Single-Cell Transcriptomics and Lineage Tracing Unveil Parallels in Lymphatic Muscle and Venous Smooth Muscle Development, Identity, and Function.

BACKGROUND: Lymphatic muscle cells (LMCs) are indispensable for lymphatic vessel contraction, and their aberrant recruitment or absence is associated with both primary and secondary lymphedema. Despite their critical role in lymphatic vessel function, the cellular and molecular bases that confer the unique contractile properties to LMCs are largely undefined, limiting the development of therapeutic interventions that precisely target LMCs. METHODS: We used single-cell RNA sequencing, genetic lineage tracing, whole mount immunostaining, and intravital imaging to investigate the basis for the h

Secondary Hyperparathyroidism in Heart Failure.

Secondary hyperparathyroidism (SHPT) is a well-known pathophysiologic feature of chronic renal failure. In recent years, SHPT has become recognized as a complication of the aldosteronism associated with congestive heart failure and where excretory Ca2+ and Mg2+ wasting results in plasma-ionized hypocalcemia and hypomagnesemia. Elevations in plasma parathyroid hormone have adverse systemic consequences, including intracellular Ca2+ overloading of myocytes and vascular smooth muscle with the induction of oxidative stress. Herein, we briefly review the presence and adverse outcomes of SHPT in per

S100B: a multifunctional role in cardiovascular pathophysiology.

S100B, a calcium-binding protein of the EF-hand type exerts both intracellular and extracellular functions. S100B is induced in the myocardium of human subjects and an experimental rat model following myocardial infarction. Forced expression of S100B in neonatal rat myocyte cultures, and high level expression of S100B in transgenic mice hearts and aortic smooth muscle cells inhibit cardiac hypertrophy and the associated phenotype, arterial smooth muscle proliferation, respectively, but demonstrate increased apoptosis following α(1)-adrenergic stimulation or myocardial infarction. [Effect of n-palmitoylethanolamine on energy-dependent transport of Ca2+ in intracellular structures of myometrium and their phospholipid composition].

N-palmitoylethanolamine (NPE) was studied for its effect on the systems of energy-dependent transport of Ca2+ in the intracell structures (mitochondria, endoplasmic reticulum) of permeabilized cells of smooth muscles of the rat uterus as well as on the lipid composition of myocytes. NPE in concentration of 10(-5) M partially (by 30-50%) inhibited energy-dependent accumulation of Ca2+ in endoplasmic reticulum and mitochondria of myometrium cells permeabilized by means of treatment of myocytes suspension by digitonin (0.1 mg/ml). NPE modifies the lipid composition of permeabilized myocytes when

Cardiotrophin-1 in cardiovascular regulation.

Cardiotrophin (CT)-1 was discovered by coupling expression cloning with an embryonic stem cell-based model of cardiogenesis. Comparison of similarity in amino acid sequence and conformational structure indicates that CT-1 is a member of the interleukin (IL)-6 type cytokine family that shares the transmembrane signaling protein, glycoprotein (gp) 130 as a receptor. These cytokines mediate overlapping pleiotropic actions on a variety of cell types including cardiac myocytes, hepatocytes, megakaryocytes, osteoclasts, and neuronal cells. CT-lmediates its hypertrophic and cytoprotective properties

Electrophysiological and computational analyses of anethole-induced inhibition of L-type calcium channels in rat tracheal smooth muscle.

Anethole, a naturally occurring phenylpropanoid, is a major constituent of several essential oils and exhibits a broad spectrum of pharmacological activities, including reduction of neuronal excitability, antispasmodic effects, and inhibition of Ca2+ influx. To elucidate the mechanism underlying calcium current inhibition, we investigated the effects of anethole on voltage-dependent calcium channels (VDCCs) in rat tracheal smooth muscle myocytes. Whole-cell patch-clamp recordings in dissociated tracheal myocytes were combined with molecular docking analyses. Ba2+ was used as the charge carrier

A pH-sensitive potassium conductance (TASK) and its function in the murine gastrointestinal tract.

The excitability of smooth muscles is regulated, in part, by background K+ conductances that determine resting membrane potential. However, the K+ conductances so far described in gastrointestinal (GI) muscles are not sufficient to explain the negative resting potentials of these cells. Here we describe expression of two-pore K+ channels of the TASK family in murine small and large intestinal muscles. TASK-2, cloned from murine intestinal muscles, resulted in a pH-sensitive, time-dependent, non-inactivating K+ conductance with slow activation kinetics. A similar conductance was found in native

Synaptopodin-2 Isoforms Have Specific Binding Partners and Display Distinct, Muscle Cell Type-Specific Expression Patterns.

Synaptopodin-2 (SYNPO2) is a protein associated with the Z-disc in striated muscle cells. It interacts with α-actinin and filamin C, playing a role in Z-disc maintenance under stress by chaperone-assisted selective autophagy (CASA). In smooth muscle cells, SYNPO2 is a component of dense bodies. Furthermore, it has been proposed to play a role in tumor cell proliferation and metastasis in many different kinds of cancers. Alternative transcription start sites and alternative splicing predict the expression of six putative SYNPO2 isoforms differing by extended amino- and/or carboxy-termini. Our a

Smooth muscle hypertrophy following partial bladder outlet obstruction is associated with overexpression of non-muscle caldesmon.

