Antibody responses to Onchocerca volvulus in Ecuadorian Indians and blacks.

In Ecuador, previous epidemiological studies of onchocerciasis affecting a population consisting of Blacks of African origin and Amerindians of the Chachi tribe, have demonstrated different frequencies of clinical disease manifestations in each racial group. To explore the possibility of differential antibody responses to O. volvulus antigens in this population, a subset of sera from these two groups of Ecuadorians with onchocerciasis was analyzed for antibody to O. volvulus antigens using ELISA and Western blot techniques. Significantly higher levels of immunoglobulin against O. volvulus were

Survival of Onchocerca volvulus in nodules implanted in immunodeficient rodents.

Onchocerca volvulus is an obligate human parasite, and its study has been difficult due to an inability to maintain it outside the human host. We report the successful transplantation of onchocercomata containing living adult O. volvulus worms into immunodeficient C.B.-17.scid/scid (scid) mice or athymic rnu/rnu (nude) rats. Living, motile worms containing viable microfilariae were present in onchocercomata recovered from scid mice or nude rats for up to 20 wk, establishing a novel animal model for future investigation of O. volvulus.

The reproductive lifespan of Onchocerca volvulus in West African savanna.

The epidemiological model ONCHOSIM--a model and computer simulation program for the transmission and control of onchocerciasis--has been used to determine the range of plausible values for the reproductive lifespan of Onchocerca volvulus. Model predictions based on different lifespan quantifications were compared with the results of longitudinal skin-snip surveys undertaken in 4 reference villages during 13 to 14 years of successful vector control in the Onchocerciasis Control Programme in West Africa. Good fits between predicted and observed trends in skin microfilarial loads could be obtaine

[Effects of diethylcarbamazine and ivermectin on the mobilization of microfilariae of Onchocerca volvulus].

Microfilaria of Onchocerca volvulus are localized in superficial dermis. However, they are sometimes noticed in the blood and urines, when there is an important infestation. Diethylcarbamazine (DEC) bring on an increase of microfilaria, successively in the blood and after, in the urines. This study of 30 patients, treated in double blind by placebo, DEC and ivermectin, a new molecule with spectacular action on this filariasis, allowed to compare effect of each one on apparition of microfilariae in the blood and urines. Ivermectin bring on very important increase of microfilaremia without micro

The paralyzing effect of midazolam on Onchocerca volvulus microfilariae in vitro.

Midazolam, a water soluble benzodiazepine derivative, inhibited the motility of Onchocerca volvulus microfilariae in vitro at concentrations of 25, 50, and 100 micrograms/ml. The effect was proportional to the concentrations used. Microfilariae regained motility after elimination of midazolam by washing. Diethylcarbamazine did not alter the midazolam effect. It is suggested that the Mazzotti reaction as the sequelae of diethylcarbamazine is caused by transient increased microfilarial migration. Combining midazolam with the diethylcarbamazine medication may prevent the Mazzotti reaction.

Cell adherence to microfilariae of Onchocerca volvulus: a comparative study.

The conditions were examined for in vitro antibody-mediated adherence of granulocytes to microfilariae of Onchocera volvulus and Dirofilaria immitis. Reactivity in human sera from patients in endemic foci in Sudan was specific for O. volvulus and no reactions were observed with heterologous Onchocerca species or with Mansonella perstans. Microfilariae from skin, nodules or adult female worms were satisfactory targets for cell adherence, and the cells involved were almost exclusively eosinophils. The reaction was inhibited by indomethacin but not by nordihydroguaiaretic acid, an inhibitor of le

A field method for the procurement of infective larvae of Onchocerca volvulus.

Natural vectors of onchocerciasis (S. yahense) were collected. Ninety-eight were intrathoracically inoculated with 30 freshly obtained skin-dwelling microfilariae. Of the 82 flies dissected, 56 were infected and 28 harbored infective larvae. A total of 193 infective O. volvulus larvae were obtained. The mean infective worm burden was 6.9.

Life span and distribution of Onchocerca volvulus microfilariae in mice.

Microfilariae of Onchocerca volvulus obtained from onchocercomas were inoculated subcutaneously into the inguinal region of mice. Microfilariae disseminated rapidly, invading the tail and lungs within 1 hr and the ears within 3 hr postinoculation. During the early period of infection microfilariae appeared in the eyes, ears, viscera, pelt, carcass, and tail; later, the microfilariae tended to accumulate in the tail. Liver microfilariae were recovered up to 12 wk postinoculation. Intraperitoneal inoculation and subcutaneous inoculation into the inguinal region resulted in the concentration of m

Development of Onchocerca volvulus in Simulium ochraceum and Simulium metallicum.

