Poxvirus complement control proteins are expressed on the cell surface through an intermolecular disulfide bridge with the viral A56 protein.

The vaccinia virus (VACV) complement control protein (VCP) is an immunomodulatory protein that is both secreted from and expressed on the surface of infected cells. Surface expression of VCP occurs though an interaction with the viral transmembrane protein A56 and is dependent on a free N-terminal cysteine of VCP. Although A56 and VCP have been shown to interact in infected cells, the mechanism remains unclear. To investigate if A56 is sufficient for surface expression, we transiently expressed VCP and A56 in eukaryotic cell lines and found that they interact on the cell surface in the absence

Role of hemolysin BL in the pathogenesis of extraintestinal Bacillus cereus infection assessed in an endophthalmitis model.

Bacillus cereus is a rare cause of serious human infection but, paradoxically, causes one of the most severe posttraumatic or endogenous infections of the eye, endophthalmitis, which frequently results in blindness. The virulence of B. cereus endophthalmitis historically has been attributed to toxin production. We therefore sought to examine the contribution of the dermonecrotic toxin, hemolysin BL, to the pathogenesis of B. cereus infection in an endophthalmitis system that is highly amenable to study. The pathogenesis of infection resulting from intravitreal injection of 10(2) CFU of either

Engineering and Adapting Disulfide-Containing Proteins to Enable Intracellular Functionality.

Disulfide bonds are key structural components ensuring proper protein folding. Those bonds, however, do not form in the reducing intracellular environment, often resulting in misfolded, unstable proteins with subpar function. To enable intracellular functionality of such proteins, disulfide-free variants with restored stability and function should be engineered. Here, we present a method to substitute the cysteine residues with optimal amino acid combinations using site saturation mutagenesis and screening for improved disulfide-free variants using yeast surface display. We provide a detailed

Functional characterization and engineering of the sugar donor specificity of a 2″-O-xylosyltransferase from Panax notoginseng for rare saponins biosynthesis.

2″-O-xylosylation is a rare glycosylation pattern conferring unique bioactivities to triterpenoid saponins in Panax species. Here, we identify a novel 2″-O-xylosyltransferase, PnUGT94X1, from P. notoginseng, which regioselectively catalyzes 2″-O-xylosylation at the C3-linked sugar of diverse saponins. PnUGT94X1 exhibits a pronounced preference for UDP-xylose and a broad specificity tolerance for sugar acceptors including protopanaxadiol-, cycloartenol-, and oleanane-type saponins. Site-directed mutagenesis revealed that the residues C140 and T141, with a distance of 22 residues downstream of t

[Protein engineering of PgsA combined with Bayesian optimization drives efficient poly-γ-glutamic acid synthesis by Corynebacterium glutamicum].

Poly-γ-glutamic acid (γ-PGA) is a biopolymer polymerized from l-glutamic acid (l-Glu) and/or d-glutamic acid monomers, exhibiting broad application prospects in pharmaceuticals, cosmetics, and other fields. The low yields remain a key factor limiting large-scale production of γ-PGA in heterologous expression systems. To enhance the biosynthetic efficiency, this study employed Corynebacterium glutamicum as the chassis cell and utilized the γ-PGA synthase PgsBCA from Bacillus licheniformis to catalyze γ-PGA synthesis. First, a polycistron expression system was constructed, increasing the γ-PGA y

CRISPR/Cas9 mediates efficient site-specific mutagenesis of the odorant receptor co-receptor (Orco) in the malaria vector Anopheles sinensis.

BACKGROUND: Anopheles sinensis is the most widely distributed mosquito species and is the main transmitter of Plasmodium vivax malaria in China. Most previous research has focused on the mechanistic understanding of biological processes in An. sinensis and novel ways of interrupting malaria transmission. However, the development of functional genomics and genetics-based vector control strategies against An. sinensis remain limited because of insufficient site-specific genome editing tools. RESULTS: We report the first successful application of the CRISPR/Cas9 mediated knock-in for highly effi

A structure-function study of C-terminal residues predicted to line the export channel in Salmonella Flagellin.

BACKGROUND: Structural studies of a Salmonella Typhimurium flagellin protein indicated that four polar or charged C-terminal amino acid residues line the inner channel of the flagellum. The hydrophilic character of these putative channel-lining residues was predicted to be essential to facilitate the transport of unfolded flagellin monomers during flagellar assembly. The structure-function relationship of these putative channel-lining residues was investigated by site-directed mutagenesis to examine effects of side chain polarity and size on flagella assembly and function. METHODS: Channel-li

Structural basis of the transactivation deficiency of the human PPARγ F360L mutant associated with familial partial lipodystrophy.

