Cutting edge: Role of C-C chemokine receptor 5 in organ-specific and innate immunity to Cryptococcus neoformans.

After intratracheal inoculation of the AIDS-associated pathogen Cryptococcus neoformans, 12-wk survival was >90% for CCR5+/+ mice but <25% for CCR5-/- mice. There were no defects in lung leukocyte recruitment (wk 5), pulmonary clearance, or delayed-type hypersensitivity in CCR5-/- mice. However, CCR5-/- mice had defects in leukocyte recruitment into the brain and, strikingly, in elimination of cryptococcal polysaccharide from the brain. In nonimmune CCR5-/- mice, there was a significant defect in macrophage recruitment after challenge with shed cryptococcal products (C. neoformans filtrate Ag)

Microbiome-orchestrated cross-organ immunity in autoimmunity: from metabolites to therapeutic targets.

Autoimmune diseases are systemic disorders in which barrier-site immune activation, especially in the gut, can reshape inflammatory programs in distant organs. This review advances a metabolite-centered, cross-organ framework for understanding how gut microbial ecology influences autoimmunity beyond individual gut-organ axes. We synthesize evidence that short-chain fatty acids, bile acid derivatives, tryptophan catabolites, polyamines and related microbial products act as mobile biochemical checkpoints linking intestinal barrier integrity, pattern-recognition signaling, immune-cell metabolism

Intestinal epithelial cell polarity defects in disease: lessons from microvillus inclusion disease.

The intestinal epithelium is a highly organized tissue. The establishment of epithelial cell polarity, with distinct apical and basolateral plasma membrane domains, is pivotal for both barrier formation and for the uptake and vectorial transport of nutrients. The establishment of cell polarity requires a specialized subcellular machinery to transport and recycle proteins to their appropriate location. In order to understand and treat polarity-associated diseases, it is necessary to understand epithelial cell-specific trafficking mechanisms. In this Review, we focus on cell polarity in the adul

Endothelium and its alterations in cardiovascular diseases: life style intervention.

The endothelium, which forms the inner cellular lining of blood vessels and lymphatics, is a highly metabolically active organ that is involved in many physiopathological processes, including the control of vasomotor tone, barrier function, leukocyte adhesion, and trafficking and inflammation. In this review, we summarized and described the following: (i) endothelial cell function in physiological conditions and (ii) endothelial cell activation and dysfunction in the main cardiovascular diseases (such as atherosclerosis, and hypertension) and to diabetes, cigarette smoking, and aging physiolog

A Multi-Modal Toolkit for Studying Neutrophils in Cancer and Beyond.

As key effector cells of the innate immune response, neutrophils are rapidly deployed to sites of inflammation where they deliver a payload of potent effector mechanisms that are essential for host defense against pathogens as well as tissue homeostasis. In addition, neutrophils are central contributors to the pathogenesis of a vast spectrum of inflammatory, degenerative, and neoplastic diseases. As our understanding of neutrophils in health and disease continually expands, so too does our appreciation of their complex and dynamic nature in vivo; from development, maturation, and trafficking t

An intimate look at LET-23 EGFR trafficking in the vulval cells of live C. elegans larvae.

Precise cell fate specification is essential for organ formation. A simple view is that one or several signal sending cells emit a ligand to a group of signal receiving cells that express the corresponding receptor, which transduces the signal through intracellular enzyme pathways. All these events must be spatio-temporally regulated to achieve the proper strength, duration and output of the signaling pathways. In particular, the production and secretion of the ligand has to be coordinated with the expression and accessibility of the receptor in the signal receiving cells. Furthermore, removal

The role of chemokines in severe malaria: more than meets the eye.

Plasmodium falciparum malaria is responsible for over 250 million clinical cases every year worldwide. Severe malaria cases might present with a range of disease syndromes including acute respiratory distress, metabolic acidosis, hypoglycaemia, renal failure, anaemia, pulmonary oedema, cerebral malaria (CM) and placental malaria (PM) in pregnant women. Two main determinants of severe malaria have been identified: sequestration of parasitized red blood cells and strong pro-inflammatory responses. Increasing evidence from human studies and malaria infection animal models revealed the presence of

Histone deacetylase inhibitor (HDACi) suberoylanilide hydroxamic acid (SAHA)-mediated correction of α1-antitrypsin deficiency.

