonadotropin-releasing hormone gene expression in the male rat brain.

It has been reported that activation of the GABAA receptor complex by different agents such as barbiturates and neurosteroids can negatively regulate the hypothalamopituitary-gonadal axis. Recently, an 86-amino acid polypeptide with high affinity for diazepam binding sites, termed diazepam-binding inhibitor (DBI), has been found in the rat brain. DBI as well as a peptide derived from DBI, the octadecaneuropeptide DBI[33-50] (ODN), can interact with the GABAA receptor complex. In order to investigate the role of these endogenous ligands for GABAA receptors on GnRH gene expression, we studied th

Involvement of benzodiazepine/GABA-A receptor complex in ethanol-induced state-dependent learning in rats.

State-dependent learning (SDL) induced by ethanol (EtOH) was investigated on the step-through passive avoidance task in rats. Pretraining injection of EtOH dose-dependently reduced step-through latency in the test session 24 h after the training. Injection of EtOH (1.0 g/kg) before both the training and test sessions, however, failed to reduce the latency. These results show that EtOH produces SDL. The failure of learning performance in SDL (dissociation in SDL) induced by EtOH was blocked by bicuculline, Ro15-4513 and picrotoxin injected before the training session. The success of learning pe

The effect of cyclopyrrolones on GABAA receptor function is different from that of benzodiazepines.

The effects of the cyclopyrrolones zopiclone and suriclone on the function of the central gamma-amino-butyric acid type A (GABAA) receptor complex in mouse brain were evaluated both in vitro and in vivo. Added in vitro to mouse cerebral cortical membranes, these compounds potently inhibited [3H]flumazenil binding with IC50 (50% inhibitory concentration) values of 35.8 nM (zopiclone) and 1.1 nM (suriclone). Similar results were obtained with cerebellar membranes, indicating that these drugs do not discriminate between putative type I and type II benzodiazepine receptors. The interaction of cycl

The influence of gamma-aminobutyric acid on hormone release by the mouse and rat endocrine pancreas.

The present study was aimed at localizing gamma-aminobutyric acid (GABA) and its enzyme of synthesis, glutamic acid decarboxylase (GAD), in the mouse pancreas by immunocytochemical methods. The influence of GABA on hormone release was also studied with normal mouse and rat islets and the isolated perfused rat pancreas. Particular attention was paid to glucagon release to test a recent hypothesis suggesting that GABA mediates the still unexplained glucose-induced inhibition of glucagon release. GABA and GAD were identified only in islet cells and never in the exocrine tissue. Exogenous GABA, ba

Spinal 5-HT2A receptors regulate cutaneous sympathetic vasomotor outflow in rabbits and rats; relevance for cutaneous vasoconstriction elicited by MDMA (3,4-methylenedioxymethamphetamine, "Ecstasy") and its reversal by clozapine.

We determined whether spinal 5-hydroxytryptamine 2A (5-HT2A) receptors contribute to resting cutaneous sympathetic vasomotor activity, and to increases in activity elicited by electrical stimulation of the medullary raphe/parapyramidal region, and whether these receptors are involved in the cutaneous vasoconstricting action of systemically administered MDMA (3,4-methylenedioxymethamphetamine, "Ecstasy") and its reversal by clozapine. Experiments were conducted in urethane-anesthetized rabbits and rats. Administration of the 5-HT2A antagonist, trans-4-((3Z)3-[(2-Dimethylaminoethyl)oxyimino]-3-(

Effects of GABAergic drugs on ethanol-induced gastric lesions.

Different classes of GABAergic drugs--baclofen, GABA, muscimol, Na-valproate, Mg-valproate and diazepam--were tested per os on ethanol-induced gastric lesions in rats. The GABAB agonist baclofen failed to affect gastric susceptibility to ethanol damage, while all the other compounds exerted a dose-dependent inhibition on haemorrhagic-necrotic lesions. This effect was not significantly reversed by the specific GABAA antagonist bicuculline, suggesting it to be independent from GABAA receptors. The blockade of prostaglandin synthesis by indomethacin significantly decreased the gastroprotective ac

Fast Compensatory Functional Network Changes Caused by Reversible Inactivation of Monkey Parietal Cortex.

The brain has a remarkable capacity to recover after lesions. However, little is known about compensatory neural adaptations at the systems level. We addressed this question by investigating behavioral and (correlated) functional changes throughout the cortex that are induced by focal, reversible inactivations. Specifically, monkeys performed a demanding covert spatial attention task while the lateral intraparietal area (LIP) was inactivated with muscimol and whole-brain fMRI activity was recorded. The inactivation caused LIP-specific decreases in task-related fMRI activity. In addition, these

Central and peripheral action of GABAA and GABAB agonists on small intestine motility in rats.

gamma-Aminobutyric acid (GABA) is known as a neurotransmitter in the central nervous system and in the enteric nervous system. The effects and the sites of action of GABA and of its GABAA (muscimol) and GABAB (baclofen) agonists were determined on intestinal motility of unanesthetized rats chronically fitted with intraparietal electrodes in the duodeno-jejunum and fasted for 8 h. GABA (6 mg/kg i.p.) induced a biphasic response i.e. a primary inhibition followed by a period of irregular spiking activity. Muscimol (4 mg/kg i.p.) inhibited the cyclic motor profile while baclofen (4 mg/kg i.p.) ha
