CD44 isoform expression in the diffuse neuroendocrine system. I. Normal cells and hyperplasia.

Isoforms of the transmembrane glycoprotein CD44, which are generated by alternative splicing of nine variant exons, have been implicated in tumor cell adhesion, invasion and metastatic spread and may be indicators of the degree of tumor differentiation. Since little is known about the distribution of CD44 in non-neoplastic neuroendocrine cell types, we systematically investigated 42 samples of tissue from different organs, including the pituitary gland, thyroid, parathyroid, adrenal gland, lung, pancreas, stomach, duodenum, jejunum, ileum, appendix, and colon, immunohistochemically for the exp

Abnormal patterns of pulmonary neuroendocrine cells in victims of sudden infant death syndrome.

Ventilatory dysfunction has become the main focus of current research in sudden infant death syndrome (SIDS). This has been correlated with structural abnormalities in the carotid body and respiratory nuclei of the brainstem. In recent studies, the denervating effect of asphyxial brainstem dysfunction on the pulmonary neuroendocrine cells, which probably function as chemoreceptors, was demonstrated and prompted the following study. The pulmonary neuroendocrine system was evaluated in 25 victims of SIDS and 20 control infants, ranging in age from 3 weeks to 7 months and 1 to 12 months, respecti

Genetic and immunohistochemical profiling of small cell and large cell neuroendocrine carcinomas of the breast.

Neuroendocrine carcinomas (NEC) of the breast are exceedingly rare tumors, which are classified in the WHO system as small cell (SCNEC) and large cell (LCNEC) carcinoma based on indistinguishable features from their lung counterparts. In contrast to lung and enteropancreatic NEC, the genomics of breast NEC have not been well-characterized. In this study, we examined the clinicopathologic, immunohistochemical, and genetic features of 13 breast NEC (7 SCNEC, 4 LCNEC, 2 NEC with ambiguous small versus large cell morphology [ANEC]). Co-alterations of TP53 and RB1 were identified in 86% (6/7) SCNEC

Neural Circuits between Nodose Ganglion and Pulmonary Neuroendocrine Cells Regulate Lung Inflammatory Responses.

The lungs interface directly with the external environment, exposing them to airborne pathogens like endotoxins. We investigated whether the vagus nerve, which innervates the lungs-detects such pathogens. Using transcriptomics, tissue clearance imaging, electrophysiology, and cell-specific knockout models, we discovered that vagal sensory endings synapse with pulmonary neuroendocrine cells (PNECs). These nerve endings detect bacterial endotoxins primarily through the pain receptor TRPA1, not via Toll-like receptor 4 (TLR4). This detection triggers electrical excitation in vagal neurons and upr

Development of anti-somatostatin receptors CAR T cells for treatment of neuroendocrine tumors.

BACKGROUND: Neuroendocrine tumors (NETs) overexpress somatostatin receptors (SSTRs). METHODS: We developed a second-generation, ligand-based, anti-SSTR chimeric antigen receptor (CAR) incorporating the somatostatin analog octreotide in its extracellular moiety. RESULTS: Anti-SSTR CAR T cells exerted antitumor activity against SSTR+NET cell linesin vitro. The killing activity was highly specific, as demonstrated by the lack of CAR T cell reactivity against NET cells engineered to express mutated variants of SSTR2/5 by CRISPR/Cas9. When adoptively transferred in NSG mice, anti-SSTR CAR T cells

Src kinases regulate de novo actin polymerization during exocytosis in neuroendocrine chromaffin cells.

The cortical actin network is dynamically rearranged during secretory processes. Nevertheless, it is unclear how de novo actin polymerization and the disruption of the preexisting actin network control transmitter release. Here we show that in bovine adrenal chromaffin cells, both formation of new actin filaments and disruption of the preexisting cortical actin network are induced by Ca2+ concentrations that trigger exocytosis. These two processes appear to regulate different stages of exocytosis; whereas the inhibition of actin polymerization with the N-WASP inhibitor wiskostatin restricts fu

Perifosine-mediated Akt inhibition in neuroendocrine tumor cells: role of specific Akt isoforms.

The majority of neuroendocrine tumors (NETs) of the gastroenteropancreatic system show aberrant Akt activity. Several inhibitors of the phosphoinositide 3-kinase (PI(3)K)-Akt-mTOR signaling pathway are currently being evaluated in clinical phase II and III studies for the treatment of NETs with promising results. However, the molecular mechanisms and particularly the role of different Akt isoforms in NET signaling are not fully understood. In this study, we examine the effect of Akt inhibition on NET cells of heterogeneous origin. We show that the Akt inhibitor perifosine effectively inhibits

Wnt-11 promotes neuroendocrine-like differentiation, survival and migration of prostate cancer cells.

BACKGROUND: Wnt-11 is a secreted protein that modulates cell growth, differentiation and morphogenesis during development. We previously reported that Wnt-11 expression is elevated in hormone-independent prostate cancer and that the progression of prostate cancer from androgen-dependent to androgen-independent proliferation correlates with a loss of mutual inhibition between Wnt-11- and androgen receptor-dependent signals. However, the prevalence of increased expression of Wnt-11 in patient tumours and the functions of Wnt-11 in prostate cancer cells were not known. RESULTS: Wnt-11 protein le

A confocal microscopic study of solitary pulmonary neuroendocrine cells in human airway epithelium.

