Glycogen storage disease in the alpha-glucosidase knockout(6neo(-)/6neo(-)) (GAA KO) mouse, a model of Pompe disease, results in the pathologic accumulation of glycogen primarily within skeletal myocytes and cardiomyocytes. Intravenous administration of recombinant human alpha-glucosidase (rhGAA, Myozyme, aglucosidase alfa) can result in significant glycogen clearance from both cardiomyocytes and skeletal myocytes, however, the degree of clearance varies from one skeletal muscle type to another. We sought to determine what role muscle fiber type predominance played in this variability. To exam

Skeletal muscles are composed of heterogeneous collections of fibers with different metabolic profiles. With varied neuronal innervation and fiber-type compositions, each muscle fulfils specific functions and responds differently to stimuli and perturbations. We assessed individual fibers by mass spectrometry to dissect protein changes after loss of neuronal innervation due to section of the sciatic nerve in mice. This proteomics approach enabled us to quantify ∼600 proteins per individual soleus and EDL (extensor digitorum longus) muscle fiber. Expression of myosin heavy chain (MyHC) in indiv

Muscle wasting is one of the major health problems in older adults and is traditionally associated to sarcopenia. Nonetheless, muscle loss may also occur in older adults in the presence of cancer, and in this case, it is associated to cancer cachexia. The clinical management of these conditions is a challenge due to, at least in part, the difficulties in their differential diagnosis. Thus, efforts have been made to better comprehend the pathogenesis of sarcopenia and cancer cachexia, envisioning the improvement of their clinical discrimination and treatment. To add insights on this topic, this

The skeletal muscle is the main organ responsible for insulin action, and glucose disposal and metabolism. Endurance and/or resistance training raises the number of mitochondria in diabetic muscles. The details of these adaptations, including mitochondrial adaptations of the slow and fast muscles in diabetes, are unclear. This study aimed to determine whether exercise training in streptozotocin (STZ)-induced mice leads to differential adaptations in the slow and fast muscles, and improving glucose clearance. Eight-week-old mice were randomly distributed into normal control (CON), diabetes (DM)

Ice hockey is an intense team sport characterized by repeated bursts of fast-paced skating, rapid changes in speed and direction and frequent physical encounters. These are performed in on-ice shifts of ~30-80 s interspersed with longer sequences of passive recovery, resulting in about 15-25 min on-ice time per player. Nearly 50% of the distance is covered at high-intensity skating speeds and with an accentuated intense activity pattern in forwards compared to defensemen. During ice hockey match-play, both aerobic and anaerobic energy systems are significantly challenged, with the heart rate i

Muscle fibers have been classified into two major forms of red (slow twitch) and white (fast twitch) muscles. The red muscle utilizes lipid as energy source through mitochondrial metabolism and function to sustain the position against gravity (sometimes called as antigravity muscle). Under microgravity the red muscle is selectively involved. In our unloading study by hindlimb suspension experiment on rats, the one of the representative red muscle of soleus muscle underwent rapid atrophy; they reduced their weights about 50% after 2 week-unloading. In addition, myofibrils were occasionally mark

PURPOSE: Increased oxidative stress and impaired antioxidant defense are important mechanisms in the pathogenesis of diabetic myopathy. Since diabetes mellitus type 1 decreases muscle regeneration capacity the present study was designed to determine the influence of alpha-lipoic acid (ALA), a potent biological antioxidant, on the process of regeneration of diabetic rat skeletal muscles.
METHODS: 40 Wistar rats were divided into three groups: control (n = 8), untreated diabetic group (n = 16) and ALA treated diabetic group (n = 16). The regeneration process was provoked in streptozotocin-induc

Muscle responses to tetanic electrical stimulation were detected by the non-invasive tensiomyographic (TMG) measuring method. The main objective of this study was to find out whether the TMG measuring method is suitable for monitoring the unfused tetanus (stimulation frequencies ranging from 1 Hz to the fusion frequency (ff)--the frequency at which tetanus occurs), and whether this monitoring provides any information on skeletal muscles' structural or functional changes. The muscle adaptation process was observed in damped unfused tetanus (DUT). The measured results in the clinical environment

We tested the hypothesis that cancer cachexia progression would induce oxidative post-translational modifications (Ox-PTMs) associated with skeletal muscle wasting, with different responses in muscles with the prevalence of glycolytic and oxidative fibers. We used cysteine-specific isotopic coded affinity tags (OxICAT) and gel-free mass spectrometry analysis to investigate the cysteine Ox-PTMs profile in the proteome of both plantaris (glycolytic) and soleus (oxidative) muscles in tumor-bearing and control rats. Histological analysis revealed muscle atrophy in type II fibers in plantaris muscl

The transcriptional coactivator peroxisome proliferator-activated receptor gamma coactivator 1alpha (PGC-1alpha) is a key integrator of neuromuscular activity in skeletal muscle. Ectopic expression of PGC-1alpha in muscle results in increased mitochondrial number and function as well as an increase in oxidative, fatigue-resistant muscle fibers. Whole body PGC-1alpha knock-out mice have a very complex phenotype but do not have a marked skeletal muscle phenotype. We thus analyzed skeletal muscle-specific PGC-1alpha knock-out mice to identify a specific role for PGC-1alpha in skeletal muscle func

INTRODUCTION: The mechanism by which weakness develops in idiopathic inflammatory myopathies (IIMs) is still unclear. In this study we investigated the maximum force of single muscle fibers from patients with IIMs.
METHODS: Permeabilized single muscle fibers from patients with IIMs and healthy controls were subjected to contractility measurements. Maximum force and specific force production (maximum force normalized to fiber size) and fiber type were determined for each isolated fiber.
RESULTS: A total of 178 fibers were studied from five patients with IIMs and 95 fibers from four controls.

