Selective Phosphonylation of 5'-Adenosine Monophosphate (5'-AMP) via Pyrophosphite [PPi(III)].

We describe here experiments which demonstrate the selective phospho-transfer from a plausibly prebiotic condensed phosphorus (P) salt, pyrophosphite [H2P2O52-; PPi(III)], to the phosphate group of 5'-adenosine mono phosphate (5'-AMP). We show further that this P-transfer process is accelerated both by divalent metal ions (M2+) and by organic co-factors such as acetate (AcO-). In this specific case of P-transfer from PPi(III) to 5'-AMP, we show a synergistic enhancement of transfer in the combined presence of M2+ & AcO-. Isotopic labelling studies demonstrate that hydrolysis of the phosphonyla

Primordial soup was edible: abiotically produced Miller-Urey mixture supports bacterial growth.

Sixty years after the seminal Miller-Urey experiment that abiotically produced a mixture of racemized amino acids, we provide a definite proof that this primordial soup, when properly cooked, was edible for primitive organisms. Direct admixture of even small amounts of Miller-Urey mixture strongly inhibits E. coli bacteria growth due to the toxicity of abundant components, such as cyanides. However, these toxic compounds are both volatile and extremely reactive, while bacteria are highly capable of adaptation. Consequently, after bacterial adaptation to a mixture of the two most abundant abiot

Production of guanine from NH(4)CN polymerizations.

The synthesis of adenine from the polymerization of concentrated ammonium cyanide solutions is well known. We show here that guanine is also produced by this reaction but at yields ranging from 10 to 40 times less than that of adenine. This synthesis is effective at both +80 and -20 degrees C. Since high concentrations of NH(4)CN are obtainable only by freezing, this prebiotic synthesis would be applicable to frozen regions of the primitive Earth, the Jovian satellite Europa and other icy satellites, and the parent body of the Murchison meteorite.

The radiolysis of aqueous ammonium cyanide: polymers of interest to chemical evolution studies.

Some of the polymers isolated from the mixture of products obtained upon irradiation of dilute oxygen-free solutions of NH4CN with gamma rays from a 60Co source were examined. Their molecular weights were estimated to be in the range 4000-20000 daltons; elements of their structure were inferred from chemical and spectroscopic studies; and their interactions with nucleic acid components were examined. Significance of these results for chemical evolution studies is summarized.

Balance Between Order and Disorder: Quantum-Epigenetic Pathways for the Emergence and Continuous Evolution of Life.

The scientific restlessness surrounding the question of what life is (expressed in the provocations of Feynman and Schrödinger) guides this review article by integrating physics, chemistry, and biology into a multidimensional and emergent approach to the origin, evolution, and recreation of life. By exploring classical and quantum concepts, we discuss how synthetic biology, combined with epigenetics, ploidy variation, and quantum effects (such as coherence and tunneling) offers new possibilities for adapting and designing living systems. This review covers a spectrum of topics, from prebiotic

Other Earths, Gaian bottlenecks and Darwinized Gaias.

In our galaxy, tens of billions of wet, rocky Earth-like planets orbit in the habitable zones of their host stars. Stellar spectra and models of devolatilization yield ranges of plausible chemistries on these Earths. The early emergence of biology on Earth from similar chemistry has a chance of being universally relevant to the origins of life elsewhere. The Gaian bottleneck suggests that most life does not establish Gaian regulation quickly enough to counter positive feedback of runaway greenhouse or runaway ice albedo. Methane (a strong greenhouse gas) was produced by methanogens and oxidize

Mathematical modeling reveals spontaneous emergence of self-replication in chemical reaction systems.

Explaining the origin of life requires us to elucidate how self-replication arises. To be specific, how can a self-replicating entity develop spontaneously from a chemical reaction system in which no reaction is self-replicating? Previously proposed mathematical models either supply an explicit framework for a minimal living system or consider only catalyzed reactions, and thus fail to provide a comprehensive theory. Here, we set up a general mathematical model for chemical reaction systems that properly accounts for energetics, kinetics, and the conservation law. We found that 1) some systems

Experimental Models on the Prebiotic Formation of Biopolymer Building Blocks.

The scientific study of the origins of life is a deep pursuit that exists at the intersection of multiple disciplines. Prebiotic chemistry focuses on understanding how biopolymer building blocks such as amino acids, nucleotides, and sugars emerged and how their chemical and structural space evolves toward life (chemical evolution). Simulation experiments have been essential for exploring plausible pathways for the origin of building blocks under early Earth conditions and planetary environments. Key examples include the seminal Miller-Urey experiment and the polymerization of hydrogen cyanide.

Prebiotic Origin of Pre-RNA Building Blocks in a Urea "Warm Little Pond" Scenario.

Urea appears to be a key intermediate of important prebiotic synthetic pathways. Concentrated pools of urea likely existed on the surface of the early Earth, as urea is synthesized in significant quantities from hydrogen cyanide or cyanamide (widely accepted prebiotic molecules), it has extremely high water solubility, and it can concentrate to form eutectics from aqueous solutions. We propose a model for the origin of a variety of canonical and non-canonical nucleobases, including some known to form supramolecular assemblies that contain Watson-Crick-like base pairs. The dual nucleophilic-elec

Mechanism for the Coupled Photochemistry of Ammonia and Acetylene: Implications for Giant Planets, Comets and Interstellar Organic Synthesis.

Laboratory studies provide a fundamental understanding of photochemical processes in planetary atmospheres. Photochemical reactions taking place on giant planets like Jupiter and possibly comets and the interstellar medium are the subject of this research. Reaction pathways are proposed for the coupled photochemistry of NH3 (ammonia) and C2H2 (acetylene) within the context Jupiter's atmosphere. We then extend the discussion to the Great Red Spot, Extra-Solar Giant Planets, Comets and Interstellar Organic Synthesis. Reaction rates in the form of quantum yields were measured for the decompositio

Cold-selective topological bias and the emergence of the first membranes.

