Hepatocyte Nuclear Factor-1β Regulates Urinary Concentration and Response to Hypertonicity.

The transcription factor hepatocyte nuclear factor-1β (HNF-1β) is essential for normal kidney development and function. Inactivation of HNF-1β in mouse kidney tubules leads to early-onset cyst formation and postnatal lethality. Here, we used Pkhd1/Cre mice to delete HNF-1β specifically in renal collecting ducts (CDs). CD-specific HNF-1β mutant mice survived long term and developed slowly progressive cystic kidney disease, renal fibrosis, and hydronephrosis. Compared with wild-type littermates, HNF-1β mutant mice exhibited polyuria and polydipsia. Before the development of significant renal str

Plasma vasopressin response to hypertonic saline infusion to assess posterior pituitary function.

Hypertonic saline was infused into 11 volunteers to osmotically stimulate vasopressin secretion. A strong positive correlation between plasma arginine vasopressin (PAVP) and plasma osmolality (Pos) was obtained, defined by the function PAVP=0.63 (Pos-284), r=+0.80, P<0.001. The sensitivity of vasopressin secretion to osmotic stimulation was represented by the slope of the expression and the theoretical threshold of vasopressin release by the abscissal intercept. Plasma osmolality at the onset of thirst was 298.5±1.1 mmol/kg. Application of hypertonic saline infusion to 10 polyuric patients cle

[The inappropriate antidiuretic hormone secretion syndrome associated with cryptococcal meningitis in a patient with the acquired immunodeficiency syndrome].

Meningitis and other infectious diseases of the central nervous system are predisposing causes of the inadequate secretion syndrome of the antidiuretic hormone (ISADH). The association between the acquired immunodeficiency syndrome (AIDS) and ISADH is recognized but little studied. In AIDS patients hyponatremia is a frequent finding and it is responsible for ISADH in most cases. However, there is little reference material concerning the incidence of presentation and the type of osmoregulation anomaly this entity presents in these patients. A case in which ISADH is associated to cryptococcal me

Function and evolution of the aquaporin IsAQP1 in the Lyme disease vector Ixodes scapularis.

Ticks are important vectors of pathogenic viruses, bacteria, and protozoans to humans, wildlife, and domestic animals. Due to their life cycles, ticks face significant challenges related to water homeostasis. When blood-feeding, they must excrete water and ions, but when off-host (for stretches lasting several months), they must conserve water to avoid desiccation. Aquaporins (AQPs), a family of membrane-bound water channels, are key players in osmoregulation in many animals but remain poorly characterized in ticks. Here, we bioinformatically identified AQP-like genes from the deer tick Ixodes

Inorganic mercury effects on biomarker gene expressions of a freshwater amphipod at two temperatures.

Mercury (Hg) is a global contaminant resulting of both natural processes and human activities. In aquatic environments, studies conducted on vertebrates highlighted changes of gene expression or activity of antitoxic and oxidative enzymes. However, although Hg is a highly toxic compound in aquatic environments, only a few studies have evaluated the lethal and sublethal effects of inorganic Hg on Gammarus sp. Therefore, this study aimed at evaluating the effects of inorganic Hg (HgCl2) on the expression of 17 genes involved in crucial biological functions or mechanisms for organisms, namely res

Atypical cilia in the choroid plexus of guineapig.

Ultrastructural study of choroid plexus of normal and kaolin induced hydrocephalic guineapigs showed clusters of cilia arising from the apical part of some of the epithelial cells. Some of these cells had typical 9 + 2 cilia, while most of the control and the hydrocephalic groups showed 9 + 0 type of cilia. These atypical (9 + 0 type) cilia of choroidal epithelium differ from the primary cilia of other mammalian tissues in that, clusters of long cilia arise from a single cell and have no associated centrioles within the cytoplasm. They may be motile and sensory (osmoregulator or pressure senso

Prevalence, predictors and patterns of postoperative polyuria and hyponatraemia in the immediate course after transsphenoidal surgery for pituitary adenomas.

