Chikungunya virus replication in skeletal muscle cells is required for disease development.

Chikungunya virus (CHIKV) is an arbovirus capable of causing a severe and often debilitating rheumatic syndrome in humans. CHIKV replicates in a wide variety of cell types in mammals, which has made attributing pathologic outcomes to replication at specific sites difficult. To assess the contribution of CHIKV replication in skeletal muscle cells to pathogenesis, we engineered a CHIKV strain exhibiting restricted replication in these cells via incorporation of target sequences for skeletal muscle cell-specific miR-206. This virus, which we term SKE, displayed diminished replication in skeletal

Mechanisms Regulating Neuromuscular Junction Development and Function and Causes of Muscle Wasting.

The neuromuscular junction is the chemical synapse between motor neurons and skeletal muscle fibers. It is designed to reliably convert the action potential from the presynaptic motor neuron into the contraction of the postsynaptic muscle fiber. Diseases that affect the neuromuscular junction may cause failure of this conversion and result in loss of ambulation and respiration. The loss of motor input also causes muscle wasting as muscle mass is constantly adapted to contractile needs by the balancing of protein synthesis and protein degradation. Finally, neuromuscular activity and muscle mass

Tumor inoculation site affects the development of cancer cachexia and muscle wasting.

The phenotype and severity of cancer cachexia differ among tumor types and metastatic site in individual patients. In this study, we evaluated if differences in tumor microenvironment would affect the development of cancer cachexia in a murine model, and demonstrated that body weight, adipose tissue and gastrocnemius muscle decreased in tumor-bearing mice. Interestingly, a reduction in heart weight was observed in the intraperitoneal tumor group but not in the subcutaneous group. We evaluated 23 circulating cytokines and members of the TGF-β family, and found that levels of IL-6, TNF-α and act

Vitamin D, muscle and bone: Integrating effects in development, aging and injury.

Beyond the established effects of muscle loading on bone, a complex network of hormones and growth factors integrates these adjacent tissues. One such hormone, vitamin D, exerts broad-ranging effects in muscle and bone calcium handling, differentiation and development. Vitamin D also modulates muscle and bone-derived hormones, potentially facilitating cross-talk between these tissues. In the clinical setting, vitamin D deficiency or mutations of the vitamin D receptor result in generalized atrophy of muscle and bone, suggesting coordinated effects of vitamin D at these sites. In this review, w

POMK mutations disrupt muscle development leading to a spectrum of neuromuscular presentations.

Dystroglycan is a transmembrane glycoprotein whose interactions with the extracellular matrix (ECM) are necessary for normal muscle and brain development, and disruptions of its function lead to dystroglycanopathies, a group of congenital muscular dystrophies showing extreme genetic and clinical heterogeneity. Specific glycans bound to the extracellular portion of dystroglycan, α-dystroglycan, mediate ECM interactions and most known dystroglycanopathy genes encode glycosyltransferases involved in glycan synthesis. POMK, which was found mutated in two dystroglycanopathy cases, is instead involv

Maternal ethanol consumption: effect on skeletal muscle development in guinea pig offspring.

The effect of ethanol on developing skeletal muscle was analyzed by examining the gastrocnemius muscle from newborn guinea pigs, exposed to ethanol during the second half of gestation. Electron microscopy revealed vacuolated sarcoplasmic reticula, enlarged lipid droplets, decreased glycogen and mitochondrial abnormalities in the skeletal muscle samples from the ethanol-exposed newborn guinea pigs. None of these abnormalities were seen in the newborn controls who were born to dams which consumed the same amount of calories as the ethanol-treated dams. The ethanol-associated abnormalities, seen

Emerging new tools to study and treat muscle pathologies: genetics and molecular mechanisms underlying skeletal muscle development, regeneration, and disease.

Skeletal muscle is the most abundant tissue in our body, is responsible for generating the force required for movement, and is also an important thermogenic organ. Skeletal muscle is an enigmatic tissue because while on the one hand, skeletal muscle regeneration after injury is arguably one of the best-studied stem cell-dependent regenerative processes, on the other hand, skeletal muscle is still subject to many degenerative disorders with few therapeutic options in the clinic. It is important to develop new regenerative medicine-based therapies for skeletal muscle. Future therapeutic strategi

Developing fluorescence sensor probe to capture activated muscle-specific calpain-3 (CAPN3) in living muscle cells.

Calpain-3 (CAPN3) is a muscle-specific type of calpain whose protease activity is triggered by Ca2+ Here, we developed CAPN3 sensor probes (SPs) to detect activated-CAPN3 using a fluorescence/Förster resonance energy transfer (FRET) technique. In our SPs, partial amino acid sequence of calpastatin, endogenous CAPN inhibitor but CAPN3 substrate, is inserted between two different fluorescence proteins that cause FRET. Biochemical and spectral studies revealed that CAPN3 cleaved SPs and changed emission wavelengths of SPs. Importantly, SPs were scarcely cleaved by CAPN1 and CAPN2. Furthermore, ou

Trichinella spiralis: inhibition of muscle larva growth and development is associated with a delay in expression of infected skeletal muscle characteristics.

Infection of skeletal muscle cells by Trichinella spiralis induces host cells to reenter the cell cycle and these cells subsequently become persistently suspended in apparent G2/M. To investigate the role of the parasite in this process, parasite growth and development within the infected cell was inhibited by irradiation of newborn larvae (48 and 80 krad) prior to infection of host muscle cells. Although larvae entered skeletal muscle cells, irradiation inhibited their subsequent growth and stichosome development, including development of alpha and beta stichocytes. A distinct delay in cell c

Muscle strength, rate of torque development and neuromuscular activation of the upper arm muscles in children and adolescents with spina bifida.