α1-Antitrypsin (α1AT) deficiency (α1ATD) is a consequence of defective folding, trafficking, and secretion of α1AT in response to a defect in its interaction with the endoplasmic reticulum proteostasis machineries. The most common and severe form of α1ATD is caused by the Z-variant and is characterized by the accumulation of α1AT polymers in the endoplasmic reticulum of the liver leading to a severe reduction (>85%) of α1AT in the serum and its anti-protease activity in the lung. In this organ α1AT is critical for ensuring tissue integrity by inhibiting neutrophil elastase, a protease that deg

Health of men, women, and children in post-trafficking services in Cambodia, Thailand, and Vietnam: an observational cross-sectional study.

BACKGROUND: Trafficking is a crime of global proportions involving extreme forms of exploitation and abuse. Yet little research has been done of the health risks and morbidity patterns for men, women, and children trafficked for various forms of forced labour. METHODS: We carried out face-to-face interviews with a consecutive sample of individuals entering 15 post-trafficking services in Cambodia, Thailand, and Vietnam. We asked participants about living and working conditions, experience of violence, and health outcomes. We measured symptoms of anxiety and depression with the Hopkins Symptom

Dynamic imaging of allogeneic mesenchymal stem cells trafficking to myocardial infarction.

BACKGROUND: Recent results from animal studies suggest that stem cells may be able to home to sites of myocardial injury to assist in tissue regeneration. However, the histological interpretation of postmortem tissue, on which many of these studies are based, has recently been widely debated. METHODS AND RESULTS: With the use of the high sensitivity of a combined single-photon emission CT (SPECT)/CT scanner, the in vivo trafficking of allogeneic mesenchymal stem cells (MSCs) colabeled with a radiotracer and MR contrast agent to acute myocardial infarction was dynamically determined. Nanoplastics as disruptors of microtubule dynamics and intracellular trafficking: implications for cellular senescence and ageing.

Nanoplastics (NPs) are increasingly recognized as persistent intracellular cargoes whose long-term effects may depend not only on chemical reactivity, oxidative stress, and inflammation, but also on how particle-containing vesicles are transported, positioned, and cleared. Because the positioning and movement of endosomes, lysosomes, autophagosomes, mitochondria, and NP-containing vesicles are extensively regulated by microtubule-based transport, prolonged NP retention has the potential to disrupt intracellular trafficking and organelle homeostasis. Here, we examine how NPs may influence micro

Non-vesicular Lipid Transport Machinery in Trichomonas vaginalis: Novel Drug Targets against Trichomoniasis.

Eukaryotic cells have separate membrane-enclosed organelles with distinct biochemical identities and specialized functions. The unique characteristics of each organelle are regulated by asymmetric distribution and intra-cellular trafficking of two important biomolecules, proteins and lipids. Non-vesicular lipid transport facilitated by lipid transfer proteins performs essential roles in intracellular lipid trafficking and homeostasis, while vesicular transport regulates protein trafficking. Comparative analysis of lipid transport machinery in protists could help us to understand the basis of p

S1P Lyase Regulation of Thymic Egress and Oncogenic Inflammatory Signaling.

Sphingosine-1-phosphate (S1P) is a potent lipid signaling molecule that regulates pleiotropic biological functions including cell migration, survival, angiogenesis, immune cell trafficking, inflammation, and carcinogenesis. It acts as a ligand for a family of cell surface receptors. S1P concentrations are high in blood and lymph but low in tissues, especially the thymus and lymphoid organs. S1P chemotactic gradients are essential for lymphocyte egress and other aspects of physiological cell trafficking. S1P is irreversibly degraded by S1P lyase (SPL). SPL regulates lymphocyte trafficking, infl

Upregulation of synapse-associated protein 97 in the spinal dorsal horn exacerbates inflammatory pain through mediating GluA1-containing AMPARs membrane trafficking.