OBJECTIVE: Disturbances of osmoregulation, leading to diabetes insipidus and hyponatraemia are well known complications after surgery in the sella region. This study was performed to examine the prevalence and predictors of polyuria and hyponatraemia after a complete and selective removal of pituitary adenomas was attempted via the transnasal-transsphenoidal approach. DESIGN: 1571 patients with pituitary adenomas (238 Cushing's disease, 405 acromegaly, 534 hormonally inactive adenomas, 358 prolactinoma, 23 Nelson's syndrome, and 13 thyrotropinoma) were daily examined within a 10-day postopera

The essential roles of metal ions in insect homeostasis and physiology.

Metal ions play distinct roles in living organisms, including insects. Some, like sodium and potassium, are central players in osmoregulation and 'blood and guts' transport physiology, and have been implicated in cold adaptation. Calcium is a key player as a second messenger, and as a structural element. Other metals, particularly those with multiple redox states, can be cofactors in many metalloenzymes, but can contribute to toxic oxidative stress on the organism in excess. This short review selects some examples where classical knowledge has been supplemented with recent advances, in order t

Hepatic Expression of Growth Hormone Receptor (GHrec) and Insulin-like Growth Factor-I (IGF-I) Genes and Cellular Location of IGF-I mRNA in Diploid and Triploid Atlantic Salmon (Salmo salar) Undergoing Parr-Smolt Transformation.

The induction of triploidy, a strategy to mitigate unwanted pre-harvest sexual maturation and a genetic containment measure for escaped farmed Atlantic salmon (Salmo salar), may give rise to challenges because of the distinct environmental and dietary requirements of sterile triploid fish. Smoltification is a critical phase in the life cycle of Atlantic salmon, so knowledge about parr-smolt transformation in triploids is important for the salmon farming industry. This study covered an investigation of hepatic expression of growth hormone receptor (GHrec) and insulin-like growth factor-I (IGF-I

Electrical phenotypes of calcium transport mutant strains of a filamentous fungus, Neurospora crassa.

We characterized the electrical phenotypes of mutants with mutations in genes encoding calcium transporters-a mechanosensitive channel homolog (MscS), a Ca(2+)/H(+) exchange protein (cax), and Ca(2+)-ATPases (nca-1, nca-2, nca-3)-as well as those of double mutants (the nca-2 cax, nca-2 nca-3, and nca-3 cax mutants). The electrical characterization used dual impalements to obtain cable-corrected current-voltage measurements. Only two types of mutants (the MscS mutant; the nca-2 mutant and nca-2-containing double mutants) exhibited lower resting potentials. For the nca-2 mutant, on the basis of

Localization and Expression of Aquaporin 1 (AQP1) in the Tissues of the Spiny Dogfish (Squalus acanthias).

Aquaporin 1 is a membrane water channel protein, which was studied here in spiny dogfish (Squalus acanthias) osmoregulatory tissues using a variety of techniques. The cloning of aquaporin 1 (AQP1) in the spiny dogfish identified a splice variant version of the mRNA/protein (AQP1SV1/AQP1SV1). Polymerase chain reaction (PCR) in a range of tissues showed AQP1 to be expressed at very high levels in the rectal gland with ubiquitous mRNA expression at lower levels in other tissues. Northern blotting showed that AQP1 had a mRNA size of 5.3 kb in kidney total RNA. The level of AQP1 mRNA was significan

Enhancement of root abscisic acid mediated osmotic regulation by macroalgal compounds promotes adaptability of rice (Oryza sativa L.) in response to progressive metal ion mediated environmental stress.

Soil deterioration is a major cause of poor agricultural productivity, necessitating sufficient nutrient inputs like fertilizers and amendments for sustainable use. As one such strategy, the current study evaluates the potential of Sargassum wightii, a brown seaweed extract, as an osmopriming agent to improve seed germination, early establishment, and competent seedling performances in acidic soil. The elemental makeup of seaweed extract (BS) showed that it included major plant macro (Potassium, Nitrogen and Phosphorous), as well as micronutrients (Magnesium and Iron) and trace elements (Zinc,

Recent Insights into NCL Protein Function Using the Model Organism Dictyostelium discoideum.

