Lactococcus lactis is widely used as a starter in the manufacture of cheese and fermented milk. Its main role is the production of lactic acid, but also contributes to the sensory attributes of cheese. Unfortunately, the diversity of suitable strains to be commercialized as dairy starters is limited. In this work, we have applied adaptive evolution under cell envelope stress (AE-CES) as means to provide evolved L. lactis strains with distinct physiological and metabolic traits. A total of seven strains, three of industrial origin and four wild nisin Z-producing L. lactis, were exposed to subin

Listeria monocytogenes is a food-borne pathogen of concern in the traditional Norwegian fermented fish product known as rakfisk. This product is made from freshwater salmonid fish through mild-salting and maturation in brine at 3-7 °C for 3-12 months. L. monocytogenes can potentially grow during this manufacturing process if initial contamination occurs, and since the product is consumed without any heat treatment, it may cause outbreaks of listeriosis. One of the most severe outbreaks of listeriosis in Norway during recent years was caused by contaminated rakfisk. Consequently, rakfisk produc

CD80 is an important co-stimulatory molecule that participates in the immune response. Soluble CD80 can induce T cell activation and overcome PDL1-mediated immune suppression. In this study, we aimed to construct recombinant Lactococcus lactis for oral delivery of the soluble CD80 (hsCD80) protein or the fusion protein containing the cholera toxin B subunit (CTB) and hsCD80 (CTB-hsCD80) under the control of the nisin-inducible expression system. The recombinant L. lactis expressed and secreted hsCD80 or CTB-hsCD80 fusion proteins after induction by nisin in vitro and in the enteric cavity. Add

Lactococcus lactis has previously been proposed as a vaccine platform for the safe delivery of heterologous antigens. Here we utilized L. lactis as a live vector for expression of listeriolysin O (LLO), a major Listeria monocytogenes antigen and virulence factor. A variety of plasmid constructs were designed to permit either constitutive or nisin-inducible expression of secreted or non-secreted LLO in L. lactis. Recombinant strains were subsequently tested in a murine model for vaccination efficacy against L. monocytogenes infection. CD8(+) T lymphocytes specific for the LLO(91-99) epitope wer

The rise of antibiotic resistance, coupled with a dwindling antibiotic pipeline, presents a significant threat to public health. Consequently, there is an urgent need for novel therapeutics targeting antibiotic-resistant pathogens. Nisin, a promising peptide antibiotic, exhibits potent bactericidal activity through a mechanism distinct from that of clinically used antibiotics. However, its cationic nature leads to hemolysis and cytotoxicity, which has limited its clinical application. Here, nanodelivery systems have been developed by mimicking the mechanisms bacteriophages use to deliver their

Variants resistant to penicillin G (RvP), streptomycin (RvS), lincomycin (RvL) and rifampicin (RvR) were developed from a colistin-sensitive isolate of Pseudomonas fluorescens LRC-R73 (P. fluorescens). Cell fatty acid composition, K(+) efflux and sensitivity to antimicrobial peptides (nisin Z, pediocin PA-1/AcH and colistin) alone or combined with antibiotics were determined. P. fluorescens was highly sensitive to kanamycin, tetracycline and chloramphenicol at minimal inhibitory concentrations of 0.366, 0.305 and 0.732 μg/ml respectively. P. fluorescens, RvP, RvS, RvL and RvR were resistant to

Fresh pork remains susceptible to psychrotrophic spoilage during superchilled storage. Although oregano essential oil (OEO), nisin, and active absorbent pads have each been studied, their effects on spoilage-community proliferation and concurrent quality loss remain poorly resolved. Here, we coupled longitudinal 16S rRNA gene profiling with conventional microbiological and quality measurements to evaluate a poly(vinyl alcohol)/soy protein isolate pad containing OEO-loaded soluble soybean polysaccharide-nisin nanoparticles (PS-NPs). Pork was stored at -1 °C for 24 days without a pad (CK), with

OBJECTIVES: Oral health's inextricable links to systemic health are highlighted by the emerging oral-gut-brain axis and other well-known axes. There is growing evidence of a complex oral-gut-brain axis linking mouth and gut microbiomes with the central nervous system. Axis disruptions, characterized as oral and gut dysbiosis or microbial imbalances, can trigger oral and systemic inflammation and neuroinflammation, contributing to diseases such as Alzheimer's disease and Parkinson's disease.
MATERIALS AND METHODS: We summarize the oral-gut-brain axis mechanistic pathways, key evidence from hum

Homogenization is used widely in the dairy industry to improve product stability and quality. High-pressure homogenization (HPH) of fluid foods up to pressures of 300 MPa has demonstrated excellent potential for microbial inactivation. Microbial inactivation can be enhanced during HPH with the inclusion of antimicrobial compounds. Escherichia coli K-12 cells, grown statically or in chemostat, were exposed to HPH processing pressures of 50 to 350 MPa in the absence or presence of the antimicrobial nisin. Valve temperature was regulated by a water bath and pressure, and temperature data were rec

The demand for natural and multifunctional ingredients in food systems has increased interest in microbial biosurfactants as potential alternatives to synthetic emulsifiers and antimicrobial agents. However, their practical functionality in food systems remains insufficiently explored. Here, a biosurfactant produced by Bacillus velezensis GJ1 was evaluated as a functional emulsifier and antibacterial agent for food applications. Whole-genome sequencing confirmed the presence of lipopeptide biosynthetic gene clusters associated with surfactin and iturin production, while no genes related to kno

