Impairment of the GABAergic system in the anterior insular cortex of heroin-addicted males.

Opioid addiction is a global problem, causing the greatest health burden among drug use disorders, with opioid overdose deaths topping the statistics of fatal overdoses. The multifunctional anterior insular cortex (AIC) is involved in inhibitory control, which is severely impaired in opioid addiction. GABAergic interneurons shape the output of the AIC, where abnormalities have been reported in individuals addicted to opioids. In these neurons, glutamate decarboxylase (GAD) with its isoforms GAD 65 and 67 is a key enzyme in the synthesis of GABA, and research data point to a dysregulation of GA

Constant neurofibrillary changes in the neocortex in progressive supranuclear palsy. Basic differences with Alzheimer's disease and aging.

Neocortical neurofibrillary tangles (NFT) revealed by Bodian technique and anti-tau immunolabelling were seen in 5/5 cases of progressive supranuclear palsy (PSP) aged 58-76 years. These lesions differed from Alzheimer's disease or age-related changes: (1) they were most frequent in the precentral gyrus (Brodmann's area 4) whereas associative areas are predominantly lesioned in Alzheimer's disease; (2) they affected mainly large pyramidal neurons and small cells, relatively sparing the cell population selectively involved in Alzheimer's disease; (3) they predominated in layers V and VI of area

Early axonal lesion and preserved microvasculature in epilepsy-induced hypermetabolic necrosis of the substantia nigra.

The time course of structural change in epilepsy-induced necrosis of the substantia nigra was studied by light and electron microscopy, and was correlated with previous metabolic studies. By light microscopy, tinctorial pallor appeared early, followed by pan-necrosis and macrophage infiltration. Mild lesions showed neuropil vacuolation but sparing of neurons, rather than a selective neuronal vulnerability. Electron microscopy of the evolving necrosis revealed an orderly sequence of structural damage involving first axons, then dendrites, neurons, and glia. No necrotic endothelial cells could b

Effects of intragastric water infusion and gastric distension on hypothalamic neuronal activity.

The effects of gastric water infusion and distension were examined in neurons from various parts of the rat brain. Neurons in the lateral preoptic-lateral hypothalamic-medial forebrain bundle (LPA-LH-MFB) neuropil were sensitive to gastric water infusion and distension. Cells randomly selected and examined in other brain areas were less sensitive to the same stimulation which indicated that the effects were relatively specific. The results, in terms of changes in neuronal discharge frequency from an established baseline, indicate that many cells in the lateral preoptic-lateral hypothalamic-med

A group of related surface glycoproteins distinguish sets and subsets of sensory afferents in the leech nervous system.

The distribution of 4 surface glycoproteins on axons of peripheral neurons was studied in the leech Hirudo medicinalis through monoclonal antibodies. All 4 glycoproteins have a similar molecular weight of 130 kDa. Immunohistochemical localization of these glycoproteins on tissue sections of nerves and neuropil reveals tracts of afferent axons organized as nested sets. Their distribution suggests a possible role for these molecules in mediating axon fasciculation.

Sarcocystosis in an aborted bovine fetus.

Sarcocystosis was diagnosed in an aborted bovine fetus. Immature and mature schizonts of Sarcocystis were disseminated in the vascular endothelium of all organs, but especially the brain. Microscopic granulomas, focal gliosis, and petechial hemorrhages in the neuropil were scattered in the brain. Multifocal collections of mononuclear cells were observed in the kidney, liver and heart. Organisms in sections of frozen tissues were demonstrated by immunofluorescent techniques to be Sarcocystis.

Aging, the brain and human prion disease.

Human prion diseases (PrD) preferentially manifest in the elderly. Their neuropathology may coexist with tau immunoreactive neuropil threads, neurofibrillary tangles, and beta-amyloid senile plaques, most likely representing an age-related change rather than a pathogenic link with Alzheimer's disease. Cerebrovascular disease with brain infarction, another malady preferring the elderly, is useful to prove the origin of PrD-associated prion protein deposition exclusively from neurons.

