Neuroimmune Cross Talk in the Gut. Neuroendocrine and neuroimmune pathways contribute to the pathophysiology of irritable bowel syndrome.

Irritable bowel syndrome (IBS) is a common disorder characterized by recurrent abdominal pain, bloating, and disturbed bowel habit, symptoms that impact the quality of life of sufferers. The pathophysiological changes underlying this multifactorial condition are complex and include increased sensitivity to luminal and mucosal factors, resulting in altered colonic transit and visceral pain. Moreover, dysfunctional communication in the bidirectional signaling axis between the brain and the gut, which involves efferent and afferent branches of the peripheral nervous system, circulating endocrine

Association of sympathetic nervous system activity with polycystic ovarian syndrome.

Polycystic ovary syndrome (PCOS) is a disease with high prevalence which has various clinical manifestations and increased risk of long-term complications, such as dyslipidemia, diabetes, metabolic syndrome, and hormone related tumor. However, the etiology of PCOS is still unclear. Consequently, the effect of symptomatic treatment is not always satisfactory and the prognosis is also unpredictable. Currently, commonly psychological syndromes and imbalance of sympathetic neuroendocrine system have been found in PCOS population, and increasing evidence highlighted the hypothesis that characterist

Neuroendocrine abnormalities associated with HIV infection.

Functional derangement of every endocrine organ system has been reported in association with HIV infection. The changes in endocrine function may be related to the viral infection of the gland, to systemic effects of HIV or an opportunistic infection, to infiltration by a neoplasm such as Kaposi's sarcoma, to a complication of treatment, or generation of cytokines. A wide spectrum of endocrine abnormalities is observed in HIV-infected patients. Some of these abnormalities are similar to those seen in other systemic illness, whereas others are unique to HIV infection. The clinical significance

Development of the neurohypophysis: A major neuroendocrine interface.

The neurohypophysis is a major central neuroendocrine interface regulating reproductive functions and water homeostasis. Distinct neurovascular cell types interact via evolutionarily conserved signaling molecules in the developing neurohypophysis, providing a model system for studying principles in neuroendocrine interface morphogenesis. This review provides an overview of neurohypophysis development with a focus on paracrine signaling and the intrinsic mechanisms that regulate the major cell types and neurovascular interface development.

Neuroendocrine control of glucose homeostasis: integrative mechanisms from the hypothalamus to the brainstem.

Glucose is vital for brain physiological function, acting as the primary energy source that supports cognitive processes, neurotransmitter production, and overall health. The brain requires a constant supply of glucose, and the body has evolved protective mechanisms to maintain this supply during hypoglycemia. Increased appetite and food intake is a fundamental protective response. The precise network of brain regions, nerves, and connections responsible for initiating and coordinating these responses has not been fully identified or mapped. Neuroendocrine centers within the hypothalamus and b

Melatonin as the Cornerstone of Neuroimmunoendocrinology.

Much attention has been recently drawn to studying melatonin - a hormone whose synthesis was first found in the epiphysis (pineal gland). This interest can be due to discovering the role of melatonin in numerous physiological processes. It was the discovery of melatonin synthesis in endocrine organs (pineal gland), neural structures (Purkinje cells in the cerebellum, retinal photoreceptors), and immunocompetent cells (T lymphocytes, NK cells, mast cells) that triggered the evolution of new approaches to the unifield signal regulation of homeostasis, which, at the turn of the 21st century, lead

Stress and food allergy: mechanistic considerations.

Recent years have seen a marked increase in food allergy prevalence among children, particularly in Western countries, that cannot be explained by genetic factors alone. This has resulted in an increased effort to identify environmental risk factors underlying food allergies and to understand how these factors may be modified through interventions. Food allergy is an immune-mediated adverse reaction to food. Consequently, considerations of candidate risk factors have begun to focus on environmental influences that perturb the healthy development of the emerging immune system during critical pe

Two dopamine receptors: biochemistry, physiology and pharmacology.

In 1979, two categories of dopamine (DA) receptors (designated as D-1 and D-2) were identified on the basis of the ability of a limited number of agonists and antagonists to discriminate between these two entities. In the past 5 years agonists and antagonists selective for each category of receptor have been identified. Using these selective drugs it has been possible to attribute the effects of DA upon physiological and biochemical processes to the stimulation of either a D-1 or a D-2 receptor. Thus, DA-induced enhancement of both hormone release from bovine parathyroid gland and firing of ne

Hormonal and Behavioral Consequences of Social Isolation and Loneliness: Neuroendocrine Mechanisms and Clinical Implications.

Social isolation and loneliness represent critical psychosocial stressors associated with profound hormonal dysregulation and adverse behavioral outcomes. This review synthesizes current evidence on neuroendocrine mechanisms linking perceived and objective social disconnection to health consequences, emphasizing hypothalamic-pituitary-adrenal axis dysfunction, altered glucocorticoid signaling, and inflammatory pathways. Loneliness activates conserved transcriptional responses with upregulated proinflammatory gene expression and downregulated antiviral responses, mediated through sustained cort

Bridging the Nervous-Endocrine System and Immune Response, in Human Chagas Disease Pathology.

