Viral attachment to target cells is the first step in infection and also serves as a determinant of tropism. Like many viruses, mammalian reoviruses bind with low affinity to cell-surface carbohydrate receptors to initiate the infectious process. Reoviruses disseminate with serotype-specific tropism in the host, which may be explained by differential glycan utilization. Although α2,3-linked sialylated oligosaccharides serve as carbohydrate receptors for type 3 reoviruses, neither a specific glycan bound by any reovirus serotype nor the function of glycan binding in type 1 reovirus infection wa

Infection with reovirus 3 (Reo-3) has been suggested as the cause of extrahepatic biliary atresia and idiopathic neonatal hepatitis, but confirmation has been lacking. Therefore we have searched for a specific anti-Reo-3 antibody response in the sera of patients with biliary atresia or neonatal hepatitis and for Reo-3 antigens in their hepatobiliary tissues. Sera from 23 infants with extrahepatic biliary atresia, 12 with neonatal hepatitis, 30 age-matched control patients with other liver diseases, and 55 control patients without liver disease were tested by an enzyme-linked immunosorbent assa

Reovirus type 3 (Reo-3) infection has recently been implicated in the pathogenesis of certain idiopathic, cholestatic liver diseases of newborns. In the present study, antibody titres to Reo-3 virus (anti-Reo-3) were determined in sera from 43 adults with idiopathic cholestatic liver disease, including 34 patients with primary biliary cirrhosis (PBC) and 9 patients with primary sclerosing cholangitis (PSC). Seventy-four adults with various other causes of chronic liver disease and 16 healthy volunteers served as controls. Geometric mean titres of anti-Reo-3 were significantly higher in PBC and

Respiratory enteric orphan virus (reovirus) has been used to study many aspects of the biology and genetics of viruses, viral infection, pathogenesis, and the immune response to virus infection. This report describes the functional activity of virus labeled with Alexa Fluor 488, a stable fluorescent dye. Matrix assisted laser desorption-time of flight analysis indicated that Alexa Fluor 488 labeled the outer capsid proteins of reovirus. Labeled virus bound to murine L929 fibroblasts as determined by flow cytometry and fluorescence microscopy, and the specificity of binding were demonstrated by

Avian reovirus (ARV) causes a range of diseases in poultry and results in significant economic losses for the poultry industry. To address the epidemic of ARV infection in yellow-feathered broilers in Xinjiang, this study conducted genetic and evolutionary analyses of a chicken-origin ARV field strain, designated ARV xj-1.1. The σC gene of this strain was expressed in vitro using both Escherichia coli and Pichia pastoris systems to compare expression characteristics and immunogenicity. Phylogenetic analysis revealed that ARV xj-1.1 belongs to genotype IV. The σC gene was successfully cloned an

Outcomes of pancreatic adenocarcinoma (PDA) remain dismal despite extensive clinical investigation. Combination chemotherapy provides modest improvements in survival above best supportive care, and immunotherapy has thus far not proven effective. Nevertheless, growing insight into antitumor immunity and the tumor microenvironment has inspired the discovery of novel agents targeting PDA. Oncolytic viruses represent an emerging class of immunotherapeutic agents that have undergone extensive preclinical investigation and warrant further investigation in well-designed clinical trials.

Avian reoviruses (ARVs) are pathogens that cause significant morbidity among commercial poultry. ARVs are prototypic representatives of non-enveloped viruses that can cause cell-cell fusion. They belong to the Reoviridae family, which contains many highly pathogenic viruses. ARVs are ubiquitous in commercial poultry and are frequently isolated from the gastrointestinal and respiratory tracts of chickens with acute infections. The virus causes a range of disease states in chicken, including viral arthritis/tenosynovitis, gastroenteritis, hepatitis, myocarditis, "pale bird syndrome," runting-stu

Muscovy duck reovirus (MDRV) causes substantial economic losses in the waterfowl industry, necessitating rapid and field-deployable diagnostic tools. In this study, we developed, for the first time, a lyophilized reagent-based reverse transcription recombinase-aided amplification (RT-RAA) assay targeting the σC gene of MDRV for point-of-care testing (POCT). The assay was optimized to operate at 39°C with a probe concentration of 100 nM. It achieved a detection limit of 1.03 × 101 copies/μL within 20 min, which is comparable to RT-quantitative polymerase chain reaction (qPCR) and superior to ex

Avian reovirus (ARV) infections, characterized by severe arthritis, tenosynovitis, pericarditis, and poor weight gain, have become increasingly serious in recent years. The economic impact is significant as it causes growth inhibition and immunosuppression. Some commercial poultry in China have been widely vaccinated with available ARV vaccines; however, infections continue to occur even after vaccination. This study aimed to isolate a novel variant, ARV-SD19/11103, from the joint tissues of infected broiler chickens vaccinated with ARV vaccines in Shandong Province. Genetic evolution analysis

Reoviruses infect cells that manifest an activated Ras-signaling pathway, and have been shown to effectively destroy many different types of neoplastic cells, including those derived from brain, breast, colon, ovaries, and prostate. In this study, we investigated the reovirus as a potential therapeutic agent against lymphoid malignancies. A total of 9 lymphoid cell lines and 27 primary human lymphoid malignancies, as well as normal lymphocytes and hematopoietic stem/progenitor cells, were tested for susceptibility to reovirus infection. For in vitro studies, the cells were challenged with reov

