Human neuromuscular organoids mimic cancer-induced muscle cachexia.

Cancer cachexia, a devastating metabolic wasting syndrome affecting up to 80% of solid cancer patients, remains incurable despite advances in tumor biology understanding. This study introduces neuromuscular organoids (NMOs) derived from human-induced pluripotent stem cells (hiPSCs) as a platform to investigate cancer-driven muscle cachexia. We found that NMOs respond well to atrophic stimuli and replicate the key features of cancer cachexia when treated with conditioned media derived from cachexia-inducing cancer cells. Specifically, cachectic NMOs showed muscle mass loss, impairment of muscle

Organoids in endometrial diseases: from bench to bedside.

The endometrium, a dynamic tissue of paramount importance to female reproductive health, undergoes cyclic remodeling governed by hormonal signals. Endometrial diseases have a relatively high incidence rate and exert a considerable influence on women's quality of life; however, their underlying mechanisms remain incompletely understood. Endometrial organoids (EOs) possess considerable potential to replicate tissue-specific functions and disease phenotypes. This review begins by outlining the physiological and pathological basis of the endometrium, then systematically reviews the latest progress

Generation and Genetic Manipulation of Human Cervical Organoids.

Cervical cancer is the fourth most common cancer in women and most frequently affects the uterine ectocervix. The uterine ectocervix is lined by stratified squamous epithelium, comprising basal cells and differentiating parabasal and suprabasal cells. The regeneration observed after cervical conization suggests the presence of robust stem cell activity. However, our understanding of the identity and regulatory mechanisms of cervical stem cells and their malignant transformation process has been limited due to the restricted access to human cervical tissues and the lack of optimal systems to as

Crosstalk between inflammation and autophagy in CeD organoids.

Celiac disease (CeD), a small bowel disorder characterized by mucosal inflammation, villous atrophy and crypt hyperplasia, is induced by gliadin peptides in genetically predisposed individuals. Alterations in inflammation, intracellular vesicular trafficking, mTOR signalling and autophagy have been reported in the intestinal epithelium of CeD patients. The aim of this manuscript was to investigate the interplay between autophagy and inflammation in intestinal mucosa of CeD patients. Increased levels of markers related to impaired autophagic flux, mTOR pathway activation and inflammatory signal

Long-term culture of patient-derived pheochromocytoma organoids.

INTRODUCTION: Pheochromocytomas (PCCs) are rare neuroendocrine tumors with limited treatment options once metastasized. Progress toward effective therapies has been hindered by their rarity, disease heterogeneity, and lack of representative preclinical models. Organoids are three-dimensional, self-renewing structures that recapitulate key features of their tissue of origin, providing valuable platforms for disease modeling, drug screening, and personalized medicine. This study aimed to establish and characterize patient-derived organoid cultures of PCCs. METHODS: Tumor tissue from two patient

Generation of T cell containing adipose tissue organoids.

T cells and especially regulatory T cells play a pivotal role in adipose tissue homeostasis. Peripheral blood T cells are well studied; however, in-depth analysis of adipose tissue T cells in their environment is challenging, and the analysis of adipose tissue-resident T cells is currently restricted to in vivo mouse models and ex vivo human donor samples. We developed a model to study T cells in adipose tissue by means of an adipose tissue organoid model. We could observe a clear T cell infiltration in these organoids. Interestingly, we observed different infiltration ratios of regulatory T c

Organoids in respiratory virus research: advances and perspectives.

The pandemics of respiratory viruses pose a worldwide public health problem and bio-safety threat. Therefore, the development of high-throughput and accurate infection models is crucial for elucidating viral pathogenesis and accelerating countermeasures to address the evolving respiratory viruses and the unexpected outbreaks of emerging variants. Compared to traditional 2D cultures, organoids exhibit pronounced intercellular interactions, extracellular matrix signaling, and tissue-specific multicellular cooperation, thereby more accurately recapitulating the in vivo microphysiological environm

MeCP2 regulates telencephalic development in human cerebral organoids.

Loss-of-function variants in the X-linked gene MeCP2 cause a severe form of syndromic autism spectrum disorder known as Rett syndrome (RTT). Although traditionally considered a postnatal disorder, increasing evidence suggests that MeCP2 plays a crucial role during prenatal brain development. Here, we used human pluripotent stem cell-derived cerebral organoids and human telencephalic assembloids to model early cortical development in the context of MeCP2 deficiency. Loss of MeCP2 led to widespread dysregulation of transcriptional programs associated with cortical excitatory neuron development,

Investigating Aging-Related Endometrial Dysfunction Using Endometrial Organoids.

