Detection and Quantification of Nocardia crassostreae, an Emerging Pathogen, in Mytilus galloprovincialis in the Mediterranean Sea Using Droplet Digital PCR.

Nocardia crassostreae is a novel pathogen responsible for infections in oysters (Crassostrea gigas) and mussels (Mytilus galloprovincialis). N. crassostreae is also responsible for nocardiosis both in immunocompetent and immunocompromised patients. We investigated N. crassostreae DNA in mussels grown in marine sites of the Mediterranean Sea in the Campania Region. We examined 185 mussel pooled samples by droplet digital PCR (ddPCR) and real-time quantitative PCR (qPCR), each pool composed of 10 mussels and 149 individual mussels. ddPCR detected N. crassostreae DNA in 48 mussel pooled samples a

The blue mussel inside: 3D visualization and description of the vascular-related anatomy of Mytilus edulis to unravel hemolymph extraction.

The blue mussel Mytilus edulis is an intensely studied bivalve in biomonitoring programs worldwide. The lack of detailed descriptions of hemolymph-withdrawal protocols, particularly with regard to the place from where hemolymph could be perfused from, raises questions regarding the exact composition of aspirated hemolymph and does not exclude the possibility of contamination with other body-fluids. This study demonstrates the use of high resolution X-ray computed tomography and histology combined with 3D-reconstruction using AMIRA-software to visualize some important vascular-related anatomic

Effects of common pharmaceutical drugs (paracetamol and acetylsalicylic acid) short term exposure on biomarkers of the mussel Mytilus spp.

Pharmaceutical drugs in the wild may pose significant risks to non-target exposed organisms. This situation is even more troublesome for coastal marine or estuarine environments, located in the vicinity of large human conglomerates, for which the putative number of pollutants is extremely high, and the regime by which wild organisms are exposed is continuous. In addition, the number of studies addressing this issue is still scarce, despite evidences that show the potential contamination profiles and adverse biological effects in organisms from such areas. In this study, the ecotoxicity of comm

The application of phage-based faecal pollution markers to predict the concentration of adenoviruses in mussels (Mytilus edulis) and their overlying waters.

AIM: This study set out to determine whether phage-based indicators may provide a 'low-tech' alternative to existing approaches that might help maintain the microbial safety of shellfish and their overlying waters. METHODS AND RESULTS: Mussels and their overlying waters were collected biweekly from an estuary in southeast England over a 2-year period (May 2013-April 2015) (n = 48). Levels of bacterial indicators were determined using membrane filtration and most probable number methods and those of bacteriophages were determined by direct plaque assay. The detection of adenovirus was determin

Sex-Dependent Changes in Quality and Flavor-Related Chemical Profiles of Mytilus coruscus During Long-Term Frozen Storage.

The effects of frozen storage on the quality and flavor of female and male Mytilus coruscus were investigated at -18 °C for 0, 90, and 150 days through analyses of physicochemical properties, free amino acids, 5'-nucleotides, fatty acids, and volatile organic compounds. Distinct sex-dependent changes were observed in the quality and flavor profiles. After 150 days, the centrifugal loss reached 1.2 and 1.5 times the fresh levels in females and males, respectively, whereas total volatile basic nitrogen reached 2.7 and 2.4 times the corresponding fresh levels. Malondialdehyde increased to approxi

Blue mussel (Mytilus edulis) water extract ameliorates intestinal immune response in high-fat diet-streptozotocin-induced diabetic mice.

Diabetes mellitus is a metabolic disease characterized by high blood glucose levels or hyperglycemia. Diabetes causes a decrease in immune function in the human body. Mytilus edulis has been identified as having anti-inflammatory properties and the ability to improve inflammation. Thus, this study aimed to investigate the function of Matsu M. edulis water extract (MWE) in mediating the regulation of immune responses and dysregulating the intestinal immune system in hyperglycemia mouse models. The mice were treated with MWE for seven weeks. The results showed that treatment with MWE has the abi

Sexual dimorphism in the gonad lipidome of blue mussels (Mytilus sp.): New insights from a global lipidomics approach.

Blue mussels (Mytilus sp.) are an economically important species for European aquaculture. Their importance as a food source is expected to increase in the coming net-zero society due to their low environmental footprint; however, their production is affected by anthropogenic stressors and climate change. During reproduction, lipids are key molecules for mussels as they are the main source of energy on which newly hatched embryos depend in the first days of their development. In this work, blue mussels of different origins are analysed, focusing on the differences in lipid composition between

Effects of ethanol pretreatment on osteogenic activity and off-flavors in blue mussel (Mytilus edulis L.) enzymatic hydrolysates.

