Automated analysis of breathing waveforms using BreathMetrics: a respiratory signal processing toolbox.

Nasal inhalation is the basis of olfactory perception and drives neural activity in olfactory and limbic brain regions. Therefore, our ability to investigate the neural underpinnings of olfaction and respiration can only be as good as our ability to characterize features of respiratory behavior. However, recordings of natural breathing are inherently nonstationary, nonsinusoidal, and idiosyncratic making feature extraction difficult to automate. The absence of a freely available computational tool for characterizing respiratory behavior is a hindrance to many facets of olfactory and respirator

Investigating plasticity of the olfactory system through the lens of specific anosmia.

The olfactory experiences vary widely among individuals. Despite having an otherwise normal sense of smell, some people exhibit specific anosmia, i.e., the inability to detect certain odorants. While genetic factors play a key role in this phenomenon, olfactory perception is also influenced by environment, and repeated exposure to odors (olfactory training) can enhance olfactory sensitivity. In this study, we examined whether olfactory training increases sensitivity towards odors participants were specifically anosmic to, and preliminary investigated whether this training-induced change is sta

[Olfactory stimulation and contingent negative variation (CNV) (author's transl)].

In addition to olfactory evoked responses (Fig. 1a) the vertex-negative voltage shift was registered in 20 normal adults. Fig. 2 shows this potential for an olfactory indicative stimulus followed by a tone burst or flash sequence, which are stopped by the test person's manual reaction. In Fig. 3 two alternating odorous stimuli are presented, one of which is followed by the tone. The selective expectancy wave is followed by the tone. The selective expectancy wave is only built up by the marked olfactory stimulus. For the first time CNV can be used as a clinical investigation tool to confirm cen

Olfaction in the context of obesity and diabetes: Insights from animal models to humans.

The olfactory system is at the crossroad between sensory processing and metabolic sensing. In addition to being the center of detection and identification of food odors, it is a sensor for most of the hormones and nutrients responsible for feeding behavior regulation. The consequences of modifications in body homeostasis, nutrient overload and alteration of this brain network in the pathological condition of food-induced obesity and type 2 diabetes are still not elucidated. The aim of this review was first to use both humans and animal studies to report on the current knowledge of the conseque

Pleasant parosmia with regard to own stool after SARS-CoV-2 infection.

Coronavirus disease 2019 (COVID-19) has had a significant global impact due to the millions of deaths it has caused secondary to respiratory failure. However, the disease has also been associated with a wide array of manifestations in other organ systems. Among them, the presence of anosmia, which occurs in up to half the patients, has become a new sign of alarm to suspect the infection. Although up to 90% of affected patients will experience an improvement of their olfactory alterations within a month after the infection, the variety and severity of olfactory disturbances clearly cannot be su

Olfactory training in specific anosmia to androstenone and its association with genetic variations of OR7D4.

Short-term, repeated exposure to odors, "olfactory training" (OT), improves olfactory function. Clinically, this works not only for trauma- or disease-related olfactory impairment but also in people with specific anosmia. Androstenone is an odorant for which the frequent occurrence of specific anosmia is already known. It is an odorous steroid derived from testosterone. Besides some people who cannot perceive the odor, it is perceived differently by different individuals in terms of odor quality. These differences in the ability to perceive androstenone as well as in the perception of its qual

Retrobulbar Amphotericin B Injection in Curbing the Progression of COVID Associated Rhino-orbital Cerebral Mucormycosis: A Retrospective Case Series.

BACKGROUND: Parosmia symptoms are difficult to quantify due to their heterogeneity among patients, and thus a clinical challenge. This study aimed to assess parosmia with Self-Administered Odor Questionnaire for Parosmia (SAOQ-P), a modification of the widely used SAOQ in Japan. The primary objective was to assess the effectiveness of SAOQ-P in identifying parosmia symptoms and its potential integration into the clinical assessment process. The study also explored traditional olfactory test differences between patients with and without parosmia. METHODS: Patients at Jikei Smell Clinic that pr

An unexpected insight into the cause of olfactory dysfunction: fibrillogenesis of odorant-binding proteins.

Olfactory dysfunction is a common complication of serious pathologies, including neurodegenerative disorders, bacterial and viral infections, including COVID-19, and others. Despite the widespread prevalence of olfactory disorders, the pathophysiological mechanisms of their development, as well as the molecular basis of their association with the underlying disease, remain incompletely understood. The current work formulates a new concept of the origin of olfactory disorders, linking a decrease in the activation of olfactory neurons and their death to the fibrillogenesis of odorant-binding pro

Diagnostics of olfactory dysfunction in Parkinson's disease - a literature overview and case series.

Olfactory dysfunction is increasingly recognised for its predictive value as an early indicator of a number of degenerative neuropathologies including Parkinson's Disease (PD). In this overview, we cover the relationship between PD and olfactory dysfunction (OD). Prodromal premotor symptoms of PD include sleep disturbances, psychiatric disorders, constipation and OD. The latter can precede motor symptoms by several years and its occurrence is frequent in Parkinson s disease with a prevalence that can range from 45%-90%. Olfactory perception in these cases can be tested using subjective and obj

When the sense of smell meets emotion: anxiety-state-dependent olfactory processing and neural circuitry adaptation.