BACKGROUND: The use of locomotive devices requires sufficient levels of upper limb strength. Therefore, it is important to evaluate the maximal isometric torque, rate of torque development and neuromuscular activation in youth with spina bifida. The objective was to investigate these parameters in the elbow muscles of youth with spina bifida versus healthy age-matched peers. METHODS: Forty-eight participants (8-17 years) were recruited: Spina Bifida (n = 23) and non-affected Controls (n = 25). Maximal isometric elbow flexor/extensor contractions were performed to assess maximal muscle strengt

Developing a ceRNA-based lncRNA-miRNA-mRNA regulatory network to uncover roles in skeletal muscle development.

The precise role of lncRNAs in skeletal muscle development and atrophy remain elusive. We conducted a bioinformatic analysis of 26 GEO datasets from mouse studies, encompassing embryonic development, postnatal growth, regeneration, cell proliferation, and differentiation, using R and relevant packages (limma et al.). LncRNA-miRNA relationships were predicted using miRcode and lncBaseV2, with miRNA-mRNA pairs identified via miRcode, miRDB, and Targetscan7. Based on the ceRNA theory, we constructed and visualized the lncRNA-miRNA-mRNA regulatory network using ggalluvial among other R packages. G

Flexible 3D-Printed Piezoelectric and Bioinstructive Platforms for Skeletal Muscle Development and Organization.

Volumetric muscle loss (VML) is a devastating injury in which extensive skeletal muscle loss overwhelms endogenous healing. Despite their therapeutic efficacy, acellular constructs fail to provide a comprehensive set of bioactive cues required for muscle development. Notably, very few constructs provide bioelectrical cues necessary for muscle development. To address that need, this study aimed to develop a myogenic platform that provides a comprehensive set of bioactive cues, including bioelectrical cues that can be delivered wirelessly, to promote skeletal muscle development. To achieve this,

Effects of Aging on Muscle Development Based on Transcriptome Sequencing in Dahen Broilers.

Skeletal muscle growth and development represent a pivotal biological process that determines both the yield and quality of poultry meat. As chickens age, there is an observable shift in their muscle structure, function and metabolic properties. However, the molecular mechanisms by which age regulates chicken muscle development and meat quality formation are not yet fully understood. The objective of this study was to elucidate the fundamental molecular mechanisms through which the aging process influences the development of chicken muscle and the quality of meat. A total of 20 female Dahen ch

RNA m5C Modifications in the Development and Prognosis of Muscle-Invasive Bladder Cancer.

The m5C RNA modifications have been implicated in the pathogenesis of urothelial carcinoma and hold potential as prognostic biomarkers for muscle-invasive bladder cancer (MIBC) patients. In this study, we developed an MIBC-risk model by integrating m5C modification-related genes and differentially expressed genes using Nanopore sequencing and a machine learning approach. Compared to our previous research, we observed that m5C modifications are more functional, with the most enriched regions being the 3'UTR and exons. Our analysis revealed differential m5C methylation sites in several well-char

Vascular Smooth Muscle Cell Migration and P70S6K: Key Players in Intimal Hyperplasia Development.

BACKGROUND: Vascular smooth muscle cell (VSMC) recruitment and activation by vessel injury cause intimal hyperplasia (IH) and restenosis. Drug-eluting stents releasing mTOR (mechanistic target of rapamycin) blockers (sirolimus, everolimus [EV]) improve surgery outcomes but exhibit nonspecific effects and poor efficacy in diseased vessels. Drug combinations targeting the multifactorial processes leading to IH could enhance efficacy and reduce toxicity. Our previous work showed that Kv1.3 channel blockers such as 5-(4-phenoxybutoxy)psoralen (PAP-1) prevented IH. Since Kv1.3 signaling works throu

Targeting Progesterone Receptor Membrane Component 1 to Improve Muscle Development and Glucose Homeostasis.

BACKGROUND: Type 2 diabetes mellitus (T2D) arises from the interplay between peripheral insulin resistance and pancreatic β-cell dysfunction, ultimately leading to impaired glucose utilization and chronic hyperglycemia. Despite therapeutic advances, the multifactorial nature of T2D continues to demand the development of novel treatment strategies. Progesterone receptor membrane component 1 (PGRMC1) has emerged as a potential modulator of metabolic function, though its role in T2D pathogenesis has not been fully elucidated. METHODS: To investigate the role of PGRMC1 in T2D, we generated skelet

Development of Soft Palatal Musculature Characterized by the Uvula Muscle of Miniature Pig.

INTRODUCTION: Surgical reconstruction is recommended for cleft palate patients. However, improper muscle reconstruction can cause scarring and velopharyngeal insufficiency. Understanding the developmental patterns of soft palatal musculature is crucial for improving treatments. Although mice are commonly used to investigate soft palate development, the uvula deficiency in mice limits their applicability. This study aimed to compare the developmental characteristics of soft palate muscles in miniature pigs and mice at various stages to better understand the patterns in large mammals. METHODS:

Development of a prognostic model for muscle-invasive bladder cancer using glutamine metabolism.

BACKGROUND: Muscle-invasive bladder cancer (MIBC) is distinguished by its pronounced invasiveness and unfavorable prognosis. Immunotherapy and targeted therapy have emerged as key treatment options for various types of cancer. Altered metabolism is a defining characteristic of cancer cells, and there is mounting evidence suggesting the important role of glutamine metabolism (GM) in tumor metabolism. Nevertheless, the relationship between GM and clinical outcomes, immune microenvironment, and immunotherapy in MIBC remains unknown. METHODS: This study employed Mendelian randomization to explore
