Oral bacteriome and mycobiome of patients with idiopathic membranous nephropathy with different tongue coatings treated with a Chinese herbal formula.

ETHNOPHARMACOLOGICAL RELEVANCE: Moshen Fuyuan Formula (MSFY) is one of the representative Chinese medicine compound for Idiopathic membranous nephropathy (IMN), that originate from Fang Ji Huang Qi decoction in the Han dynasty. IMN is usually accompanied by different tongue coatings in traditional Chinese medicine (TCM), and tongue microorganisms are important factors affecting the formation of the tongue coating. Recently, oral microbiomes, including bacteria and fungi, have been identified as pivotal factors that contribute to disease development. However, the regulation of oral microbiomes

Exogenous ketones exert antiseizure effects and modulate the gut microbiome and mycobiome in a clinically relevant murine model of epilepsy.

OBJECTIVE: Despite growing interest in the potential use of exogenous ketones for the treatment of epilepsy, their impact on seizures and the gut microbiome and mycobiome remain unclear. METHODS: Here, we examined the effects of both oral gavage and subcutaneous (SC) injection of a ketone ester (KE) in spontaneously epileptic Kcna1-null (KO) mice that model seminal aspects of human temporal lobe epilepsy. Electroencephalographic recordings and biochemical analyses were performed in KE-treated KO mice. Fecal microbial and fungal communities were profiled to determine whether the antiseizure ac

Gut mycobiome in the spectrum of Metabolic-dysfunction Associated Steatotic Liver Disease (MASLD): from MASH to hepatocellular carcinoma.

UNLABELLED: Gut Microbiome (GM) dysbiosis is associated with the development and progression of Metabolic-dysfunction Associated Steatotic Liver Disease (MASLD) and steatohepatitis (MASH). Human GM contains bacteria, fungi, archaea, and viruses. Gut mycobiome (GMyco) is still poorly studied in patients with MASLD. Herein we investigated GMyco composition at different MASLD stages. METHODS: 23 patients were recruited: controls (n = 5); MASH (n = 6); cirrhosis (n = 7); and Hepatocellular Carcinoma (HCC) (n = 5). Stool samples were collected and stored in -80 °C for subsequent DNA extraction and

Gut mycobiome maturation and its determinants during early childhood: a comparison of ITS2 amplicon and shotgun metagenomic sequencing approaches.

INTRODUCTION: Microbial colonization of the gut in early life is important for the development of metabolism, immunity, and the brain. Fungi and bacteria both colonize the human infant gut. The relatively smaller contribution of fungi to the gut microbiome, as compared to bacteria, has posed technical challenges for the precise characterization of fungal communities (mycobiomes) and limited the ability to longitudinally examine mycobiome development. BACKGROUND: The aims of this study were to (1) characterize mycobiome maturation and identify clinical determinants of mycobiome compositional v

ITS2 amplicon profiling reveals site-specific mycobiomes in Avicennia marina rhizospheres along the red sea coast.

Despite their ecological significance, the rhizosphere mycobiomes of Egyptian Red Sea Avicennia marina remain largely unexplored. This study presents the first ITS2 amplicon-based characterization of these mangrove rhizosphere fungi at two sites: Al-Kilo 17 south of Safaga (K17) and Ras Mohamed National Park (RM). Alpha diversity showed higher richness in K17, while PCoA showed clear beta diversity differences. Taxonomic profiling indicated significant site-specific communities; Linear Discriminant Analysis Effect Size identified significant biomarkers across all hierarchical levels Ascomycota

Metagenome-based characterization of the gut bacteriome, mycobiome, and virome in patients with chronic hepatitis B-related liver fibrosis.

INTRODUCTION: The gut microbiota is believed to be directly involved in the etiology and development of chronic liver diseases. However, the holistic characterization of the gut bacteriome, mycobiome, and virome in patients with chronic hepatitis B-related liver fibrosis (CHB-LF) remains unclear. METHODS: In this study, we analyzed the multi-kingdom gut microbiome (i.e., bacteriome, mycobiome, and virome) of 25 CHB-LF patients and 28 healthy individuals through whole-metagenome shotgun sequencing of their stool samples. RESULTS: We found that the gut bacteriome, mycobiome, and virome of CHB-

ITS1-based profiling of the skin mycobiome in truncal acne reveals altered baseline ecology and heterogeneous doxycycline-associated patterns.