The neuronal ceroid lipofuscinoses (NCLs) are a group of devastating neurological disorders that have a global distribution and affect people of all ages. Commonly known as Batten disease, this form of neurodegeneration is linked to mutations in 13 genetically distinct genes. The precise mechanisms underlying the disease are unknown, in large part due to our poor understanding of the functions of NCL proteins. The social amoeba Dictyostelium discoideum has proven to be an exceptional model organism for studying a wide range of neurological disorders, including the NCLs. The Dictyostelium genom

Hybrid histidine kinases and antifungal warfare in thermal dimorphic fungi.

Thermal dimorphic fungal pathogens are fungi that infect humans, often through the inhalation of asexual conidia, and which transition from hyphae to yeast in the human body. These fungi cause severe or chronic mycoses and are typically treated with azoles or amphotericin B. Ambruticin, a polyketide antifungal, shows promise as an alternative therapy. It targets hybrid histidine kinases (HHKs), which are fungal-specific proteins essential for osmoregulation and parasitic morphology and are conserved across thermal dimorphic species. Targeting HHKs suggests that ambruticin may therapeutically t

Vasopressin function in hypercalcaemia.

Vasopressin function and thirst were studied in fourteen hypercalcaemic patients (ten hyperparathyroid and four disseminated malignant disease). Ten patients had decreased renal concentrating ability which reversed within a few days in the majority of patients whose hypercalcaemia was corrected by parathyroidectomy. Although eight patients complained of thirst, none showed a lowered threshold of thirst appreciation during hypertonic saline infusion. Osmoregulation of vasopressin secretion was not reduced in any patient, but the hyperparathyroid group had an exaggerated vasopressin response to

Adipsic diabetes insipidus following pituitary surgery for a macroprolactinoma.

Adipsic diabetes insipidus (ADI) is a rare condition in which thirst, an essential clinical feature for the prevention of hypernatraemic dehydration, is absent. We report the first case of adipsic diabetes insipidus to occur following surgery for a pituitary macroprolactinoma, with loss of both osmoregulated and baroregulated vasopressin release. Following extensive surgery for a vision threatening macroprolactinoma a 14-year-old boy developed profound hypernatraemia with absent thirst sensation. Detailed investigation, with hypertonic saline infusion and trimetaphan infusion, revealed absence

Gene encoding prolactin in cinnamon clownfish Amphiprion melanopus and its expression upon acclimation to low salinities.

BACKGROUND: Prolactin (PRL) is a key hormone for osmoregulation in fish. Levels of PRL in the pituitary gland and plasma ion composition of clownfish seem to change to regulate their hydromineral balance during adaptation to waters of different salinities. In order to understand osmoregulatory mechanism and its association with growth performance and PRL in fish, the gene encoding PRL and its expression level in cinnamon clownfish Amphiprion melanopus upon acclimation to low salinity was analyzed. RESULTS: The PRL gene of A. melanopus encoded a protein of 212 amino acid residues comprised of

Synergistic melatonin and myo-inositol application reinforces antioxidant defence, osmotic homeostasis, and membrane stability in Western Himalayan apple rootstocks under drought stress.

Drought stress induces profound biochemical and physiological disturbances in apple (Malus × domestica Borkh.), including oxidative imbalance, chlorophyll degradation, osmotic disruption, and membrane injury. This study evaluated the individual and synergistic effects of two key metabolites, melatonin (MT) and myo-inositol (MI) in enhancing drought resilience across five genetically diverse apple rootstocks (MM 111, MM 106, Bud 118, MM 116, and M9). A two-phase experimental framework was implemented, beginning with metabolite dose optimisation under 40% field capacity using the Response Index

The transcriptomic response of adult salmon lice (Lepeophtheirus salmonis) to reduced salinity.

Salmon lice (Lepeophtheirus salmonis) are marine parasitic copepods living on salmonids and are challenging for salmon aquaculture. One of several treatment methods is the application of freshwater to the fish which can lead to lice loss. However, lab experiments have shown that salmon lice, acclimated to seawater, are capable of surviving for several weeks in freshwater, when attached to a host. If not attached to a host, they die within a few hours in freshwater but can survive a longer time in brackish water. The molecular mechanisms involved in the adaptation to low salinity of the louse h
