Glutamate decarboxylase (GadB) is a pyridoxal phosphate (PLP)-dependent enzyme that contributes to intracellular pH homeostasis and supports the survival of Mycobacterium tuberculosis (Mtb) in lung tissue, granulomas, and within host macrophages. Here, we demonstrate that glutamate decarboxylase from Mycobacterium tuberculosis (MtbGadB) is a multimeric enzyme exhibiting maximal activity under acidic conditions. MtbGadB is inhibited by the clinical antibiotic d-cycloserine as well as l-cycloserine in vitro. Both cycloserine enantiomers break the internal aldimine bond between PLP and the conser

Rv3787c is a putative S-adenosylmethionine (SAM)-dependent methyltransferase from Mycobacterium tuberculosis, the main agent of human tuberculosis. Researchers identified a conserved non-synonymous polymorphism within the highly transmissible Latin American-Mediterranean lineage, suggesting that Rv3787c may play a role in host-pathogen interactions. In this study, we phenotypically characterized an M. tuberculosis H37Rv mutant lacking Rv3787c and evaluated the complementation of this mutant with strains carrying either the wild-type or mutant alleles of Rv3787c. The mutant strain showed no sig

Future research on Mycobacterium tuberculosis (Mtb) proteins is essential for advancing tuberculosis (TB) diagnosis, treatment, and control amid rising drug resistance and global health challenges. Recent studies focus on elucidating protein structures and interactions critical to bacterial survival, virulence, and drug resistance, enabling the identification of novel therapeutic targets. Cutting-edge technologies such as single-cell proteomics, cryo-electron microscopy, and spatial proteomics provide unprecedented resolution of protein localization, dynamics, and host-pathogen interactions. The inner membrane domain (IMD) is a metabolically active and laterally discrete membrane domain initially discovered in Mycobacterium smegmatis. The IMD correlates both temporally and spatially with the polar cell envelope elongation in M. smegmatis. Whether or not a similar membrane domain exists in pathogenic species remains unknown. Here, we show that the IMD is a conserved membrane structure found in Mycobacterium tuberculosis. We used two independent approaches, density gradient fractionation of membrane domains and visualization of IMD-associated proteins through fluorescence microscopy

The present paper describes the synthesis, characterization, and biological activity of five quinoline derivatives, DCQ; ACQ12; ACQ13; ACQ14; ACQophen, and their respective copper-(II) complexes. The class of organic compounds is composed of 4,7-dichloroquinoline (DCQ) and its derivatives containing aliphatic diamines, 1,2-ethanediamine (ACQ12); 1,3-propanediamine (ACQ13); 1,4-butanediamine (ACQ14); and an aromatic diamine, o-phenylenediamine (ACQophen), as a side chain at the 4-position of the quinoline ring. Single-crystal X-ray diffraction was used to determine the structure of the Cu-DCQ c

Extrapulmonary tuberculosis (EPTB) accounts for approximately 17% of all Mycobacterium tuberculosis (M.tb) infections globally. Immunocompromised individuals, such as those with HIV infection or type 2 diabetes mellitus (T2DM), are at an increased risk for EPTB. Previous studies have demonstrated that patients with HIV and T2DM exhibit diminished synthesis of glutathione (GSH) synthesizing enzymes. In a murine model, we showed that the diethyl maleate (DEM)-induced depletion of GSH in the lungs led to increased M.tb burden and an impaired pulmonary granulomatous response to M.tb infection. How

Despite the therapeutic benefit of immune checkpoint blockade in cancers, there is no consensus on its effect in infectious diseases. Here we investigated whether blocking the immune checkpoint T cell immunoreceptor with immunoglobulin and immunoreceptor tyrosine-based inhibitory motif domains (TIGIT) increases T cell immunity in active Mycobacterium tuberculosis infection. TIGIT expression in both peripheral blood and lung lesions in tuberculosis patients was assessed, and the correlation with clinical features analyzed. The functional status of TIGIT+ and TIGIT-CD8+ T cell subsets in tubercu