The peroxisome proliferator-activated receptors (PPARs) are transcription factors that regulate glucose and lipid metabolism. The role of PPARs in several chronic diseases such as type 2 diabetes, obesity and atherosclerosis is well known and, for this reason, they are the targets of antidiabetic and hypolipidaemic drugs. In the last decade, some rare mutations in human PPARγ that might be associated with partial lipodystrophy, dyslipidaemia, insulin resistance and colon cancer have emerged. In particular, the F360L mutant of PPARγ (PPARγ2 residue 388), which is associated with familial partia

Calcineurin-binding protein Cbp1 directs the specificity of calcineurin-dependent hyphal elongation during mating in Cryptococcus neoformans.

Mating and virulence of the human fungal pathogen Cryptococcus neoformans are controlled by calcineurin, a serine-threonine-specific calcium-activated phosphatase that is the target of the immunosuppressive drugs cyclosporine A and FK506. In previous studies, a calcineurin binding protein (Cbp1, Rcn1, Dscr1/Csp1-3/MCIP1-3) that is conserved from yeasts to humans has been identified, but whether this protein functions to regulate calcineurin activity or facilitate calcineurin function as a signaling effector has been unclear. Here we show that, like calcineurin, Cbp1 is required for mating in C

C-terminus mutations of Acremonium chrysogenum deacetoxy/deacetylcephalosporin C synthase with improved activity toward penicillin analogs.

Deacetoxy/deacetylcephalosporin C synthase (acDAOC/DACS) from Acremonium chrysogenum is a bifunctional enzyme that catalyzes both the ring-expansion of penicillin N to deacetoxycephalosporin C (DAOC) and the hydroxylation of the latter to deacetylcephalosporin C (DAC). Three residues N305, R307 and R308 located in close proximity to the C-terminus of acDAOC/DACS were each mutated to leucine. The N305L and R308L mutant acDAOC/DACSs showed significant improvement in their ability to convert penicillin analogs. R308 was identified for the first time as a critical residue for DAOC/DACS activity. K

Matrix metalloproteinase degradation of extracellular matrix: biological consequences.

Targeted mutagenesis has allowed investigators to perform controlled experiments in mammals and determine the contribution of individual proteins to physiologic and pathologic processes. Recent lessons learned from matrix metalloproteinase gene targeted mice and other in vivo observations have given new life to old concepts regarding the role of proteolytic fragments of extracellular matrix proteins in regulating a variety of critical processes in cell biology.

Partially purified Echis carinatus venom cleaves active-site-mutated bovine prothrombin at two sites.

Partially purified snake venom of Echis carinatus (ECV) is a well-known enzyme used to generate meizothrombin from prothrombin. In this reaction, meizothrombin is slowly converted to two smaller fragments. It is not certain if this slow reaction is catalyzed by meizothrombin or by ECV. Using previously constructed active site mutant bovine prothrombin, we eliminated meizothrombin autolysis and examined ECV reactions. ECV did slowly generate two fragments during activation of the active site mutant prothrombin. This result indicates that ECV cleaved bovine prothrombin at two sites: at Arg323-Il

Trifluoperazine binding to mutant calmodulins.

Trifluoperazine (TFP) binding by 14 calmodulins, including 12 produced by site-directed mutagenesis, was determined. While vertebrate calmodulin binds 4.2 +/- 0.2 equiv of TFP, Escherichia coli expressed but unmutated calmodulins bind about 5.0 +/- 0.5 equiv of TFP. The cause for this difference is not known. The E. coli expressed proteins consist of two different series expressed from different calmodulin genes, CaMI and SYNCAM. The wild-type genes code for proteins that differ by nine conservative amino acid substitutions. Both these calmodulins bind 5 equiv of TFP with similar affinities, t

Path dependence theory-inspired rational design of toad tryptophan hydroxylase 1 via engineering of non-active substrate binding pockets.

