New surgical paradigm for open neural tube defects.

INTRODUCTION: An open neural tube defect (ONTD) features an exposed, unclosed neural plate in the form of an expanded, flat, and frequently hefty neural placode. Traditional philosophy of ONTD repair aims at preserving function at any cost, which often means stuffing the entire thick and unwieldy but non-functional placode into a tight dural sac, increasing the likelihood of future tethering of the spinal cord. The same philosophy of attempting to save the whole perimetry of the placode also sometimes leads to inadvertent inclusion of parts of the squamous epithelial membrane surrounding the p

[Pathogenesis and genetics of neural tube defects].

After congenital heart defects neural tube defects (NTDs) is the second most prevalent congenital malformation among birth defects. The average rate of isolated NTDs is 1.4-2.0 per 1000 live births worldwide. The etiology of isolated (nonsyndromic) NTDs is believed to be the result of a combination of genetic predisposition and environmental factors. Over 80 genes believed to be engaged in the neurulations have been identified during the investigation and research of the mouse models. Despite exhaustive research efforts, now spanning several decades, little is known about the actual genetic me

The ontogenesis of cranial neuromeres in the rat embryo. II. A transmission electron microscope study.

The morphogenesis of rhombomeres (neuromeres) caudal to the preotic sulcus during neurulation in rat embryos is described. A model is proposed to explain the development of the characteristic neuromeric sulci and interneuromeric gyri based on the cytoskeletal elements and the kinetic behaviour of the neural epithelium. Evidence obtained from a study of control, cytochalasin D-treated and colchicine-treated embryos, at the electron microscopic level, supports the proposed model. The longitudinally expanding cranial neural epithelium bulges between microtubule blocks present within the interneur

The role of chrysanthemum phytochemicals in neural tube development: a narrative review of underlying mechanisms.

BACKGROUND: Neural tube defects (NTDs) have long been attributed to genetic and hereditary factors, but emerging evidence highlights the significant role of pharmacological and environmental agents. The global integration of traditional medicine systems, including Traditional Chinese Medicine, has introduced diverse bioactive compounds with potential to influence neurodevelopmental processes. PURPOSE: This narrative review aims to comprehensively summarize and critically evaluate the current evidence on the bioactivity of chrysanthemum (Chrysanthemum morifolium and related species) extracts a

Retinoid signaling is required to complete the vertebrate cardiac left/right asymmetry pathway.

Vitamin A-deficient (VAD) quail embryos have severe abnormalities, including a high incidence of reversed cardiac situs. Using this model we examined in vivo the physiological function of vitamin A in the left/right (L/R) cardiac asymmetry pathway. Molecular analysis reveals the expression of early asymmetry genes activin receptor IIa, sonic hedgehog, Caronte, Lefty-1, and Fgf8 to be unaffected by the lack of retinoids, while expression of the downstream genes nodal-related, snail-related (cSnR), and Pitx2 is altered. In VAD embryos nodal expression in left lateral plate mesoderm (LPM) is seve

Histone H2A deubiquitinase BAP1 is required for human neuronal progenitor cell formation.

Formation of neuronal progenitor cells, neural crest cells and surface ectoderm cells is a complex process and differential gene expression allows for generation of these three cell types during neurulation process. Histone modifiers such as writers and erasers may play an important role in achieving the differential gene expression during neurulation event, similar to how DNMTs and TET enzymes function in germ cell development. We had previously reported that Polycomb group (PcG) protein RING1B which catalysed repressive H2AK119ub1 mark and prevented precocious activation of neuronal genes in

Polystyrene nanoplastics exposure caused defective neural tube morphogenesis through caveolae-mediated endocytosis and faulty apoptosis.