We evaluated the vaccine properties of a novel attenuated strain of M. tuberculosis BN (Mtb BN) and its impact on the gut microbiota in inbred female mice in comparison with a virulent strain Mtb H37Rv and a vaccine strain BCG. The Mtb BN strain demonstrated the highest anti-tuberculosis vaccine effect in I/St mice highly susceptible to tuberculosis infection and the same effect as BCG in mice of the recombinant strain B6.I-100 and in β2 microglobulin gene knockout mice. No adverse effects of the new Mtb BN strain on the gut microbiota of BALB/c mice were revealed. The virulent strain Mtb H37R

Mycobacterium tuberculosis kills more people than any other bacterial pathogen and is becoming increasingly untreatable due to the emergence of resistance. Verapamil, an FDA-approved calcium channel blocker, potentiates the effect of several antituberculosis (anti-TB) drugs in vitro and in vivo This potentiation is widely attributed to inhibition of the efflux pumps of M. tuberculosis, resulting in intrabacterial drug accumulation. Here, we confirmed and quantified verapamil's synergy with several anti-TB drugs, including bedaquiline (BDQ) and clofazimine (CFZ), but found that the effect is no

Extrapulmonary tuberculosis refers to Mycobacterium tuberculosis involving organs other than the lungs (eg, pleura, lymph nodes, genitourinary tract, abdomen, skin, joints and bones, or meninges). In non-HIV-endemic areas, where reactivation is the predominant mechanism of tuberculosis, pleural involvement occurs in 4% of cases. We present an extremely rare case of a 62-year-old immunocompetent patient with pleural tuberculosis confirmed by surgical pleural biopsies, who presented with a large mediastinal mass and evidence of pulmonary artery invasion on CT scanning and endobronchial ultrasono

Haemophagocytic lymphohistiocytosis (HLH) is a rare, potentially fatal condition that can be primary or secondary. Secondary HLH can occur in association with infections, most commonly viral infections, but has also been reported in association with Mycobacterium tuberculosis (TB). Prompt identification of the underlying cause of HLH is important as it guides treatment decisions. Early initiation of appropriate treatment (eg, anti-TB treatment) reduces morbidity and mortality. We present a case of HLH associated with TB infection. Initial TB investigations were negative and standard combinatio

Mycobacterium tuberculosis (MTB) is a highly successful pathogen because of its ability to persist in human lungs for long periods of time. MTB modulates several aspects of the host immune response. Lymphocyte-activation gene 3 (LAG3) is a protein with a high affinity for the CD4 receptor and is expressed mainly by regulatory T cells with immunomodulatory functions. To understand the function of LAG3 during MTB infection, a nonhuman primate model of tuberculosis, which recapitulates key aspects of natural human infection in rhesus macaques (Macaca mulatta), was used. We show that the expressio

We aimed to estimate the global occurrence of zoonotic tuberculosis (TB) caused by Mycobacterium bovis or M. caprae infections in humans by performing a multilingual, systematic review and analysis of relevant scientific literature of the last 2 decades. Although information from many parts of the world was not available, data from 61 countries suggested a low global disease incidence. In regions outside Africa included in this study, overall median proportions of zoonotic TB of ≤1.4% in connection with overall TB incidence rates ≤71/100,000 population/year suggested low incidence rates. For c

Pulmonary tuberculosis is currently considered a serious health problem worldwide. To understand tuberculosis infection we need to know the interaction between the host's immune response and the microorganism causing tuberculosis. Ample research has been carried out to identify new molecules and genes involved in the evasion mechanism of the host's immune response. This knowledge has generated the possibility that future therapeutic schemes will be designed to fight these particular molecules or genes. We reviewed recent experimental evidence on the mechanisms that will successfully fight agai

TB is still a global health problem. The selection and spread of multidrug-resistant and extensively drug-resistant Mycobacterium tuberculosis strains represents a threat for global TB control. The reappearance of TB has driven an increased interest in understanding the mechanisms of drug action and drug resistance, which could provide a significant contribution in the development of new antimicrobials. In this article, the authors describe the mode of action and the resistance mechanisms of the principal first- and second-line antitubercular agents, namely isoniazid, ethionamide, ethambutol,