The emergence of the first membranes requires more than the synthesis of amphiphiles; it requires a mechanism that stabilizes bilayer-forming geometries within chemically diverse prebiotic mixtures. We present a minimal thermodynamic model coupling amphiphile topology to self-assembly, incorporating packing, curvature, and steric contributions. Changes in temperature alter the balance between ordering and disorder within amphiphile aggregates, thereby modifying the relative stability of lamellar, micellar, and dispersed states. The model identifies a bounded temperature regime in which geometr

Prebiotic Nucleoside Phosphorylation in a Simulated Deep-Sea Supercritical Carbon Dioxide-Water Two-Phase Environment.

Prebiotic synthesis of complex organic molecules in water-rich environments has been a long-standing challenge. In the modern deep sea, emission of liquid CO2 has been observed in multiple locations, which indicates the existence of benthic CO2 pools. Recently, a liquid/supercritical CO2 (ScCO2) hypothesis has been proposed that a two-phase ScCO2-water environment could lead to efficient dehydration and condensation of organics. To confirm this hypothesis, we conducted a nucleoside phosphorylation reaction in a hydrothermal reactor creating ScCO2-water two-phase environment. After 120 h of uri

Salinity Effects on the Adsorption of Nucleic Acid Compounds on Na-Montmorillonite: a Prebiotic Chemistry Experiment.

Any proposed model of Earth's primitive environments requires a combination of geochemical variables. Many experiments are prepared in aqueous solutions and in the presence of minerals. However, most sorption experiments are performed in distilled water, and just a few in seawater analogues, mostly inconsistent with a representative primitive ocean model. Therefore, it is necessary to perform experiments that consider the composition and concentration of dissolved salts in the early ocean to understand how these variables could have affected the absorption of organic molecules into minerals. An investigation of prebiotic purine synthesis from the hydrolysis of HCN polymers.

The polymerization of concentrated NH4CN solutions has been studied at various temperatures and ammonia concentrations. The products of the oligomerization of ammonium cyanide include adenine and guanine, as well as trace amounts of 2,6-diaminopurine. Our results indicate that the adenine yield is not strongly dependent on temperature. Guanine is produced in lower yield. The original studies by Oró and Kimball (1961) showed that the 6 N HCl hydrolysis of the NH4CN polymerization supernatant greatly increased the adenine yield. However, this hydrolysis also decomposes adenine and other purines.

Chemical reduction of phosphate on the primitive earth.

If phosphorus played a role in the origin of life, some means of concentrating micromolar levels of phosphate (derived from the calcium phosphate mineral apatite), must first have been available. Here we show that simulated (mini)lightning discharges in model prebiotic atmospheres, including only minimally reducing ones, reduce orthophosphates, including apatite, to produce substantial yields of phosphite. Electrical discharges associated with volcanic eruptions could have provided a particularly suitable environment for this process. Production of relatively soluble and reactive phosphite sal

Reactions of molybdenum-sulphur compounds with cyanide: chemical evolution and deactivation of molybdoenzymes.

Reactions of molybdenum-sulphur compounds with cyanide are reported which may be relevant to (1) the chemical evolution of molybdoenzymes and (2) deactivation of molybdoenzymes by cyanide. (1) With aqueous cyanide MoS2 gave thio-bridged complex anions [(Mo(CN)6)2(mu-S)]6- and [(Mo(CN)4(mu-S))2]6-. Under prebiotic conditions such complexes could have been formed similarly from molybdenite and may have been precursors of molybdoenzymes. (2) Only those compounds which contained terminal sulphur bound to molybdenum (i.e., Mo = S groups), viz. oxothiomolybdates and the complex [(Mo(mu-S)(S)(Et2NCS2

Continuous UV irradiation played an inconclusive role in selecting the canonical genetic alphabet.

An abundance of heterocyclic molecules analogous to canonical nucleobases (i.e., adenine, thymine, guanine, cytosine, and uracil) used as the genetic alphabet in DNA and RNA could have been present in various prebiotic environments on early Earth. However, extant biology generally uses the canonical nucleobases and nucleosides as the genetic alphabet, and not any number of possible noncanonical counterparts. Ultraviolet (UV) irradiation was likely abundant on the surface of the early Earth and has been suggested to play a role in the selection of the canonical nucleobases and nucleosides as th

The Prebiotic Provenance of Semi-Aqueous Solvents.

The numerous and varied roles of phosphorylated organic molecules in biochemistry suggest they may have been important to the origin of life. The prominence of phosphorylated molecules presents a conundrum given that phosphorylation is a thermodynamically unfavorable, endergonic process in water, and most natural sources of phosphate are poorly soluble. We recently demonstrated that a semi-aqueous solvent consisting of urea, ammonium formate, and water (UAFW) supports the dissolution of phosphate and the phosphorylation of nucleosides. However, the prebiotic feasibility and robustness of the U

Cometary Delivery of Hydrogen Cyanide to the Early Earth.

Delivery of water and organics by asteroid and comet impacts may have influenced prebiotic chemistry on the early Earth. Some recent prebiotic chemistry experiments emphasize hydrogen cyanide (HCN) as a feedstock molecule for the formation of sugars, ribonucleotides, amino acids, and lipid precursors. Here, we assess how much HCN originally contained in a comet would survive impact, using parametric temperature and pressure profiles together with a time-dependent chemistry model. We find that HCN survival mainly depends on whether the impact is hot enough to thermally decompose H2O into reacti