Molecules involved in trafficking and organization of glutamate receptors at the synapses between primary afferents and spinal dorsal horn neurons play an important role in the regulation of pain-related synaptic transmission. Synapse-associated protein 97 (SAP97) interacts directly with the AMPARs subunit GluA1 and drives GluA1-containing AMPARs trafficking from the Golgi network to the plasma membrane. However, the role of SAP97 in inflammatory pain remains unclear. In this study we found that intraplantar injection of complete Freund's adjuvant (CFA) in male and female rats led to increase

Contribution of vesicle trafficking dysregulation to the pathomechanism of mucopolysaccharidosis.

Although mucopolysaccharidoses (MPS) are monogenic diseases, caused by mutations in genes coding for enzymes involved in degradation of glycosaminoglycans (GAGs), recent studies suggested that changes in expressions of various genes might cause secondary and tertiary cellular dysfunctions modulating the course of these diseases. In this report, we demonstrate that vesicle trafficking regulation is affected in fibroblasts derived from patients suffering from 11 different types of MPS due to changes in levels of crucial proteins (estimated by automated Western-blotting) involved in this process,

Leukocyte trafficking to the lungs and beyond: lessons from influenza for COVID-19.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the causative agent of coronavirus disease 2019 (COVID-19). Understanding of the fundamental processes underlying the versatile clinical manifestations of COVID-19 is incomplete without comprehension of how different immune cells are recruited to various compartments of virus-infected lungs, and how this recruitment differs among individuals with different levels of disease severity. As in other respiratory infections, leukocyte recruitment to the respiratory system in people with COVID-19 is orchestrated by specific leukocyte tra

Key role for scavenger receptor B-I in the integrative physiology of host defense during bacterial pneumonia.

Scavenger receptor B-I (SR-BI) is a multirecognition receptor that regulates cholesterol trafficking and cardiovascular inflammation. Although it is expressed by neutrophils (PMNs) and lung-resident cells, no role for SR-BI has been defined in pulmonary immunity. Herein, we report that, compared with SR-BI(+/+) counterparts, SR-BI(-/-) mice suffer markedly increased mortality during bacterial pneumonia associated with higher bacterial burden in the lung and blood, deficient induction of the stress glucocorticoid corticosterone, higher serum cytokines, and increased organ injury. SR-BI(-/-) mic

CAMKK2-CAMK4 signaling regulates transferrin trafficking, turnover, and iron homeostasis.

BACKGROUND: Circulatory iron is a hazardous biometal. Therefore, iron is transported in a redox-safe state by a serum glycoprotein - transferrin (TF). Different organs acquire iron from the systemic circulation through a tightly regulated mechanism at the blood-tissue interface which involves receptor-mediated internalization of TF. Thus, abnormal TF trafficking may lead to iron dyshomeostasis associated with several diseases including neurodegeneration. Iron -induced toxicity can cause neuronal damage to iron-sensitive brain regions. Recently, it was discovered that CAMKK2, a calcium (Ca2+)/c

Targeting immune cell migration as therapy for inflammatory disease: a review.

Immune cell migration plays a pivotal role in coordinating inflammatory responses and maintaining immune surveillance. Here, we provide a comprehensive overview of the migratory behaviors of key immune cell subsets, including Th1, Th2, regulatory T cells, macrophages, dendritic cells, and neutrophils and the molecular mechanisms that guide their trafficking from lymphoid organs to inflamed tissues. We highlight the stepwise migration cascade: priming in secondary lymphoid organs, trafficking through blood vessels, adhesion to endothelium, and extravasation into tissues. Each step is critically

Zonulin and its regulation of intestinal barrier function: the biological door to inflammation, autoimmunity, and cancer.

The primary functions of the gastrointestinal tract have traditionally been perceived to be limited to the digestion and absorption of nutrients and to electrolytes and water homeostasis. A more attentive analysis of the anatomic and functional arrangement of the gastrointestinal tract, however, suggests that another extremely important function of this organ is its ability to regulate the trafficking of macromolecules between the environment and the host through a barrier mechanism. Together with the gut-associated lymphoid tissue and the neuroendocrine network, the intestinal epithelial barr