Phylogenetically the most ancient sense, olfaction is characterized by a unique intimacy with the emotion system. However, mechanisms underlying olfaction-emotion interaction remain unclear, especially in an ever-changing environment and dynamic internal milieu. Perturbing the internal state with anxiety induction in human subjects, we interrogated emotion-state-dependent olfactory processing in a functional magnetic resonance imaging (fMRI) study. Following anxiety induction, initially neutral odors become unpleasant and take longer to detect, accompanied by augmented response to these odors

Perceptual differences in olfactory fat discrimination are not detected in neural activation.

Olfaction is involved in detecting, identifying, and discriminating dietary fat within foods, yet the underlying neural mechanisms remain uncharted. Our functional magnetic resonance imaging (fMRI) study investigated the neural correlates of olfactory fat perception and their association with discrimination ability in a complex food matrix. We measured brain activation resulting from orthonasal exposure to an ecologically relevant fat-related odor source-dairy milk, manipulated to contain 0%, 3.5%, or 14% fat. Twenty-six healthy, normosmic adults underwent olfactory fat content discrimination

Changes in gustatory perceptions of patients with major depression treated with vagus nerve stimulation (VNS).

BACKGROUND: Olfactory and gustatory functions were investigated before and during vagus nerve stimulation (VNS) in a group of 9 patients with therapy-resistant depression, implanted with a VNS system. METHODS: Gustation and olfaction were tested using standard sniffing tests. Subjects participated in 2 sessions with the vagal stimulator switched on and off, respectively. RESULTS: Under conditions of stimulation of the VNS, there were statistically significant differences of the threshold of perception, with an intensification of the taste "sweet" (Z = -2.0; p = 0.048) and "bitter" (Z = - 2.5

Two classic OBPs modulate the responses of female Holotrichia oblita to three major ester host plant volatiles.

Insects possess a fairly sophisticated olfactory system in their antennae to detect odorants essential for their survival and reproduction. Among them, insect first perceives odour sources by odorant-binding proteins (OBPs) to locate host-plants. Methyl salicylate, (Z)-3-hexenyl acetate and dibutyl phthalate are major volatile components of Ulmus pumila and Ricinus communis and elicit strong responses of the scarab beetle Holotrichia oblita adults. However, olfactory perception of the scarab beetle to these odorant compounds is unclear. In the current study, we cloned the OBP6 and OBP7 of H. o

Early Odorant Exposure Increases the Number of Mitral and Tufted Cells Associated with a Single Glomerulus.

UNLABELLED: The highly specific organization of the olfactory bulb (OB) is well known, but the impact of early odorant experience on its circuit structure is unclear. Olfactory sensory neurons (OSNs) project axons from the olfactory epithelium to the OB, where they form spherical neuropil structures called glomeruli. These glomeruli and the postsynaptic targets of OSNs, including mitral and tufted cells (M/TCs) and juxtaglomerular cells, form glomerular modules, which represent the basic odor-coding units of the OB. Here, we labeled M/TCs within a single glomerular module of the mouse OB and s

Rapid temporal processing in the olfactory bulb underlies concentration-invariant odor identification and signal decorrelation.

In a dynamic environment, sensory systems must filter out irrelevant information to construct a stable percept. Animals who rely on smell need to identify and discriminate odors despite fluctuations in concentration, yet odor receptor activation is strongly concentration dependent. Here we explored how odor signals are transformed within the mouse olfactory bulb (OB) by developing an all-optical approach to identify the connectivity between odor receptor channels (glomeruli) and the mitral and tufted cells (MTCs), while monitoring their odor responses. We found that the glomeruli and MTCs acti

Gray matter alterations in parosmia.

Parosmia is a common olfactory disorder. In this condition, odors are perceived in a different quality than usual. This distorted olfactory percept is typically reported to be unpleasant. Little is known about the pathophysiology of this phenomenon. Previous studies demonstrated smaller volumes of the olfactory bulbs in patients with parosmia compared to subjects without parosmia. In order to investigate structural brain alterations in areas beyond the olfactory bulb, in the current study voxel-based morphometry was applied. A group of 22 parosmic patients was compared with control subjects ma

[Impairment of Odor Recognition in Myotonic Dystrophy Type 1].

There is evidence that impaired human cognitive abilities are reflected in loss of olfactory abilities. Declining olfactory perception may be a biomarker for impairment of cognitive function and of impending illnesses in neurodegenerative disorders such as Parkinson's disease (PD) and Alzheimer's disease (AD). Previously, we reported that patients with myotonic dystrophy type 1 (DM 1) had lower sensitivity to emotional facial expressions as well as abnormal olfactory threshold or recognition level. In DM 1, pathological studies have reported neurofibrillary tangles in several temporal areas in

Olfactory receptor and neural pathway responsible for highly selective sensing of musk odors.

Musk odorants are used widely in cosmetic industries because of their fascinating animalic scent. However, how this aroma is perceived in the mammalian olfactory system remains a great mystery. Here, we show that muscone, one musk odor secreted by various animals from stink glands, activates a few glomeruli clustered in a neuroanatomically unique anteromedial olfactory bulb. The muscone-responsive glomeruli are highly specific to macrocyclic ketones; interestingly, other synthetic musk odorants with nitro or polycyclic moieties or ester bonds activate distinct but nearby glomeruli. The Olfactory Nerve: Anatomy and Pathology.

The human sense of smell is the unique sense through which the olfactory system can identify aromatic molecules within the air and provide a taste sensation. Still, also it plays an essential role in several other functions, warning about environmental safety and even impacts our emotional lives. Recently, olfactory impairment has become an issue of interest due to the COVID-19 pandemic. The dysfunction may vary from only reduced smell detection (hyposmia) to complete loss of it (anosmia) but also includes changes in the normal perception of odors (parosmia).