Bacteria-derived natural antimicrobial compounds such as bacteriocins, reruterin, and organic acids have recently received substantial attention as food preservatives or therapeutic alternatives in human or animal sectors. This study aimed to evaluate the antimicrobial activity of different bacteria-derived antimicrobials, alone or in combination, against a large panel of Gram-negative and Gram-positive bacteria. Bacteriocins, including microcin J25, pediocin PA-1, nisin Z, and reuterin, were investigated alone or in combination with lactic acid and citric acid, using a checkerboard assay. Con

The main cause of non-typhoidal Salmonella (NTS) infection in humans is ingestion of contaminated animal-derived foods such as eggs, poultry and dairy products. These infections highlight the need to develop new preservatives to increase food safety. Antimicrobial peptides (AMPs) have the potential to be further developed as food preservative agents and join nisin, the only AMP currently approved, for use as a preservative in food. Acidocin J1132β, a bacteriocin produced by probiotic Lactobacillus acidophilus, displays no toxicity to humans, however it exhibits only low and narrow-spectrum ant

The exchange of bacterial virulence factors driven by lateral gene transfer (LGT) can help indicate possible bacterial transmission among different hosts. Specifically, overlaying the phylogenetic signal of LGT among bacteria onto the distribution of respective isolation sources (hosts) can indicate patterns of transmission among these hosts. Here, we apply this approach towards a better understanding of patterns of bacterial transmission between humans and livestock. We utilize comparative genomics to trace patterns of LGT for an 11-gene operon responsible for the production of the antibiotic

A gene associated with lipopolysaccharide (LPS) transport was cloned from a local clinical Vibrio cholerae O1 strain of the Ogawa serotype by using the Lactococcus lactis nisin-controlled expression (NICE) system. The V. cholerae wzm gene, which codes for an integral membrane transporter protein, was expressed and targeted to the cytoplasmic membrane, and was crudely isolated through simple centrifugation and SDS solubilization. To examine seroreactivity of this construct, rabbits were orally fed with 10(9) cfu/ml of live, recombinant L. lactis carrying the wzm gene, induced with nisin prior t

UNLABELLED: Microorganisms exhibit remarkable adaptability to diverse environmental stresses through transcriptional reprogramming. Here, we focused on the MarR family regulator RmaH in Lactococcus lactis, which orchestrated a pivotal trade-off: enhancing tolerance to the bacteriocin nisin and the antibiotic vancomycin at the expense of acid resistance. RmaH directly activated genes for cell wall and membrane lipid biosynthesis, leading to a thicker cell wall, and repressed the arginine deiminase pathway, attenuating acid tolerance. Integrated transcriptomic, chromatin immunoprecipitation-sequ

Compound seasoning sauce is susceptible to microbial spoilage during storage, which can lead to package swelling, thereby posing a serious threat to both product shelf life and food safety. High-throughput sequencing, traditional culture isolation, and inhibition experiments were employed to systematically investigate the gas-producing spoilage bacteria in compound seasoning sauce and its control strategies. The results demonstrated that, compared with normal sauces, the swollen bagged samples exhibited a significant increase in TCA-soluble peptide and reducing sugars content, along with a sha

Concerns with fungal plant pathogen resistance to fungicides require sustainable alternatives. Model yeast lipid membranes were used to assess the role of ergosterol in the activity of antimicrobials known to affect biomembranes. Results showed that nystatin decreased yeast liposome stability as ergosterol content increased, whereas iturin effects were consistently high, but did not appear to be affected by ergosterol content. Nisin tended to decrease yeast liposome stability as ergosterol content increased, albeit much less than nystatin, and caused increased liposome size at lower ergosterol

BACKGROUND/OBJECTIVES: Listeria monocytogenes is a major foodborne pathogen responsible for listeriosis, a serious illness with high morbidity and mortality, particularly in vulnerable populations. Its persistence in food processing environments and resistance to conventional preservation methods pose significant food safety challenges. Lactic acid bacteria (LAB) offer a promising natural alternative due to their antimicrobial properties, especially through the production of bacteriocins. This study investigates the competitive interactions between Lactococcus lactis and L. monocytogenes under

The Brucella abortus ribosomal protein L7/L12 is an immunodominant antigen and an interesting candidate for the development of oral live vaccines against brucellosis. Here, a recombinant Lactococcus lactis strain producing L7/L12 under the control of nisin inducible promoter was orally administered to BALB/c mice. Significant levels of anti-L7/L12 specific IgA detected in feces revealed an induced local humoral immune response. However, serum analysis did not reveal any anti-L7/L12 antibodies suggesting the absence of a systemic response. Nevertheless, the vaccinated mice showed a partial prot

Flow cytometry in combination with fluorescent molecular markers 5- (and 6-) carboxyfluorescein succinimidylester (CFSE) and propidium iodide (PI) have been applied to determine lag times, numbers of cell divisions and injury after mild heat (50 degrees C, 5 min) and nisin treatments (0.1 and 1.0 microgram ml-1) of Lactobacillus plantarum. Initial labelling with covalently bound dye CFSE (20 and 100 micrograms ml-1) allowed determination of lag times and cell proliferation for up to eight generations. Double-labelling with CFSE and PI (5 micrograms ml-1) provided additional information about d