The brain and the corresponding sense organs in calanoid copepods - Evidence of vestiges of compound eyes.

Copepoda is one of the crustacean taxa with still unresolved phylogenetic relationships within Tetraconata. Recent phylogenomic studies place them close to Malacostraca and Cirripedia. Little is known about the morphological details of the copepod nervous system, and the available data are sometimes contradictory. We investigated several representatives of the subgroup Calanoida using immunohistochemical labeling against alpha-tubulin and various neuroactive substances, combining this with confocal laser scanning analysis and 3D reconstruction. Our results show that the studied copepods exhibi

Experimental chronic wasting disease (CWD) in the ferret.

Chronic wasting disease (CWD), a prion disease of North American deer, elk and moose, affects both free-ranging and captive cervids. The potential host range for CWD remains uncertain. The susceptibility of the ferret to CWD was examined experimentally by administering infectious brain material by the intracerebral (IC) or oral (PO) route. Between 15 and 20 months after IC inoculation, ferrets developed neurological signs consistent with prion disease, including polyphagia, somnolence, piloerection, lordosis and ataxia. Upon first sub-passage of ferret-adapted CWD, the incubation period decrea

Glutamate-like immunoreactivity marks compartments of the mushroom bodies in the brain of the cricket.

In the mushroom bodies of the brain of the cricket Gryllus bimaculatus, the distribution of glutamate-like immunoreactivity is shown by using several immunocytochemical staining protocols and confocal and conventional microscopy. Glutamate-like staining of intrinsic cells of mushroom bodies (Kenyon cells), their axons and projections, is demonstrated for the first time. Two types of Kenyon cells constituting distinct, separated populations within the perikaryal layer and in prominent neuropilar subcompartments exhibit strong (type III cells) or medium (type II cells) glutamate-like immunoreact

Intracisternal enzyme replacement therapy in lysosomal storage diseases: routes of absorption into brain.

AIMS: The research concerns enzyme replacement therapy in lysosomal storage diseases with central nervous system involvement. The principle aim was to understand the routes of entry of enzyme into the brain when delivered directly into the cerebrospinal fluid (CSF) via the cerebellomedullary cistern. METHODS: Pathways for absorption of replacement enzyme were investigated in dogs with mucopolysaccharidosis IIIA (MPSIIIA) following intracisternal injections of human recombinant N-sulphoglucosamine sulphohydrolase (rhSGSH, EC3.10.1.1) by light and confocal microscopy using chromogenic and fluor

Brain development in the yellow fever mosquito Aedes aegypti: a comparative immunocytochemical analysis using cross-reacting antibodies from Drosophila melanogaster.

Considerable effort has been directed towards understanding the organization and function of peripheral and central nervous system of disease vector mosquitoes such as Aedes aegypti. To date, all of these investigations have been carried out on adults but none of the studies addressed the development of the nervous system during the larval and pupal stages in mosquitoes. Here, we first screen a set of 30 antibodies, which have been used to study brain development in Drosophila, and identify 13 of them cross-reacting and labeling epitopes in the developing brain of Aedes. We then use the identi

MRI volumes of amygdala and hippocampus in non-mentally retarded autistic adolescents and adults.

OBJECTIVE: To determine whether volumes of hippocampus and amygdala are abnormal in people with autism. BACKGROUND: Neuropathologic studies of the limbic system in autism have found decreased neuronal size, increased neuronal packing density, and decreased complexity of dendritic arbors in hippocampus, amygdala, and other limbic structures. These findings are suggestive of a developmental curtailment in the maturation of the neurons and neuropil. METHODS: Measurement of hippocampus, amygdala, and total brain volumes from 1.5-mm coronal, spoiled gradient-recalled echo MRI scans in 14 non-ment

Ultrastructural organisation of the projection from the superior colliculus to the ventral lateral geniculate nucleus of the rat.