Partial bladder outlet obstruction (PBOO) induces remodeling of urinary bladder smooth muscle (detrusor). We demonstrate an increase in bladder wall mass, muscle bundle size, and a threefold increase in the cross-sectional area of detrusor myocytes following PBOO in male New Zealand White rabbits compared to that of controls. Some bladders with detrusor hypertrophy function close to normal (compensated), whereas others were dysfunctional (decompensated), showing high intravesical pressure, large residual urine volume, and voiding difficulty. We analyzed the expression of smooth muscle-specific

From Invaginating Site to Deep Lesion: Spatial Transcriptomics Unravels Ectopic Endometrial Penetration Features in Adenomyosis.

Adenomyosis, characterized by clinical intractability, significantly impacts female fertility and life quality due to the absence of definitive diagnostic markers and effective treatment options. The invagination theory is a primary hypothesis for adenomyosis, but the underlying molecular mechanisms remain unclear. In this study, a spatial transcriptional landscape of adenomyosis with an evident invagination structure is mapped from the endometrial invaginating site to ectopic lesions utilizing spatial transcriptomics and single-cell RNA sequencing. In addition, the authors employ bulk RNA seq

A Polyphenol-Network-Mediated Coating Modulates Inflammation and Vascular Healing on Vascular Stents.

Localized drug delivery from drug-eluting stents (DESs) to target sites provides therapeutic efficacy with minimal systemic toxicity. However, DESs failure may cause thrombosis, delay arterial healing, and impede re-endothelialization. Bivalirudin (BVLD) and nitric oxide (NO) promote arterial healing. Nevertheless, it is difficult to combine hydrophilic signal molecules with hydrophobic antiproliferative drugs while maintaining their bioactivity. Here, we fabricated a micro- to nanoscale network assembly consisting of copper ion and epigallocatechin gallate (EGCG) via π-π interactions, metal c

Long Noncoding RNA FTX Reduces Hypertrophy of Neonatal Mouse Cardiac Myocytes and Regulates the PTEN/PI3K/Akt Signaling Pathway by Sponging MicroRNA-22.

BACKGROUND Cardiac myocyte hypertrophy results from clinical conditions that include hypertension and valvular heart disease, and can result in heart failure. This study aimed to investigate the expression and role of the long noncoding RNA FTX (lnc-FTX), an X-inactive-specific transcript (XIST) regulator transcribed from the X chromosome, in hypertrophy of neonatal mouse cardiac myocytes induced by angiotensin II (Ang II) in vitro. MATERIAL AND METHODS Cardiac myocytes were isolated from neonatal mice and cultured with and without Ang II. Immunofluorescence, with localization of an antibody t

Ultrastructural changes in resistance arterioles of normotensive and hypertensive premenopausal women with uterine fibroids.

The association between uterine smooth muscle fibroids and systemic arterial hypertension is poorly understood. Therefore, we explored possible concomitant myocyte pathology of systemic resistance-sized arterioles (200 to 400 μm), isolated from omental samples donated by 19 women (mean age 42 y; SE 1) undergoing fibroid surgery. Vessels of 17 women (8 hypertensives) were available for transmission electron microscopy. We found ultrastructural vessel wall abnormalities in all patients, albeit with greater severity in hypertensives. In the endothelium, this ranged from moderate mitochondrial and

Stromatoxin-sensitive, heteromultimeric Kv2.1/Kv9.3 channels contribute to myogenic control of cerebral arterial diameter.

Cerebral vascular smooth muscle contractility plays a crucial role in controlling arterial diameter and, thereby, blood flow regulation in the brain. A number of K(+) channels have been suggested to contribute to the regulation of diameter by controlling smooth muscle membrane potential (E(m)) and Ca(2+) influx. Previous studies indicate that stromatoxin (ScTx1)-sensitive, Kv2-containing channels contribute to the control of cerebral arterial diameter at 80 mmHg, but their precise role and molecular composition were not determined. Here, we tested if Kv2 subunits associate with 'silent' subuni

On the wide spectrum of abnormalities in the coronary arteries of Whipple's disease.

BACKGROUND: There is growing interest in the role of microbes in the pathogenesis of coronary atherosclerosis but most of the evidence has been seroepidemiologic. It would be useful to know more about the cytology and histology of coronary lesions containing clearly depicted microbes. OBJECTIVE: To define carefully the assorted abnormalities apparent in the coronary arteries of individuals dying with Whipple's disease. METHODS: Myocardial tissue from 12 cases of Whipple's disease was studied by light microscopy. Slides were stained routinely (in sequence) with either the periodic-acid-Schiff

Molecular Mechanisms and Therapeutic Targeting of the STAT3 Signaling Axis in Vascular Smooth Muscle Cell Phenotypic Switching and Vascular Remodeling.

Vascular smooth muscle cells (VSMCs) exhibit remarkable phenotypic plasticity, dynamically transitioning from a quiescent contractile state to a dedifferentiated synthetic phenotype that constitutes the fundamental cytological driver of pathological vascular remodeling in cardiovascular diseases (CVDs) including atherosclerosis, vascular restenosis, aortic dissection (AD), and vascular calcification (VC). Signal transducer and activator of transcription 3 (STAT3) operates as a master transcriptional and functional convergence node, integrating diverse upstream biochemical stimuli, neurohumoral
