Tobacco use is a major cause of preventable morbidity and mortality globally. Tobacco products, including smokeless tobacco (ST), generally contain tobacco-specific N-nitrosamines (TSNAs), such as N'-nitrosonornicotine (NNN) and 4-(methylnitrosamino)-1-(3-pyridyl)-butanone (NNK), which are potent carcinogens that cause mutations in critical genes in human DNA. This review covers the series of biochemical and chemical transformations, related to TSNAs, leading from tobacco cultivation to cancer initiation. A key aim of this review is to provide a greater understanding of TSNAs: their precursors

In this work, a low-cost alternative method (i.e., adding nitrate into WAS) to significantly enhance hydrogen production was reported. Experimental results showed that with an increase of nitrate addition from 0 to 362 mg/L, the maximal hydrogen production from acidic (pH 5.5) fermentation of WAS obviously increased from 12.6 ± 0.5 to 19.3 ± 0.9 mL per gram volatile suspended solids (VSS). The mechanism investigations illustrated more substrates were provided for subsequent hydrogen production. Although the nitrate added inhibited all the biological processes, its inhibition to the hydrogen co

Protein tyrosine (Tyr) nitration is a post-translational modification yielding 3-nitrotyrosine (NO2 -Tyr). Formation of NO2 -Tyr is generally considered as a marker of nitro-oxidative stress and is involved in some human pathophysiological disorders, but has been poorly studied in plants. Leghemoglobin (Lb) is an abundant hemeprotein of legume nodules that plays an essential role as an O2 transporter. Liquid chromatography coupled to tandem mass spectrometry was used for a targeted search and quantification of NO2 -Tyr in Lb. For all Lbs examined, Tyr30, located in the distal heme pocket, is t

BACKGROUND: The efficacy of dietary nitrate supplementation to lower blood pressure (BP) in pregnant women is highly variable. We aimed to investigate whether differences in oral microbiota profiles and oral nitrate-reducing capacity may explain interindividual differences in BP lowering following nitrate supplementation.
METHODS: Participants recruited for this study were both pregnant and nonpregnant women, with or without hypertension (n=55). Following an overnight fast, plasma, saliva, and tongue scraping samples were collected for measurement of nitrate/nitrite concentrations, oral NaR (

Nitric oxide (NO) is a vital immune molecule eliciting resistance to diverse microbial pathogens in humans and animals. However, its functional integration into plant immune networks remains incompletely characterized. In this study, we reveal that both endogenous induction and exogenous supplementation of NO significantly enhance resistance to rice stripe virus (RSV), a Bunyavirus that poses a huge threat to rice production. The nitrate transporter OsNPF6.1 potentiates virus resistance by upregulating the expression of nitrate reductase (OsNR2) and subsequent NO biosynthesis. Functional analy

Elevated nitrate in the environment inhibits sulfate reduction by important microorganisms of sulfate-reducing bacteria (SRB). Several SRB may respire nitrate to survive under elevated nitrate, but how SRB that lack nitrate reductase survive to elevated nitrate remains elusive. To understand nitrate adaptation mechanisms, we evolved 12 populations of a model SRB (i.e., Desulfovibrio vulgaris Hildenborough, DvH) under elevated NaNO3 for 1000 generations, analyzed growth and acquired mutations, and linked their genotypes with phenotypes. Nitrate-evolved (EN) populations significantly (p < 0.05)

Intertidal macroalgae Fucus and Laminaria experience seasonally fluctuating inorganic N supply. This study examined the effects of long-term N deprivation, recovery following N resupply, and effects of elevated ammonium and nitrate exposure on N acquisition in intertidal algae using manipulations of N supply in tank culture. Over 15 weeks of N deprivation, internal N and nitrate reductase activity (NRA) declined, but maximum quantum yield of PSII was unaffected in Fucus serratus and Fucus vesiculosus. Low NRA was maintained despite no external nitrate availability and depletion of internal poo

BACKGROUND: Xanthine oxidoreductase (XOR) is a multi-functional enzyme that metabolises purines generating uric acid and generates reactive oxygen species. Both functions have been implicated in the pathogenesis of cardiovascular disease. More recently, a third function of XOR as a nitrite reductase has been identified. This nitrite reductase activity has been proposed to play a key role in the benefits of targeting the non-canonical pathway for nitric oxide (NO) generation in the cardiovascular disease setting; an effect specifically attributed to XOR-dependent recovery of NO levels. However,

Lowpermeability zone (LPZ) can play an important role as a sink or secondary source in contaminant transport in groundwater system. This study investigated the rate and end product of nitrate bioreduction in LPZ sediments. The sedimentswere fromthe U.S. Department of Energy's Hanford Site, where nitrate is a groundwater contaminant as a by-product of radionuclide waste discharges. The LPZ at the Hanford site consists of two layerswith an oxidized layer on top and reduced layer below. The oxidized layer is directly in contact with the overlying contaminated aquifer, while the reduced layer is in

The soil-water interface is replete with photic biofilm and iron minerals; however, the potential of how iron minerals promote biotic nitrate removal is still unknown. This study investigates the physiological and ecological responses of photic biofilm to hematite (Fe2O3), in order to explore a practically feasible approach for in-situ nitrate removal. The nitrate removal by photic biofilm was significantly higher in the presence of Fe2O3 (92.5%) compared to the control (82.8%). Results show that the presence of Fe2O3 changed the microbial community composition of the photic biofilm, facilitat

