Impacts of CuO nanoparticles on nitrogen removal in sequencing batch biofilm reactors after short-term and long-term exposure and the functions of natural organic matter.

The impacts of CuO nanoparticle (NP) exposure on total nitrogen (TN) removal in a sequencing batch biofilm reactor (SBBR) as well as the effects of natural organic matter (NOM) in wastewater were studied. Short-term exposure (8 h) to 1 and 50 mg/L CuO NPs induced negligible influence on the nitrogen removal efficiency, and biofilms could recover from the slight damage caused by the prolonged exposure (45 days) to 1 mg/L CuO NPs. On the other hand, long-term exposure to 50 mg/L CuO NPs notably decreased the nitrogen removal efficiencies to 47.74 and 59.04 % in the absence and presence of bovine

Effects of Chewing Gum on Nitric Oxide Metabolism, Markers of Cardiovascular Health and Neurocognitive Performance after a Nitrate-Rich Meal.

OBJECTIVES: Cardiovascular and neurocognitive responses to chewing gum have been reported, but the mechanisms are not well understood. Chewing gum after a nitrate-rich meal may upregulate the reduction of oral nitrate to nitrite and increase nitric oxide (NO), a molecule important to cardiovascular and neurocognitive health. We aimed to explore effects of chewing gum after a nitrate-rich meal on nitrate metabolism (through the enterosalivary nitrate-nitrite-NO pathway), endothelial function, blood pressure (BP), neurocognitive performance, mood and anxiety. METHODS: Twenty healthy men (n = 6)

Oxygen control of nitrogen oxide respiration, focusing on α-proteobacteria.

Denitrification is generally considered to occur under micro-oxic or anoxic conditions. With this in mind, the physiological function and regulation of several steps in the denitrification of model α-proteobacteria are compared in the present review. Expression of the periplasmic nitrate reductase is quite variable, with this enzyme being maximally expressed under oxic conditions in some bacteria, but under micro-oxic conditions in others. Expression of nitrite and NO reductases in most denitrifiers is more tightly controlled, with expression only occurring under micro-oxic conditions. A possi

Nitrate bioreduction in redox-variable low permeability sediments.

Lowpermeability zone (LPZ) can play an important role as a sink or secondary source in contaminant transport in groundwater system. This study investigated the rate and end product of nitrate bioreduction in LPZ sediments. The sedimentswere fromthe U.S. Department of Energy's Hanford Site, where nitrate is a groundwater contaminant as a by-product of radionuclide waste discharges. The LPZ at the Hanford site consists of two layerswith an oxidized layer on top and reduced layer below. The oxidized layer is directly in contact with the overlying contaminated aquifer, while the reduced layer is in

The interplay of hematite and photic biofilm triggers the acceleration of biotic nitrate removal.

The soil-water interface is replete with photic biofilm and iron minerals; however, the potential of how iron minerals promote biotic nitrate removal is still unknown. This study investigates the physiological and ecological responses of photic biofilm to hematite (Fe2O3), in order to explore a practically feasible approach for in-situ nitrate removal. The nitrate removal by photic biofilm was significantly higher in the presence of Fe2O3 (92.5%) compared to the control (82.8%). Results show that the presence of Fe2O3 changed the microbial community composition of the photic biofilm, facilitat

In vitro evaluation of a new treatment for urinary tract infections caused by nitrate-reducing bacteria.

Dietary and endogenous nitrates are excreted in urine, and during infection with nitrate-reducing bacteria they are reduced to nitrite. At a low pH nitrite is converted to a variety of nitrogen oxides that are toxic to bacteria. We hypothesized that acidification of nitrite-rich infected urine would result in the killing of the nitrate-reducing bacteria. An Escherichia coli control strain and a mutant lacking nitrate reductase activity were preincubated in urine supplemented with sodium nitrate (0 to 10 mM) at pH 7.0. Then, the nitrite-containing bacterial culture was transferred (and diluted

Comparative genomic analysis of hydrogen peroxide and nitric oxide metabolic pathways in Limosilactobacillus fermentum.

Limosilactobacillus fermentum strains influence human health through distinct metabolic pathways; however, the genetic basis of these strain-specific functions remains unclear. This study investigated phenotypic divergence between two L. fermentum strains isolated from the human oral microbiome. Despite 98.2% average nucleotide identity, comparative genomics revealed substantial strain-specific gene repertoires (485 and 542 unique genes in DM072 and DM075, respectively). Functional characterization demonstrated that DM072 synthesizes hydrogen peroxide via pyruvate oxidase (EC 1.2.3.3; K00158),

Nitrate Sensing and Metabolism Inhibit Biofilm Formation in the Opportunistic Pathogen Burkholderia pseudomallei by Reducing the Intracellular Concentration of c-di-GMP.

The opportunistic pathogen Burkholderia pseudomallei is a saprophytic bacterium and the causative agent of melioidosis, an emerging infectious disease associated with high morbidity and mortality. Although melioidosis is most prevalent during the rainy season in endemic areas, domestic gardens and farms can also serve as a reservoir for B. pseudomallei during the dry season, in part due to irrigation and fertilizer use. In the environment, B. pseudomallei forms biofilms and persists in soil near plant root zones. Biofilms are dynamic bacterial communities whose formation is regulated by extrac

ArnR binds a [4Fe-4S] cluster and indirectly senses anaerobicity in Corynebacteria.