Apoptosis is a common host response to virus infection. The extent and timing of apoptosis following infection is controlled by the balance between the strength of signals that activate death-inducing and survival-promoting pathways in cells. In many cell types, infection with mammalian orthoreovirus (reovirus) results in induction of cell death by apoptosis late in infection. In this study, we uncovered that WD repeat-containing protein 81 (WDR81) is required for apoptosis induction after reovirus infection. The requirement for WDR81 for apoptosis induction is not unique to reovirus because c

Novel duck reovirus (NDRV) is a major pathogen that causes immunosuppression and secondary infections in ducklings, resulting in elevated mortality and severe economic losses to the global duck industry. However, there is currently no licensed vaccine available in China for the effective prevention and control of NDRV infections. To effectively control NDRV outbreaks, we developed a live-attenuated vaccine candidate against NDRV by serially passaging the NDRV E232 strain in specific-pathogen-free (SPF) chicken embryos and DF-1 cells. The resulting E232-P100 strain replicated efficiently in DF-

Avian reoviruses are ubiquitous in poultry production worldwide and can be transmitted vertically or horizontally among chickens. The pathogenicity of reoviruses can range from very pathogenic viruses that affect multiple tissues and organs to apathogenic. Avian reoviruses have been associated with many disease presentations, and two of the most economically significant diseases are viral arthritis/tenosynovitis and viral enteritis. Viral arthritis/tenosynovitis has been recognized since the 1950s and essentially disappeared after development of attenuated live and inactivated vaccines in the

Interferons (IFNs) are key host cytokines in the innate immune response to viral infection, and recent work has identified unique roles for IFN subtypes in regulating different aspects of infection. Currently emerging is a common theme that type III IFNs are critical in localized control of infection at mucosal barrier sites, while type I IFNs are important for broad systemic control of infections. The intestine is a particular site of interest for exploring these effects, as in addition to being the port of entry for a multitude of pathogens, it is a complex tissue with a variety of cell type

Freon 113 (Freon) is an essential component used in some viral purification methods to separate virus from infected cell debris. With its environmental and toxic hazards, Freon's availability is limited and more tightly regulated. Several organic solvent substitutes were selected to identify a suitable Freon replacement for the purification of both cultivable reovirus and fastidious calicivirus. Reovirus was extracted from tissue cultured cells with each solvent tested and purified in cesium chloride gradients by standard techniques. Purified virions were analyzed for conservation of physical

Viral Arthritis (VA), a disease caused by Avian Reovirus (ARV), has emerged as a significant cause of economic losses in broiler chicken flocks in Western Canada. These outbreaks were characterized by 4-13% morbidity, followed by a spike in mortality/culling that in extreme cases required total flock depopulation. From 2012-2017, 38 ARV isolates were recovered. Molecular characterization of a partial segment of the sigma (σ)C gene shows all six previously known ARV clusters in Western Canadian broiler chickens. The most numerous clusters were Cluster#4 and Cluster #5 while the most variable cl

An 8-week-old female infant presented with a history of active varicella complicated by Escherichia coli sepsis, oral thrush, hypoalbuminemia, intermittent fevers, diarrhea and feeding intolerance. Rhesus monkey kidney cells inoculated with cerebrospinal fluid revealed reovirus-like particles by electron microscopy. Virus neutralization and RNA-gel electrophoresis studies identified the isolated pathogen as reovirus serotype 2. This report represents one of only a few to isolate reovirus from the central nervous system in humans.

Reovirus genome segment S1 encodes protein final sigma1, which is the receptor binding protein, modulates tissue tropism, and specifies the nature of the antiviral immune response. It makes up less than 2% of reovirus particles and is synthesized in very small amounts in infected cells. Any antiviral strategy aimed at reducing specifically the expression of this genome segment should, in principle, reduce the infectivity of the virus. To test this hypothesis, we have assembled two hammer-head motif-containing ribozymes (Rzs) targeted to cleave at the conserved B and C domains of the reovirus s

The sensitivity of the mouse antibody production test with intraperitoneal or intrasplenic inoculation of mice with reovirus type 3, minute virus of mice, lymphocytic choriomeningitis virus, or mouse hepatitis virus was compared with that of direct isolation in cultured cells. The mouse antibody production test for detection of mouse hepatitis virus was significantly more sensitive than virus isolation in permissive cells, but differences in sensitivity were less marked for the other three viruses. The intrasplenic route of inoculation did not yield seroconversion that occurred earlier or more

To investigate the prevalence and pathogenic characteristics of avian reovirus (ARV) in the Xinjiang region of China, this study collected suspected joint tissue samples from broiler farms across different areas of the Taikun Group. The samples were subjected to virus isolation, RT-PCR analysis, sequence analysis, in vitro replication assays, and pathogenicity assessments in specific pathogen-free (SPF) chicken embryos and chickens. The results revealed the isolation of a chicken-derived ARV epidemic strain, designated as ARV xj-1.1. The virus-induced cytopathic effects (CPEs) in LMH cells and