Growing evidence demonstrated that bioaccumulation of polystyrene nanoplastics (PS-NPs) in various organisms including human beings caused destructive effects on health. Nanoplastics may adversely affect fetal development potentially since they can pass through the placental barrier. However, very little has been known about the embryonic toxicity of polystyrene nanoplastics, especially in embryonic neurulation, the early developmental stage of the fetus, as well as the corresponding mechanisms. In this study, we first observed that 60- or 900-nm PS-NPs (especially 60-nm PS-NPs) could cross mo

Bioelectric signalling via potassium channels: a mechanism for craniofacial dysmorphogenesis in KCNJ2-associated Andersen-Tawil Syndrome.

KEY POINTS: Xenopus laevis craniofacial development is a good system for the study of Andersen-Tawil Syndrome (ATS)-associated craniofacial anomalies (CFAs) because (1) Kcnj2 is expressed in the nascent face; (2) molecular-genetic and biophysical techniques are available for the study of ion-dependent signalling during craniofacial morphogenesis; (3) as in humans, expression of variant Kcnj2 forms in embryos causes a muscle phenotype; and (4) variant forms of Kcnj2 found in human patients, when injected into frog embryos, cause CFAs in the same cell lineages. Forced expression of WT or variant

Tethered spinal cord related to caudal spinal dysraphism in a tailless Holstein calf.

A rare dysraphic caudal spinal anomaly, or caudal agenesis, comprising a tethered spinal cord, was found in a tailless Holstein calf that presented ataxia and paresis with analgesia of the hind limbs. The gently and slimly tapered conus medullaris was poorly formed between S2 and S3 which indicated that it was lying more caudally. The caudal end of the filum terminale adhered to the inner periosteum of the vertebral arch at S4, which is compatible with tethering of the spinal cord. The dysraphic changes from the secondary neurulation error and the longitudinal deranged cord morphology that may

[Neural tube dysraphia at the level of the midbrain in alcoholic embryopathy].

Forty-four 5-12-week embryos obtained from mothers who abused alcohol during pregnancy were examined for brain development. Six cases of dysraphia of the neural tube at the midbrain level were revealed. In two cases, it was defined as primary. This was confirmed by the impairment of neurulation in the neural tube, by the growth of the ends of the neural plate, not yet grown together (hyperplasia), and derangement of the mesoderm (partial cranioschisis). The use of alcohol might be the most probable cause that gave rise to dysraphia. In these cases, the teratogenic termination period was referr

Retinoid receptors and vitamin A deficiency: differential patterns of transcription during early avian development and the rapid induction of RARs by retinoic acid.

The functional links of specific retinoid receptors to early developmental events in the avian embryo are not known. Before such studies are undertaken, knowledge is required of the spatiotemporal expression patterns of the receptor genes and their regulation by endogenous retinoic acid levels during the early stages of development. Here, we report the expression patterns of mRNAs for RARalpha, RARalpha2, RARbeta2, RARgamma, RARgamma2, RXRalpha, and RARgamma from neurulation to HH10 in the normal and vitamin A-deficient (VAD) quail embryo. The transcripts for all retinoid receptors are detecta

Congenital anomalies of the cervical spine.

A wide variety of congenital pathology affects the cervical spine and underlying spinal cord. This article discusses commonly encountered conditions under the categories of osseous anomalies, Chiari malformations, dysraphic conditions, and skeletal dysplasias. Osseous anomalies tend to involve the upper cervical spine to a greater degree than the lower cervical spine. Chiari malformations warrant inclusion on the basis that they have many manifestations within the cervical spine. Dysraphic conditions resulting from failure of either primary or secondary neurulation are distinctly uncommon in t

From conception to the child.

An attempt to present all the data that account for the progressive transformation of a fertilized zygote into a child at birth is obviously a herculean work. In this review, I focus on four aspects of central nervous system development in which our understanding has considerably been enhanced during the last century. The first part concerns the phenomenon of neural induction, a process devoted to the acquisition of neural identity. It was classically considered that active molecules were secreted by the organizer (the dorsal lip of the blastopore in amphibians and the node in amniotes) and th

Spinal congenital dermal sinus associated with upper thoracic meningocele. Case Report.