The development of Onchocerca volvulus was compared in Simulium ochraceum and S. metallicum by maintaining infected flies under uniform conditions. Larval development in S. ochraceum was synchronous and orderly so that nearly all larvae reaching the thoracic muscles from a single blood meal matured to third-stage infective larvae within 192 hours. In contrast, development in S. metallicum was asynchronous, slower, and retarded. In this species many first-larvae were malformed and stunted after the 5th day of development; and microfilariae and first- and second-stage larvae were still present 8

Seasonal variation in the concentration of Onchocerca volvulus microfilariae in the skin?

Repeated multiple skin snips in 18 persons with onchocerciasis from the Sudan-savanna of West Africa suggested the possibility of a seasonal variation in microfilarial concentrations. This variation may be an evolutionary adaptation of the parasite to the climate conditions that affect the seasonal distribution of the vector, but a migration of the microfilariae in the skin layers caused by the Simulium bites cannot be excluded.

Antigenic similarity of Onchocerca volvulus to other helminths examined by monoclonal antibodies against O. volvulus.

Antigens in Onchocerca volvulus were characterized using anti-O. vovulus monoclonal antibodies (McAb). A total of 17 McAb which were not species-specific to O. volvulus was examined for the antigenic similarity of O. volvulus with 12 parasitic helminths using indirect immunofluorescence. The range of reactivity of these McAb was wider in nematodes in contrast to species of cestoda or trematoda. The antigenic similarity within helminths examined was generally in agreement with their taxonomic relationships but was not shown in the lower taxonomic hierarchy. The helminth which holds the antigens

Nodding syndrome (NS) and Onchocerca Volvulus (OV) in Northern Uganda.

Nodding Syndrome (NS) is a childhood neurological disorder characterized by atonic seizures, cognitive decline, school dropout, muscle weakness, thermal dysfunction, wasting and stunted growth. There are recent published information suggesting associations between Nodding Syndrome (NS) with cerebrospinal fluid (CSF) VGKC antibodies and serum leiomidin-1 antibody cross reacting with Onchocerca Volvulus (OV). These findings suggest a neuro-inflammatory cause of NS and they are important findings in the search for the cause of Nodding Syndrome. These observations perhaps provide further, the uniq

Genetic selection of low fertile Onchocerca volvulus by ivermectin treatment.

BACKGROUND: Onchocerca volvulus is the causative agent of onchocerciasis, or "river blindness". Ivermectin has been used for mass treatment of onchocerciasis for up to 18 years, and recently there have been reports of poor parasitological responses to the drug. Should ivermectin resistance be developing, it would have a genetic basis. We monitored genetic changes in parasites obtained from the same patients before use of ivermectin and following different levels of ivermectin exposure. METHODS AND FINDINGS: O. volvulus adult worms were obtained from 73 patients before exposure to ivermectin a

Angiogenic activity of an Onchocerca volvulus Ancylostoma secreted protein homologue.

Angiogenesis is an important step in the development of ocular onchocercaisis. In previous studies, it has been demonstrated that Onchocerca volvulus homologues of the Ancylostoma secreted protein family have pronounced angiogenic activity. The overall goal of the current study was to determine if this angiogenic effect is exerted through a direct or indirect mechanism. These studies focused on one member of this family, OvASP-2, as this protein is expressed in microfilaria, the stage of the parasite that causes ocular onchocercaisis. Clones encoding truncated and full length open reading fram

Immune mechanisms in Onchocerca volvulus-mediated corneal disease (river blindness).

Infection with the parasitic nematode Onchocerca volvulus can lead to severe visual impairment and blindness. In an effort to characterize the molecular basis for the inflammatory response in the cornea, we have developed a murine model for O. volvulus-mediated keratitis in which parasite antigens are injected into the corneal stroma of sensitized mice. This model reproduces the two main clinical features of human disease, corneal opacification and neovascularization. Histological analysis of corneas from these mice reveals a biphasic recruitment of neutrophils and eosinophils to the central c

Strain differentiation of Onchocerca volvulus from Uganda using DNA probes.

Polymerase chain reaction (PCR) combined with non-radioactive DNA hybridization was applied for the detection and characterization of a 150 bp tandem repeat of Onchocerca volvulus. DNA of worms from western Uganda was amplified and then probed with a digoxygenin-labelled oligonucleotide, specific for the forest form of O. volvulus and compared to samples from various African countries. Hybridization was only observed with PCR products from the forest in Liberia, south-eastern Ghana, Benin and southern Cameroon, but not with worms from Uganda or the savannah in Burkina Faso and northern Ghana.

Onchocerciasis in Ecuador: ocular findings in Onchocerca volvulus infected individuals.

Little is known of the epidemiology and clinical picture of ocular onchocerciasis in South America. A survey of onchocercal eye disease was performed in the hyperendemic area of a rain forest focus of onchocerciasis in Esmeraldas Province in Ecuador. A total of 785 skin snip positive individuals from black and Chachi Amerindian communities were examined. The blindness rate attributable to onchocerciasis was 0.4%, and 8.2% were visually impaired. Onchocercal ocular lesions were seen in a high proportion of the study group: 33.6% had punctate keratitis, microfilariae in the anterior chamber and