Establishing efficient 5-hydroxytryptophan (5-HTP) biosynthesis systems remains industrially challenging. Conceptually guided by Path Dependence Theory, which suggests that initial states can influence subsequent developments, we hypothesized that optimizing substrate routing away from non-active substrate binding pockets would enhance catalytic efficiency by reducing kinetic trapping. Using site-directed mutagenesis on Bufo bufo gargarizans tryptophan hydroxylase 1 (BbgTPH1), we engineered mutants Y59R-S120R and A198R to weaken substrate L-tryptophan binding to these non-active pocket. Cytoplasmic CopZ-Like Protein and Periplasmic Rusticyanin and AcoP Proteins as Possible Copper Resistance Determinants in Acidithiobacillus ferrooxidans ATCC 23270.

Acidophilic organisms, such as Acidithiobacillus ferrooxidans, possess high-level resistance to copper and other metals. A. ferrooxidans contains canonical copper resistance determinants present in other bacteria, such as CopA ATPases and RND efflux pumps, but these components do not entirely explain its high metal tolerance. The aim of this study was to find other possible copper resistance determinants in this bacterium. Transcriptional expression of A. ferrooxidans genes coding for a cytoplasmic CopZ-like copper-binding chaperone and the periplasmic copper-binding proteins rusticyanin and A

Studies on the specificity of unprocessed and mature forms of phytanoyl-CoA 2-hydroxylase and mutation of the iron binding ligands.

The mature form of phytanoyl-coenzyme A 2-hydroxylase (PAHX), a nonheme Fe(II)- and 2-oxoglutarate-dependent oxygenase, catalyzes the alpha-hydroxylation of phytanoyl-CoA within peroxisomes. Mutations in PAHX result in some forms of adult Refsum's disease. Unprocessed PAHX (pro-PAHX) contains an N-terminal peroxisomal targeting sequence that is cleaved to give mature PAHX (mat-PAHX). Previous studies have implied a difference in the substrate specificity of the unprocessed and mature forms of PAHX. We demonstrate that both forms are able to hydroxylate a range of CoA derivatives, but under the

Establishment of a Plasmid-Based Reverse Genetics System for the Cell Culture-Adapted Hepatitis E Virus Genotype 3c Strain 47832c.

The hepatitis E virus (HEV) causes acute and chronic hepatitis in humans. Investigation of HEV replication is hampered by the lack of broadly applicable, efficient cell culture systems and tools for site-directed mutagenesis of HEV. The cell culture-adapted genotype 3c strain 47832c, which represents a typical genotype predominantly detected in Europe, has previously been used for several basic and applied research studies. Here, a plasmid-based reverse genetics system was developed for this strain, which efficiently rescued the infectious virus without the need for in vitro RNA transcription.

Posttranslational Chemical Mutagenesis: To Reveal the Role of Noncatalytic Cysteine Residues in Pathogenic Bacterial Phosphatases.

The field of chemical site-selective modification of proteins has progressed extensively in recent decades to enable protein functionalization for imaging, drug delivery, and functional studies. In this Perspective, we provide detailed insight into an alternative use of site-selective protein chemistry to probe the role(s) of unpaired Cys residues in the structure and function of disease relevant proteins. Phosphatases are important players in the successful infection of pathogenic bacteria, which represent a significant health burden, particularly in multi-drug-resistant strains. Therefore, a

Discovery and characterization of a covalent inhibitor of New Delhi metallo β-lactamase.

New Delhi metallo β-lactamase (NDM-1) grants bacteria resistance to a broad spectrum of β-lactam antibiotics, including the last resort carbapenems. The absence of a nucleophilic serine in the active site of NDM-1 renders it resistant to canonical β-lactamase inhibitors that are used to treat β-lactam resistant bacteria. Herein, we report two new covalent irreversible inhibitors that target two highly conserved nucleophilic residues in the active site of NDM-1. N-Acryloyl-N-(isoquinoline-1-yl)acrylamide (2) inhibits NDM-1 in a time- and concentration-dependent manner without showing saturation

Cyclin-dependent kinase 12 influences protein kinase D1 kinase activity.

A unique and aggressive molecular subtype of prostate cancer is driven by recurrent mutations in the cyclin-dependent kinase 12 (CDK12) gene, which occur exclusive of other common genetic alterations. Protein Kinase D1 (PrKD1) is a well-established tumor suppressor in prostate cancer. Phosphoproteomics studies have identified serines 681 and 685 as putative PrKD1 phosphorylation sites in CDK12-mutated tumors; however, whether these proteins interact directly or the potential impact on either protein's function remains unknown. In this study, we demonstrate a direct interaction between PrKD1 an