BACKGROUND: Alcoholic myopathy is known to primarily affect type II muscle fibers (glycolytic, fast-twitch, anaerobic), whereas type I fibers (oxidative, slow-twitch, aerobic) are relatively protected.
OBJECTIVE: We investigated whether aldehyde-derived adducts of proteins with malondialdehyde and acetaldehyde are formed in muscle of rats as a result of acute exposure to ethanol and acetaldehyde. The differences between type I muscle, type II muscle, and liver tissue were also assessed.
DESIGN: The formation and distribution of malondialdehyde- and acetaldehyde-protein adducts were studied w

The effects of thyrotoxicosis on the contractile properties of soleus muscle were examined in rats given 3 mg of T4 and 1 mg of T3 per kg of diet for 6--8 wk. Thyrotoxicosis induced significant decreases in isometric twitch contraction time (CT), one-half relaxation time, and peak twitch tension. The Ca2+ uptake activity of the sarcoplasmic reticulum (SR) was increased in the thyrotoxic muscles; this adaptation in the SR provides a possible mechanism for the alterations in isometric contractile properties. Thyrotoxicosis induced a large increase in fibers classified as type 2, on the basis of

Mitsugumin 29 (MG29), a major protein component of the triad junction in skeletal muscle, has been identified to play roles in the formation of precise junctional membrane structures important for efficient signal conversion in excitation-contraction (E-C) coupling. We carried out several experiments to not only study the role of MG29 in normal muscle contraction but also to determine its role in muscle fatigue. We compared the in vitro contractile properties of three muscles types, extensor digitorum longus (EDL) (fast-twitch muscle), soleus (SOL) (slow-twitch muscle), and diaphragm (DPH) (mi

We report a patient with myoclonic epilepsy who underwent muscle biopsy for suspected mitochondrial disease (myoclonic epilepsy with ragged-red fibers, MERRF). In spite of normal histochemical studies and of the absence of a severe COX deficiency, the molecular analysis showed the common MERRF mutation (A8344G) in the tRNA(Lys) gene on mitochondrial DNA. The case serves to illustrate the importance of pursuing the proposed mitochondrial genetic abnormality, even in patients with normal biopsy findings.

Several variations in the muscle fiber distribution and differentiation have been described. Different hypotheses, including maturational dysfunction of motor neurons have been postulated. We present a 28 year-old male with proximal weakness, and negative family history. Motor strength was 4-/5 proximally and 4+/5 distally in the extremities. He was areflexic with trace triceps jerks. Electrophysiological studies showed motor neuron dysfunction. Marked type II fiber predominance 97% was noted without group atrophy on a vastus-lateralis muscle biopsy. Type 2A were the largest with type 2B and 2

Sarcopenia is the age-related atrophy and weakening (decrease in force per cross-sectional area) of muscle fibers. In rat diaphragm muscle (DIAm), sarcopenia primarily affects type IIx/IIb fibers, which generate greater specific force, have the fastest maximum shortening velocities (Vmax), but display greater fatigue with repeated activation. We hypothesized that, because of the selective effect of sarcopenia on type IIx/IIb DIAm fibers, Vmax and peak power output of the DIAm decrease in old age, whereas endurance during repetitive isovelocity shortening is unaffected. Midcostal DIAm strips we

BACKGROUND: Chronic alcohol intake induces atrophy of type II (anaerobic, fast-twitch) muscle fibers in preference to type I (aerobic, slow-twitch) muscle fibers. However, the molecular mechanism underlying the preferential atrophy of type II muscle fibers remains unclear. The ubiquitin-proteasome system (UPS) mediates the degradation of myofibrillar proteins in skeletal muscle loss. This study investigated whether E3 ubiquitin ligases, including UBR2 and MuRF1, are involved in ethanol-induced type II muscle fiber-specific atrophy.
METHODS: In a chronic alcohol intake rat model, 4- to 5-week-

This study aimed to evaluate the influence of combined intermittent fasting (IF) and high-intensity interval training (HIIT) on morphology, caspase-independent apoptosis signaling pathway, and myostatin expression in soleus and gastrocnemius (white portion) muscles from healthy rats. Sixty-day-old male Wistar rats (n = 60) were divided into four groups: control (C), IF, high-intensity-interval training (T), and high-intensity-interval training and intermittent fasting (T-IF). The C and T groups received ad libitum chow daily; IF and T-IF received the same standard chow every other day. Animals

Three-dimensional engineered muscle (3D-EM) provides a physiologically relevant model for examining skeletal muscle function. Tumor necrosis factor-α (TNF-α), a pro-inflammatory cytokine elevated in chronic conditions such as sarcopenia and cachexia, has been linked to muscle weakness. However, the mechanism underlying this effect remains unclear. In this study, we used a 3D-EM system to evaluate the direct impact of TNF-α on muscle contractile force. 3D-EM was produced by seeding C2C12 myoblasts with type I collagen on a culture device, followed by 15 days of differentiation. Constructs were