Dietary and endogenous nitrates are excreted in urine, and during infection with nitrate-reducing bacteria they are reduced to nitrite. At a low pH nitrite is converted to a variety of nitrogen oxides that are toxic to bacteria. We hypothesized that acidification of nitrite-rich infected urine would result in the killing of the nitrate-reducing bacteria. An Escherichia coli control strain and a mutant lacking nitrate reductase activity were preincubated in urine supplemented with sodium nitrate (0 to 10 mM) at pH 7.0. Then, the nitrite-containing bacterial culture was transferred (and diluted

Glycyrrhiza uralensis is a highly valued medicinal species worldwide. However, a paradox arises in its cultivation in that high nitrogen fertilization boosts yield at the expense of root quality, a problem linked to nitrogen's regulation of tricarboxylic acid (TCA) cycle-driven respiration. It remains unclear how different nitrogen forms coordinate respiratory and primary metabolism. We examined the regulatory mechanisms of nitrate (NO3-) versus ammonium (NH4+) on these processes in cultivated G. uralensis by supplying seedlings with varying concentrations of K15NO3 or (15NH4)2SO4 in a modifie

Cyclodextrins (CDs) have been widely used due to the excellent solubilization of hydrophobic organics. However, their effect on the biotransformation process of hydrophilic pollutants is unclear. This study first evaluated the effect and mechanism of CDs on nitrate reduction by S.oneidensis.MR-1. The three CDs (α-CD, β-CD and γ-CD) all accelerated nitrate reduction, among which β-CD had the best effect. The nitrate reduction rate was increased by 21.8% with 0.5 mM β-CD. As for the mechanism, β-CD increased the biomass, membrane permeability and EPS of S.oneidensis.MR-1. The nitrate reductase a

The impacts of CuO nanoparticle (NP) exposure on total nitrogen (TN) removal in a sequencing batch biofilm reactor (SBBR) as well as the effects of natural organic matter (NOM) in wastewater were studied. Short-term exposure (8 h) to 1 and 50 mg/L CuO NPs induced negligible influence on the nitrogen removal efficiency, and biofilms could recover from the slight damage caused by the prolonged exposure (45 days) to 1 mg/L CuO NPs. On the other hand, long-term exposure to 50 mg/L CuO NPs notably decreased the nitrogen removal efficiencies to 47.74 and 59.04 % in the absence and presence of bovine

OBJECTIVES: Cardiovascular and neurocognitive responses to chewing gum have been reported, but the mechanisms are not well understood. Chewing gum after a nitrate-rich meal may upregulate the reduction of oral nitrate to nitrite and increase nitric oxide (NO), a molecule important to cardiovascular and neurocognitive health. We aimed to explore effects of chewing gum after a nitrate-rich meal on nitrate metabolism (through the enterosalivary nitrate-nitrite-NO pathway), endothelial function, blood pressure (BP), neurocognitive performance, mood and anxiety.
METHODS: Twenty healthy men (n = 6)

Nitrate contamination from human activities (e.g., domestic pollution, livestock breeding, and fertilizer application) threatens marine ecosystems and net primary productivity. As the main component of primary productivity, diatoms can adapt to high nitrate environments, but the mechanism is unclear. We found that electron transfer from marine colloids to diatoms enhances nitrogen uptake and assimilation under visible-light irradiation, providing a new pathway for nitrogen adaptation. Under irradiation, marine colloids exhibit semiconductor properties (e.g., the separation of electron-hole pai

It was found by the fluorimetric method using 2,3-diaminonaphthalene that moderate heating of water (60-80°C, for up to 4 h) leads to the fixation of atmospheric nitrogen with the formation of nitrite. The kinetic parameters of this process were determined. The energy of activation of [Formula: see text]formation was estimated to be 139 kJ/mol. It was found that the amount of nitrite formed depends on the concentration of dissolved oxygen and nitrogen. It was shown by two independent methods (Griess reagent/VCl3 and 2,3-diaminonaphthalene/nitrate reductase) that heating of water (80°C, 1 h) re

The key challenge in current biological technologies for the control of harmful algal blooms (HABs) is that algicidal bacteria's lytic mechanisms during bloom treatment results in structural rupture of algal cells. This results in the massive release of intracellularly stored nutrients (such as nitrogen/phosphorus compounds) and secondary metabolites such as microcystins into aquatic environments, causing secondary pollution. This study isolated two Bacillus strains (Bacillus haynesii and Bacillus tequilensis) from phycosphere microbiota, applying them synergistically against Microcystis aerug

Corynebacteria are commercially and medically important Gram-positive bacteria that can switch from aerobic to anaerobic respiration in response to low O2 and the availability of nitrate as an alternative electron acceptor. The narKGHJI operon encoding the respiratory nitrate reductase is under the control of a novel regulator, ArnR, which plays a major role in the aerobic/anaerobic respiratory switch. ArnR was previously shown to be an iron-sulfur cluster protein that modulates its DNA binding according to availability of O2. However, previous data suggest that it does not do this directly in

This review describes the unique links of the functioning of the nitric oxide cycle in the respiratory tract in normal and pathological conditions. The concept of a nitric oxide cycle has been expanded to include the NO-synthase and NO-synthase-independent component of its synthesis and the accompanying redox cascades in varying degrees of reversible reactions. The role of non-NO-synthase cycle components has been shown. Detailed characteristics of substrates for the synthesis of nitric oxide (NO) in the human body, which can be nitrogen oxides, nitrite and nitrate anions, and organic nitrates
