The objective of this study was to analyse in vivo the effect of oxygen on the nitrogenase of Bacillus polymyxa. The culture technique employed in this study prevented spore formation by B. polymyxa during the entire period of exposure to acetylene. Under these conditions the acetylene-reduction assay allowed quantification of nitrogenase activity over long incubation periods (44 h). Nitrogenase activity was highest in cells harvested in the late logarithmic phase. At PO2 of 0.19 and 0.37 kPa, acetylene reduction was inhibited by 80 and 100%, respectively. This switch-off effect could be rever

Grasslands are vital for providing wildlife habitats, essential ecosystem services, and food to humanity. Elevated atmospheric CO2 concentration (eCO2), as a key driver of climate change, has substantial effects on grasslands. Nonetheless, a holistic quantification on the response of the nitrogen cycle to eCO2 in global grasslands is lacking. Here, we show that solely eCO2 could increase the net primary productivity (NPP) in grasslands by 10% (95% confidence interval, 8-12%), yet simultaneously decrease leaf nitrogen content by 8% (95% confidence interval, 5-10%). When considering the effects

The nitrogen transformation during sludge pyrolysis is affected by the dewater conditioner. However, the comparative analysis of the conditioner under identical pyrolysis conditions has been previously absent. In this study, Ca-, Fe- and Al-based conditioners were selected as the representatives. A comprehensive evaluation considering the cost of the conditioners and the product characteristics was conducted. Additionally, the in-situ fixation mechanism of the conditioner on nitrogen-containing gas was concurrently revealed. Among the six conditioners, CaO and AlCl3 were identified as the top

Marine diazotrophic cyanobacteria play a crucial role in oceanic nitrogen cycling, supporting primary production and ecosystem balance. Crocosphaera watsonii WH8501 exemplifies this capability by temporally separating photosynthesis and diazotrophy. To investigate the regulatory mechanisms underlying this process, we employed mass spectrometry-based proteomics in a high-resolution diel culturing experiment, revealing tightly coordinated protein abundance patterns. Unsupervised clustering of diel protein abundance profiles identified six groups of co-regulated proteins characterized by distinct

Dissimilatory nitrate reduction to ammonium (DNRA), denitrification, anaerobic ammonium oxidation (anammox), and biological N2 fixation (BNF) can influence the nitrogen (N) use efficiency of rice production. While the effect of N application on BNF is known, little is known about its effect on NO3- partitioning between DNRA, denitrification, and anammox. Here, we investigated the effect of N application on DNRA, denitrification, anammox, and BNF and on the abundance of relevant genes in three paddy soils in Australia. Rice was grown in a glasshouse with N fertilizer (150 kg N ha-1) and without

Symbiosomes are plant membrane-bound, organelle-like structures that enclose single or multiple bacteroids (differentiated rhizobia) within legume cells, serving specifically as the site for N2 fixation. Although isolated symbiosomes have been used to study nitrogen fixation, ion transport, and metabolite translocation, conventional density gradient-based isolation methods for their isolation are labor-intensive. Here, we describe a simplified, density gradient-free protocol for rapid enrichment of symbiosome-containing fractions from mature legume nodules using the Medicago truncatula-Sinorhi

A unique Klebsiella species strain, 10982, was cultured from a perianal swab specimen obtained from a patient in the University of Maryland Medical Center intensive care unit. Klebsiella sp. 10982 possesses a large IncA/C multidrug resistance plasmid encoding a novel FOX AmpC β-lactamase designated FOX-10. A novel variant of the LEN β-lactamase was also identified. Genome sequencing and bioinformatic analysis demonstrated that this isolate contains genes associated with nitrogen fixation, allantoin metabolism, and citrate fermentation. These three gene regions are typically present in either K

Legumes are important for nitrogen cycling in the environment and agriculture due to the ability of nitrogen fixation by rhizobia. In this review, we introduce an important and potential role of legume-rhizobia symbiosis in aiding phytoremediation of some metal contaminated soils as various legumes have been found to be the dominant plant species in metal contaminated areas. Resistant rhizobia used for phytoremediation could act on metals directly by chelation, precipitation, transformation, biosorption and accumulation. Moreover, the plant growth promoting (PGP) traits of rhizobia including n

Diazotrophic cyanobacteria fix both atmospheric carbon (C) and nitrogen (N) into biomass, but the two assimilation pathways are not compatible. Species like Anabaena sp. PCC 7120 physically separate C and N assimilation in different cell types. Even if separated, they are strongly intertwined, as N assimilation relies on the C skeletons and reducing power from photosynthesis, that in turn depends on N-rich molecules as pigments and proteins. Whereas the two pathways have been extensively studied individually, this work investigates their interaction by analysing photosynthetic properties upon

Biological nitrogen (N) fixation is an important process supporting primary production in ecosystems, especially in those where N availability is limiting growth, such as peatlands and boreal forests. In many peatlands, peat mosses (genus Sphagnum) are the prime ecosystem engineers, and like feather mosses in boreal forests, they are associated with a diverse community of diazotrophs (N2-fixing microorganisms) that live in and on their tissue. The large variation in N2 fixation rates reported in literature remains, however, to be explained. To assess the potential roles of habitat (including n