<p>Background: Chronic Chagas disease can affect multiple organs, most notably the heart and gastrointestinal tract, and in some cases, the nervous system. However, the underlying pathophysiological mechanisms of this parasitic infection remain incompletely understood. Summary: Evidence from studies in both mice with acute Trypanosoma cruzi (T. cruzi) infection and in patients with Chagas disease has revealed a range of immune-neuroendocrine alterations and metabolic disruptions. In this review, we highlight key findings in human Chagas disease related to these abnormalities and discuss their

[Perinatal stress in brain programming and pathogenesis of psychoneurological disorders].

We analyzed data from our studies and other author's works concerning the influence of stress stimuli on the development of neuroendocrine and immune systems. The results of our analysis suggest that "immunological stress" induced by viral or bacterial infection, an unhealthy diet, and the mother's behavior during pregnancy and lactation are significant risk factors for the developing offspring. The brain is the key target for these factors. The continuous effect of stress stimuli during critical periods of brain development induces hyperactivity and increased response of hypothalamic-pituitar

Conversation galante: how the immune and the neuroendocrine systems talk to each other.

The generation of endogenous adjuvants and the clearance of apoptotic cells occur at the intersection between the neuroendocrine and the immune systems. Recent data suggest that autoimmunity associates with a communication breakdown between the two systems and that events taking place in lymphoid organs and in peripheral inflamed tissues shape the response to tissue damage. Autonomic nerve endings release norepinephrine and acetylcholine, whereas sensitive fibers release neuropeptides. Moreover, nervous endings in the tissues control the secretory activity of neuroendocrine cells, which are di

The possible role of the gut neuroendocrine system in diabetes gastroenteropathy.

Gastrointestinal symptoms such as nausea and vomiting, heartburn, abdominal pain, diarrhoea, constipation and faecal incontinence are common in patients with diabetes. Diabetes gastroenteropathy is a clinically relevant problem. In addition to the increased morbidity it causes, it results in severely impaired metabolic control, which in turn increases the risk of hyper-/hypoglycaemia. Moreover, the poorly controlled blood glucose level increases the risk of secondary diabetes complications, namely, retinopathy, nephropathy, neuropathy and cardiovascular affection. Gastrointestinal symptoms may

Chemokines and their receptors in the central nervous system.

Chemokines are a family of proteins associated with the trafficking of leukocytes in physiological immune surveillance and inflammatory cell recruitment in host defence. They are classified into four classes based on the positions of key cystiene residues: C, CC, CXC, and CX3C. Chemokines act through both specific and shared receptors that all belong to the superfamily of G-protein-coupled receptors. Besides their well-established role in the immune system, several recent reports have demonstrated that these proteins also play a role in the central nervous system (CNS). In the CNS, chemokines

Neuroendocrine abnormalities in patients with traumatic brain injury.

This article provides an overview of hypothalamic and pituitary alterations in brain trauma, including the incidence of hypothalamic-pituitary damage, injury mechanisms, features of the hypothalamic-pituitary defects, and major hypothalamic-pituitary disturbances in brain trauma. While hypothalamic-pituitary lesions have been commonly described at postmortem examination, only a limited number of clinical cases of traumatic hypothalamic-pituitary dysfunction have been reported, probably because head injury of sufficient severity to cause hypothalamic and pituitary damage usually leads to early

The hypothalamic magnocellular system in the domestic fowl. Study on semithin sections.

Neuronal characteristics and location of the neurosecretory, magnocellular, fuchsin-paraldehyde-positive (FA+) system of the fowl are described at the light-microscopic level on serial semithin sections. Three nuclei make up this system, the nucleus supraopticus, n. magnocellularis interstitialis and n. paraventricularis. These nuclei display magnocellular neurons, not showing a parvocellular component. The neurons of the three nuclei showed a scattered pattern of distribution and a dense surrounding neuropil. Groups formed by magnocellular neurons were found in the three nuclei and groups for

Distribution and functional significance of Met-enkephalin-Arg6-Phe7- and Met-enkephalin-Arg6-Gly7-Leu8-like peptides in the blowfly Calliphora vomitoria. I. Immunocytochemical mapping of neuronal pathways in the brain.

Neuronal pathways immunoreactive to antisera against the extended-enkephalins, Met-enkephalin-Arg6-Phe7 (Met-7) and Met-enkephalin-Arg6-Gly7-Leu8 (Met-8), have been identified in the brain of the blowfly Calliphora vomitoria. Co-localisation with other enkephalins in certain neurons suggests that a precursor similar to preproenkephalin A exists in insects and that differential enzymatic processing occurs as in vertebrates. Co-localisations of the extended-enkephalin-like peptides with other vertebrate-type peptides, including cholecystokinin and pancreatic polypeptide, also occur. The enkephal