Truncal acne represents a biologically distinct manifestation of acne vulgaris, yet its fungal ecology remains incompletely characterized. Previous work using internal transcribed spacer 2 (ITS2) sequencing suggested that truncal acne is associated with altered fungal richness and Malassezia species composition; however, fungal marker choice may influence ecological inference, particularly in sebaceous skin dominated by Malassezia. In this study, we characterized the truncal skin mycobiome of patients with truncal acne and healthy controls using internal transcribed spacer 1 (ITS1) amplicon se

Vaginal mycobiome characteristics and therapeutic strategies in vulvovaginal candidiasis (VVC): differentiating pathogenic species and microecological features for stratified treatment.

SUMMARYVulvovaginal candidiasis (VVC) is a prevalent global health burden, particularly among reproductive-aged women. Recurrent VVC affects a significant proportion of this population, presenting therapeutic challenges. The predominant pathogen, Candida albicans, opportunistically transitions from a commensal organism to a pathogen when microenvironmental conditions become dysregulated. Recently, non-albicans Candida species have gained attention for their reduced antifungal susceptibility and recurrence tendencies. Diagnosis is constrained by the limitations of conventional microbiological t

Characterization of Intestinal Mycobiome in Surgical Resections from Inflammatory Bowel Disease Patients: A Deeper Analysis in Complicated Crohn's Disease Phenotypes.

BACKGROUND: Inflammatory bowel disease (IBD), which encompasses ulcerative colitis (UC) and Crohn's disease (CD), is a chronic condition characterized by recurrent intestinal inflammation and complications. Despite extensive research on bacterial dysbiosis in IBD, the role of the gut mycobiome remains largely unexplored, particularly in surgical tissue specimens. METHODS: In this study, we performed a comprehensive analysis of the intestinal fungal communities in surgical resections obtained from 20 patients with UC and 30 patients with CD, with non-IBD resections serving as controls. Dynamic Observation of Colonic Lesions, Commensal Microbiome, and Mycobiome Variations in Trinitrobenzene Sulfonic Acid-Induced Experimental Crohn's Disease in Rats.

INTRODUCTION: Crohn's disease (CD) is an inflammatory bowel disease characterized by chronic inflammation of the entire digestive lining. Although the pathogenesis of CD remains unclear, multiple factors, especially altered microbiota, are among its causes. METHODS: In this study, an experimental CD model was established by trinitrobenzene sulfonic acid (TNBS) enema. Then the dynamic changes of colonic tissue lesions, tight junctions, inflammation response, and oxidative stress are respectively tested by hematoxylin and eosin staining, immunofluorescence staining, and commercial kits. 16S rRN

Fungal Mycobiome of Mature Strawberry Fruits (Fragaria x ananassa Variety 'Monterey') Suggests a Potential Market Site Contamination with Harmful Yeasts.

An amplicon metagenomic approach based on the ITS2 region of fungal rDNA was used to investigate the diversity of fungi associated with mature strawberries collected from a volcanic orchard and open-air market stands. Based on the Kruskal-Wallis test, no statistically significant differences were observed in both non-phylogenetic and phylogenetic alpha diversity indices. According to beta diversity analyses, significant differences in fungal communities were found between groups (orchard vs. market). Taxonomic assignment of amplicon sequence variables (ASVs) revealed 7 phyla and 31 classes. Th

Across Kingdoms: The Bacteriome, Mycobiome, and Virome in Autoimmune Diseases: Mechanistic Insights, Therapeutic Perspectives, and the Emerging Role of COVID-19.

Autoimmune and immune-mediated inflammatory diseases (IMIDs) develop when genetically and environmentally susceptible hosts lose stable immune tolerance. The gut ecosystem is increasingly recognized as a biologically active interface in this process. Its bacterial, fungal, and viral components may shape mucosal and systemic immunity through antigenic stimulation, barrier regulation, and metabolite-dependent signaling, although the strength of evidence is uneven: bacteriome data are currently the most mature, whereas mycobiome, virome, and phageome findings remain more disease-specific and emer

Evaluating the impact of immunization with the "pan-fungal" vaccine, NXT-2, on the gut mycobiome and microbiome in non-human primates (NHPs).