Corynebacteria are commercially and medically important Gram-positive bacteria that can switch from aerobic to anaerobic respiration in response to low O2 and the availability of nitrate as an alternative electron acceptor. The narKGHJI operon encoding the respiratory nitrate reductase is under the control of a novel regulator, ArnR, which plays a major role in the aerobic/anaerobic respiratory switch. ArnR was previously shown to be an iron-sulfur cluster protein that modulates its DNA binding according to availability of O2. However, previous data suggest that it does not do this directly in

Nitric oxide levels in Behçet's disease.

Behçet's disease (BD) is a chronic multisystemic disorder which is characterized by a relapsing systemic inflammatory process. In certain inflammatory conditions such as rheumatoid arthritis, over production of nitric oxide (NO) could damage host cells and tissues, either directly and/or following reaction with other free radicals, such as superoxide anion to form species including peroxynitrite or hydroxyl radicals. Excessive superoxide radical production and impaired antioxidant mechanism in both the neutrophils and plasma of patients with BD have been reported. Our study was designed to inv

Heat-induced formation of nitrogen oxides in water.

It was found by the fluorimetric method using 2,3-diaminonaphthalene that moderate heating of water (60-80°C, for up to 4 h) leads to the fixation of atmospheric nitrogen with the formation of nitrite. The kinetic parameters of this process were determined. The energy of activation of [Formula: see text]formation was estimated to be 139 kJ/mol. It was found that the amount of nitrite formed depends on the concentration of dissolved oxygen and nitrogen. It was shown by two independent methods (Griess reagent/VCl3 and 2,3-diaminonaphthalene/nitrate reductase) that heating of water (80°C, 1 h) re

Co-culturing fungus Penicillium citrinum and strain Citrobacter freundii improved nitrate removal and carbon utilization by promoting glyceride metabolism.

Exploring the interaction between denitrifying microbial species is significant for improving denitrification performance. In this study, the effects of co-culturing fungus Penicillium citrinum and strain Citrobacter freundii on denitrification were investigated. Results showed that the maximum nitrate removal and carbon utilization in co-culture were 68.0 and 14.1 mg·L-1·d-1, respectively. The total adenosine triphosphatase activity was increased to 9.87 U‧mg-1 protein in co-culture, and nicotinamide adenine dinucleotide production was 1.7-2.3 times that of monoculture, attributing to increas

The State of the Nitric Oxide Cycle in Respiratory Tract Diseases.

This review describes the unique links of the functioning of the nitric oxide cycle in the respiratory tract in normal and pathological conditions. The concept of a nitric oxide cycle has been expanded to include the NO-synthase and NO-synthase-independent component of its synthesis and the accompanying redox cascades in varying degrees of reversible reactions. The role of non-NO-synthase cycle components has been shown. Detailed characteristics of substrates for the synthesis of nitric oxide (NO) in the human body, which can be nitrogen oxides, nitrite and nitrate anions, and organic nitrates

Nitrogen Assimilation and Protein Synthesis in Wheat Seedlings as Affected by Mineral Nutrition. II. Micronutrients.

Activity of nitrate reductase from Triticum aestivum L. seedlings was decreased by deficiencies of molybdenum, zinc, and chlorine. Nitrate accumulated in molybdenum-deficient seedlings, declined in zinc-deficient seedlings, and was unaffected by the other micronutrient treatments. Glutamic acid dehydrogenase activity was decreased by deficiency of molybdenum, the only nutrient that affected the enzyme. Glutamine synthetase activity was decreased only by copper deficiency, and glutamic-oxaloacetic transaminase was not affected by any micronutrient deficiencies. Incorporation of (14)C-leucine in

Iron oxide minerals promote simultaneous bio-reduction of Cr(VI) and nitrate: Implications for understanding natural attenuation.

Nitrate and Cr(VI) coexist in aquifers, posing a potential threat to ecological environment and public health. Iron oxide minerals (hematite and magnetite) exist ubiquitously in groundwater, which are hot spots for biogeochemical transformation. However, there is still a knowledge gap anout the effect of iron oxide minerals on bioreduction of nitrate and Cr(VI). Here we observed that iron oxide minerals can significantly improve the ability of microorganisms to simultaneously reduce nitrate and Cr(VI), the reduction rates of nitrate and Cr(VI) increased by 7.3 and 8.5 times, respectively. The

Reexamining the risks of drinking-water nitrates on public health.

BACKGROUND: Nitrates in drinking water are generally considered the sole source of nitrite poisoning with methemoglobinemia in infantile methomoglobinemia (IM). However, IM, which occurs during the first 4 months of life, is actually a constellation of cyanosis and hypoxia associated with methemoglobinemia that can result from several other causes. METHODS: This review reexamines the role of nitrate levels in drinking water as a cause of IM and identifies other sources of nitrates that can affect public health and cause chronic diseases. RESULTS: Causes of IM include nitrites in foods, envir

Salmonella uses sulfate reductases with unique catalytic activity to promote gut colonization in mice.

Non-typhoidal Salmonella use molybdenum cofactor-containing MopB- or DMSO reductase-family members to respire chemically diverse substrates, including formate, nitrate and methionine sulfoxide, during infection. The DmsABC enzymatic complex encodes one such DMSO reductase to promote oxidative stress resistance. The Salmonella genome encodes several gene paralogues but their role in virulence is unclear. Here we characterize three Salmonella MopB-family extracytoplasmic sulfate reductases, which we call Xsr1A, Xsr2A and Xsr3A. Infection experiments in mice and macrophages show that these sulfat