Aquatic protein hydrolysates have many biological activities, but the off-flavor seriously decreases their commercial acceptability. Therefore, it is important to invest in finding an effective deodorization of aquatic hydrolysates that do not affect activities. In this study, ethanol pretreatment of mussel was applied to establish a new method to deodorize the blue mussel (Mytilus edulis L.) hydrolysates. LC-MS and GC-MS analysis results showed that 87.34% of fatty acids, 83.94% of aldehydes, most volatile flavor compounds including aldehydes, ketones, alcohols, acids, and hydrocarbons were d

Low oxygen stress alters response to sublethal copper exposure without inducing cross-tolerance in the blue mussel Mytilus edulis.

The Baltic Sea's limited water exchange and densely populated catchment area contribute to the frequent co-occurrence of hypoxia and chemical pollutants such as copper (Cu), raising concerns about the effects of sequential stressors on marine organisms. This study tested whether pre-exposure to intermittent oxygen stress alters the physiological and immune responses of Mytilus edulis to subsequent Cu exposure. Mussels were preconditioned for 21 days to either mild intermittent hypoxia (7 h/day at ∼2 mg/L DO) or severe intermittent anoxia (2 days/week at <0.01 mg/L DO), followed by a 7-day expo

Sex-specific lipidomic profiling of the marine mussel Mytilus coruscus: A comparative analysis of male and female lipid metabolism.

Sex-dependent lipid metabolism plays a pivotal role in physiological adaptation and reproduction in marine bivalves, yet its molecular basis remains poorly understood. Here, UPLC-HRMS-based lipidomics was applied to characterize sex-specific lipid profiles in mussels. Females showed higher levels of glycerophospholipids, particularly phosphatidylcholine and phosphatidylethanolamine enriched with polyunsaturated fatty acids, whereas males contained more glycerolipids, especially saturated fatty acid-rich triacylglycerols. Principal component analysis and orthogonal partial least squares-discrim

Benefit or burden? Effects of repeated heat exposure on thermal tolerance, energetics, and immunity in the blue mussel Mytilus edulis.

Irregular heat spikes associated with marine heat waves may either induce thermal hardening and enhance resilience or exceed physiological limits and compromise future stress tolerance in marine ectotherms. Understanding the mechanisms underlying these contrasting responses is critical for predicting the impacts of increasing thermal variability. We investigated whether prior heat exposure induces protective responses in Baltic blue mussels (Mytilus spp.). Mussels were exposed to repeated heat stress (RHS; four cycles of 2.5 h at 27°C followed by 21.5 h recovery at 15°C) and subsequently allow

Rayon microfibers exacerbate the toxic effects of polychlorinated biphenyls on foot integrity and byssal performance in the mussel Mytilus coruscus.

Rayon microfibers (RY) and polychlorinated biphenyls (PCBs) are common emerging contaminants in marine environments, and their ecological impacts have attracted increasing research attention. Although these contaminants frequently coexist and pose potential risks to aquatic organisms, their combined toxicity remains poorly understood. This research examined the effects of PCBs, RY, and their combined exposure on foot structure, byssus thread functionality, and the expression of byssus-related genes in the mussel Mytilus coruscus. Mussels were exposed for 14 days to PCBs at low (100 ng/L) and h

Molecular responses of Mytilus coruscus hemocytes to lipopolysaccharide and peptidoglycan as revealed by 4D-DIA based quantitative proteomics analysis.

Mytilus are sessile filter feeders that live in close contact with numerous marine microorganisms. Hemocytes, the immunocompetent cells of Mytilus, participate in the immune response in a very efficient manner. Lipopolysaccharide (LPS) and peptidoglycan (PGN) follow specific microbe/pathogen-associated molecular patterns (MAMPs or PAMPs) and are involved in immune stimulation in host cells. This study evaluated the molecular profiles and reactions at protein level of Mytilus hemocytes after stimulation with LPS and PGN. Mytilus coruscus was challenged in vivo with LPS and PGN. The hemocytes we

Genomic analysis reveals the presence of hypervirulent and fluoroquinolone-resistant Clostridioides difficile in farmed mussels (Mytilus galloprovincialis) in Slovenia.

Clostridioides difficile is one of the leading causes of antibiotic-associated diarrhea. In this study, we characterized 76C. difficile isolates, obtained from three Mediterranean mussel (Mytilus galloprovincialis) farms in Slovenia from November 2014 to October 2015 (sampling period 1) and from January to December 2021 (sampling period 2). The overall isolation rate of C. difficile from all the examined mussels was 59.8 %. A statistically significant trend of seasonal variation was observed, with a higher isolation rate in the colder months of the year (87.9 %; sea temperature ≤ 15 °C) compar

Cloning and expression of a GST-pi gene in Mytilus galloprovincialis. Attempt to use the GST-pi transcript as a biomarker of pollution.