Retinorecipient regions of the ventral lateral geniculate nucleus of the thalamus and the superior colliculus of the midbrain are linked by reciprocal axonal projections. In this study we have investigated the ultrastructural characteristics, the distribution, and the postsynaptic targets of the terminals of axons projecting to the ventral lateral geniculate nucleus from the superior colliculus. Horseradish peroxidase was injected into the superior colliculi of adult albino rats, and the Hanker-Yates method was used to visualize anterogradely and retrogradely transported peroxidase in the vent

Clinical, biochemical, neuropathological and molecular findings of the first Polish case of adenylosuccinase deficiency.

Adenylosuccinase (ADSL) deficiency is an autosomal recessive disorder affecting mainly the nervous system. The disease causes psychomotor retardation, frequently with autistic features and epilepsy. ADSL deficiency may be diagnosed by detection of two abnormal metabolites in body fluids--succinyladenosine (S-Ado) and succinylaminoimidazole carboxamide riboside (SAICAr). It is assumed that the former metabolite is neurotoxic. We present clinical, biochemical and neuropathological findings of a child affected by a severe form of ADSL deficiency. She had progressive neurological symptoms that sta

[Pathogenesis and prevention of Alzheimer's disease: when and in what way does the pathological process begin?].

Abnormal tau lesions (e.g., pretangles, neuropil threads, and neurofibrillary tangles) that develop in a few types of nerve cells in the central nervous system are essential to the pathogenesis of Alzheimer's disease. Pretangles begin to occur in puberty and even during early childhood in the locus coeruleus. Evolutionally speaking, the propensity to develop tau lesions may be related to late developing and maturing nerve cell types in the human brain.

Characterization and tissue distribution of neuropeptide F in the eyestalk and brain of the male giant freshwater prawn, Macrobrachium rosenbergii.

We previously analyzed the central nervous system (CNS) transcriptome and found three isotypes of long neuropeptide F (MrNPF-I, -II, -III) and four isoforms of short NPF (sMrNPF) in the giant freshwater prawn, Macrobrachium rosenbergii. We now validate the complete sequences of the MrNPF-I and -II precursor proteins, which show high similarity (91-95 %) to NPFs of the penaeus shrimp (PsNPF). MrNPF-I and -II precursors share 71 % amino acid identity, whereas the mature 32-amino-acid MrNPF-I and 69-amino-acid MrNPF-II are identical, except for a 37-amino-acid insert within the middle part of the

Acute Wernicke's encephalopathy: comparison of magnetic resonance images and autopsy findings.

This is the first report of acute Wernicke's encephalopathy in which characteristic magnetic resonance (MR) findings have been verified by postmortem histopathological examination. The high-signal areas surrounding the third and the fourth ventricles and the aqueduct on the T2-weighted images reflected the spongy disintegration of the neuropil, which is the typical pathological finding in Wernicke's encephalopathy. We confirmed that MR imaging is essential to the early diagnosis of Wernicke's encephalopathy.

Population morphology implies a common developmental blueprint for Drosophila motion detectors.

T4 and T5 neurons are the first direction-selective neurons in the visual pathway. They are the most numerous cell types in the fly brain (~6000 within each optic lobe) and, as a population, their compact dendritic arbours span the entire visual field. They are classified into four subtypes (a, b, c, and d). Each subtype encodes one of four orthogonal motion directions (up, down, forwards, backwards). Crucially, the dendrites of these neurons are oriented inversely to the functional direction of motion which they encode. This dendritic orientation is what ultimately determines their functional

Comparative Neuroanatomy of Hydrothermal Vent Shrimps: Ecological Differentiation Among Four Alvinocaridid Species.

Alvinocaridid shrimps colonize hydrothermal vent fields along the Mid-Atlantic Ridge (MAR), where they experience extreme environmental conditions including high hydrostatic pressure, steep thermal and chemical gradients, and dim light. While various aspects of the biology of the dominant vent shrimp Rimicaris exoculata, such as general anatomy, microhabitat, symbiotic relationships, or trophic networks, have been extensively studied, little is known about how brain organization varies among alvinocaridid species occupying different ecological niches. Here, we investigated whether vent shrimps