The congenital dermal sinus is an abnormal epithelium-lined sinus tract between the skin surface and deeper tissues. It occurs during neurulation when the neural groove closes to form the neural tube on Day 26 of gestation and results from a failure of neuroectoderm to separate from the cutaneous ectoderm. The most frequent location is the lumbosacral area; an upper thoracic location is quite rare. This 37-year-old man presented with headache and numbness in both arms. No specific neurological findings were observed. Physical examination revealed a dimple at T-2. Radiography and magnetic reson

Embryogenesis. Why do we need a new explanation for the emergence of spina bifida with lipoma?

Spina bifida with lipoma is a human malformation that most often affects the lumbosacral area. It is a complex morphological type. Its origin is controversial, and none of the previous hypotheses can be retained in view of the most recent advances in experimental embryogenesis. Contrary to earlier opinions, adipocytes cannot arise from meninges, vessels or glial cells and spina bifida cannot be explained by incarceration of mesodermal tissues during primary neurulation or developmental defect at the level of the tail bud. Spina bifida with lipoma, which actually involves all the derivatives of

Discovering the unexpected: Insights into the dynamics of mouse neural tube closure revealed by time-lapse imaging.

The use of time-lapse imaging to study neural tube closure in mouse embryos has provided unexpected insights into the complex morphogenetic processes involved. When neural tube closure is disrupted, it leads to neural tube defects (NTDs), which are among the most common structural birth defects in humans, associated with long-term disabilities and death. This review explores the growing body of research on time-lapse imaging experiments conducted in mice, emphasizing discoveries of the dynamic cellular movements and changes that enable neural tube formation. Advances in mouse embryo culture an

Mediolateral neural patterning during gastrulation precedes dorsoventral patterning in chick and frog.

INTRODUCTION: dorsoventral polarity of the neural tube is based on molecular patterning established during neurulation and is suggested to be induced by fine-tuned interpretation of opposing morphogen gradients in the neural tube: a ventral-to-dorsal gradient is formed by shh secreted from floor plate and notochord whereas a dorsal-to-ventral gradient is created by BMP and Wnt originating from the roof plate. Recent results revealed an unexpected early induction of Shh in the chick prospective floor plate prior to notochord formation. Hence, we addressed the questions as to whether key compone

Neural tube defects and sex ratios.

The sex ratio of 147 fetuses with presumed multifactorial neural tube defect (NTD) was studied. Overall, the ratio (males:females) was 0.73 with expected female excess. However, when the NTDs were subdivided according to the site of the lesion, the sex ratios varied. Total craniorachischisis, anencephaly with cervical spina bifida, holoacrania, and thoracic spina bifida showed a greater female excess than that overall; the sex ratio for meroacrania was close to unity, while that for low spinal lesions, particularly those involving the sacrum, showed an extreme bias towards males. These finding

Insights into metabolic mechanisms underlying folate-responsive neural tube defects: a minireview.

Neural tube defects (NTDs), including anencephaly and spina bifida, arise from the failure of neurulation during early embryonic development. Neural tube defects are common birth defects with a heterogenous and multifactorial etiology with interacting genetic and environmental risk factors. Although the mechanisms resulting in failure of neural tube closure are unknown, up to 70% of NTDs can be prevented by maternal folic acid supplementation. However, the metabolic mechanisms underlying the association between folic acid and NTD pathogenesis have not been identified. This review summarizes ou

Planar cell polarity and the pathogenesis of Tourette Disorder: New hypotheses and perspectives.

Planar cell polarity (PCP) signaling plays a fundamental role in shaping the development and ongoing function of the nervous system, beginning from early stages of neural tube closure and spanning the maintenance of functional synapses in adults. While mutations in core PCP signaling proteins have long been suspected to underlie neural tube closure defects in humans, recent findings also implicate their potential involvement in neurodevelopmental and neuropsychiatric disorders. Missense and loss-of-function mutations in CELSR3, a core component of PCP signaling complexes, are highly associated