Glutathione S-transferase is involved in the detoxication of many chemical compounds. Northern blot analysis of mRNA GST gene from the mussel Mytilus galloprovincialis shows the presence of a transcript of 850 bp (GenBank accession no. Gi:22094808 and AF527010). The cDNA cloned sequence is constituted by an ORF of 621 bp encoding for a protein of 23,700 Da present in the gills and digestive gland of M galloprovincialis. The sequence, called Mg (M. galloprovincialis) GST-p, is clearly related to the pi class GST. M. galloprovincialis treated with Cd (200 microg/L) and BaP (100 microg/L) or co-t

Production of reactive oxygen species by hemocytes from the marine mussel, Mytilus edulis: lysosomal localization and effect of xenobiotics.

Hemolymph of M. Edulis is rich in phagocytic hemocytes. Hemocytes contain numerous lysosomes which, in turn, contain various hydrolytic enzymes. Phagocytic activity of M. edulis hemocytes is thought to be associated with NAD(P)H-oxidase activity of the plasma membrane. The laser dye, dihydrorhodamine 123 (DHR), was used for cytochemical and biochemical detection of the generation of reactive oxygen species (ROS) by isolated M. edulis hemocytes. Hemocytes readily take up DHR from the suspension medium and selectively concentrate it in the lysosomes, wherein DHR is oxidized to fluorescent rhodam

Vibrio infection compromises byssal attachment in the mussel Mytilus coruscus by modulating energy homeostasis and byssal protein gene expression.

Mussels are important bivalve species that are cultured worldwide and attach to substrate by byssal threads. Mass detachment of mussels poses a significant threat to the sustainability of mussel aquaculture. It remains unclear whether biological factors such as infections caused by marine pathogenic bacteria affect byssal attachment in mussels. In this study, two strains each of Vibrio splendidus and V. anguillarum were isolated from seawater, and mussels were exposed to different concentrations (104, 105, 106, and 107 CFU/mL) of each bacterial strain for seven days. After exposure, we analyze

Multi-Level Toxicity of 6PPD-Q and TiO2 Nanoparticles in Mytilus coruscus: Links among Neurochemical, Circadian and Metabolic Responses.

Tire-derived 6PPD-quinone (6PPD-Q) and titanium dioxide nanoparticles (TiO2 NPs) are increasingly detected in aquatic environments, yet their combined effects on marine bivalves remain poorly understood. In this study, the thick-shell mussel Mytilus coruscus was exposed to 6PPD-Q, TiO2 NPs, and their mixtures to evaluate multi-level physiological, biochemical, histological, and molecular responses. The results demonstrated that 6PPD-Q and TiO2 NPs, particularly under high-concentration co-exposure, significantly impaired mussel physiological performance, as indicated by reduced clearance rate

Novel immunomodulatory peptides identified from Mytilus edulis: Molecular docking, immunomodulatory effects on RAW264.7 cells and molecular dynamics simulation studies.

This study sought to screen novel immunomodulatory peptides from Mytilus edulis hydrolysates (MEHs) after simulated gastrointestinal digestion (SGD) and to clarify their regulatory mechanisms. Nano-liquid chromatography-tandem mass spectrometry (LC-MS/MS) identified 99 peptides with molecular weights ranging from 186.10 to 1197.59 Da. In silico screening combined with molecular docking analyses led to the selection of five novel candidates: FPR, FRE, KFP, LLPK, and PDRM. Among these, FPR and LLPK exhibited the most potent immunomodulatory activities, significantly enhancing RAW264.7 macrophage

Combined exposure to nano-TiO₂ and carbamazepine exacerbates reproductive toxicity in mussels Mytilus coruscus by disrupting steroid hormone homeostasis.

Titanium dioxide nanoparticles (nano-TiO₂) and carbamazepine (CBZ) are both emerging contaminants of concern in coastal environments. Current monitoring data indicate that nano-TiO₂ occur in nearshore seawater affected by land-based inputs, and CBZ is persistently detected in estuarine and nearshore waters; therefore, these contaminants may co-occur at a regional scale. In this study, the thick-shelled mussel (Mytilus coruscus) was used as a model organism. Nominal concentrations of nano-TiO₂ (100 μg/L) and CBZ (10 μg/L) were applied in single and combined exposure experiments over 14 days. Pa
