Substance P in the serum of patients with rheumatoid arthritis.

Serum substance P was assayed in rheumatoid arthritis patients and healthy controls to evaluate whether neurogenic inflammation with substance P release is significant in rheumatoid arthritis. A very sensitive competitive immunoenzymetric assay was used. Mean serum substance P level was significantly higher in rheumatoid arthritis patients than in controls and was not correlated with disease duration, morning stiffness duration, Thompson's articular index, Larsen's radiographic score, or the following laboratory indices of inflammation: erythrocyte sedimentation rate, C-reactive protein, and a

alpha-eudesmol, a P/Q-type Ca(2+) channel blocker, inhibits neurogenic vasodilation and extravasation following electrical stimulation of trigeminal ganglion.

In this study, we investigated the effect of alpha-eudesmol, which potently inhibits the presynaptic omega-agatoxin IVA-sensitive (P/Q-type) Ca(2+) channel, on neurogenic inflammation following electrical stimulation of rat trigeminal ganglion. Treatment with alpha-eudesmol (0.1-1 mg/kg. i.v.) dose-dependently attenuated neurogenic vasodilation in facial skin monitored by a laser Doppler flowmetry. In addition, alpha-eudesmol (1 mg/kg. i.v.) significantly decreased dural plasma extravasation in analysis using Evans blue as a plasma marker. On the other hand, alpha-eudesmol (1 mg/kg, i.v.) did

Brain inflammation and hypertension: the chicken or the egg?

Inflammation of forebrain and hindbrain nuclei controlling the sympathetic nervous system (SNS) outflow from the brain to the periphery represents an emerging concept of the pathogenesis of neurogenic hypertension. Angiotensin II (Ang-II) and prorenin were shown to increase production of reactive oxygen species and pro-inflammatory cytokines (interleukin-1 beta (IL-1β), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α)) while simultaneously decreasing production of interleukin-10 (IL-10) in the paraventricular nucleus of the hypothalamus and the rostral ventral lateral medulla. Role of ocular surface neurobiology in neuronal-mediated inflammation in dry eye disease.

Inflammation is the consequence of dry eye disease regardless of its etiology. Several injurious or harmless processes to the ocular surface neurons promote ocular surface neurogenic inflammation, leading to the vicious cycle of dry eye disease. These processes include the regular release of neuromediators during the conduction of ocular surface sensations, hyperosmolarity-induced ocular surface neuronal damage, neuro-regenerative activities, and neuronal-mediated dendritic cell activities. Neurogenic inflammation appears to be the main culprit, instigating the self-perpetuating inflammation o

Re-sensitization of neuropeptide receptors: should we stop the recycling?

Neurogenic inflammation, an important component of many disease states, is mediated by the release of neuropeptides from sensory nerves. To date, it has been possible to inhibit neurogenic inflammation using neuropeptide receptor blockers or by prevention of neuropeptide release. In the current edition of the British Journal of Pharmacology, Cattaruzza and co-workers discuss a novel way of blocking the action of neuropeptides. They have shown that the re-sensitization of the substance P neurokinin-1 receptor and the substance P-induced pro-inflammatory effects are mediated by the enzyme, endot

Treatment resistance in inclusion body myositis: the role of mast cells.

Inclusion body myositis is the commonest acquired myopathy in those over 50 years of age. Although it is classified as an idiopathic inflammatory myopathy and the most frequent finding on muscle biopsy in inclusion body myositis is an endomysial inflammatory infiltrate, it is clinically distinct from other myositis, including a lack of response to immunosuppressive medication. Neurogenic changes are commonly reported in inclusion body myositis and inflammatory changes are observed in muscle following neurogenic injury. The objective of our study was to explore whether neurogenic inflammation p

Neurogenic vasodilatation and plasma leakage in the skin.

1. Primary afferent nerve fibers control cutaneous blood flow and vascular permeability by releasing vasoactive peptides. These vascular reactions and the additional recruitment of leukocytes are commonly embodied in the term neurogenic inflammation. 2. Calcitonin gene-related peptide (CGRP) acting via CGRP1 receptors is the principal transmitter of neurogenic dilatation of arterioles whereas substance P (SP) and neurokinin A (NKA) acting via NK1 receptors mediate the increase in venular permeability. 3. Neurogenic vasodilatation and plasma protein leakage play a role in inflammation because m

Diabetes mellitus prevents capsaicin from inducing hyperaemia in the rat sciatic nerve.