Ageing of the endometrium is a critical factor that affects reproductive health, yet its intricate mechanisms remain poorly explored. In this study, we performed transcriptome profiling and experimental verification of endometrium and endometrial organoids from young and advanced age females, to elucidate the underlying mechanisms and to explore novel treatment strategies for endometrial ageing. First, we found that age-associated decline in endometrial functions including fibrosis and diminished receptivity, already exists in reproductive age. Subsequently, based on RNA-seq analysis, we ident

Cholangiocyte organoids for disease, cancer, and regenerative medicine.

The biliary tract is a ductal network comprising the intrahepatic (IHBDs) and extrahepatic bile duct (EHBDs). Biliary duct disorders include cholangitis, neoplasms, and injury. However, the underlying mechanisms are not fully understood. With advancements in 3D culture technology, cholangiocyte organoids (COs) derived from primary tissues or induced pluripotent stem cells (iPSCs) can accurately replicate the structural and functional properties of biliary tissues. These organoids have become powerful tools for studying the pathogenesis of biliary diseases, such as cystic fibrosis and primary s

Brain Organoids: A Game-Changer for Drug Testing.

Neurological disorders are the second cause of death and the leading cause of disability worldwide. Unfortunately, no cure exists for these disorders, but the actual therapies are only able to ameliorate people's quality of life. Thus, there is an urgent need to test potential therapeutic approaches. Brain organoids are a possible valuable tool in the study of the brain, due to their ability to reproduce different brain regions and maturation stages; they can be used also as a tool for disease modelling and target identification of neurological disorders. Recently, brain organoids have been us

Establishing 3D Endometrial Organoids from the Mouse Uterus.

Endometrial tissue lines the inner cavity of the uterus and is under the cyclical control of estrogen and progesterone. It is a tissue that is composed of luminal and glandular epithelium, a stromal compartment, a vascular network, and a complex immune cell population. Mouse models have been a powerful tool to study the endometrium, revealing critical mechanisms that control implantation, placentation, and cancer. The recent development of 3D endometrial organoid cultures presents a state-of-the-art model to dissect the signaling pathways that underlie endometrial biology. Establishing endomet

Long-Term Expansion of Murine Primary Hepatocyte Organoids.

The in vitro maintenance and expansion of primary hepatocytes provide immense opportunities for disease modeling and other biological, viral, and toxicological studies, as well as for applications in regenerative medicine such as cell transplantation for the treatment of metabolic liver diseases. Here, we describe a protocol for the isolation and in vitro culture of primary hepatocytes in a three-dimensional extracellular matrix gel. The inflammatory cytokine tumor necrosis factor alpha (TNFα) is crucial for the long-term expansion of mouse hepatocyte organoids by mimicking the regenerative re

Lung organoids: current strategies for generation and transplantation.

Lung diseases occupy a leading position in human morbidity and are the third leading cause of death. Often the chronic forms of these diseases do not respond to therapy, so that lung transplantation is the only treatment option. The development of cellular and biotechnologies offers a new solution-the use of lung organoids for transplantation in such patients. Here, we review types of lung organoids, methods of their production and characterization, and experimental works on transplantation in vivo. These results show the promise of work in this direction. Despite the current problems associat

Organoids as Novel Models for Embryo Implantation Study.

In the last decade, organoids have become emerging novel models for biomedical research. Organoids are small, self-organized three-dimensional (3D) tissue cultures derived from stem cells that mimic certain tissues or organs. In reproductive medicine, researchers have generated numerous organoids including blastoid (blastocyst organoid), endometrial organoid, and trophoblast organoid. These organdies provide useful models for studying the embryo implantation mechanism through observation of cell differentiation, gene expression, and epigenetic profiles at the implantation stage. As in vitro ti

Human SPG11 cerebral organoids reveal cortical neurogenesis impairment.

Spastic paraplegia gene 11(SPG11)-linked hereditary spastic paraplegia is a complex monogenic neurodegenerative disease that in addition to spastic paraplegia is characterized by childhood onset cognitive impairment, thin corpus callosum and enlarged ventricles. We have previously shown impaired proliferation of SPG11 neural progenitor cells (NPCs). For the delineation of potential defect in SPG11 brain development we employ 2D culture systems and 3D human brain organoids derived from SPG11 patients' iPSC and controls. We reveal that an increased rate of asymmetric divisions of NPCs leads to p