BACKGROUND: Pulmonary neuroendocrine cells (PNEC) are specialized epithelial cells that are thought to play important roles in lung development and airway function. PNEC occur either singly or in clusters called neuroepithelial bodies. Our aim was to characterize the three dimensional morphology of PNEC, their distribution, and their relationship to the epithelial nerves in whole mounts of adult human bronchi using confocal microscopy. METHODS: Bronchi were resected from non-diseased portions of a lobe of human lung obtained from 8 thoracotomy patients (Table 1) undergoing surgery for the rem

Immunohistochemical distribution of bombesin-positive pulmonary neuroendocrine cells in a congenital diaphragmatic hernia.

Morphometrical analyses of the immunohistochemical expression of bombesin, which is one of the peptides produced by pulmonary neuroendocrine (PNE) cells, were carried out on the bronchioles of human congenital diaphragmatic hernia (CDH) neonates, and the findings were then compared with those in a gestational and postnatal age-matched control group. As a result, no difference was found in the number of bombesin-positive cells between the lungs of the control group and the unaffected side lungs in the CDH group except for the ratio of the bombesin-positive cells per unit of the bronchiolar surf

Neuroendocrine marker substances in human Leydig cells--changes by disturbances of testicular function.

A number of neuroendocrine and neuronal markers were demonstrated in Leydig cells of the testes of 18 men aged between 20 and 81 years. Tissue sections were divided into five groups, i.e. carcinoma of the prostate (control cases; n = 4), seminoma (n = 8), anti-androgen therapy (n = 3), oestradiol therapy (n = 2) and cryptorchidism (n = 1). The following substances were immunocytochemically tested: the monoamine synthesizing enzymes tyrosine hydroxylase, aromatic L-amino acid decarboxylase, dopamine-beta-hydroxylase and phenylethanolamine-N-methyltransferase, the indolamine serotonin, the calci

Primary small cell neuroendocrine carcinoma of the liver: a case report.

This study presents a case of primary hepatic small cell neuroendocrine carcinoma (PHNEC). The patient was a 74-year-old male with a history of hepatitis C virus (HCV) infection, who presented with persistent right upper quadrant and lower back pain for two weeks. Imaging studies revealed multiple nodules and masses in the left hepatic lobe, accompanied by enlargement of lymph nodes at the hepatic hilum and in the retroperitoneum. Subsequent liver biopsy was performed, and to rule out potential origins from the gastrointestinal tract or lungs, gastrointestinal endoscopy and chest computed tomo

Clinical characteristics of diffuse idiopathic pulmonary neuroendocrine cell hyperplasia.

RATIONALE: Diffuse idiopathic pulmonary neuroendocrine cell hyperplasia (DIPNECH) is characterised by diffuse bronchial hyperplasia of pulmonary neuroendocrine cells, which are situated within the walls of bronchi and bronchioles. Presenting symptoms are nonspecific and the clinical course varies, making diagnosis challenging. We sought to describe the clinical characteristics of patients with DIPNECH in a large multinational case series to guide and inform future care and research. METHODS: Data were collated from 18 international centres. Information collected included disease presentation,

A case of Large Cell Neuroendocrine Carcinoma (LCNEC) treated with tarlatamab.

UNLABELLED: Transgender and gender-diverse young adults, most of whom identify as lesbian, gay, bisexual, or queer, encounter considerable health, social, and systemic inequalities, leading to higher rates of trauma exposure, mental health difficulties, and suicidal thoughts and behaviours compared to their heterosexual and cisgender peers. The complex relationship between sexual orientation, gender identity, and suicide has received considerable attention, primarily from quantitative research focused on risk narratives, rather than an individual's lived experiences or specific healthcare need

Intranodular Thyroid Metastasis of a Large Cell Neuroendocrine Carcinoma of the Endometrium.

A 64-year-old woman underwent initial 18 F-FDG PET/CT staging for a suspicious endometrial mass, which showed high uptake in the endometrial mass and a focal uptake in a known left thyroid nodule. Histology revealed a high-grade large cell neuroendocrine carcinoma of the endometrium with FIGO (International Federation of Gynecology and Obstetrics) stage Ib. Further explorations revealed a synchronous thyroid metastasis. Patient then received adjuvant immunochemotherapy. This case showed a potential pitfall in the initial assessment of this rare and aggressive malignancy on which the therapeuti

Long-Term Survivor of Laryngeal Small Cell Neuroendocrine Carcinoma.

Small cell neuroendocrine carcinoma (SCNC) of the larynx is an exceptionally rare subset of laryngeal carcinoma with a clinically aggressive course and poor prognosis. Long-term survival is rare, and treatment strategies remain nonstandardized due to limited data. In this report, the authors present a case of a 51-year-old male who presented with stage IVa laryngeal SCNC and no distant metastasis, treated with a combination of platinum-based chemotherapy and radiation, who has survived for more than 5 years. This case represented 1 of few reported instances of extended disease-free survival in