Loss of neurogenic inflammation in response to tissue injury may be an important complication of diabetes mellitus. We studied local neurogenic inflammation in the peripheral nerve trunk of Sprague-Dawley rats 4 months following the induction of diabetes by streptozotocin injection. To assess neurogenic inflammation, the epineurial plexus of the sciatic nerve was exposed to topical capsaicin, an agent that releases vasoactive neuropeptides from perivascular afferent terminals. Under normal circumstances, local vasodilation results in endoneurial hyperaemia or a 'flare'. We evaluated the influe

Functional Upregulation of TRPM3 Channels Contributes to Acute Pancreatitis-associated Pain and Inflammation.

Transient receptor potential melastatin M3 (TRPM3) channels have been recognized as a pain transducer in dorsal root ganglion (DRG) neurons in recent years. TRPM3 activation initiates neurogenic inflammation and is required for the development of inflammatory hyperalgesia. We aimed to evaluate the role of TRPM3 in pancreas sensory afferents in pancreatic nociception, neurogenic inflammation, and acute pancreatitis (AP)-associated pain. AP was induced by intraperitoneal (i.p.) injection of L-arginine in rats. TRPM3 expression in pancreatic DRG neurons, spontaneous or mechanical-stimulation-evok

[Sensory and inflammatory reaction mechanisms in the dental pulp].

Laser-Doppler-flowmetry is used to show whether electrical stimulation of the rats tooth pulp leads to reactions such as neurogenic inflammations. Following electrical stimulation, a flow reduction with a subsequent flow increase can be observed After experimental elimination it can be shown that both sympathetic and nociceptive fibres are involved in this reaction. Thus the neurogenic inflammation of the dental pulp is a mechanism allowing early and localised reactions to noxious stimuli.

Substance P release from rat dura mater is inversely correlated with CGRP release- experiments using glycerol trinitrate and anti-CGRP antibodies.

BACKGROUND: The neuropeptides calcitonin gene-related peptide (CGRP) and substance P are important mediators of neurogenic inflammation when they are released from activated primary nociceptive afferents. It is long evident that neuropeptides play an important role in migraine pathophysiology, but the significance of neurogenic inflammation is still debated. METHODS: In an approved hemisected rodent head preparation, we measured CGRP release from the cranial dura mater in parallel with substance P release using animals pre-treated with anti-CGRP antibodies or control solutions. RESULTS: Apar

Bronchial asthma as neurogenic paroxysmal inflammatory disease: do some antiepileptic drugs have antiasthmatic properties?

Bronchial asthma is an inflammatory disease, and neurogenic inflammation may play important role in asthma mechanisms. But asthma also is an inflammatory disease with paroxysmal clinical picture. It is known that migraine and trigeminal neuralgia also are inflammatory diseases with paroxysmal clinical picture, and neurogenic inflammation plays important role in their mechanisms. Antiepileptic drugs are highly effective in pharmacotherapy of migraine and trigeminal neuralgia, and these drugs completely prevent attacks of migraine and trigeminal pain in great majority of cases. Due to this it is

Updates in the Treatment of Rosacea with γ-Aminobutyric Acid Derivatives.

Neurogenic inflammation plays an important role in the pathogenesis of rosacea. Recently, several clinical studies have found that γ-aminobutyric acid (GABA) derivatives can effectively improve the erythema and burning symptoms of rosacea. The underlying mechanisms may relate to the ability of GABA derivatives to inhibit the transient receptor potential channel pathway, subsequently reducing abnormal neuronal activation, inhibiting neuropeptide release, and suppressing neurogenic inflammation. This paper aimed to comprehensively review the progress of fundamental and clinical research on the r

Electroacupuncture at Neurogenic Spots in Referred Pain Areas Attenuates Hepatic Damages in Bile Duct-Ligated Rats.