Fungal infections remain a significant public health concern with high mortality, morbidity, and increasing associated health costs. This burden is projected to rise due to expansion of at-risk populations, limited therapeutics, increasing drug resistance, and the emergence of new fungal pathogens. Even with these challenges, there are currently no approved vaccines. We previously developed a "pan-fungal" vaccine candidate, NXT-2, that confers protection against multiple invasive fungal infections such as pulmonary aspergillosis, pneumocystosis, and invasive candidiasis, as well as non-invasiv

Associations of the gut mycobiome and its cross-kingdom interactions with estrus return in post-weaning sows revealed by metagenomic analysis.

Post-weaning estrus return is a critical determinant of reproductive efficiency in the swine industry. While the gut microbiome, particularly bacteria, has been significantly associated with estrus return in sows, the role of the gut mycobiome and its cross-kingdom interactions with bacteria in this context remains largely unexplored. Here, we employed fecal metagenomics to characterize the gut mycobiome in 85 sows and investigated its association with post-weaning estrus return. A total of 22 fungal species were significantly associated with estrus return. Normal-return sows were characterize

Age-specific alterations of the gut mycobiome in patients with myalgic encephalomyelitis/chronic fatigue syndrome and identification of potential diagnostic biomarkers.

BACKGROUND: While bacterial dysbiosis and metabolic disruptions have been widely implicated in the pathogenesis of myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS), the contribution of the gut mycobiome remains largely unresolved, particularly in the context of host age. RESULTS: Here, high-throughput internal transcribed spacer (ITS) sequencing was applied to fecal samples from 118 individuals (59 ME/CFS patients and 59 healthy matched controls), stratified into three age cohorts: young (18–34 years), middle-aged (35–55 years), and elderly (56–85 years). ME/CFS patients demonstrate

Amphotericin B ameliorates obesity through modulating gut mycobiome dysbiosis and brown adipose tissue thermogenesis in high-fat diet-induced mice.

OBJECTIVES: The gut mycobiome influences adipose tissue thermogenesis and obesity. Amphotericin B (AmB) has been shown to improve steatohepatitis in mice fed a high-fat diet (HFD). The objective of this study was to examine the effects of AmB on diet-induced obesity and the resulting gut mycobiome dysbiosis in mice. METHODS: Male C57BL/6 mice were fed a normal-fat diet or an HFD, with HFD-mice receiving either no treatment or AmB gavage. After 11 weeks, adipose indexes, lipid levels, tissue histology, gene expression, and the gut mycobiome composition were analysed. Key fungal species in lipi

High humidity reprograms the gut mycobiome to promote Meyerozyma caribbica-derived syringic acid and attenuate sepsis-induced acute kidney injury.

Sepsis-induced acute kidney injury (SAKI) remains a major contributor to mortality, yet the impact of environmental factors-particularly humidity-on disease progression is poorly understood. Here, we investigated how short-term high-humidity exposure shapes host susceptibility to SAKI and explored the underlying microbiota- and metabolite-mediated mechanisms. Mice pre-exposed to high humidity exhibited markedly attenuated renal injury and improved survival following cecal ligation and puncture (CLP). Notably, this protective effect persisted after bacterial depletion, but was abolished by amph

Adipose tissue-derived stem cell exosomes enhance skin barrier function and show exploratory associations with the skin mycobiome in aging skin.

Skin aging increases transepidermal water loss (TEWL), reduces elasticity, and perturbs the skin microbiome. Adipose tissue-derived stem cell exosomes (ASCE) show regenerative potential; however, their clinical effects on skin physiology and microbiome remain unclear. We conducted a split-face, randomized controlled trial in 16 adults aged ≥ 40 years with visible facial aging. One facial side received ultrasound-assisted transdermal delivery of a human ASCE-containing solution (HACS), whereas the other side received normal saline, at two-week intervals for three sessions.