The industrial production of conifer seedlings in nurseries uses large amounts of fertilizers to ensure their proper growth and accurate nutrient status. However, inorganic nitrogen fertilization leads to nitrate leaching, which has negative environmental consequences. An alternative solution could be the use of controlled-release fertilizers that supply nutrients over longer periods and hence have a lower environmental impact. This study analysed the performance of a novel arginine-iron-hexametaphosphate complex on Scots pine (Pinus sylvestris) seedlings. The complex was characterized using a

Titanium dioxide is the most studied photocatalytic material and has been reported to be active for a wide range of reactions, including the oxidation of hydrocarbons and the reduction of nitrogen. However, the molecular-scale interactions between the titania photocatalyst and dinitrogen are still debated, particularly in the presence of hydrocarbons. Here, we used several spectroscopic and computational techniques to identify interactions among nitrogen, methanol, and titania under illumination. Electron paramagnetic resonance spectroscopy (EPR) allowed us to observe the formation of carbon r

Deep-sea hydrothermal vents and cold seeps, typical ecosystems characterized by abundant carbon but limited nitrogen, support thriving microbial activities. The source of nitrogen has long been a key focus in scientific research. Although cultivation experiments have confirmed the presence of diverse nitrogen-fixing microorganisms, direct in situ evidence has been lacking. Therefore, a deep-sea in situ surface-enhanced Raman scattering sensor has been developed. It can adapt to extreme environment (≥350°C and ≥2000-m depth and LOD < 10-7 M) and has excellent deep-sea application potential. Cru

The cyanosphere is composed of non-cyanobacterial microorganisms living within the exopolysaccharide sheath of cyanobacteria, interacting with the cyanobacterial hosts and their surrounding environment. Understanding the interactions between cyanobacteria and their cyanospheres can help predict the success of terrestrial cyanobacteria in providing ecosystem services in nutrient-poor environments. However, knowledge of the microbial diversity and functions within the cyanosphere remains limited. Here, we used metagenomic sequencing to reconstruct 415 metagenome-assembled genomes (MAGs) from cya

To further understand the nitrogen metabolism disrupted by anthropogenic activities, 2.5 g/L of ferrihydrite were added into cathodic chamber of bioelectrochemical system to expediate the nitrogen removal process. It was found that the nitrate removal constant was significantly improved and maintained at around 0.09 h-1 with ferrihydrite addition, while the control group maintained only at around 0.05 h-1. Besides, it seemed that the addition of ferrihydrite lead to less biomass accumulation but higher biofilm viability. Meanwhile, ferrihydrite selectively enriched OTUs capable of participatin

Soybean (Glycine max L. [Merr.]) nitrogen fixation is sensitive to soil drying and is an important target for improving soybean drought resilience. Transcriptional regulation of the nodule drought response and the influence of symbiotic rhizobia are not well understood. We measured whole-plant responses to mild and moderate soil moisture deficit (41% and 20% of field capacity) alongside the nodule transcriptome, including soybean and Bradyrhizobium diazoefficiens transcripts, in two soybean genotypes ('Benning' and PI 471938) that were previously found to have diverging nitrogen fixation droug

Thismia species are non-photosynthetic plants entirely dependent on fungal partners for carbon and nutrients. While their arbuscular mycorrhizal associations are well-documented, bacterial symbiont roles remain unexplored. Using 16S rRNA gene amplicon sequencing, we investigated endophytic bacterial communities in T. gardneriana, T. javanica, and T. mirabilis from geographically distinct locations in Thailand. Despite geographic separation, Thismia spp. consistently harbored bacterial compositions taxonomically and functionally distinct from surrounding soil microbiomes. Root endospheres were

Geochemical evidence invokes anoxic deep oceans until the terminal Neoproterozoic ~0.55 Ma, despite oxygenation of Earth's atmosphere nearly 2 Gyr earlier. Marine sediments from the intervening period suggest predominantly ferruginous (anoxic Fe(II)-rich) waters, interspersed with euxinia (anoxic H(2)S-rich conditions) along productive continental margins. Today, sustained biotic H(2)S production requires NO(3)(-) depletion because denitrifiers outcompete sulphate reducers. Thus, euxinia is rare, only occurring concurrently with (steady state) organic carbon availability when N(2)-fixers domin

Increase in perception of solar ultraviolet-B (UV-B) radiation on Earth's surface due to anthropogenic activities has potential in causing detrimental effects on plants. The present study was performed to evaluate the effect of elevated UV-B on Pisum sativum L., a leguminous plant with emphasis on nitrogen metabolism, flavonoids and hormonal changes. Elevated UV-B (ambient+7.2 kJ m(-2) day(-1)) negatively affected the growth, biomass, yield and its quality by generating oxidative stress directly or due to elevation of salicylic acid in two cultivars with higher magnitude being observed in HUP-

Nitrogen (N2) fixation is essential for sustaining life but is challenging under ambient conditions. Industrial N2 fixation is energy-intensive and exacerbates carbon dioxide (CO2) emissions, while high concentration of atmospheric CO2 urgently requires scalable mitigation strategies. In this study, we report that simultaneous spontaneous activation of N2 and CO2 under ambient conditions in microdroplets, producing ammonium (NH4 +), nitrite (NO2 -), and formate (HCO2 -), as confirmed by isotopic labelling experiments and theoretical calculations. This can clarify the doubts about environmental