Visceral pain frequently produces referred pain at somatic sites due to the convergence of somatic and visceral afferents. In skin overlying the referred pain, neurogenic spots characterized by hyperalgesia, tenderness and neurogenic inflammation are found. We investigated whether neurogenic inflammatory spots function as acupoints in the rat model of bile duct ligation-induced liver injury. The majority of neurogenic spots were found in the dorsal trunk overlying the referred pain and matched with locations of acupoints. The spots, as well as acupoints, showed high electrical conductance and

Molecular mechanisms of pain in acute pancreatitis: recent basic research advances and therapeutic implications.

Although severe abdominal pain is the main symptom of acute pancreatitis, its mechanisms are poorly understood. An emerging body of literature evidence indicates that neurogenic inflammation might play a major role in modulating the perception of pain from the pancreas. Neurogenic inflammation is the result of a crosstalk between injured pancreatic tissue and activated neurons, which leads to an auto-amplification loop between inflammation and pain during the progression of acute pancreatitis. In this review, we summarize recent findings on the role of neuropeptides, ion channels, and the endo

In Situ Hybridisation Study of Neuronal Neuropeptides Expression in Models of Mandibular Denervation with or without Inflammation: Injury Dependant Neuropeptide Plasticity.

UNLABELLED: Neuronal expression of neuropeptides is altered following peripheral tissue injury associated with inflammation or nerve injury. This results in neuropathic pain with or without neurogenic inflammation which is a major health problem regularly seen in trigeminal neuralgia. Activation of the trigeminal system results in the release of vasoactive neuropeptides substance P and Calcitonin Gene-related Peptide (CGRP) which contribute to nociception, pain and neurogenic inflammation in injured tissues. AIM: To study the alterations in the neuronal neuropeptides expressions in models of

[Innervation of the airways in asthma bronchiale and chronic obstructive pulmonary disease (COPD)].

Airway nerves have the capacity to control airway functions via neuronal reflexes and through neuromediators and neuropeptides. Neuronal mechanisms are known to play a key role in the initiation and modulation of airway hyperresponsiveness and inflammation. Therefore, the nerve fibres may contribute to airway narrowing in asthma and COPD. In addition to the traditional transmitters such as norepinephrine in postganglionic sympathetic nerve fibres and acetylcholine in parasympathetic nerve fibres, a large number of neuropeptides have been identified to have different pharmacological effects on

Dental pulp neuropathophysiology.

Mechanisms of pulpal pathophysiology are complex and the low compliance environment in which the dental pulp is allocated, further enhances the complexity of this process. Although it is known that it involves the interaction of the immune cells, pulpal cells, cytokines, chemokines and multiple neuropeptides but still there are many gaps in our current knowledge. The understanding of the biochemical and molecular pathways involved in the pulpal inflammation is important so that it can be used clinically to keep the dental pulp vital and healthy. It may thus provide an opportunity to develop po

Astrocyte-Microglia Crosstalk: A Novel Target for the Treatment of Migraine.

Migraine is a pervasive neurologic disease closely related to neurogenic inflammation. The astrocytes and microglia in the central nervous system are vital in inducing neurogenic inflammation in migraine. Recently, it has been found that there may be a crosstalk phenomenon between microglia and astrocytes, which plays a crucial part in the pathology and treatment of Alzheimer's disease and other central nervous system diseases closely related to inflammation, thus becoming a novel hotspot in neuroimmune research. However, the role of the crosstalk between microglia and astrocytes in the pathog

Naltrexone attenuates airway remodeling by modulating neuromediators and sensory ion channels in cigarette smoke-induced COPD pathophysiology.

Chronic obstructive pulmonary disease (COPD) accompanied by inflammation contributes to airway obstruction, mucus hypersecretion, and other characteristic symptoms. The airways densely innervated by subsets of nerve fibers highlight the critical role of neuroimmune interactions in disease pathogenesis. The present study aimed to investigate the role of neurogenic inflammation in COPD and evaluate the therapeutic potential of naltrexone (NTX) at ultra-low doses (10  μg/kg 1  μg/kg 0.1  μg/kg) in modulating neuroimmune signaling in cigarette smoke (CS) induced COPD.
